        Biodegradable cross-linked poly(L-lactide-co-e-caprolactone) networks for ureteral stent formed by gamma irradiation under vacuum

        The poly(L-lactide-co-e-caprolactone) (PLCL) ureteral stent creeps and loses shape stability, increasingthe risk of stent tube dislocation. The rubbery biodegradable cross-linked PLCL networks were preparedthrough gamma irradiation under vacuum in the presence of trimethylolpropane triacrylate (TMPTA),pentaerythritol tetraacrylate (PET4A), and pentaerythritol triacrylate (PETA). At a standard sterilizationdose of 25 kGy, the gel content and network density of PLCL networks increased with increasingcrosslinking agent content (1, 3, 5, 7 wt%), and crosslinking efficiency decreased in the order ofPETA > PET4A > TMPTA. The average molecular weight (Mc ) between two crosslinks ranged from 2000to 105 g/mol. To perform the beneficial semi-interpenetrated polymer network and characterized bythe principle, the networks were processed in several doses (25, 50, 75, 100, and 125 kGy). In place ofthe Charlesby-Pinner equation, the irradiation cross-linking followed the Chen-Liu-Tang equation. ThePLCL network with 7 wt% PETA had a gel fraction of 83%, tensile strength of 34.7 MPa, and tensile setvalue as low as 5%. Furthermore, degradation in vitro was slowed down. Thus, PLCL networks with appropriateelasticity and flexibility, inherent biodegradability, and excellent biocompatibility can provide apromising alternative method for soft tissue repair engineering, such as ureteral stents.

        Kinetics of Nitrogen Absorption in Molten AISI 316 Stainless Steel During Immersion Nitrogen Blowing

        Nitrogen absorption in molten metal for stainless steel AISI316 has been investigated by immersion nitrogen blowing through an immersed alumina nozzle with an internal diameter of 3 mm. Based on these experimental data, some kinetic parameters of nitrogen absorption, such as reaction order, rate constant and apparent activation energy of nitrogen absorption reaction, have been obtained. Effect of stirring by immersion nitrogen blowing through an immersed alumina nozzle on nitrogen absorption reaction has been observed. Results show the following: (1) Nitrogen absorption reaction is the −1.5th order reaction. The rate constant N is of the order of 10−5wt%2.5·min −1. Nitrogen absorption reaction for AISI 316 has negative apparent activation energy of −92.40 kJ·mol −1. This indicates that the nitrogen absorption reaction has a complex and multistep reaction mechanism. (2) The rate of nitrogen absorption reaction in molten stainless steel is mixture control by the adsorption of monatomic nitrogen on the surface of molten stainless steel and mass transfer in molten metal. (3) A rate equation of nitrogen absorption reaction has been derived based on a mixed control mechanism by both the -1st order nitrogen absorption reaction and mass transfer in molten metal.

        Extracorporeal Shock Wave Combined with Teriparatide-Loaded Hydrogel Injection Promotes Segmental Bone Defects Healing in Osteoporosis

        BACKGROUND: Osteoporosis is a systemic bone disease characterized by decreased bone density and deterioration of bone microstructure, leading to an increased probability of fragility fractures. Once segmental bone defect occurs, it is easy to cause delayed union and nonunion. METHODS: The aim of this study is to investigate the efficacy of extracorporeal shock wave (ESW) and teriparatide-loaded hydrogel (T-Gel) combined strategy on the cell activity and differentiation of osteoporosis derived bone marrow mesenchymal stem cells (OP-BMSCs) in vitro and bone regeneration in osteoporotic segmental bone defects in vivo. RESULTS: In vitro, the strategy of combining ESW and T-Gel significantly enhanced OP-BMSCs proliferation, survival, migration, and osteogenic differentiation by up-regulating the alkaline phosphatase activity, mineralization, and expression of runt-related transcription factor-2, type I collagen, osteocalcin, and osteopontin. In the segmental bone defect models of osteoporotic rabbits, Micro-CT evaluation and histological observation demonstrated this ESW-combined with T-Gel injection significantly induced bone healing by enhancing the osteogenic activity of the local microenvironment in osteoporotic defects. CONCLUSION: In conclusion, ESW-combined with T-Gel injection could regulate the poor osteogenic microenvironment in osteoporotic defects and show potential for enhancing fragility fractures healing.       • AtHAP3b Plays a Crucial Role in the Regulation of Flowering Time in Arabidopsis during Osmotic Stress

        The HAP complex has been found in many eukaryotic organisms. HAP recognizes the CCAAT box present in the promoters of 30% of all eukaryotic genes. The HAP complex consists of three subunits - HAP2, HAP3 and HAP5. In this paper, we report the biological function of the AtHAP3b gene that encodes one of the HAP3 subunits in Arabidopsis. Compared with wild-type plants, hap3b-1 and hap3b-2 mutants exhibited a delayed flowering time under long-day photoperiod conditions. Moreover, the transcription levels of FT were substantially lower in the mutants than in the wild-type plants. These results imply that AtHAP3b may function in the control of flowering time by regulating the expression of FT in Arabidopsis. In a subsequent study, AtHAP3b was found to be induced by osmotic stress. Under osmotic stress conditions, the hap3b- 1 and hap3b-2 mutants flowered considerably later than the wild-type plants. These results suggest that the AtHAP3b gene plays more important roles in the control of flowering under osmotic stress in Arabidopsis.

      • Conjugated linoleic acid (CLA) regulates female reproduction via sex pheromone regulation without affecting larval development in Drosophila melanogaster

        Knowledge on the potential role of conjugated linoleic acid (CLA) on reproduction and development is currently limited. Therefore, the goal of the current study was to determine the effects of CLA on reproduction and development using the Drosophila model. In this study, adult and larva Drosophila melanogaster were fed fly food with 0.5% water (blank), CLA (50:50 of cis-9, trans-11 and trans-10, cis-12 CLA isomers in triglyceride form) or safflower oil (69% linoleic acid, LA, as control) to examine the effects of CLA on fecundity, sex pheromones, transcription level of oogenesis, larval body composition, and eclosion time. CLA-treated females had lowered brood size without changes in hatchability, along with decreased 7,11-heptacosadiene and 7,11-nonacosadiene, the principle female pheromones, when compared to LA and blank. Moreover, CLA reduced transcription level of lipid storage droplet-2 (lsd-2) compared to LA. CLA did not influence larval composition nor eclosion time. In conclusion, CLA inhibited reproduction capability in D. melanogaster in part via reduced fatty acid-derived signaling pheromone and oogenesis modulations. As key biological processes are conserved between humans and flies, knowledge from this research may provide valuable insight into reproduction and development responses to CLA.

        Antimicrobial effect and proposed action mechanism of cordycepin against Escherichia coli and Bacillus subtilis

        The detailed antibacterial mechanism of cordycepin efficacy against food-borne germs remains ambiguous. In this study, the antibacterial activity and action mechanism of cordycepin were assessed. The results showed that cordycepin effectively inhibited the growth of seven bacterial pathogens including both Gram-positive and Gram-negative bacterial pathogens; the minimum inhibitory concentrations (MIC) were 2.5 and 1.25 mg/ml against Escherichia coli and Bacillus subtilis, respectively. Scanning electron microscope and transmission electron microscope examination confirmed that cordycepin caused obvious damages in the cytoplasmatic membranes of both E. coli and B. subtilis. Outer membrane permeability assessment indicated the loss of barrier function and the leakage of cytoplasmic contents. Propidium iodide and carboxyfluorescein diacetate double staining approach coupled with flow cytometry analysis indicated that the integrity of cell membrane was severely damaged during a short time, while the intracellular enzyme system still remained active. This clearly suggested that membrane damage was one of the reasons for cordycepin efficacy against bacteria. Additionally, results from circular dichroism and fluorescence analysis indicated cordycepin could insert to genome DNA base and double strand, which disordered the structure of genomic DNA. Basis on these results, the mode of bactericidal action of cordycepin against E. coli and B. subtilis was found to be a dual mechanism, disrupting bacterial cell membranes and binding to bacterial genomic DNA to interfere in cellular functions, ultimately leading to cell death.

      • Prevalence and Genotype Distribution of Human Papillomavirus Infections in Women Attending Hospitals in Chaozhou of Guangdong Province

        Background: Human papillomavirus (HPV) infection is the main cause of cervical cancer. Limited epidemiologic data of HPV prevalence are available for women attending hospitals in southern China. This study aimed to evaluate the profiles of HPV infection and cytology status in gynecological outpatients in Chaozhou City. Methods: A total of 2833 eligible women were enrolled. The HPV GenoArray test was used for HPV detection and genotyping. Nearly one half of the HPV positive women received liquid-based cytology test. Logistic regression analysis was performed to assess the predictable effects of age and genotype for categories of abnormal cytology. Results: The prevalence of overall, high-risk, and low-risk HPV infection were 24.5%, 19.5% and 8.4%, respectively. A U-shaped age-specific prevalence curve was observed in overall HPV and high-risk HPV, but not in low-risk HPV, which declined with age increasing. The 6 most common high-risk HPV type in descending order, were types 52, 16, 58, 18, 68, and 33. Age and HPV genotype were both important determinants of abnormal cytology incidence, the older women (>45 years) and those infected with HPV type 16 and/or 18 having the highest risk for abnormal cytology. Conclusion: Our findings support the hypothesis that second-generation HPV prophylactic vaccines including HPV-52 and -58 may offer higher protection for women residing in Chaozhou and neighboring cities in Guangdong.

        Predictive Current Control for Multilevel Cascaded H-Bridge Inverters Based on a Deadbeat Solution

        Finite-set predictive current control (FS-PCC) is advantageous for power converters due to its high dynamic performance and has received increasing interest in multilevel inverters. Among multilevel inverter topologies, the cascaded H-bridge (CHB) inverter is popular and mature in the industry. However, a main drawback of FS-PCC is its large computational burden, especially for the application of CHB inverters. In this paper, an FS-PCC method based on a deadbeat solution for three-phase zero-common-mode-voltage CHB inverters is proposed. In the proposed method, an inverse model of the load is utilized to calculate the reference voltage based on the reference current. In addition, a cost function is directly expressed in the terms of the voltage errors. An optimal control actuation is selected by minimizing the cost function. In the proposed method, only three instead of all of the control actuations are used for the calculations in one sampling period. This leads to a significant reduction in computations. The proposed method is tested on a three-phase 5-level CHB inverter. Simulation and experimental results show a very similar and comparable control performance from the proposed method compared with the traditional FS-PCC method which evaluates the cost function for all of the control actuations.
