Serum Periplakin as a Potential Biomarker for Urothelial Carcinoma of the Urinary Bladder

The objectives of this study were to examine serum periplakin expression in patients with urothelial carcinoma of the urinary bladder and in normal controls, and to examine relationships with clinicopathological findings. Detection of serum periplakin was performed in 50 patients and 30 normal controls with anti-periplakin antibodies using the automatic dot blot system, and a micro-dot blot array with a 256 solid-pin system. Levels in patients with urothelial carcinoma of the urinary bladder were significantly lower than those in normal controls (0.31 and 5.68, respectively; p<0.0001). The area under the receiver-operator curve level for urothelial carcinoma of the urinary bladder was 0.845. The sensitivity and specificity, using a cut-off point of 4.045, were 83.7% and 73.3%, respectively. In addition, serum periplakin levels were significantly higher in patients with muscle-invasive cancer than in those with nonmuscle-invasive cancer (P = 0.03). In multivariate Cox proportional hazards regression analysis, none of the clinicopathological factors was associated with an increased risk for progression and cancer-specific survival. Examination of the serum periplakin level may play a role as a non-invasive diagnostic modality to aid urine cytology and cystoscopy.

Evenki is a language belonging to the Tungus language family and prevailing through East Siberia widely. In this paper, I argue that the suffix -yA in Evenki has an ergativelike feature. This suffix generally appears in next two situations: one is as what is usually called ‘indefinite accusative’, and the other is with nouns before the negative noun ??in. In earlier literatures some treated this suffix as an identical suffix, and others as two different suffixes. There is no common view. I selected up all the examples including the suffix -yA from some texts, and found a peculiar distribution. It appears as next two roles: the one is as an object of transitive verbs, and the other as a subject in the sentences expressing non-existence especially with ??in. In addition it appears as an object more often in the negative sentences with indefinite pronouns. From this distribution I redefine the suffix -yA as the case marker expressing objects not existing before a speaker, or indefinite objects existing before a speaker. It is able to say that the suffix -yA in Evenki, which is typically an accusative language, plays a role as a subsidiary ergative system to express indefiniteness or non-existence. In conclusion the suffix -yA should be treated as an identical suffix.

Background/Aims Food interaction, including food hypersensitivity, plays a key role in the pathogenesis of irritable bowel syndrome with diarrhea (IBS-D). Since only a few studies have been reported about the relationship between food hypersensitivity and IBS-D, we elucidate the prevalence of serological food hypersensitivity in patients with IBS-D and the characteristics of gastrointestinal symptoms and serum cytokine profiles in patients with IBS-D and serological food hypersensitivity. Methods Immunoglobulin E (Ig E)-mediated serological food hypersensitivity and serum cytokine levels were evaluated using the multiple allergen simultaneous test evaluating food allergen-specific serum IgE and Luminex Milliplex Panel containing multiple fluorescencelabeled beads. Class 2 or above was considered as IgE-mediated food hypersensitivity positive. The gastrointestinal symptom rating scale was used to evaluate symptoms. Results We enrolled 92 subjects, including 60 with IBS-D and 32 healthy controls. The percentages of patients with IgE-mediated serological food hypersensitivity were not significantly different between the groups (controls = 28.1% and IBS-D = 33.3%). Serum IL-1β, IL- 6, IL-8, macrophage inflammatory protein-1alpha, and TNF-α levels were higher in patients with IBS-D than in controls. Serum concentration of TNF-α (43.4 vs 21.8 pg/mL, P = 0.009) was higher in patients with IBS-D without IgE-mediated serological food hypersensitivity than those with food hypersensitivity. Conclusions One-third of Japanese patients with IBS-D showed IgE-mediated serological food hypersensitivity. The serum cytokine profile differed and was characterized by lower inflammatory cytokine levels in IBS-D with IgE-mediated serological food hypersensitivity. Serological test regarding IgE-mediated food hypersensitivity can detect a certain cluster of IBS-D. (J Neurogastroenterol Motil 2018;24:415-421)

Background/Aims: We applied a back light system (BLS) with a magnifying glass to improve the ability to assess the adequacy ofspecimen sampling using endosonography. We conducted this study to evaluate the effcacy of the BLS in sampling of specimens byendoscopic ultrasound-guided fine needle aspiration of solid pancreatic masses. Methods: This was a prospective, randomized, crossover, single-center clinical trial. An endosonographer evaluated adequacy on grossvisual inspection and identified whitish specimen sampling sites with and without the BLS according to a randomization sequence inthe first and second passes with a 25-G needle. On cytological evaluation, the presence of well-defined pancreatic ductal epithelium wasevaluated by a cytopathologist who was blinded to any clinical information. Results: A total of 80 consecutive patients were eligible during the study period. Adequacy was observed for 52 specimens (65%) withthe BLS and 54 (68%) without the BLS (p=0.88). In assessment of specimen adequacy on gross examination, only fair agreement wasobserved both with and without BLS (kappa score 0.40 and 0.29, respectively). Conclusions: The BLS did not influence the ability to identify specimen sampling sites or reliable assessment of specimen site adequacyusing gross visual inspection.

Background: Uroplakins have been widely investigated as potential markers in patients with bladder cancer because these proteins are specific to the urothelium. However, the role of uroplakin proteins in bladder cancer remains unknown. In this study, preoperative serum levels of uroplakin III were measured in patients with urothelial carcinoma of the urinary bladder and examined for possible association with clinicopathological features and clinical outcomes. Materials and Methods: This study included 52 bladder cancer patients at various stages and 28 healthy controls. Uroplakin III levels were detected in preoperative sera using an automated dot blot system and a micro-dot blot array. Results: There was a significant increase in serum uroplakin III levels in patients with bladder cancer as compared to healthy controls (p<0.05). In addition, serum uroplakin III levels were associated with muscle-invasive status, high grade and lymphovascular invasion (p<0.02). Log-rank tests indicated high serum uroplakin III to be significantly associated with cancer-specific mortality. Conclusions: Determination of serum uroplakin III level could be valuable for identifying patients with biologically aggressive bladder cancer.
Mechanical Optimization of Series Hybrid Electric Vehicle with Plant Oil Electric generator

We propose a new hybrid electric vehicle which uses natural energy, consumes little fuel, and operates over a long distance. We are designing a series hybrid system, in which a diesel engine operating on plant oil rotates an electric generator, and runs the vehicle using the generated electricity. The generated electricity may also be stored in batteries, which are also able to power the electric vehicle. The general intention of the proposed system is to reduce the use of non-renewable fossil fuels, and to reduce overall costs. Plant oil is ecological, and allows for the creation of a carbon neutral system in its production. Some forms of plant oils, such as non-esterified straight vegetable oil (NE-SVO) and waste vegetable oil (NE-WVO), are promising alternatives used in order to reduce fuel costs. In this paper, we will explain the concept of the proposed series hybrid electric vehicle system using plant oil, and will provide some experimental results of mechanical optimization in the electric generation system and the vehicle drive system. Considering fuel efficiency and the drive torque, optimum reduction ratios were obtained in the experimental results.
Effects of Vitamin E Derivative TMG on the Radiation Protector and Tumor Growth during Radiotherapy

Background: The purpose of this study is to evaluate the immunosuppressive and antioxidant effects of a novel radioprotective agent using the vitamin E derivative 2-(alpha-D-glucopyranosyl) methyl-2,5,7,8-tetramethylchroman-6-ol (TMG) and its effect on tumors, and to study its usefulness. Materials and Methods: In this study, C57BL/6NCrSlc mice were divided into four groups (control, TMG, radiation therapy [RT], and RT+TMG), using 10 mice in each group. In the TMG and 2 Gy+TMG groups, 500 mg/kg TMG was administered. Two groups (2 Gy and 2 Gy+ TMG) among RT and RT+TMG groups were irradiated with 2 Gy in a single fraction, while the other two groups (6 Gy and 6 Gy+TMG) were irradiated locally with 6 Gy in three fractions. Results and Discussion: TMG positively affected CD4+ and CD8+ T lymphocytes. Tumor volumes and growth inhibition rates were compared. In order to evaluate how TMG administration affected tumor growth, Ehrlich cancer cells were injected into the thigh of mice, and the tumor volume and growth suppression rate were compared. Not only RT but also TMG alone inhibited tumor growth. If RT conducted to the mice with TMG, TMG could increase the number of leukocytes, primarily that of lymphocytes. TMG also inhibited tumor growth in addition to RT. Tumor growth was significantly inhibited in the 6 Gy+TMG group. Conclusion: In conclusion, TMG exerted an immunopotentiating effect mainly by increasing the white blood cell numbers including that of lymphocytes. In addition to RT, TMG also inhibited tumor growth. Therefore, TMG is considered to be a useful radioprotective agent in radiotherapy without tumor growth induction.

<P><B>Abstract</B></P> <P>We present an investigation of the alloying effects of 3d transition metals with palladium, and on their activity towards the oxygen reduction reaction (ORR) in alkaline media, as well as the durability of the electrocatalysts over time. Bimetallic Pd-<I>M</I> (<I>M</I> = Fe, Co and Ni) nanoparticles (NPs) were prepared by an impregnation method followed by reduction under forming gas and a subsequent annealing treatment. In order to enhance the catalytic activity towards the ORR, the bimetallic Pd-<I>M</I> nanoparticles were electrochemically dealloyed. The dealloyed Pd-<I>M</I> nanoparticles had a core-shell structure with a Pd<SUB>3</SUB> <I>M</I>-core and a Pd-shell about 1 nm in thickness. The core-shell materials exhibited higher electrocatalytic activity towards the ORR, and longer-term durability when compared to pure Pd.</P>
Aspect-Ratio Effects and Unsteady Pressure Measurements inside a Cross-Flow Impeller

In the present experimental study, the authors try to clarify the characteristics of the flow around and inside a cross-flow impeller in a typical geometry, over a wide parameter range of an aspect ratio L/D2. In order to eliminate the complicated casing factors, the impeller rotates in open space without any casings. As a result, by using hot wire anemometer measurements and by conventional flow visualisations with a particle image velocimetry technique, the authors show that both the outflow rate and the maximum vorticity attain the maximum for L/D2 = 0.6. In order to investigate the aspect-ratio effect, we further reveal minute fluctuating pressures on an impeller end wall for a singular L/D2 = 0.6. Especially in these pressure measurements, the eccentric vortex is prevented to revolute by the insertion of a tongue, in order to consider the spatial structure of flow more precisely.
