      • Comparative analysis of polyspecificity of the endogenous tRNA synthetase of different expression host towards photocrosslinking amino acids using an in silico approach

        Photo-induced covalent crosslinking has emerged as the powerful strategy for analyzing and characterizing the protein-protein interaction and mapping protein 3D conformations. In the last decades, a number of photocrosslinking amino acids have been reported but only a few have been efficiently utilized for photocrosslinking purposes. Recently, incorporation of diazirine containing photoactivatable analogs such as photo-methionine, photo-leucine, photo-isoleucine and photo-lysine into target proteins were accomplished in live cells (Human A549cells, HEK 293) by depleting corresponding natural amino acid and supplementing these analogs in the medium. Likewise, incorporation of photo-methionine and photo-leucine is also reported in E. coli. Incorporation of these unnatural amino acids were demonstrated only in a limited number species, thereby conventional methods have been utilized for the protein-protein interaction study in other species. With this in mind, we studied in silico analysis of polyspecificity of four endogenous tRNA synthetases (LeuRS, IleRS, MetRS, and LysRS) from six different species such as Escherichia coli, Pseudomonas fluorescens, Corynebacterium glutamicum, Saccharomyces cerevisiae, Aspergillus oryzae and Homo sapiens towards its photocrosslinking amino acids. In addition, here we describe the active site similarity of different protein bio-factories. Based on the active site similarity and similar binding mode, we predicted that the endogenous tRNA synthetases of all the species are reactive to corresponding photoactivatable analogs. This is the first in silico study to demonstrate that the photocrosslinking unnatural amino acids are recognized by the endogenous tRNA synthetases of different protein expression biofactories.

        Protective Effect of Panax ginseng extract on Renal Functions Altered by Mercuric Chloride in Albino Rats

        Liver and kidney are specific organs which play an active role in biotransformation and detoxification mechanisms. Ant adverse effect of chemicals or heavy metal can cause the delay or fade in these mechanisms. Present study was designed to find out the protective effect of Panax ginseng extract on renal functions altered by mercuric chloride (heavy metal) in albino rat. Fifty albino rats were divided into 10 groups. Five groups for acute study and five groups for sud-acute study viz. control group (Tween 20 and distilled water), mercuric chloride treated group (0.926 ㎎/㎏ body wt. for acute and 0.044 ㎎/㎏ body wt. for sub-acute group after calculated LD?? (9.26 ㎎/㎏ body wt.) by probit analysis (Finney, 1971)¹, Panax ginseng extract treated group (10 ㎎/㎏ body wt. for acute and sub-acute sets), mercuric chloride treated followed by Panax ginseng extract and Panax ginseng extract followed by mercuric chloride group. All doses were given orally by gavage tube. The result revealed that the serum urea and creatinine significantly increased in mercuric chloride treated group, while significantly decreased (p<0.01) in Panax ginseng extract group after acute and sub-acute treatment. The biochemical estimation is also confirmed by nephropathological aspect. However, the Panax ginseng extract treated followed by mercuric chloride group is more prominent than the mercuric chloride treated followed by Panax ginseng extract group. It can be concluded that Panax ginseng extract had a protective nature on renal functions against mercuric chloride toxicity in albino rats.

        Genetic diversity and population structure assessment of Hellenia speciosa from Indian agro-ecological regions using inter-simple sequence repeat markers

        Hellenia speciosa (J. Koenig) S. R. Dutta (Syn: Costus speciosus (J. Koenig) Sm.) is well known for its anti-diabetic proper- ties and is in high demand for its steroidal sapogenin, diosgenin, which is used in the pharmaceutical industry for manufac- turing steroidal drugs. The increasing demand for this plant has led to the depletion of its wild population. This study was conducted to reveal the genetic diversity of H. speciosa collected from various agro-ecological regions of India for use in devising conservational strategies, and cultivation and breeding programs. Twenty inter-simple sequence repeat markers yielded 304 products with 100% polymorphism demonstrating high genetic diversity. Nei′s gene diversity and Shannon information indexes were estimated among the populations as 0.19 ± 0.23 and 0.28 ± 0.25, respectively. High genetic dif- ferentiation ( GST = 0.40) and low gene fl ow ( Nm = 0.74) were observed, which corroborated with the analysis of molecular variance revealing maximum (86.75%) variance within populations. The unweighted pair group method with arithmetic mean analysis grouped all accessions into three major clusters. Principal component analysis showed that the fi rst three compo- nents accounted for 30.66%, 4.54%, and 3.98% variation, respectively, and Bayesian clustering through structure showed the presence of two genetic population (K = 2). This study revealed high genetic diversity among the populations collected from high altitude compared to those from the plain. A positive but low correlation among genetic and geographic distances was obtained ( r = 0.282, p < 0.0001). Identifi cation of genetic diversity and intra population variation of a species are prime factors to take defi ned conservation strategies.

        The simulations on dynamic stability of Sports Utility Vehicles using Carsim/Vehsim software are presented on this paper with the aim of studying the rollover process occurring in such vehicles. Carsim/Vehsim is an integrated simulation package that has been developed for chassis dynamics controls and integration. The simulation procedure makes use of dynamic testing maneuvers (Fishhook tests, J-Turn test and Double Lane Change test) recommended by NHTSA to evaluate the antirollover propensities of SUVs on road. The simulation results emphasize the need for better understanding of SUV rollover performance and characteristics that are very essential in enhancing vehicle design. Furthermore, the simulation dramatically shows less development time and cost reduction compared to field testing. This paper, in general, investigates the dynamic rollover characteristics of 4 SUVs in 2 different road conditions.

        Influence of 60Co gamma irradiation on the structural and optical properties of 2-aminopyridinium 4-nitrophenolate 4-nitrophenol crystals

        In this article, effect of gamma irradiation on the structural and optical properties of 2-aminopyridinium 4- nitrophenolate 4-nitrophenol (2AP4N) has been reported. The grown crystals of 2AP4N were exposed to 60Co gamma rays with a dose of 50 kGy and 100 kGy. The radiation-induced effects were analyzed using X-ray diffraction, FT-IR, UV–visible, photoluminescence techniques. The refractive index was determined using a long arm spectrometer. The structural properties of the pristine and irradiated crystals were studied using powder XRD. The peak intensity decrease after irradiation may be attributed to the formation of point defects. The UV visible study reveals that the energy gap has decreased after irradiation and then has increased for the higher dose. The intensity variation in the PL spectra is due to colour center mechanism. The SHG efficiency of 2AP4N crystals was found to be unaffected by gamma irradiation.

      • A Mathematical Model Development of Automotive 42-Volt Battery System and a Starter-Generator

        The proposed mathematical model of the starter-generator system incorporates the motor speed, battery voltage and the desired current to estimate the moment generation capabilities of the starter-generator and the actual current of the battery system. The fundamentals for this mathematical modeling are the simulated results of the experimental data. These pertinent data are used in establishing the governing equations for the determination of motor moments, actual battery currents and efficiencies of the system's operation at different loading characteristics and speed regions. The derived equations are projected to incorporate into simulation programs which can be used in further study of the system's operations.

        Anesthetic considerations for a patient with myasthenia gravis undergoing deep sedation in an outpatient oral surgery setting

        Myasthenia gravis (MG) is a neuromuscular autoimmune disorder which clinically presents as muscular weakness and fatigue due to autoantibody formation against acetylcholine receptors (AChR), leading to their subsequent destruction. Due to the neuromuscular implications of MG, certain considerations must be taken into account when providing anesthesia to MG patients. In the following case report, we have outlined procedural considerations for the anesthetic management of a patient with MG undergoing deep sedation for an elective oral surgery in an outpatient setting, as well as a discussion of relevant literature.

        Scavenging Strategy of Panax ginseng Against Formed Free Radicals Under Stress of Mercuric Chloride in Rattus norvegicus

        Twenty five albino rats were divided into five groups for conducting this experiment. The first group was for positive control (Vitamin C, ascorbic acid), the second group was of Panax ginseng (10 ㎎/㎏ body weight) treated group after bio-activity assay, the third group was of mercuric chloride treated group (0.033 ㎎/㎏ body weight) based on calculating LD?? 9.26 ㎎/㎏ body weight by probit analysis, the fourth group was of mercuric chloride (0.033 ㎎/㎏ body weight) followed by Panax ginseng (10 ㎎/㎏ body weight) and the fifth group was Panax ginseng (10 ㎎/㎏ body weight) followed by mercuric chloride (0.033 ㎎/㎏ body weight) treated group. The interval between intake of Panax ginseng and mercuric chloride was of 2 hours in groups, fourth and fifth respectively. Comparative free radical scavenging property of Panax ginseng was studied under three in vitro models (role model for calculating scavenging activity) viz. DPPH method (hydroxyl free radicals), Nitric oxide method (nitrile free radicals) and Lipid peroxidation (mercury free radicals).
