We have examined the feasibility of using the specific interaction between mistletoe lectin I (ML I) and ${\beta}$-Dgalactose instead of the anti-ML I antibody in developing a homogeneous type competitive binding assay for ML I. We also have examined the feasibility of adapting the biotin/avidin mediated homogeneous assay for this system. Alkaline phosphatase (AKP) was employed as a single substrate enzyme label. The dose-response curve shows a detection range of 1-25 ${\mu}$g/mL and a linear response with a correlation coefficient of 0.99. To demonstrate the analytical utility of this method, 10 ${\mu}$g/mL of ML I was spiked into distilled water. The results show that the mean recovery was 10.03 ${\mu}$g/mL with an SD of 0.18. The difference between the spiked value and the mean recovery was 0.03 ${\mu}$g/mL, with a relative error of 0.3 and 1.6 % of RSD.

Stepwise dechlorination of 1,2,4-trichlorobenzene was observed at a glassy carbon electrode in dimethylformamide containing 0.1 M tetraethylammonium perchlorate. Especially, dechlorination to dichlorobenzene and further to monochlorobenzene or benzene was successfully demonstrated with a porous reticulated vitreous carbon electrode. Electrochemical dechlorination of polychlorobenzenes employing a flow cell with a reticulated vitreous carbon working electrode is also described. Preliminary experiments with a flow cell showed that dechlorination of trichlorobenzene to dichlorobenzene was partially completed while dechlorination to benzene or monochlorobenzene was not successful, suggesting that a flow rate and electrolysis time should be further optimized for the complete electrolysis.
Resonance Raman Study on Authrornyces Ramosus Peroxidase and its CO adducts

We observed the v (FeCO) and v (CO) on ARPCO for the first time and δ (FeCO) was calculated using normal coordinate calculation under a bending angle of δ= 165°. The v (FeCO) and v (CO) on ARPCO was obtained at 519 cm^(-1) and 1928 cm^(-1) for the ^(12)C^(16)O derivative, and shifted to 516 cm^(-1) and 1884 cm^(-1) with ^(13)C^(16)O, and again to 510 cm^(-1) and 1885 cm^(-1) with ^(12)C^(18)O, and then to 506 cm^(-1) and 1840 cm^(-1) with ^(13)C^(18)O, respectively. Among various heme-CO, the v(CO) is the lowest value and v (FeCO) is the highest value. The CO-isotope-sensitive- band at 583 cm^(-1) was detected as seen for other heme protein-CO.

High-performance resonance Raman/ absorption simultaneous determination technique for enzymatic reaction intermediate, compound Ⅱ, was performed upon addition of hydrogen peroxide to ferric Arthrornyces Ramosus Peroxidase (ARP) using microcirculating system. The Fe=O stretching, v(Fe=O), frequency of compound Ⅱ was observed at 788 cm^(-1) for the v(Fe=^(16)O) mode of ARP Ⅱ and at 750 cm^(-1) for the v (Fe=^(18)O) mode (△v = 35 cm^(-1)). The v(Fe=O) value found in compound Ⅱ of ARP is coincidently same as compound Ⅱ of HRP in alkaline pH. Also, it is shown that a trans ligand effect [v(Fe-N_(his))] is one of the important factors to influence to the v(Fe=O) frequency of compound Ⅱ.
Mono-, Di-fluoro 5, 10, 15, 20-tetraphenylporphyrin Iron(Ⅲ)의 분광학적 연구

Resonance Raman(RR) spectra of 5,10, 15, 20-tetraphenylporphyriniron(III) difluoride, [Fe(TPP)F_2], which was formed by the reaction between Fe(TPP)Cl and tetrabutylammonium fluoride(TBAF 3H_2O) in various non-aqueous solvents, were obtained with the 441.6nm(He-Cd laser) excitation. [Fe(TPP)F_2] is six-coordinate iron porphyrin with two polar Fe-F bonds. The porphyrin skeletal modes can be correlated with previously recorded mode of Ni(TPP) and [FeTPP]_2O. And these assignments were confirmed by observed wavenumber shifts due to ??Fe/^54Fe and TPP/TOO-d_8/TPP-N_15 isotopic substitutions. Difluoroiron complex is an iron(III) high-spin complex with the oxidation sensitive band at 1347cm^-1 for v_4 and core size/spin state sensitive band at 1541cm^-1 for v_2.

A variety of labels have been used in the development of immunoassay. Compared to the other detection system, bioluminescence offers numerous advantages, such as high sensitivity, stability, and low detection limit. A sensitive competitive heterogeneous bioluminescence immunoassay for serotonin measurements was developed using a photoprotein, aequorin as a label. Avidin/biotin mediated assay was also performed for signal amplification and activity conservation of aequorin. Serotonin-avidin conjugate was prepared by Mannich reaction and biotin-aequorin conjugate was prepared through NHS-biotin. The resulting dose-response curve showed that the detection range for serotonin was 10^(-6) to 10^(-10)M, and the detection limit was 7.9 x 10^(-10)M. Cross reactivity test indicated that serotonin cross-reacted with 5-methoxytryptamine (28%) but not with 3-methylindole, melatonin, nor with 5-hydroxyindole-3-acetic acid.
Fast mixing device를 이용한 Protein sample의 Time-Resolved Resonance Raman Spectroscopy

The application of fast mixing devices on the heme protein samples was described. Obviously, it is important to determine whether protein samples are stable under conditions encountered with new devices. Human Hemoglobin (Hb) solution was introduced into Platinum sphere mixing (PSM) system at 20 lb/in² gas pressure. The results were checked by UV-Vis spectroscopy and resonance Raman spectroscopy, and the spectra are superimposable upon those obtained from the initial Hb solution, respectively. The ligand binding reaction (oxygenation) of deoxy Hb was foIIowed by PSM. Time-resolved resonance Raman spectroscopy shows that the oxygenation reaction of Hb can be measured with PSM device for resonance Raman studies. Also, resonance Raman spectrum of product of the reaction between met Hb and HzOz supports that this Hb-H₂O₂ complex is ferryl Hb.
