        Investigation on the Preparation and Adsorption Performance of Bamboo Fiber Based Activated Carbon

        In this work, an approach was explored to make natural fiber based activated carbon with high adsorption capacity for treating dyeing wastewater. Bamboo fiber based activated carbon (BFAC) was prepared by bamboo fiber extracted from bamboo strips by extracting technique. An orthogonal experiment was carried out to obtain optimum preparation condition for the best dye adsorption. The microstructures of BFAC were characterized by scanning electron microscopy, N2 adsorption-desorption analysis, X-ray diffraction, and Fourier transform infrared spectroscopy. Adsorption mechanism of BFAC to methylene blue (MB) was studied and the adsorption isotherm followed the Langmuir model, with maximum monolayer adsorption capacity of 651.7 mg g-1. The adsorption kinetic was better described by the pseudo-second-order model. Microstructure characterization showed lots of opened pores and cracks with different size on BFAC, with Brunauer-Emmett-Teller (BET) surface area of 1370 m2 g-1 and total pore volume of 0.9411 cm3 g-1. The analysis indicated that cellulose crystal of bamboo fiber was severely destroyed in the preparation process, which could contribute to the formation of pores. Moreover, -OH and -C=O functional groups on BFAC are beneficial for adsorbing MB in aqueous solution. As a result, adopting bamboo fiber may help form BFAC with high BET surface area, which proved to be an effective idea to prepare activated carbon with high adsorption performance.

        Myeloid-Derived Suppressor Cells Are Associated with Viral Persistence and Downregulation of TCR ζ Chain Expression on CD8<sup>+</sup> T Cells in Chronic Hepatitis C Patients

        Myeloid-derived suppressor cells (MDSCs) play an important role in impairing the function of T cells. We characterized MDSCs in two chronic hepatitis C (CHC) cohorts: a cross-sectional group that included 61 treatment-naive patients with CHC, 14 rapid virologic response (RVR) cases and 22 early virologic response (EVR) cases; and a longitudinal group of 13 cases of RVR and 10 cases of EVR after pegylated-interferon-${\alpha}$/ribavirin treatment for genotype 1b HCV infection. Liver samples from 32 CHC patients and six healthy controls were subjected to immunohistochemical analysis. MDSCs frequency in treatment-naive CHC was significantly higher than in RVR, EVR, or healthy subjects and was positively correlated with HCV RNA. Patients infected with HCV genotype 2a had a significantly higher frequency of MDSCs than those infected with genotype 1b. Decreased T cell receptor (TCR) ${\zeta}$ expression on $CD8^+$ T cells was significantly associated with an increased frequency of MDSCs in treatment-naive CHC patients and was restored by L-arginine treatment in vitro. Increased numbers of liver arginase-$1^+$ cells were closely associated with the histological activity index in CHC. The TCR ${\zeta}$ chain was significantly downregulated on hepatic $CD8^+$ T cells in CHC. During antiviral follow up, MDSCs frequency in peripheral blood mononuclear cells was directly correlated with the HCV RNA load in the plasma and inversely correlated with TCR ${\zeta}$ chain expression in $CD8^+$ T cells in both RVR and EVR cases. Notably, the RVR group had a higher frequency of MDSCs at baseline than the EVR group. Collectively, this study provides evidence that MDSCs might be associated with HCV persistence and downregulation of CD8 ${\zeta}$ chain expression.

        Clinical Model for Predicting Hepatocellular Carcinomas in Patients with Post-Sustained Virologic Responses of Chronic Hepatitis C: A Case Control Study

        Background/Aims: No clinical model exists to predict the occurrence of hepatocellular carcinoma in sustained virologic response-achieving (HCC after SVR) patients with chronic hepatitis C (CHC). Methods: We performed a case-control study using a clinical database to research the risk factors for HCC after SVR. A predictive model based on risk factors was established, and the area under the receiver operating characteristic curve (AUC) was calculated. Results: In the multivariate model, an initial diagnosis of compensated cirrhosis and post-SVR albumin reductions of 1 g/L were associated with 21.7-fold (95% CI, 4.2 to 112.3; p< 0.001) and 1.3-fold (95% CI, 1.1 to 1.7; p=0.004) increases in the risk of HCC after SVR, respectively. A predictive model based on an initial diagnosis of compensated cirrhosis (yes, +1; no, 0) and post- SVR albumin ≤36.0 g/L (yes, +1; not, 0) predicted the occurrence of HCC after SVR with a cutoff value of >0, an AUC of 0.880, a sensitivity of 0.833, a specificity of 0.896, and a negative predictive value of 0.956. Conclusions: An initial diagnosis of compensated cirrhosis combined with a post-SVR albumin value of ≤36.0 g/L predicts the occurrence of HCC after SVR in patients with CHC.         Ionic liquid extraction of silkworm pupa protein and its biological characteristics

        Silkworm (Bombyx mori) pupa protein (SPP) is a high-quality source of animal protein with substantial nutri tional benefits and health value. To develop an efficient extraction method for SPP that is environmentally friendly, we selected choline hydroxide ionic liquid (CH-IL) as the extraction solvent and performed orthogonal experiments to optimize the extraction conditions. We demonstrated that 3% CH-IL, a solid-to-liquid ratio (g/mL) of 1:30, an extraction temperature of 40 ◦ C, and an extraction time of 1 h facilitated the most efficient extraction. Compared to the conventional alkali solubilization–acid precipitation method, the CH-IL extraction increased protein content by 12.14%. Protein structure analysis showed that the β-sheet content increased by 10.98% and that of disulfide bonds reduced by 16.4%. The processing properties of the CH-IL extracted protein showed that the solubility, emulsification, and foaming capacity were enhanced by 82.87%, 15.44%, and 18.97%, respec tively. The physical properties of SPP remarkably improved relative to the increased stretching of the poly peptide chains. The findings of this study provide technical knowledge that will enhance the processing performance of pupal proteins.

        Sublethal effects of beta-cypermethrin on the bird cherry-oat aphid Rhopalosiphum padi (Hemiptera: Aphididae)

        Rhopalosiphum padi is a sap-sucking aphid and an important pest of wheat that causes considerable yield loss. Beta-cypermethrin, a synthetic pyrethroid pesticide, has a broad insecticide spectrum and is considered effective for aphid control, while its residual concentrations may have sublethal effects on R. padi. Here, the sublethal effects of beta-cypermethrin on R. padi were conducted under laboratory conditions. The acute toxicity test showed that LC 10 , LC 20 , and LC 25 of beta-cypermethrin to R. padi adults were 0.003, 0.031 and 0.079 mg L −1 , respectively. The pre-adult survival rate was significantly reduced by all three concentrations. LC 20 significantly extended the development duration of 1st instar nymphs, pre-oviposition period, and oviposition period of R. padi. The adult longevity was also reduced by LC 25 . However, the fecundity did not differ between the betacypermethrin treatment and control. For life table parameters, both the finite rate (λ) and intrinsic rate of increase (r) decreased at LC 10 and LC 20 , as well as the net reproductive rate (R 0 ) reduced at LC 10 and LC 25 , while mean generation time (T) increased at LC 20 . Thus, at the concentrations of beta-cypermethrin tested here, there were negative impacts on R. padi fitness by decreased pre-adult survival rate, λ, r, and R 0 , and delayed the development of some stages and increased T.

        Two New Species and a New Chinese Record of Hypocreaceae as Evidenced by Morphological and Molecular Data

        To explore species diversity of Hypocreaceae, collections from Guangdong, Hubei, and Tibet of China were examined and two new species and a new Chinese record were discovered. Morphological characteristics and DNA sequence analyses of the ITS, LSU, EF-1a, and RPB2 regions support their placements in Hypocreaceae and the establishments of the new species. Hypomyces hubeiensis sp. nov. is characterized by occurrence on fruitbody of Agaricus sp., concentric rings formed on MEA medium, verticillium-like conidiophores, subulate phialides, rod-shaped to narrowly ellipsoidal conidia, and absence of chlamydospores. Trichoderma subiculoides sp. nov. is distinguished by effuse to confluent rudimentary stromata lacking of a well-developed flank and not changing color in KOH, subcylindrical asci containing eight ascospores that disarticulate into 16 dimorphic part-ascospores, verticilliumlike conidiophores, subcylindrical phialides, and subellipsoidal to rod-shaped conidia. Morphological distinctions between the new species and their close relatives are discussed. Hypomyces orthosporus is found for the first time from China.

        Surfactant Effect on Formation of CaWO<sub>4</sub>:Eu<sup>3+</sup> Crystals with Distinguished Morphologies in Hydrothermal Ambient

        <P>Metal tungstates, expressed by the general formula of MWO4, have important properties and applications in photoluminescence, microwave applications, optical fibers, scintillator materials, humidity sensors, magnetic properties, and catalysts. In this paper, we report a successful synthesis of CaWO4: Eu3+ crystals with various morphologies in mild hydrothermal conditions with surfacntant including sodium citrate, CTAB, PEG and citrate acid (CA). The formation of the crystals are strongly dependent on the employment of surfactant. The surfactant concentration has been found significant influence in the resulting morphologies due to different properties of each one. Extensive characterization have been performed by using X-ray diffraction (XRD), field emission scanning electron microscope (FE-SEM) in search of the formation mechanism of multi-morphological CaWO4: Eu3+ crystals. The growth mechanism of monodispersed CaWO4: Eu3+ crystal are proposed. And the photoluminescence properties were investigated.</P>

        Evaluation of three vacuolar ATPase genes as potential RNAi target in Henosepilachna vigintioctopunctata

        Henosepilachna vigintioctopunctata, a major threat to commercial potato production, is sensitive to RNA inter ference (RNAi), a gene-silencing mechanism triggered by double stranded RNA (dsRNA) matching a specific target gene sequence. Vacuolar ATPases (vATPases) are ATP-driven proton pumps involved in numerous bio logical processes. A holoenzyme vATPase consists of at least 14 different subunits and accessory proteins. Pre vious results reveal that silencing vATPaseB decreases survival rate, reduces food consumption and inhibits development in H. vigintioctopunctata. In this report, we identified two other subunit genes (vATPasea and vATPased) and compared the RNAi efficacy of vATPasea and vATPased with that of vATPaseB. We found the highest expression level of HvvATPasea, HvvATPaseB and HvvATPased occurred in the hindgut, followed by those in the foregut, Malpighian tubules and midgut, and lowest in the epidermis and fat body. When the fourth-instar larvae were allowed to ingest corresponding dsRNA for three days, the relative transcript levels of HvvATPasea, HvvATPaseB and HvvATPased were significantly decreased by 79%, 83% and 73%, respectively. Knockdown of HvvATPasea, HvvATPaseB and HvvATPased caused 50%, 85% and 100% larval lethality. After the third-instar larvae had exposed to corresponding dsRNA for three days, the levels of HvvATPasea and HvvATPased signifi cantly reduced by 91% and 96% respectively. All the treated larvae were dead. Our results demonstrate that RNAi efficiencies varied among different vATPase subunit genes and various development stages. Moreover, our data imply the feasibility of RNAi as an alternative method for controlling this critical potato pest.
