A cDNA fragment encoding the chloroplastic fructose-1, 6-bisphosphatase (FBPase) was cloned via PCR from the cDNA library of pea leaves. The cloned cDNA, about 1.05 kbp without signal sequence, was introduced into a pET-28a vector for expression in E. coli
Field emission from the film of the finely dispersed arc discharge black core material
Hahn, Joeoong,Yoo, Jae-Eun,Han, Jaeik,Kwon, Hyok Bo,Suh, Jung Sang Elsevier 2005 Carbon Vol.43 No.5
<P><B>Abstract</B></P><P>We have studied, for the first time, the field emission from the film, prepared by a spray method, of the finely dispersed black core material, including multi-walled carbon nanotubes (MWNTs), fabricated by arc discharge. We dispersed the black core material by using an ultrasonic processor and found that the dispersed ones were much finer than those observed when treated with a ball mill and normal ultra-sonic bath. By SEM, HRTEM and Raman analyses, the MWNTs were almost not deformed and damaged during ultra-sonication. The field emission current density measured from the film of the dispersed black core material was about 15mA/cm2 at an applied field of 8V/μm, which was about 23 times higher than that found by a ball mill. A current density of 1mA/cm2, which is required basically for flat panel display, has been obtained at 5.3V/μm. The lifetime test of the dispersed black core material showed that the current density was almost unchanged while the field was applied. Therefore, it is concluded that a black core material fabricated by arc discharge could be used to flat panel displays as field emitters by dispersing with an ultrasonic processor, without further treatment like extraction or purification.</P>
Gene editing platform in Cattle

The CRISPR/Cas9 system is proved to be a powerful tool for knock-out and knock-in in various species. By introducing genetic materials of two components (Cas9 and small guide (sg) RNA) into cells or pronuclear of the fertilized embryo, gene editing occurs. Some studies reported that efficiency of gene editing would be increased as Cas9 was integrated into cells or animals since Cas9 is indispensable in the CRISPR/Cas9 system. Accordingly, the production of Cas9 expressing cattle may provide the broadly used gene editing platform in cattle. For this study, Cas9 and RFP genes were cloned into PiggyBac (PB) transposon system. PB-Cas9-RFP and transposase were microinjected into 1436 in vitro fertilized embryos and 241 blastocysts were formed. Blastocysts with RFP expression accounting for 14.1% of total formed blastocysts were selected and transferred into 5 recipient cow. After gestation periods, four transgenic cattle were delivered without any veterinary assistance. From a transgenic cattle, ear skin tissue was collected for primary culture. On those primary cells, sgRNAs in DNA form for various genes such as PRNP, RB1 and BLG were transfected as 2ug of sgRNA per 5x105 cells using Nucleo factor system (Neon®, invitrogen, program#16). As expected, every group of each sgRNA delivered was confirmed to be mutated by T7E1assay. Those data demonstrated that for the first time, transgenic cattle with Cas9 expression were born, grown up to date and will be avaluable resource for genome-editing in cattle. Intraventricular Vancomycin Therapy for Intractable Bacillus cereus Ventriculitis
Hahn, Jong Woo,Ju, Hee young,Park, Meerim,Yi, Eun Sang,Park, Byung-Kiu,Shin, Sang-Hoon,Lee, Sang-Hyun,Park, Hyeon Jin,Kang, Ji-Man The Korean Society of Pediatric Infectious Disease 2019 Pediatric Infection and Vaccine Vol.26 No.2
Bacillus cereus causes serious central nervous system infections, especially in immunocompromised patients. Successful treatment requires adequate antimicrobial concentrations in the cerebrospinal fluid; however, in some cases, achieving this with systemic treatment alone is difficult. We treated intractable B. cereus ventriculitis with intraventricular vancomycin, with no major adverse events.
A Simple Method for Generation of Homologous Internal Standards for Competitive PCR
Eun A Choi,Sung Sik Hahn,Kyung Hee Choi,Doe Sun Na 생화학분자생물학회 1996 BMB Reports Vol.29 No.5
In competitive PCR, which is used to quantify target DNA, an internal standard is needed. Here we present a simple method to construct homologous competitive standards. The method, which is based upon deletion of a portion of the target DNA, does not require any additional primers. This is the simplest method developed thus far to construct a competitive standard. The whole procedure, from construction of a competitive standard to quantitation by PCR, can be completed within a single day.
