      • Chemoprevention in gastrointestinal physiology and disease. Targeting the progression of cancer with natural products: a focus on gastrointestinal cancer

        <P>The last decade has witnessed remarkable progress in the utilization of natural products for the prevention and treatment of human cancer. Many agents now in the pipeline for clinical trial testing have evolved from our understanding of how human nutritional patterns account for widespread differences in cancer risk. In this review, we have focused on many of these promising agents arguing that they may provide a new strategy for cancer control: natural products once thought to be only preventive in their mode of action now are being explored for efficacy in tandem with cancer therapeutics. Natural products may reduce off-target toxicity of therapeutics while making cancers more amenable to therapy. On the horizon is the use of certain natural products, in their own right, as mitigants of late-stage cancer, a new frontier for small-molecule natural product drug discovery.</P>

        Association of a Single Nucleotide Polymorphism (SNP) in Porcine MYC Gene with Economic Traits in Pigs

        MYC (v-myelocytomatosis viral oncogene homologue) is a regulator gene that encodes for a nuclear phosphoprotein. Porcine MYC gene was mapped on chromosome SSC 4p13 and is associated with a variety of functions such as cell proliferation and cell growth. MYC expression is coupled to a multitude of physiological processes and is regulated by hormones, growth factors, cytokines, lymphokines, nutritional status, development and differentiation. MYC is also involved in myogenesis, muscle hyperplasia and adipogenesis. In this study, we investigated SNPs in MYC gene and their association with economic traits in Duroc, Landrace and Yorkshire populations. We detected a single point mutation in exon 3 of porcine MYC gene as a change of T to C at 906 base (amino acid position 302, nonsynomous mutation of alanine) in MYC-N domain. MYC mutation (T906C) was significantly associated with age at 90 kg in these breeds, signifying that this mutation can serve as a selection marker for growth traits in pigs.

      • Prognostic Value of a CYP2B6 Gene Polymorphism in Patients with Acute Myeloid Leukemia

        Background: The objectives of this study aimed to detect a CYP2B6 polymorphism in de novo cases of acute myeloid leukemia patients and identify any role in disease progression and outcome. Materials and Methods: DNA was isolated from peripheral blood of 82 newly diagnosed acute myeloid leukemia cases and the CYP2B6 G15631T gene polymorphism was assayed by PCR restriction fragment length polymorphism (PCR-RFLP). Results: The frequency of the GG genotype (wild type) was 48 (58.5%) and that of the mutant type T allele was 34 (41.9%). GT genotype heterozygous variants were found in 28 (34%), and TT genotype homozygous variants in 6 (7.3%) cases. We found no significant association between the CYP2B6 G15631T polymorphism and complete response (CR) (p-value=0.768), FAB classification (p-value=0.51), cytogenetic analysis (p-value=0.673), and overall survival (p-value=0.325). Also, there were no significant links with early toxic death (p-value=0.92) or progression-free survival (PFS) (p-value=0.245). Conclusions: Our results suggest that the CYP2B6 polymorphism has no role in disease progression, therapeutic outcome, patient free survival, early toxic death and overall survival in acute myeloid leukemia patients.

        Pumpkin Seed Extracts Inhibit Proliferation and Induce Autophagy in PC-3 Androgen Insensitive Prostate Cancer Cells

        Pumpkin seed is a rich source of polyphenols and other bioactive compounds that may act as chemopreventive agents against cancer. In this study, five different extracts of pumpkin seeds were evaluated for their ability to affect proliferation and autophagy on PC-3 prostate cancer cells. All extracts (water [W], methanolic, acetone, ethylacetate, and polar lipid [PL]) inhibited cell proliferation in a dose-dependent manner. Treatment of cells with the PL extract increased cell distribution in the S phase, whereas PL and W extracts induced autophagy significantly. Moreover, PL extract induced a remarkable increase of glutathione and oxidized glutathione levels, whereas nitrite and hydrogen peroxide levels were not altered. In conclusion, pumpkin seed extracts affect PC-3 cell viability, oxidative parameters, and autophagic mechanism, thus demonstrating their potential pharmacological use.

      • Significance of Proliferation Markers and Prognostic Factors in Egyptian Patients with Multiple Myeloma

        Background: Multiple myeloma (MM) is influenced by genetic and micro-environmental changes. Malignant plasma cells produce an abnormal monoclonal immunoglobulin, as well as cytokines, such as IL-10 and IL-6 which stimulate cells of the bone marrow microenvironment (BMM) and cause dysfunction and failure of many organs. B cell activating factor (BAFF), IL6 and IL10 are known to influence the growth and survival of malignant clones. Aim: The objectives of the present study were to investigate the circulating levels of BAFF, IL-10 and IL-6, correlate them with well-known parameters of disease activity in patients with MM, and to detect their impact on patients' survival. Materials and Methods: This study was conducted on 89 newly diagnosed MM patients and seventy apparently healthy volunteers as a normal control group. BAFF, IL6, IL10 were measured by ELISA for both groups and survival analysis was performed for all patients. Results: Studied markers were higher in the MM patients compared to the normal control subjects. Patients survival was improved by high serum BAFF levels. Conclusions: High levels of BAFF were found to improve patients' survival. BAFF and IL-6 can be considered probable diagnostic markers for MM.

        Cervical cancer in the screening era: who fell victim in spite of successful screening programs?

        Objective: To compare profiles of a prescreening and screening cohort of women with cervical cancer regarding histopathology and clinical variables in order to identify those remaining at risk despite successful screening programs. By analyzing these profiles we hope to improve future screening methods. Methods: The prescreening and screening cohorts consisted of 5,046 and 1,174 women, respectively, treated for cervical cancer at the Department of Gynecological Oncology at Radiumhemmet, Karolinska University Hospital, during the periods 1944-1957 and 1990-2004. Results: Mean age increased from 48.9 years to 55.3 years in the cohorts treated 1944-1957 and 1990-2004, respectively. The percentage of patients older than 69 years was 5.4% and 27.3% in the prescreening and screening period, respectively. A shift towards earlier stages at diagnosis, a reduction of squamous cervical cancer and an increase of adenocarcinoma were observed in the screening cohort. The percentage of adenocarcinoma was about 6 times higher among younger patients. Cases of stump cancer and cervical cancer associated with pregnancy have declined. Eighty-seven women in the screening cohort had a history of treatment for in situ carcinoma by conization; 28% of these cases developed cervical cancer within one year after conization. Conclusion: The profile changed in the screening era indicating a need to refine screening for improved detection of in older women. This study, one of the largest clinical series of cervical cancer, provides an important baseline with which later studies can be compared to evaluate the effects of human papillomavirus vaccine and other important changes in this field.       • Fostering life skills development in studentathletes: The experiences of high school sport psychology consultants

        School sport provides a structured setting for teaching life skills. Although the research has focused mainly on coaches, sport psychology consultants (SPCs) can also foster positive development in student-athletes. The objective of this study was to examine the experiences of SPCs in teaching life skills development and transfer to student-athletes in sport-study programs specific to high school settings. A basic qualitative study consistent with a constructivist paradigm was used. Semi-structured interviews were held with seven SPCs employed in a sport-study program. First, results revealed that all SPCs held a holistic philosophy and aimed for student-athletes’ positive development. Second, SPCs incorporated some life skills development and transfer strategies into their interventions (e.g., workshops, group discussions). Third, SPCs encountered certain challenges in teaching life skills development and transfer, and particularly in following up on life skills transfer and working with other stakeholders (e.g., psychoeducators, social workers). Despite the SPCs’ efforts to teach life skills development and transfer, the results highlight the need for better training in effective interventions.
