Mechanical evaluation of polymer microneedles for transdermal drug delivery: In vitro and in vivo

In this study, we reported two types of PMNs based on polylactic acid (PLA) and polyvinyl alcohol (PVA),respectively. Parafilm M film, porcine skin, and rats’ models were operated to evaluate the mechanicalproperties in vitro and in vivo to find optimal parameters for efficient insertion. Insertion depth was measuredusing Digital Force Gauge by changing insertion force and speed, respectively. Results showed thatincreasing the insertion force and speed used for PMNs application led to a significant increase in thedepth of insertion. A force of 18 N under a speed of 330 mm/min was the optimal condition for insertingPMNs array into ParafilmM film and porcine skin. In addition, PLA-MNs exhibited higher robustness andenhanced homogeneity in insertion depth compared with PVA-MNs, but PVA-MNs were able to reachmuch deeper insertion depth. Moreover, Sprague Dawley (SD) rat experiments confirmed the effectivenessof optimal insertion parameters for transdermal drug delivery. This study illustrated not only thedevelopment of novel PMNs but also the mechanical evaluation for the design of PMNs.

Molecular dynamics, an eff ective method to gain an insight into nanometric behaviour of materials, was employed to studythe nano-cutting behaviour of single crystal Ni 3 Al in nanometric scale. In this paper, comparisons were made for compressive/tensile stress, subsurface damage and surface roughness with three rake angles of a diamond tool. Subsurface damage waspartitioned by region and studied with work hardening in detail. A model for precise characterization of surface roughnesswas established with consideration of local surface fl uctuation. Simulation results showed that the chip thickness increasedas rake angle changed from negative to positive, and the boundary formed between tensile and compressive stress was inconsistent with the glide direction of stacking fault. Subsurface damage decreased as the increase of rake angle, and regularglide planes of stacking faults were found in front of the cutting tool. Further, the pinned dissociated 1/2 < 110 > superpartialdislocation with anti-phase boundary was demonstrated. The model was tested and characterized by implanted pits onperfect surface. Results showed that surface roughness can be well characterized, and an evident discrepancy was observedamong three rake angles, especially for 30° rake angle, which showed an distinct smooth surface compared with the others.

The horizontal stability of a Single-Point Moored (SPM) Floating Liquefied Natural Gas (FLNG) facility is investigated. Both numerical and experimental studies have been conducted for this SPM FLNG. The numerical simulations feature well the experimental data. The effects of the turret locations are studied based on the validated numerical model. Statistic results of the vessel’s motions with different turret locations are conducted and compared. The results show that the longitudinal location of the turret has a significant influence on the horizontal stability, which has a strong relationship with the yaw and roll motions. The calculated top tensions on the hawsers also develop a regular change as changing the turret’s location. The investigation will provide a brief of principles with more details for the design of the ongoing project.
Historical Long-term Exposure to Pentachlorophenol Causing Risk of Cancer - A Community Study

Background: Pervious studies suggested occupational workers exposure to pentachlorophenol (PCP) might contribute to increased risk of cancer. However, few studies have focused on associations between PCP and cancer risk at the community level. Objective: The present study was to explore the cancer risk for the community population living long-term in a PCP contaminated area. Methods: All the cancer cases diagnosed in 2009-2011 in Tongling City were collected. The cancer patients' residencies were geo-referenced in each district. The historical PCP usage for each district of Tongling was calculated as the PCP pollution index, which was further used to divide into PCP exposure categories. Standardized rate ratios (SRRs) of cancer incidence were applied to detect the cancer risk as exposure grade elevated. Correlation analysis was performed to analyze the relationship between PCP pollution and cancer incidence. Results: A total of 5,288 cancer cases (3,451 male and 1,837 female) were identified. PCP usage was correlated with the incidence of leukemia (r=0.88, P=0.002) for males, and with cancer of the esophagus for males (r=0.83, P=0.008) and females (r=0.71, P=0.020). Compared with the low exposure category, significant SRRs for total cancer sites was obtained for high PCP exposure category (SRR=1.61, 95%CI=1.59-1.62). Most SRR values of the cancer sites were significantly increased as exposure grade elevated and exposure time extended. Conclusion: The present study found that community residents living in the PCP contaminated area had increased risk of cancers. Leukemias, lymphomas and nasopharyngeal and esophageal cancers are most possibly associated with PCP exposure.

Background: Genetic polymorphisms of TP63 have been suggested to influence susceptibility to lung adenocarcinoma development in East Asian populations. This study aimed to investigate the relationship between common polymorphisms in the TP63 gene and the risk of lung adenocarcinoma, as well as interactions of the polymorphisms with environmental risk factors in Chinese non-smoking females. Methods: A case-control study of 260 cases and 318 controls was conducted. Data concerning demographic and risk factors were obtained for each subject. The genetic polymorphisms were determined by Taqman real-time PCR and statistical analyses were performed using SPSS software. Results: For 10937405, carriers of the CT genotype or at least one T allele (CT/TT) had lower risks of lung adenocarcinoma compared with the homozygous wild CC genotype in Chinese nonsmoking females (adjusted ORs were 0.68 and 0.69, 95%CIs were 0.48-0.97 and 0.50-0.97, P values were 0.033 and 0.030, respectively). Allele comparison showed that the T allele of rs10937405 was associated with a decreased risk of lung adenocarcinoma with an OR of 0.78 (95%CI=0.60-1.01, P=0.059). Our results showed that exposure to cooking oil fumes was associated with increased risk of lung adenocarcinoma in Chinese nonsmoking females (adjusted OR=1.58, 95%CI=1.11-2.25, P=0.011). However, we did not observe a significant interaction of cooking oil fumes and TP63 polymorphisms. Conclusion: TP63 polymorphism might be a genetic susceptibility factor for lung adenocarcinoma in Chinese non-smoking females, but no significant interaction was found with cooking oil fume exposure.

Cold water paddy field soils are relatively unproductive, but can be ameliorated by supplementing with inorganic fertilizer from animal waste-based composts. The yield of two rice cultivars was significantly raised by providing either chicken manure or cow dung-based compost. The application of these composts raised the soil pH as well as both the total nitrogen and ammonium nitrogen content, which improved the soil’s fertility and raised its nitrification potential. The composts had a measurable effect on the abundance of nitrogencycling- related soil microbes, as measured by estimating the copy number of various bacterial and archaeal genes using quantitative real-time PCR. The abundance of ammonia oxidizing archaea and bacteria was markedly encouraged by the application of chicken manure-based compost. Supplementation with the composts helped promote the availability of soil nitrogen in the cold water paddy field, thereby improving the soil’s productivity and increasing the yield of the rice crop.

Introduction: Although most prostate cancers initially respond to castration with luteinizing hormonereleasing analogues or bilateral orchiectomy, progression eventually occurs. Based on the exciting results of several randomized controlled trials (RCTs), it seems that patients with metastatic castration-resistant prostate cancer (mCRPC) might benefit more from treatment withabiraterone. Therefore we conducted a systematic review to evaluate the efficacy and toxicity of abiraterone in the treatment of mCRPC. Methods: Literature was searched from Embase, PubMed, Web of Science, and Cochrane Library up to July, 2013. Quality of the study was evaluated according to the Cochrane's risk of bias of randomized controlled trial (RCT) tool, then the Grading of Recommendations Assessment, Development and Evaluation (GRADE) System was used to rate the level of evidence. Stata 12.0 was used for statistical analysis. Summary data from RCTs comparing abiraterone plus prednisone versus placebo plus prednisone for mCRPC were meta-analyzed. Pooled hazard ratios (HRs) for overall survival (OS), radiographic progression-free survival (RPFS) and time to PSA progression (TTPP); Pooled risk ratios (RR) for PSA response rate, objective response rate and adverse event were calculated. Results: Ten trials were included in the systematic review; Data of 2,283 patients (1,343 abiraterone; 940 placebo) from two phase 3 trials: COU-AA-301 and COU-AA-302 were meta-analyzed. Compared with placebo, abiraterone significantly prolonged OS (HR, 0.74; 95% confidence interval [CI], 0.66 to 0.84), RPFS (HR, 0.59; 95% CI, 0.48 to 0.74) and time to PSA progression (HR, 0.55; 95% CI, 0.43 to 0.70); it also significantly increased PSA response rate (RR, 3.63; 95% CI, 1.72 to 7.65) and objective response rate (RR, 3.05; 95% CI, 1.51 to 6.15). This meta-analysis suggested that the adverse events caused by abiraterone are acceptable and can be controlled. Conclutios: Abiraterone significantly prolonged OS, RPFS and time to progression patients with mCRPC, regardless of prior chemotherapy or whether chemotherapy-na$\ddot{i}$ve, and no unexpected toxicity was evident. Abiraterone can serve as a new standard therapy for mCRPC.
Research of the Interconnection of Workflow System Based on Web Service

In order to achieve the interconnection between different workflow management systems, it was proposed that all the distributed workflow systems would be encapsulated as web services to perform the entire business process collaboratively by the way of processes’ composition in this paper. By analyzing the comparison between the composition of processes and ordinary Web service, we studied interactive control, the parameters required to be passed through the distributed workflow systems, the workflow system service’s interfaces and its packaging. Furthermore we put forward a general method of the workflow systems interactive interfaces’ extension and the way of the workflow service’s encapsulating and invoking. By this approach, it can easily combine the processes or process fragments which deployed on different workflow systems without other agents and components. It also provides support for the interconnection of the workflow systems in distributed environment, and ultimately achieves a coordinated operation between different workflow engines.

We report Ambient pressure X-ray Photoelectron Spectroscopy Endstation with an integrated chemical analytical system that consists of a residual gas analyzer in the 2nd differential pumping stage and a new low-reactive sample holder heating assembly. This system has a linear response to the reaction chamber gas pressure. The sample heating assembly also has a low dark reaction rate up to 400 ℃ for H2 and CO oxidation. We expect this chemical analytical system will expand our capabilities in conducting inoperando catalysis research.
