        The analytical data of general components and minerals in milk, chocolate milk and yoghurts were as follows; 1) Milk; Water content was about 89.2% and ash 0.73g%, lactose 4.7g%, protein 3.4g%, lipid 2.3g%, iron 0.09mg%, calcium 167.2mg% and magnesium 13.4mg% were obtained. 2) Chocolate milk: Water content was about 87.0% and ash 0.5g%, lactose 6.2g%, protein 2.4g%, lipid 1.4mg%, iron 0.1mg% calcium 118.6mg%, and magnesium 8.1mg% were obtained. 3) Yoghurt: Water content was about 81.2% and ash 0.22g%, lactose 3.7g%, protein 1.1g%, lipid 0.3g%, iron 0.06mg%, calcium 112.0mg% and magnesium 5.4mg% were obtained.

      • Involvement of S6K1 in mitochondria function and structure in HeLa cells

        The major biological function of mitochondria is to generate cellular energy through oxidative phosphorylation. Apart from cellular respiration, mitochondria also play a key role in signaling processes, including aging and cancer metabolism. It has been shown that S6K1-knockout mice are resistant to obesity due to enhanced beta-oxidation, with an increased number of large mitochondria. Therefore, in this report, the possible involvement of S6K1 in regulating mitochondria dynamics and function has been investigated in stable lenti-shS6K1-HeLa cells. Interestingly, S6K1-stably depleted HeLa cells showed phenotypical changes in mitochondria morphology. This observation was further confirmed by detailed image analysis of mitochondria shape. Corresponding molecular changes were also observed in these cells, such as the induction of mitochondrial fission proteins (Drp1 and Fis1). Oxygen consumption is elevated in S6K1-depeleted HeLa cells and FL5.12 cells. In addition, S6K1 depletion leads to enhancement of ATP production in cytoplasm and mitochondria. However, the relative ratio of mitochondrial ATP to cytoplasmic ATP is actually decreased in lenti-shS6K1-HeLa cells compared to control cells. Lastly, induction of mitophagy was found in lenti-shS6K1-HeLa cells with corresponding changes of mitochondria shape on electron microscope analysis. Taken together, our results indicate that S6K1 is involved in the regulation of mitochondria morphology and function in HeLa cells. This study will provide novel insights into S6K1 function in mitochondria-mediated cellular signaling.

        The muscle protein polymorphisms in korean native cattle, holstein, and imported beefs from Australia and U.S.A. was investigated to identify breed-specific muscle proteins, especially korean native cattle specific proteins. Muscle proteins was divided into three fractions; fraction A which contained mainly blood proteins, fraction B which contained mainly enzyme and soluble proteins and fraction C which contained mainly structural proteins. On the analysis of fraction A by SDS-polyacrylamide gel electrophoresis, the molecular weight (mw) 218k of protein band was appeared in only imported beefs from Australia not in korean native cattle, holstein, and imported beefs from U.S.A. Similarly, the mw 117k of protein band was appeared in only imported beefs from U.S.A. not in korean native cattle, holstein, and imported beefs from Australia. But the mw 35k of protein band was appeared in korean native cattle, holstein and imported beefs from Australia not in U.S.A. In fraction B, the mw 92k of protein band was appeared in only imported beefs from U.S.A. not in korean native cattle, holstein, and imported beefs from Australia. However, the mw 33k of protein band was appeared in korean native cattle, holstein and imported beefs from Australia not in U.S.A. In fraction C, the mw 155k of protein band was appeared in korean native cattle, holstein and imported beefs from Australia not in U.S.A. However the mw 120k of protein band was appeared in only imported beefs from U.S.A. not in korean native cattle, holstein, and imported beefs from Australia. Similarly the mw 110k of protein band was appeared in only imported beefs from Australia not. in korean native cattle, holstein, and imported beefs from U.S.A. Furthermore, the mw 54k and 28.5K of protein bands were not appeared in only imported beefs from Australia but in korean native cattle, holstein, and imported beefs from U.S.A on the analysis of muscle proteins by secondary electrophoresis, lot of breed-specific muscle proteins were identified. Therefore, the results of this study suggested that the breed-specific beefs can lie identified by the analysis of muscle proteins.

      • The microRNA miR-124 inhibits vascular smooth muscle cell proliferation by targeting S100 calcium-binding protein A4 (S100A4)

        <P>S100 calcium-binding protein A4 (S100A4) induces proliferation and migration of vascular smooth muscle cells (VSMCs). We aimed to find the microRNA regulating S100A4 expression. S100A4 transcripts are abruptly increased in the acute phase of carotid arterial injury 1 day later (at day 1) but gradually decreases at days 7 and 14. Bioinformatics analysis reveals that miR-124 targets S100A4. VSMC survival is attenuated by miR-124 mimic but increased by miR-124 inhibitor. miR-124 decreases immediately after carotid arterial injury but dramatically increases at days 7 and 14. miR-124 inhibitor-induced cell proliferation is blocked by S100A4 siRNA, whereas miR-124-induced cell death is recovered by S100A4. Our findings suggest that miR-124 is a novel regulator of VSMC proliferation and may play a role in the development of neointimal proliferation.</P>

      • Modeling and simulation of HTS cables for scattering parameter analysis

        <P>Most of modeling and simulation of high temperature superconducting (HTS) cables are inadequate for high frequency analysis since focus of the simulation's frequency is fundamental frequency of the power grid, which does not reflect transient characteristic. However, high frequency analysis is essential process to research the HTS cables transient for protection and diagnosis of the HTS cables. Thus, this paper proposes a new approach for modeling and simulation of HTS cables to derive the scattering parameter (S-parameter), an effective high frequency analysis, for transient wave propagation characteristics in high frequency range. The parameters sweeping method is used to validate the simulation results to the measured data given by a network analyzer (NA). This paper also presents the effects of the cable-to-NA connector in order to minimize the error between the simulated and the measured data under ambient and superconductive conditions. Based on the proposed modeling and simulation technique, S-parameters of long-distance HTS cables can be accurately derived in wide range of frequency. The results of proposed modeling and simulation can yield the characteristics of the HTS cables and will contribute to analyze the HTS cables. (C) 2016 Elsevier B.V. All rights reserved.</P>

        Full-length genomic analysis of porcine G9P[23] and G9P[7] rotavirus strains isolated from pigs with diarrhea in South Korea

        Group A rotaviruses (RVAs) are agents causing severe gastroenteritis in infants and young animals. G9 RVA strains are believed to have originated from pigs. However, this genotype has emerged as the fifth major human RVA genotype worldwide. To better understand the relationship between human and porcine RVA strains, complete RVA genome data are needed. For human RVA strains, the number of complete genome data have grown exponentially. However, there is still a lack of complete genome data on porcine RVA strains. Recently, G9 RVA strains have been identified as the third most important genotype in diarrheic pigs in South Korea in combinations with P[7] and P[23]. This study is the first report on complete genome analyses of 1 G9P[7] and 3 G9P[23] porcine RVA strains, resulting in the following genotype constellation: G9-P[7]/P[23]-I5-R1-C1-M1-A8-N1-T1-E1-H1. By comparisons of these genotype constellations, it was revealed that the Korean G9P[7] and G9P[23] RVA strains possessed a typical porcine RVA backbone, similar to other known porcine RVA strains. However, detailed phylogenetic analyses revealed the presence of intra-genotype reassortments among porcine RVA strains in South Korea. Thus, our data provide genetic information of G9 RVA strains increasingly detected in both humans and pigs, and will help to establish the role of pigs as a source or reservoir for novel human RVA strains.

        Cytogenetic heterogeneity and their serial dynamic changes during acquisition of cytogenetic aberrations in cultured mesenchymal stem cells

        To minimize the risk of tumorigenesis in mesenchymal stem cells (MSCs), G-banding analysis is widely used to detect chromosomal aberrations in MSCs. However, a critical limitation of G-banding is that it only reflects the status of metaphase cells, which can represent as few as 0.01% of tested cells. During routine cytogenetic testing in MSCs, we often detect chromosomal aberrations in minor cell populations. Therefore, we aimed to investigate whether such a minority of cells can expand over time or if they ultimately disappear during MSC passaging. We passaged MSCs serially while monitoring quantitative changes for each aberrant clone among heterogeneous MSCs. To investigate the cytogenetic status of interphase cells, which represent the main population, we also performed interphase FISH analysis, in combination with G-banding and telomere length determination. In human adipose tissue-derived MSCs, 4 types of chromosomal aberrations were found during culturing, and in umbilical cord MSCs, 2 types of chromosomal aberrations were observed. Sequential dynamic changes among heterogeneous aberrant clones during passaging were similar to the dynamic changes observed in cancer stem cells during disease progression. Throughout all passages, the quantitative G-banding results were inconsistent with those of the interphase FISH analysis. Interphase FISH revealed hidden aberrations in stem cell populations with normal karyotypes by G-banding analysis. We found that telomere length gradually decreased during passaging until the point at which cytogenetic aberrations appeared. The present study demonstrates that rare aberrant clones at earlier passages can become predominant clones during later passages. Considering the risk of tumorigenesis due to aberrant MSCs, we believe that our results will help to establish proper safety guidelines for MSC use. In particular, we believe it is critical to test for chromosomal aberrations using both G-banding and FISH to ensure the safety of human stem cells that are manufactured in vitro for clinical applications.

      • 간호대학생의 이론・실습 성적과 임상수행능력의 관련성 : 성인간호학 교과목을 중심으로

        Purpose: This study aimed to explore the relationship among score of theory course, score of practicum course and clinical practice ability by the course, Adult Health Nursing. Methods: A descriptive research was conducted using questionaries. Data were collected from September 25 to October 2, 2019. Participants were 67 nursing students at one university in Seoul on their junior year who have experienced clinical settings. The collected data were analyzed by t-test, ANOVA, Cohen’s Kappa, and Scheffe’s test using SPSS 25.0 version program. Results: Participants were given a quartile number from 1 to 4 respectively according to their scores of theory course those of practicum course. The total mean score of clinical practice ability was 3.61, with a perfect score of 5. The scores of theory course and practicum course showed slight correlation(Cohen’s Kappa=.168). However, significant correlation did not existed among score of theory course, score of practicum course and clinical practice ability. Conclusion: The result showed that score of theory course, score of practicum course and clinical practice ability have no significant correlation. However, its finding suggests the necessity of further research about various approach and new evaluation methods needed in order to improve nursing students’ competency.
