        황색포도상구균 신속 검출용 간이 진단키트

        This study was performed to develop a rapid test kit for pathogenic Staphylococcus in various samples. The rapid detection kit has been fabricated based on nitrocellulose lateral-flow strip. Colloidal gold and Staphylococcus antibodies were used as a tag and a receptor, respectively. Manually spotted Staphylococcus antibody and anti-mouse antibody on the surface of nitrocellulose membrane were used as test and control lines, espectively. Feasibility of the rapid kit to detect Staphylococcus aureus in samples were evaluated. The intensity of the color of the tes line started to increase with the samples in which higher concentration of the cells were contained. The sensitivity of the sensor was 106 cfu/mL Staphylococcus spiked in PBS. Also, the rapid test kit could detect 105 cfu/mL of Staphylococcus in chicken meat extract.

        Altered PLCβ-1 expression in the gerbil hippocampal complex following spontaneous seizure

        Although the phospholipase C (PLC)β-1 isoform is associated with spontaneous seizure and distinctively expressed in the telencephalon, the distribution of PLCβ-1 expression in the epileptic gerbil hippocampus remains controversial. Therefore, we determined whether PLCβ-1 is associated with spontaneous seizure in an animal model of genetic epilepsy. In the present study, PLCβ-1 immunoreactivity was down-regulated in seizure-sensitive (SS) gerbils more than in seizure-resistant (SR) gerbils. The expression of PLCβ-1 within calretinin (CR)-positive neurons was rarely detected within the dentate hilar region of SS gerbils. PLCβ-1 immunoreactivity in the hippocampus was significantly elevated as compared to that in pre-seizure SS gerbil 3 h post-ictal. These findings suggest that alterations in PLCβ-1 immunoreactivity in the SS gerbil hippocampus may be closely related to the epileptic state of the gerbil brain and transiently elevated PLCβ-1 protein levels following seizure episodes. Such alterations may be compensatory responses in the SS gerbil hippocampus. [BMB reports 2011; 44(9) : 566-571]

        Shinzami Korean purple‐fleshed sweet potato extract prevents ischaemia–reperfusion‐induced liver damage in rats

        <P><B>Abstract</B></P><P><B>BACKGROUND</B></P><P>This study was designed to investigate the hepatoprotective effect of an extract from Shinzami, a variety of purple sweet potato, in rats injured by hepatic ischaemia–reperfusion (I/R).</P><P><B>RESULTS</B></P><P>Pretreatment with Shinzami extract decreased the aspirate aminotransferase and alanine aminotransferase serum levels in our hepatic I/R rat model. The glutathione level and superoxide dismutase activity level were significantly higher in the rats pretreated with the Shinzami extract compared with the hepatic I/R rats, and the glutathione peroxidase activity level was higher in pretreated rats. The total anthocyanins extracted from Shinzami, however, only increased the superoxide dismutase activity level in the hepatic I/R rats. Rats pretreated with the Shinzami extract or anthocyanins demonstrated attenuated hepatic pathological changes, such as hepatic distortion, haemorrhage, necrosis and inflammatory cell infiltration compared with the hepatic I/R control rats.</P><P><B>CONCLUSION</B></P><P>Shinzami extract and anthocyanins have a hepatoprotective effect on the liver damage induced by hepatic I/R by improving antioxidant status. Furthermore, the Shinzami extract may have a more potent effect on the antioxidant status compared with the Shinzami anthocyanins alone. © 2014 Society of Chemical Industry</P>

        Direct Conversion of Adult Skin Fibroblasts to Endothelial Cells by Defined Factors

        <P><B>Background—</B></P><P>Cell-based therapies to augment endothelial cells (ECs) hold great therapeutic promise. Here, we report a novel approach to generate functional ECs directly from adult fibroblasts.</P><P><B>Methods and Results—</B></P><P>Eleven candidate genes that are key regulators of endothelial development were selected. Green fluorescent protein (GFP)–negative skin fibroblasts were prepared from Tie2-GFP mice and infected with lentiviruses allowing simultaneous overexpression of all 11 factors. Tie2-GFP<SUP>+</SUP> cells (0.9%), representing Tie2 gene activation, were detected by flow cytometry. Serial stepwise screening revealed 5 key factors (Foxo1, Er71, Klf2, Tal1, and Lmo2) that were required for efficient reprogramming of skin fibroblasts into Tie2-GFP<SUP>+</SUP> cells (4%). This reprogramming strategy did not involve pluripotency induction because neither Oct4 nor Nanog was expressed after 5 key factor transduction. Tie2-GFP<SUP>+</SUP> cells were isolated using fluorescence-activated cell sorting and designated as induced ECs (iECs). iECs exhibited endothelium-like cobblestone morphology and expressed EC molecular markers. iECs possessed endothelial functions such as <I>Bandeiraea simplicifolia</I>-1 lectin binding, acetylated low-density lipoprotein uptake, capillary formation on Matrigel, and nitric oxide production. The epigenetic profile of iECs was similar to that of authentic ECs because the promoters of VE-cadherin and Tie2 genes were demethylated. mRNA profiling showed clustering of iECs with authentic ECs and highly enriched endothelial genes in iECs. In a murine model of hind-limb ischemia, iEC implantation increased capillary density and enhanced limb perfusion, demonstrating the in vivo viability and functionality of iECs.</P><P><B>Conclusions—</B></P><P>We demonstrated the first direct conversion of adult fibroblasts to functional ECs. These results suggest a novel therapeutic modality for cell therapy in ischemic vascular disease.</P>

        고등학교 가정과 메이커 수업의 효과

        본 연구는 메이커 교육의 3요소(창작자, 창작활동, 창작공간)를 갖추어 고등학교 가정과 수업시간에 실행한 메이커 교육의 사례를 고찰해 가정과 교육과정에서의 메이커 교육의 가능성과 수업 효과를 탐색한 연구이다. 연구자는 대구 S고등학교 1학년 240명을 대상으로 가정과 수업시간에 12차시의 가정과 메이커 수업을 실행한 뒤, 9명(남학생 2명, 여학생 7명)의 연구 참여자와 심층 인터뷰를 실행하였다. 연구 결과, 가정과 메이커 수업의 효과를 1) 인지 발달 2) 자아 성장 3) 사회성 향상 3가지 주제로 범주화 할 수 있었다. 학습자들은 가정과 메이커 수업이 수업 내용의 이해 및 자신들의 고차원적 사고 발달에 도움이 되었으며 주체성, 자신감과 자존감, 도전 정신 등 자아 성장에도 도움이 된다고 인식하였다. 또한 학습자들은 가정과 메이커 수업을 통해 친구 관계의 발전 뿐만 아니라 사회에 관한 인식의 향상을 보여주었다. 이러한 연구 결과를 기반으로 연구자는 가정과 메이커 수업에서 교사의 역할과 가정 수업시간을 활용한 메이커 교육 방안에 관한 시사점을 제안하였고, 가정과 메이커 교육을 학내 교우 관계 등 사회 정서적 측면의 문제를 예방하는 데 효과적인 교육의 한 방안으로 학교 정규 교육으로서의 활용을 강조하였다. Maker education is a process-oriented education method that gives learners various educational effects while completing a specific outcome to actively resolve individual interests or community issues in a class. Although researches to introduce maker education into school curriculums are recently conducted, there are still insufficient studies examining the effects of maker education as a school subject to meet the standards of achievement in a regular curriculum. Another limitation is that most studies are focused on quantitative research. Thus, this study conducted a home economics class in Chapter 6 of high school home economics course to 240 first-year student at S high school in Daegu, as well as had an in-depth interview with 9 participants (2 male students and 7 female students). Through the results, this research could categorize the meaning of home economics maker education into three: 1) cognitive development; 2) Self-growth; 3) social improvement. The students perceived that home economics maker class helped them to develop higher-dimensional thinking, understand their class contents, and grow themselves such as independence, confidence, self-esteem, and challenging spirit. In addition, the students showed not only the development of friendships but also the improvement of their awareness on society through the home economics maker class. Based on these study findings, the researcher proposed implications for the role of teachers in home economics maker class and a curriculum, and emphasized the application of home economics maker class in school education as an effective means to prevent problems in relations and emotions at school.

        고등학생을 위한 ‘떡의 세계화’ 실습 프로그램 개발과 평가

        본 연구는 고등학교 가정과의 ‘떡의 세계화’ 실습 프로그램을 개발·실행·평가하여 효과를 살펴본 연구이다. 연구결과, 총 9차시의 실습 프로그램을 개발되었다. 그 내용은 우리나라 떡에 대해 알기, 백설기/절편/화전/경단 만들기, 떡의 세계화 수행평 가이다. 개발된 프로그램은 대구광역시 소재 인문계 고등학생 1학년 278명에게 2017 년 9월 20일부터 11월 6일까지 연구자가 직접 실행하였다. 수업의 효과는 2019년 1 월 2일부터 1월 8일까지 연구자가 개발한 온라인 설문지를 통해 평가하였다. 최종 수집된 102부의 설문지를 분석한 결과, 학생들의 떡의 가치에 대한 인식(4.25), 떡의 세계화 인식(4.22), 한식의 세계화 인식(4.18), 떡만들기에 대한 자신감(4.00), 한식 조리에 대한 자신감(3.72), 생활 자립 능력(3.90)이 향상되었다고 인식하였다. 개방형 질문에서 학생들은 이 수업을 통하여 창의적 표현능력이 향상되었고, 수업에 집중이 잘되었으며, 기회가 생기면 스스로 혹은 가족과 함께 창조적인 방법으로 떡만들기를 수행하고 싶다고 하였다. 이에 가정과에서는 중·고등학생을 대상으로 ‘떡의 세계화’ 실습 프로그램 외에도 한식의 가치와 우수성을 인식시키고 계승·발전시킬 수 있는 다양한 실습 프로그램을 개발할 필요가 있다. This study was designed to explore the effects of developing, implementing and evaluating the Globalization of Rice cake practice programs applicable to Home Economics course. A total of nine practice programs were developed, and the researcher directly carried out the practice program towards 278 students of a high school located in Daegu Metropolitan City from September 20 th to November 6 th in 2017. We conducted a survey to evaluate the effectiveness of the practice program in January 2019. With 102 samples, it has shown that the awareness of the value of rice cake (4.25), the globalization of rice cake (4.22), and globalization of Korean food (4.18) were improved. In addition, it was confirmed that confidence in making rice cake (4.00) and cooking Korean cuisine (3.72) as well as self-sufficiency (3.90) were increased. Furthermore, students answered that the practice program made them have creative expression and focus more on the class. They wanted to make rick cake in a creative way with their family later. Therefore, it is necessary to develop a variety of practice programs for middle and high school students to recognize the value and excellence of Korean food and to inherit and develop it.

        고등학생의 친환경 의생활을 위한 업사이클링 가정과 교수·학습 과정안 개발과 평가

        본 연구의 목적은 실습법을 적용하여 「기술·가정」교과의 가정 분야에 적용할 수 있는 친환경 의생활을 위한 업사이클링 교수·학습 과정안을 개발·실행·평가하여 청소년의 친환경 의생활 실천을 돕는 데 있다. 본 연구는 ADDIE 교수설계모형의 5단계의 과정을 밟아 진행되었다. 분석 단계에서는 2009 개정 고등학교 가정 교육과정의 성취기준, 교과서, 친환경 의생활, 업사이클링, 실습법 등의 문헌과 선행 연구를 분석하였고, 설계 단계에서는 수업 주제, 내용, 방법, 평가 요소 및 방법을 설계하였다. 개발 단계에서는 ‘실습법을 적용한 업사이클링 교수·학습 과정안’을 예비 개발한 후 업사이클링 전문가의 피드백을 받아, 최종적으로 총 9차시의 교수·학습 과정안을 개발하였다. 실행 단계에서는 고등학교 1학년 278명에게 연구자가 직접 최종 개발된 교수·학습 과정안에 따라 수업을 실행하였다. 평가 단계에서는 학생 대상 온라인 설문 조사를 통해 이 수업을 평가하였다. 이 수업에 대한 학생들의 평가 결과, 학생들은 이 수업이 평균 5점 만점에서 4점 이상으로 친환경 의생활 인식(4.15)과 업사이클 링에 자신감(4.06)을 향상시켜준다고 인식하였다. 또한 그들은 실습법을 적용한 업사이클링 수업이 새로운 경험이라 신기하였고, 자신이 만들어서 가치가 있다고 인식하였으며, 재봉틀이라는 기계를 다룰 수 있다는 자신감이 생겼고, 수업의 과정에 더 잘 집중하여 오래 기억에 남는다고 평가하였다. 학생들은 가정교과에서 더 다양한 실습수업을 마련할 것과 블록타임제 운영을 통한 충분한 실습 시간의 확보, 그리고 실습을 제대로 할 수 있는 가정실의 시설 정비를 요구하였다. The purpose of this study was to develop and evaluate the up-cycling Home Economics teaching·learning process plans for high school students eco-friendly clothing life applying the practice methods. To achieve the purpose of the study, the ADDIE instructional design model was used. In the analysis stage, literature and previous researches related to up-cycling, eco-friendly lifestyle, and the practice method were reviewed and the 2009 revised high school 「Technology·Home Economics」 curriculum and Home Economcis textbooks were analyzed. At the design stage, subject, learning content, learning process applying the practice method, teaching strategies of the lesson plans were designed. At the development stage, preliminary up-cycling teaching·learning process plans applying the practice method were developed, and then the final plans were developed as the total of 9 classes after up-cycling expert s feedback of preliminary plans. In the implementation stage, the researcher of Home Economics teacher personally implemented the up-cycling Home Economics lessons applying the practice method for the 278 high school freshman students attending S high school in Daegu city. In the evaluation stage, these lessons were evaluated through student online questionnaires. As a result of the students evaluation of the up-cycling Home Economics lessons for eco-friendly lifestyle applying the practice method, the students recognized that self-confidence of environment-friendly lifestyle and up-cycling was improved through these lessons. The students also felt that the lessons applying the practice method were a new experience, they recognized that they were worth making. And students responded that they devoted themselves on the lessons more and the lessons were memorable for a long time. The students needed more lessons applying practice methods in home economics, to secure sufficient time for practice through block-time operation, and to improve the facilities of the home economics class room for practice.

        2015 개정 고등학교 가정 교과서의 ‘생활문화’ 핵심개념 단원 분석

        본 연구의 목적은 2015 개정 교육과정의 고등학교 가정 교과서(「기술·가정」 교과서의 가정생활 영역(2018학년도 발간)) 내용 중 ‘생활문화’ 핵심개념 단원의 텍스 트와 활동과제를 분석하는 것이다. 분석대상은 텍스트의 경우, 연구자가 pdf 혹은 hwp 텍스트 원문을 수집할 수 있었던 2015 개정 고등학교 가정과 교과서 5종을 대상으로 하고, 활동과제는 12종을 대상으로 하였다. 텍스트 분석은 전통 식생활문화, 전통 의생활 문화, 전통 주생활 문화의 주제로 나누어 TEXTOM프로그램의 텍스트 마이닝을 활용해 각각 분석하고 활동과제 분석의 경우 연구자가 개발하고 가정과 전문가의 피드백을 받아 수정·보완한 분석틀을 활용해 분석하였다. 2015 개정 고등학교 「기술·가정」 교과서의 가정 분야 ‘생활문화’ 의 핵심개념에 대한 텍스트 분석결 과, ‘전통 식생활문화’주제는 식생활 문화를 음식을 통해 다루고 있었으며, ‘전통 의생활문화’주제는 전통 의생활문화의 현대 의생활의 창의적 활용을, ‘전통 주생활문 화’주제는 한옥의 가치와 다른 나라의 주생활문화이해를 통한 친환경적인 주생활의 실천을 중점적으로 다루고 있었다. 활동과제의 경우는 ‘생활문화’의 내용 요소에 따라 분류된 활동과제는 총 171개이며, 가치 51개(29.8%), 문화 44개(25.7%), 실천 76개 (44.4%)로, 전통생활문화의 가치와 다른 나라의 생활문화의 이해를 바탕으로 현대의 생활문화에 창의적으로 실천하는 활동과제가 많이 제시되어 텍스트와 활동과제가 일관성 있게 구성되어 있음을 확인할 수 있었다. The purpose of this study is to analyze the texts and activity tasks of the core concept section, Living Culture among the contents of the Home Economics curriculum from high school textbooks revised in 2015 and published in 2018. We had five Home Economics textbooks of high school to analyze the texts, and we investigated twelve high school textbooks for analyzing activity tasks. In the texts of high school Home Economics textbooks, we divided them into three topics, Traditional Korean Food Culture , Traditional Korean Clothing Culture , and Traditional Korean Housing Culture . We used TEXTOM to analyze the texts as well as analysis framework proposed and developed by the researchers of this study for activity tasks. The results indicates that the theme, Traditional Korean Food Culture mainly deals with the eating culture, and another theme, Traditional Korean Clothing Culture includes the way of creatively using the traditional clothing culture in contemporary fashion. The last theme, Traditional Korean Housing Culture focuses on the practice of environment-friendly housing through understanding the value of Traditional Korean Housing and housing culture in other countries. In the case of activity tasks, there are 171 activity tasks classified according to the content-elements of life culture , and the ‘value’ is 51 (29.8%), ‘culture’ is 44 (25.7%) and ‘practice’ is 76 (44.4%). It was confirmed that text and activity tasks had consistently similar contents in that each textbook mainly presented not only the importance to understand the value of traditional living culture of Korea and other countries, but they provided activity tasks to practice the traditional and foreign living culture in modern living culture.
