        일반논문 : 판소리 수궁가 의학기사(醫學記事)에 내포된 역사성과 조선후기 민중 의학지식의 보급 -김연수 창본 수궁가의 사설을 대상으로-

        <수궁가>는 구토설화에서 파생된 판소리 창본으로 내용 중 醫學記事가 자주 등장한다. 醫學記事는 신재효의 창본정리 과정에서 <수궁가> 전반에 삽입된 것으로 보이는데, 지방관청에 소속된 官藥房을 했던 부친의 영향으로 『東醫寶鑑』과 여러 의서들, 藥器를 쉽게 접할 수 있었던 까닭에 <수궁가> 내에 그 내용을 충분히 실어 넣은 것으로 보인다. 본고에서 진행한 <수궁가>의 의학적 분석을 통한 연구는 <수궁가>에서 절반 가까이 차지하는 藥性歌 및 醫學記事의 많은 부분이 한의학 지식과 매우 일치하며, 내용전반에 민간에서 쓸 수 있는 한의학 지식을 바탕에 깔고 있음을 확인할 수 있었다. 특히 『東醫寶鑑』이 점차 보편화되면서 <수궁가> 속의 醫學記事도 전문성을 더해갔다. <수궁가>에서 도가적 양생사상의 신선술과 주술에 의거한 치료형태를 풍자하고 희화시켰던 양상은, 동시기에 『東醫寶鑑』이 여러 의서를 종합하면서 도가적인 주술이나 미신적인 요소를 배제하고, 체계적이고 임상의학적으로 재정비했다는 점과 맥을 같이한다. 18세기중반 이후로 19~20세기 양반지식층의 애호참여로 유교사상을 재정비하면서 醫學記事가 전문지식의 과시수단으로 사용되기도 하였으나, 한편으로는 전문성있는 의학지식을 판소리내에 삽입함으로써 판소리에 사실성을 부여하고, 청자들로 하여금 판소리의 현실감을 극대화하는 역할로 작용하였다. 그로인해 <수궁가>내에서 더욱 전문화된 醫藥記事들은 이후 판소리, 국문한문소설로 변화되어 보급되었고, 19세기부터는 라디오매체와 음반사의 판소리 취입으로 더욱 보편화되면서 서민들의 입과 귀로 널리 전해졌다. 비록 醫學記事가 서민들에게 의학지식의 보급을 목적으로 작성된 것은 아니었다 하더라도, 의료정책이 미비했던 당시 『東醫寶鑑』을 바탕으로 보완되었던 <수궁가>의 보급은 원래의 목적이상으로 민중들에게 민간의학지식을 파급시키는 역할을 수행했다. Sugungga is a Pansori[recite a dramatic song] story derived from a tale about a terrapin and a rabbit, and Yakseongga appears in its first part. Sugungga has been handed down through a lineage of singers: Song Heung-Rok, Song Gwang-Rok, Song Woo-Ryong, Ryu Seong-Jun, and Kim, Yeon-Su. Yakseongga completely settled in Sugungga thanks to adaptation of Shin Jae-Hyo, whose medicinal knowledge was mostly from his father, Shin Gwang-Heup, an official pharmacist in a local government office. Between the 17th century and the mid-18th century, Sugungga was evolved from ancient taoistic idea of health to Confucianistic rational subject. This was similar in nature that Korean medicine was overhauled to a more scientific and clinical practice in the same period by putting together diverse medicinal books such as DongeuiBogam, Huangdi Neijing, Hyang-yak Jipseongbang, Euibang Ryuchwi, and Euibang Chalyo, while eliminating elements of taoistic charms or superstitions. This study confirmed that from the study of oriental medicine, Yakseongga in Sugungga was based on oriental medicinal knowledge, not simple humor. It seems that Yakseongga was heavily influenced by oriental medicinal books such as Huangdi Neijing, Hyang-yak Jipseongbang, Dongeui Bogam, and Jejung Sinpyeon. After the mid-18th century, deconfucianistic Sugungga was modified again. Participation of aristocratic intellectuals from the 19th and 20th Century tarnished lively popularity of Sugungga, while Confucianism was reintroduced and “ostentation of expertise” became more noticeable in Yakseongga. Such trends of pansori became more popular following the introduction of modern novels as well as radios and phonograph records. Although Yakseongga was not made to teach medicinal knowledge, it performed a role of distributing folk remedies to the public far beyond its original intention when it had been compiled based on Dongeui Bogam and Jejung Sinpyeon.

        Background : To understand Korean medicine it is crucial to first understand medical aspect of ‘DongUiBoGam’. It is also meaningful that it became one of the influential book even to posterity. Also based on the apprehension, we can take a view of development of Korean medicine led by ‘DongUiBoGam’. Objectives : This study aims to review the status, study field, specialist of DongUiBoGam. In the process, this review will grasp trends in this field of studies and will direct further researches into the right direction. Method : The computerized Korean databases were searched from their respective inceptions up to December 2008. The search terms used were ‘DongUiBoGam’ and random or Korean language terms related to DongUiBoGam. Several specialized journals were also manually searched for relevant articles. Result : Since the 2000s, DongUiBoGam papers in the Korean Literature is increased. Published 58 papers on The Korean Journal of Oriental Medical Prescription were more than the other journals. 58 papers published in The Korean Journal of Oriental Medical Prescription are the best in many areas related to Korean medicine. Most people have submitted papers related to DongUiBoGam was Woo-yeal Jeong. Conclusions : ‘DongUiBoGam’ research is conducted and current tendency and outlook for ‘DongUiBoGam’ is carried out focusing on several associations.

        Investigation on vascular cytotoxicity and extravascular transport of cationic polymer nanoparticles using perfusable 3D microvessel model

        <P><B>Abstract</B></P> <P>Vascular networks are the first sites exposed to cationic polymer nanoparticles (NPs) administered intravenously, and thus function as a barrier for NPs reaching the target organ. While cationic polymer NPs have been intensively studied as non-viral delivery systems, their biological effects in human microvessels have been poorly investigated due to a lack of appropriate <I>in vitro</I> systems. Here, we employed a three-dimensional microvessel on a chip, which accurately models <I>in vivo</I> conditions. An open and perfused microvessel surrounded by pericytes was shown to reproduce the important features of living vasculature, including barrier function and biomarkers. Using this microvessel chip, we observed contraction of the microvascular lumen induced by perfused polyethylenimine (PEI)/DNA NPs. We demonstrated that the oxidative stress present when microvessels were exposed to PEI NPs led to rearrangement of microtubules resulting in microvessel contraction. Furthermore, the transcytotic behavior of PEI NPs was analyzed in the microvessel by monitoring the escape of PEI NPs from the microvascular lumen into the perivascular region, which was not possible in two-dimensional culture systems. With our new understanding of the different behaviors of cationic polymer NPs depending on their transcytotic route, we suggest that caveolae-mediated transcytosis is a powerful route for efficient extravascular transport.</P> <P><B>Statement of Significance</B></P> <P>Microvascular networks are not only biological system constituting largest surface area in the body and but also first site exposed to nanoparticle <I>in vivo</I>. While cationic polymer NPs have been intensively studied as non-viral delivery systems, its biological effects in human microvessel have been poorly investigated due to lack of appropriate <I>in vitro</I> systems. Here, we microengineered an open and perfused 3D pericyte incorporated microvessel model which possesses same morphological characteristic of <I>in vivo</I>. Using the microengineered model, this study represents the first report of transcytotic behavior of NPs in 3D microvessel, and its effect on extravasation efficiency. Our study lays the groundwork for the integration of innovative technologies to examine blood vessel-nanoparticle interaction, which a critical but ill-defined phenomenon.</P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        Phosphorylation of rpS3 by Lyn increases translation of Multi-Drug Resistance (MDR1) gene

        Lyn, a tyrosine kinase that is activated by double-stranded DNA-damaging agents, is involved in various signaling pathways, such as proliferation, apoptosis, and DNA repair. Ribosomal protein S3 (RpS3) is involved in protein biosynthesis as a component of the ribosome complex and possesses endonuclease activity to repair damaged DNA. Herein, we demonstrated that rpS3 and Lyn interact with each other, and the phosphorylation of rpS3 by Lyn, causing ribosome heterogeneity, upregulates the translation of p-glycoprotein, which is a gene product of multidrug resistance gene 1. In addition, we found that two different regions of the rpS3 protein are associated with the SH1 and SH3 domains of Lyn. An in vitro immunocomplex kinase assay indicated that the rpS3 protein acts as a substrate for Lyn, which phosphorylates the Y167 residue of rpS3. Furthermore, by adding various kinase inhibitors, we confirmed that the phosphorylation status of rpS3 was regulated by both Lyn and doxorubicin, and the phosphorylation of rpS3 by Lyn increased drug resistance in cells by upregulating p-glycoprotein translation.         Changes of Ganglioside GM3 Expression in Porcine Oocyte Maturation and Early Embryonic Development In Vitro

        Gangliosides exist in glycosphingolipid-enriched domains on the cell membrane and regulate various functions such as adhesion, differentiation, and receptor signaling. Ganglioside GM3 by ST3GAL5 enzyme provides an essential function in the biosynthesis of more complex ganglio-series gangliosides. However, the role of gangliosides GM3 in porcine oocytes during in vitro maturation and early embryo development stage has not yet understood clear. Therefore, we examined ganglioside GM3 expression patterns under apoptosis stress during maturation and preimplantation development of porcine oocytes and embryos. First, porcine oocytes cultured in the NCSU-23 medium for 44 h after H2O2 treated groups (0.01, 0.1, 1 mM). After completion of meiotic maturation, the proportion MII (44 h) was significantly different among control and the H2O2 treated groups (76.8±0.3 vs 69.1±0.4; 0.01 mM, 55.7±1.0; 0.1 mM, 38.2±1.6%; 1 mM, P<0.05). The expressions of ST3GAL5 in H2O2 treated groups were gradually decreased compared with control group. Next, changes of ST3GAL5 expression patterns were detected by using immunofluorescene (IF) staining during preimplantation development until blastocyst. As a result, we confirmed that the expressions of ST3GAL5 in cleaving embryos were gradually decreased (P<0.05) according to the early embryo development progress. Based on these results, we suggest that the ganglioside GM3 was used to the marker as pro-apoptotic factor in porcine oocyte of maturation and early embryo production in vitro, respectively. Furthermore, our findings will be helpful for better understanding the basic mechanism of gangliosides GM3 regulating in oocyte maturation and early embryonic development of porcine in vitro.

        Tat-ATOX1 inhibits streptozotocin-induced cell death in pancreatic RINm5F cells and attenuates diabetes in a mouse model

        <P>Antioxidant 1 (ATOX1) functions as an antioxidant against hydrogen peroxide and superoxide, and therefore may play a significant role in many human diseases, including diabetes mellitus (DM). In the present study, we examined the protective effects of Tat-ATOX1 protein on streptozotocin (STZ)-exposed pancreatic insulinoma cells (RINm5F) and in a mouse model of STZ-induced diabetes using western blot analysis, immunofluorescence staining and MTT assay, as well as histological and biochemical analysis. Purified Tat-ATOX1 protein was efficiently transduced into RINm5F cells in a dose-and time-dependent manner. Additionally, Tat-ATOX1 protein markedly inhibited reactive oxygen species (ROS) production, DNA damage and the activation of Akt and mitogen activated protein kinases (MAPKs) in STZ-exposed RINm5F cells. In addition, Tat-ATOX1 protein transduced into mice pancreatic tissues and significantly decreased blood glucose and hemoglobin A1c (HbA1c) levels as well as the body weight changes in a model of STZ-induced diabetes. These results indicate that transduced Tat-ATOX1 protein protects pancreatic beta-cells by inhibiting STZ-induced cellular toxicity in vitro and in vivo. Based on these findings, we suggest that Tat-ATOX1 protein has potential applications as a therapeutic agent for oxidative stress-induced diseases including DM.</P>
