      • Growth, Clonability, and Radiation Resistance of Esophageal Carcinoma-derived Stem-like Cells

        Objective: To separate/enrich tumor stem-like cells from the human esophageal carcinoma cell line OE-19 by using serum-free suspension culture and to identify their biological characteristics and radiation resistance. Methods: OE-19 cells were cultivated using adherent and suspension culture methods. The tumor stem-like phenotype of CD44 expression was detected using flow cytometry. We examined growth characteristics, cloning capacity in soft agar, and radiation resistance of 2 groups of cells. Results: Suspended cells in serum-free medium formed spheres that were enriched for CD44 expression. CD44 was expressed in 62.5% of suspended cells, but only in 11.7% of adherent cells. The suspended cells had greater capacity for proliferation and colony formation in soft agar than the adherent cells. When the suspended and adherent cells were irradiated at 5 Gy, 10 Gy, or 15 Gy, the proportion of CD44+ suspended cells strongly and weakly positive for CD44 was 77.8%, 66.5%, 57.5%; and 21.7%, 31.6%, 41.4%, respectively. In contrast, the proportion of CD44+ adherent cells strongly positive for CD44 was 18.9%, 14.%, and 9.95%, respectively. When the irradiation dose was increased to 30 Gy, the survival of the suspended and adherent cells was significantly reduced, and viable CD44+ cells were not detected. Conclusion: Suspended cell spheres generated from OE-19 esophageal carcinoma cells in serum-free stem medium are enriched in tumor stem-like cells. CD44 may be a marker for these cells.

        Coenzyme Q10 Production by Sphingomonas sp. ZUTE03 with Novel Precursors Isolated from Tobacco Waste in a Two-Phase Conversion System

        Coenzyme Q10 (CoQ10) is a widely used supplement in heart diseases treatment or antioxidative dietary. The microbial production of CoQ10 was enhanced by addition of solanesol and novel precursors recovered from waste tobacco. The novel precursors were separated by silica gel and identified as α-linolenic acid (LNA) and butylated hydroxytoluene (BHT) based on the effect on CoQ10 production and GC-MS. The effects of novel precursors on CoQ10 production by Sphingomonas sp. ZUTE03 were further evaluated in a two-phase conversion system. The precursor`s combination of solanesol (70mg/l) with BHT (30mg/l) showed the best effect on the improvement of CoQ10 yield. A maximal CoQ10 productivity (9.5 mg l-1 h-1) was achieved after 8h conversion, with a molar conversion rate of 92.6% and 92.4% on BHT and solanesol, respectively. The novel precursors, BHT and LNA in crude extracts from waste tobacco leaves, might become potential candidates for application in the industrial production of CoQ10 by microbes.

        Characterization and function of a short peptidoglycan recognition protein from the Chinese oak silkworm, Antheraea pernyi

        Peptidoglycan recognition proteins (PGRPs) are conserved proteins in animals from insects to mammals and play an important role in immune response by recognizing peptidoglycan on microbial surfaces. In this study, a PGRP gene from Antheraea pernyi, named Ap-PGRP-A, was isolated and characterized. Sequence analysis revealed that the full-length cDNA of Ap-PGRP-A was 961 bp, containing a 40 bp 5′-untranslated sequence, a 339 bp 3′-untranslated region and an open reading frame of 582 bp. This gene encoded a putative protein of 193 amino acid residues. Phylogenetic analysis indicated that Ap-PGRP-A had the closest protein sequence similarity to Antheraea mylitta PGRP. The recombinant protein was successfully expressed in Escherichia coli cells and a rabbit anti-Ap-PGRP-A antibody was also prepared. Real-time quantitative reverse transcription-PCR analysis showed that Ap-PGRP-A was extensively expressed in the fat body, midgut, hemocyte,malpighian tubule and epidermis of A. pernyi, especially in the fat body and midgut. The expression levels of Ap-PGRP-A were significantly up-regulated by three types of microorganisms, and Ap-PGRP-A expression was more sensitive in response to the Gram-negative bacterium E. coli than the Gram-positive bacterium Bacillus subtilis or the fungus Beauveria bassiana. These data indicate that Ap-PGRP-A may play a role in the innate immune responses of A. pernyi.

      • Growth and Differentiation Effects of Homer3 on a Leukemia Cell Line

        The Homer protein family, also known as the family of cytoplasmic scaffolding proteins, which include three subtypes (Homer1, Homer2, Homer3). Homer3 can regulate transcription and play a very important role in the differentiation and development for some tissues (e.g. muscle and nervous systems). The current studies showed that Homer3 abnormal expression changes in acute myeloid leukemia (AML). Forced expression of Homer3 in transfected K562 cells inhibited proliferation, influenced the cell cycle profile, affected apoptosis induced by $As_2O_3$ through inhibition of Bcl2 expression, and also promoted cell differentiation induced by 12-O-tetra decanoylphorbol-acetate (TPA). These results showed that Homer3 is a novel gene which plays a certain role in the occurrence and development of AML.

        Research on Gain Scheduling Controller of the Networked Control System with Long Delay

        This paper addresses the stabilization and optimization problem of networked control systems (NCSs) with long time delays and parameter scheduling. According to the actual network conditions, the network time delay is divided into the fixed time delay and the random time delay. Then, the stabilization controller and the gain scheduling controller are constructed, where the stabilizing control parameters are obtained with cone complementary linearization (CCL) approach and the optimizing control parameters are solved with estimation of distribution algorithm (EDA). Simulation results demonstrate the effectiveness of the proposed methods.

        Preliminary numerical study of single bubble dynamics in swirl flow using volume of fluid method

        Spacer grid with mixing vane had been widely used in nuclear reactor core. One of the main feather of spacer grid with mixing vane was that strong swirl flow was formed after the spacer grid. The swirl flow not only changed the bubble generation in the near wall field, but also affected the bubble behaviors in the center region of the subchannel. The interaction between bubble and the swirl flow was one of the basic phenomena for the two phase flow modeling in fuel assembly. To obatin better understanding on the bubble behaviors in swirl flow, full three dimension numerical simulations were conducted in the present paper. The swirl flow was assumed in the cylindral calculation domain. The bubble interface was captured by Volume Of Fluid (VOF) method. The properties of saturated water and steam at different pressure were applied in the simulation. The bubble trajectory, motion, shape and force were obtained based on the bubble parameters captured by VOF. The simulation cases in the present study included single bubble with different size, at different angular velocity conditions and at different pressure conditions. The results indicated that bubble migrated to the center in swirl flow with spiral motion type. The lateral migration was mainly related to shear stress magnitude and bubble size. The bubble moved toward the center with high velocity when the swirl magnitude was high. The largest bubble had the highest lateral migration velocity in the present study range. The effect of pressure was small when bubble size was the same. The prelimenery simulation result would be beneficial for better understanding complex two phase flow phenomena in fuel assembly with spacer grid.

        Novel Method for Circulating Current Suppression in MMCs Based on Multiple Quasi-PR Controller

        An improved circulating current suppression control method is proposed in this paper. In the proposed controller, an outer loop of the average capacitor voltage control model is used to balance the sub-module capacitor voltage. Meanwhile, an individual voltage balance controller and an arm voltage balance controller are also used. The DC and harmonic components of the circulating current are separated using a low pass filter. Therefore, a multiple quasi-proportional-resonant (multi-quasi-PR) controller is introduced in the inner loop to eliminate the circulating harmonic current, which mainly contains second-order harmonic but also contains other high-order harmonics. In addition, the parameters of the multi-quasi-PR controller are designed in the discrete domain and an analysis of the stability characteristic is given in this paper. In addition, a simulation model of a three-phase MMC system is built in order to confirm the correctness and superiority of the proposed controller. Finally, experiment results are presented and compared. These results illustrate that the improved control method has good performance in suppressing circulating harmonic current and in balancing the capacitor voltage.

        Seismic behavior of properly designed CBFs equipped with NiTi SMA braces

        Shape memory alloys (SMA) exhibit superelasticity which refers to the capability of entirely recovering large deformation upon removal of applied forces and dissipating input energy during the cyclic loading reversals when the environment is above the austenite finish temperature. This property is increasingly favored by the earthquake engineering community, which is currently developing resilient structures with prompt recovery and affordable repair cost after earthquakes. Compared with the other SMAs, NiTi SMAs are widely deemed as the most promising candidate in earthquake engineering. This paper contributes to evaluate the seismic performance of properly designed concentrically braced frames (CBFs) equipped with NiTi SMA braces under earthquake ground motions corresponding to frequently-occurred, design-basis and maximum-considered earthquakes. An ad hoc seismic design approach that was previously developed for structures with idealized SMAs was introduced to size the building members, by explicitly considering the strain hardening characteristics of NiTi SMA particularly. The design procedure was conducted to compliant with a suite of ground motions associated with the hazard level of design-basis earthquake. A total of four six-story CBFs were designed by setting different ductility demands for SMA braces while designating with a same interstory drift target for the structural systems. The analytical results show that all the designed frames successfully met the prescribed seismic performance objectives, including targeted maximum interstory drift, uniform deformation demand over building height, eliminated residual deformation, controlled floor acceleration, and slight damage in the main frame. In addition, this study indicates that the strain hardening behavior does not necessarily impose undesirable impact on the global seismic performance of CBFs with SMA braces.

        Gene microarray analysis revealed a potential crucial gene RACK1 in oral squamous cell carcinoma (OSCC)

        Oral squamous cell carcinoma (OSCC) is the sixth most common cancer worldwide, which appears as a consequence of multiple molecular genetic events in various chromosomes and genes. In order to unveil the possible mechanisms underlying OSCC tumorigenesis, the OSCC-related gene expression variance and the gene interaction network should be further investigated. Herein, we conducted the NimbleGen Human Gene Expression Microarray to analyze expression heterogeneity between OSCC primary tumor tissue and its adjacent normal tissue from two patients. A total number of 7872 out of 32,448 detected genes are differentially expressed in OSCC. Gene ontology (GO) analysis demonstrated that these differentially expressed transcripts were critical in a series of metabolic processes, cancer-related signal pathways, and biological regulations. KEGG signaling pathway enrichment suggested a number of pathways (metabolic process and immune response) which are frequently enrolled during cancer progression. 15 most differential regulated genes between OSCC tumor and non-tumor were confirmed by quantitative reverse transcription polymerase chain reaction (qRT-PCR). Furthermore, the interaction network analysis of these confirmed genes by STRING database showed the two subunits of RACK1 had direct interaction with 14 differential proteins. This bioinformatics research lends support about the critical role of RACK1 which functions as a key node protein driving OSCC development.
