        Study on the activation mechanism of lead ions in the flotation of ilmenite using benzyl hydroxamic acid as collector

        The activation mechanism of Pb2+ ions to ilmenite and the subsequent interaction with benzyl hydroxamic acid (BHA) were systematically investigated. The Pb2+ ions modification occurred via a chemical adsorption process, in which lead species interacted with iron hydroxyl compounds to form lead-containing complexes on the ilmenite surface. After lead ions activation, the surface of ilmenite became more active, and iron species and lead-containing complexes served as the main active sites to covalently bond with BHA in the form of metal-BHA chelate complexes. As a result, the BHA adsorption increased, giving rise to a concomitant increase in the floatability of ilmenite.

        Obstacle crossing and traction performance of active and passive screw pipeline robots

        Pipelines are important tools for transporting fluids. The regular use of pipeline robots to inspect pipelines is an important safety measure. Passive and active screw pipeline robots have been developed and have various characteristics. In this work, the structure and driving method of screw pipeline robots were introduced. An obstacle crossing model of active and passive screw pipeline robots was established and compared using the developed screw pipeline robots. A traction model of the active and passive screw pipeline robots that considers lateral and longitudinal slip characteristics was obtained. Then, the proposed pipeline robot traction model with lateral and longitudinal slips was verified through experiments. A traction experiment on the active screw pipeline robot with improved contact confirmed that an increase in the adhesion coefficient and reduction in driving wheel slip can increase traction force.

        Laser direct patterning induced the tunable optical properties of indium tin oxide micro-hole arrays films

        Here we introduce a facile method to fabricate patterned indium tin oxide (ITO) thin films via selective laser ablation at ambient conditions. By scanning the ITO thin films with focused Nd: YAG pulsed laser, the ITO thin films were selective ablated and patterned without using any conventional chemical etching or photolithography steps. Then we investigated the effects of scanning rate for the structure, morphology and optical properties of patterned ITO thin film. These results indicate that the epsilon-near-zero (ENZ) wavelength of ITO thin films can be tuned from 1100 nm to 1340 nm by adjusting the period of the micro-hole array in microstructure. The nonlinear absorption response of patterned ITO films was about 2.85 time than of the as-deposited ITO thin film. Additionally, the results of the Finite-Difference Time-Domain (FDTD) simulation are in good agreement with those of the experiments.

        An Improved Tet-on System to Tightly Conditionally Regulate Reporter Gene Expression

        Reporter genes are often used as markers totrack the integration and expression of target genes inanimal genetic engineering. To avoid potential side effectsfrom reporter genes, in this study an improved Tet-onsystem was developed to control reporter gene expression,and its effectiveness was explored in transgenic cells. First,the rtTA protein was fused with Tat and NLS proteins toobtain the prokaryotic expression vector pET32a-Tat-rtTANSL. A eukaryotic transgenic vector was constructed, p-HS4-BPA-TmA-HS4, in which the reporter (mCherry) andtarget (PRL) genes were promoted by TRE and BCN,respectively. After confirming the functionality of thetransgenic vector, purified rtTA protein and Dox wereadded to induce expression of the mCherry gene. Theoptimal amount of purified rtTA protein, its influence ontarget gene expression, and the time of rtTA protein actionwere each investigated separately. The results showed thatrtTA protein was expressed in transformed E. coli with IPTGinduction. TRE could promote mCherry gene expressionby co-transfecting the constructed transgenic vector andprtTA plasmid. When purified rtTA protein and Dox wereadded, red fluorescence was observed in Bcap-37 cellstransfected with the p-HS4-BPA-TmA-HS4 vector, and theexogenous PRL gene was expressed regardless of mCherrygene expression. The optimal amount of rtTA protein was16 μg/mL, and it needed about 6 h to promote mCherrygene expression in transfected cells. These resultsdemonstrate that the expression of the mCherry reportergene can be tightly and conditionally regulated by our Tetonsystem.

        Epidemiology and Clinical Management of Pulmonary Hypertension in Children

        Pulmonary hypertension (PH) is a clinical hemodynamic syndrome characterized by increased pulmonary artery pressure and pulmonary vascular resistance. It can cause right ventricular failure, and even death. Pediatric PH is not very common, but is a greatly hazardous dis-ease that leads to a high mortality rate. Therefore, many registry organizations have been established in the world to strengthen the study of diagnosis and treatment of the disease, and improve the understanding of pediatric PH. This article reviewed recently published re-searches, as well as presented a comprehensive understanding of PH, including definition, classification, epidemiology, prognosis and treatment.

        Expression pattern of prohibitin, capping actin protein of muscle Z-line beta subunit and tektin-2 gene in Murrah buffalo sperm and its relationship with sperm motility

        Objective: The aim of the current study is to investigate the relationship between prohibitin (PHB), capping actin protein of muscle Z-line beta subunit (CAPZB), and tektin-2 (TEKT2) and sperm motility in Murrah buffalo. Methods: We collected the high-motility and low-motility semen samples, testis, ovary, muscle, kidney, liver, brain and pituitary from Murrah buffalo, and analysed the expression of PHB, CAPZB, and TEKT2 in mRNA (message RNA) and protein level. Results: Quantitative reverse transcription-polymerase chain reaction (qRT-PCR) result showed that the expression of PHB was higher and CAPZB, TEKT2 were specifically expressed in testis as compared to the other 6 tissues, and that in testis, the expression of TEKT2 was higher than that of CAPZB and PHB. Immunohistochemistry test revealed that all three genes were located on the convoluted seminiferous tubule and enriched in spermatogenic cells. Both qRT-PCR and Western Blot results showed that the expression levels of PHB, CAPZB, and TEKT2 were significantly lower in the low-motility semen group compared to the high-motility semen group (p<0.05). Conclusion: The expression levels of PHB, CAPZB, and TEKT2 in Murrah buffalo sperm have a high positive correlation with sperm motility. And the three genes may be potential molecular markers for the decline of buffalo sperm motility.

        The multidirectional vibration and coupling dynamics of drill string and its influence on the wellbore trajectory

        For improving the accuracy of wellbore trajectory prediction in complex wellbore structures, especially in the kick-off section, a longitudinal, torsional and radial coupled drill string dynamics model is proposed. At the same time, the coupling dynamics model is solved based on the energy method, and the torque, weight on bit (WOB), the longitudinal displacement and velocity, the torsional angular displacement and angular velocity of the drill bit are obtained during the normal drilling and the stick-slip vibration phase. Combining the numerical example and experimental test, the differences between different models and actual drilling trajectories are compared and analyzed. The research results indicate that the torsional vibration and the longitudinal vibration have a great influence on each other in the coupling model, and the longitudinal vibration displacement increases, which can effectively reduce the torque variation range of the torsional vibration. The operation of the bit stagnation is weakened, maintaining a relatively stable rotational angular velocity of rotation. The calculation results of the wellbore trajectory are more consistent with the experimental results, and can accurately reflect the real situation of the drilling process. The theoretical model has an important reference for predicting the new well structure, especially the judgment of wellbore trajectory of complex wellbore structures such as extended reach wells, horizontal wells, deep wells and high deep wells of coalbed methane, and the prediction of downhole dynamics results.

        Whole-genome identification and salt- and ABA-induced expression trends of the Nicotiana tabacum CKX gene family

        Cytokinin hormones are indispensable for plant growth and development. Cytokinin oxidase/dehydrogenase (CKX) helps regulate the dynamic balance of endogenous cytokinin levels. However, little is known about the CKX genes of Nicotiana tabacum (NtCKXs). In silico analyses were used to isolate, characterize, and elucidate the phylogenetic relationships of 15 NtCKX genes. Multi-species phylogenetic tree construction placed NtCKX1–15 on five of the eight branches of the CKX phylogenetic tree. Protein structure and network analyses revealed that NtCKX genes located on the same phylogenetic branch generally contained several conserved motifs and have highly similar structures, with structural domains related to flavin adenine dinucleotide (FAD) and cytokinin-binding found on all of the predicted NtCKX-encoded proteins. The upstream promotor region of NtCKX genes contained many abiotic stress-responsive cis-acting elements, including DRE, ERE, MBS, MYB, and MYC. Gene expression analysis revealed that each NtCKX gene responded differently to salt stress and exogenous abscisic acid (ABA) treatment. Four NtCKX genes exhibited ABA-induced expression trends with varying peak times. Under salt stress, NtCKX expression was significantly suppressed in two genes and upregulated in five others. In summary, we provided basic information regarding the CKX gene family of N. tabacum and elucidated their gene expression patterns under abiotic stresses, including ABA treatment and salt stress. The findings of this work can serve as a foundation for future study of the functions of NtCKX genes.
