Background: A Poly-L-lactic acid (PLLA) is a soft tissue filler known to stimulate collagen production. Objectives: The purpose of this study is to analyze objective volumetric effect of PLLA in different setting of injection schedule on the cheek. Methods: A split-face, evaluator-blind randomized study in 24 volunteers was conducted. One side was injected 3 times with dose of 4cc per visit at week 0, 4, and 8, and the other side was injected 2 times with dose of 6cc per visit at week 4 and 8, respectively. Volunteers were evaluated with facial volume loss scale (FVLS) from 1 to 5(most severe) and objective volumetric values using Vectra-CR 3D camera system. Results: Improvement of FVLS was statistically significant (p<0.05) and maintained until 12 months in both injection side. Vectra showed volumetric difference (cc) between before and after injection. In 3 times injection side, it was increased from 2.12cc (after 1 month) to 3.17cc (after 12 months). In 2 times injection side, it was increased from 2.26cc (after 1 month) to 3.19cc (after 12 months). Gradual volumetric improvement over 12 months was statistically significant in both sides (P<0.05, generalized linear model). There was not statistically significant difference between three and two times injection side in both FVLS and Vectra. There was no severe adverse event. Conclusion: PLLA filler has continuous volumetric effect and there was not a significant difference by injection times at the same total injection volume.

The structure development and dynamic properties of fibers produced by high-speed spinning of P(EN-ET) random copolymers were investigated. The as-spun fibers were found to remain amorphous up to the spinning speed of 1500 m/min, and subsequent increases in speed resulted in the crystalline domains containing primarily $\alpha$ crystalline modification of PEN. The f modification was not found up to spinning speeds of 4500 m/min. On the other hand, annealing of constrained fibers spun at the 2100 m/min at 180,200, and 240^{\circ}C$ exhibited $\beta$-form crystalline structure, while the annealed fibers spun in 600-1500 m/min range exhibited dominantly $\alpha$-form. However $\beta$-form crystals disappeared above the spinning speed of 3000 m/min. With increasing spinning speeds from 600 to 4500 m/min, the storage modulus of as-spun fibers increased continuously and reached a value of about 10.4 spa at room temperature. The tan $\delta$curves showed the $\alpha$-relaxation peak at about 155-165^{\circ}C$, which is considered to correspond to the glass transition. The $\alpha$-relaxation peaks became smaller and broader, and shift to higher temperatures as the spinning speed increases, meaning that molecular mobility in the amorphous region is restricted by increased crystalline domain.

The aim of this study was to investigate the effect of charge carrier lipid on the skin penetration, retention, and hair growth of topically applied finasteride-containing liposomes. Finasteride-containing liposomes were prepared by traditional thin film hydration method using Phospholipon(R) 85 G and cholesterol with or without charge carrier lipid (1,2 dimyristoyl-sn-glycero-3-phosphate or 1,2-dioleoyl-trimethylammonium-propane for anionic and cationic charge, respectively). Freshly prepared finasteride-containing liposome suspension was applied on the hairless mouse skin, and skin penetration and retention were measured using Keshary-Chien diffusion cell. Non-liposomal formulation (ethanol 10% solution containing 0.5 mg/ml of FNS) was also used as a control. The amount of finasteride in the diffusion cell and mouse skin was measured by HPLC. The hair growth was evaluated using depilated male C57BL/6N mice. Mean particle size of all finasteride-containing liposomes was less than a micron, and polydispersity index revealed size homogeneity. Skin penetration and retention studies showed that significantly less amount of finasteride was penetrated when applied as anionic liposome while more amount of the drug was retained. Specifically, in liposome prepared with 10% anionic charge carrier lipid, penetration was 12.99 μg/cm2 while retention was 79.23 μg/cm2 after 24 h of application. In hair growth study, finasteride-containing anionic liposomes showed moderate efficacy, but the efficacy was not found when applied as cationic liposomes. In conclusion, topical application of finasteride using anionic liposome formulation appears to be useful option for the treatment of androgenetic alopecia to avoid systemic side effects of the drug.
Effects of Proton on Lysolipid-induced Actions in OGR1-subfamily GPCRs

Lysolipids such as lysophosphatidylcholine (LPC), sphingosylphosphorylcholine (SPC), galactosyl-sphingosine (psychosine) have been matched as ligands for OGRI-subfamily G-protein-coupled receptors (GPCR), consisted of OGRl, GPR4, G2A, and TDAG8. Recently, those members of GPCRs have been reported as proton-sensing GPCRs. We used Jurkat T cells, which express four members of OGRI subfamily GPCRs endogenously to investigate effects of proton on lysolipid-induced several cellular events. We found no significant effect of proton on the lysolipid-induced Ca2+ increase and ROS production in Jurkat T cells. Further investigation is necessary to clarify the relationship of lysolipid and proton on the OGRI-subfamily GPCRs.
