      • MiR-454 Prompts Cell Proliferation of Human Colorectal Cancer Cells by Repressing CYLD Expression

        Previous studies have shown that miR-454 plays an important role in a variety of biological processes in various human cancer cells. However, the underlying mechanisms of this microRNA in colorectal cancer (CRC) cells remain largely unknown. In the present study, we investigated the miR-454 role in CRC cell proliferation. We found that miR-454 expression is markedly upregulated in CRC tissues and CRC cells compared with the matched tumor adjacent tissues and the FHC normal colonic cell line. Ectopic expression of miR-454 promoted the proliferation and anchorage-independent growth of CRC cells, whereas inhibition of miR-454 reduced this effect. Bioinformatics analysis further revealed cylindromatosis (CYLD), a putative tumor suppressor as a potential target of miR-454. Data from luciferase reporter assays showed that miR-454 directly binds to the 3'-untranslated region (3'-UTR) of CYLD mRNA and repressed expression at both transcriptional and translational levels. In functional assays, CYLD-silenced in miR-454-in-transfected SW480 cells have positive effect to promote cell proliferation, suggesting that direct CYLD downregulation is required for miR-454-induced CRC cell proliferation. In sum, our data provide compelling evidence that miR-454 functions as an onco-miRNA, playing a crucial role in the promoting cell proliferation in CRC, and its oncogenic effect is mediated chiefly through direct suppression of CYLD expression.

        The Quantitative Evaluation of Aging State of Field Composite Insulators Based on Trap Characteristics and Volume Resistivity-Temperature Characteristics

        In order to obtain a better understanding of the ageing process of the field composite insulators, it is necessary to explore a quantitative-valuation method for the aging state evaluation. And the linear relationship between volume resistivity and temperature is proposed. In this paper, the composite insulators with different lengths of operating lives from two manufacturers were tested. The relationship between trap characteristics and volume resistivity-temperature characteristics were analyzed based on Thermal Stimulated Current (TSC), volume resistivity-temperature test, Scanning Electron Microscope (SEM) and Fourier Transform Infrared Spectroscopy (FTIR). Furthermore, the application of trap characteristics in the quantitative evaluation of aging state of composite insulators was discussed. The results showed that there was a general negative correlation between the relative variation ratio of trap charges and the volume resistivity-temperature characteristics. Meanwhile, the physicochemical properties would change with the aging time, which would result in the increasing of electron traps. Combined with the TSC and volume resistivity test results, the trap characteristic thresholds which indicated the serious age of the composite insulators had been proposed.

        Enhanced Room-Temperature Thermoelectric Performance of In-Doped ZnO:Al Thin Films through Prefabricated Layer Doping Method

        In this study, AZO thin films prepared by direct current reactive magnetron sputtering using a Zn-Al alloy target and In with varied content were doped through the prefabricated layer doping method in order to optimize their thermoelectric properties. The effects of In content on the room temperature microstructure and thermoelectric properties of the AZO thin films were investigated. It was found that the absolute value of the Seebeck coefficient of the thin films increases stably after In doping and reaches 153 μV·K−1 when the In content is 0.71%. Though the electrical conductivity of In-doped thin films is smaller than those of the un-doped films, the power factor of the thin films shows a significant increase after In doping with a maximum value of 2.22 × 10−4 W·m−1·K−2, which is several times that of the un-doped films.

        Compound glycyrrhizin injection for improving liver function in children with acute icteric hepatitis: A systematic review and meta-analysis

        Background: Compound glycyrrhizin injection (CGI) is a preparation with glycyrrhizin as the main active ingredient extracted from licorice. As clinical trials suggest that CGI is effective in improving liver function for acute icteric hepatitis in children (AIHC), this systematic review aimed to evaluate and verify its therapeutic effects and safety. Methods: Six electronic databases were searched from their inception to 15 May 2021. Randomized controlled trials (RCTs) assessing therapeutic effects and safety of CGI for AIHC were included. The risk of bias for each trial was assessed using the Cochrane Risk of Bias Tool 2.0. Primary outcomes were indexes related to liver function, including total bilirubin (TBiL), alanine aminotransferase (ALT) and aspartate transaminase (AST). RevMan 5.4 software was used for data analyses. The certainty of the evidence was assessed using the online GRADEpro tool. Results: Six RCTs involving 608 children were included. The overall bias was assessed as having “high risk of bias” in all trials. All trials compared the combination of CGI and conventional western medicine (CWM) with CWM alone. Regarding the effects of CGI for AIHC, results showed that CGI plus CWM was superior to CWM alone in reducing the levels of TBiL (mean difference (MD) = -8.19 mmol/L, 95% CI -9.86 to -6.53), ALT (MD = -24.09 U/L, 95% CI -30.83 to -17.34) and AST (MD = -18.67 U/L, 95% CI -21.88 to -15.45). No trial reported adverse events. The certainty of the evidence for outcomes were all evaluated as low or very low. Conclusion: CGI may have adjuvant therapeutic effects on improving the liver function of children with AIHC. There is no evidence to determine the safety of CGI for AIHC. As current evidence is weak, further well-designed RCTs are required for verification of the therapeutic effects of CGI.       • The Lymphotoxin-α 252 A>G Polymorphism and Breast Cancer: A Meta-analysis

        Objective: The aim of this meta-analysis is to evaluate associations between LTA-252 A>G and breast cancer (BC). Methods: Electronic searches of several databases were conducted for all online publications. A total of 7 studies involving 4,625 BC patients and 4,373 controls were identified. Results: This meta-analysis showed no significant association between the LTA-252 A>G polymorphism and BC in overall or Caucasian populations. However, a positive association was found limited to Asian populations. Conclusion: Although there was no significant association found between the LTA-252 A>G polymorphism and BC overall, a positive association was found in Asian populations.

        Regulation of meilingmycin in Streptomyces nanchangensis: Effect of ammonium ion

        Effects of different ammonium sulfate concentrations on meilingmycin biosynthesis were studied in this research. The results show that a lower concentration of ammonium ions stimulates the biosynthesis of meilingmycin,while a concentration higher than 5mmol/L inhibits the mycelial growth and the biosynthesis of the products. However,increased sugar consumption rate with the elevated concentration of ammonium sulfate was observed during the fermentation process. On this basis, six enzymes, which are responsible for the meilingmycin biosynthesis and the glucose metabolism, were measured and analyzed during the bioprocess. The results suggest that glucose-6-phosphate dehydrogenase, citrate synthase, succinate dehydrogenase and fatty acid synthase are stimulated by higher concentration of ammonium ions, while valine dehydrogenase and methylmalonyl-CoA carboxyltransferase are inhibited. From the results it follows that the precursor supply was restricted with the higher concentration of ammonium ions, which results in the lower production of meilingmycin. Thus, the strategy of maintaining a low level of FAS activity is a critical factor for high yield of meilingmycin.

        A novel method for the synthesis of nano-sized MgAl2O4 spinel ceramic powde

        This study describes the preparation and characterization of MgAl2O4 spinel ceramic powders by a polyacrylamide gel method with Al(NO3)3·9H2O and Mg(NO3)3·6H2O as the raw materials, acrylamide as the monomer, N,N-mehtylenebisacrylamide as the cross-linking agent, and deionized water as the solvent. The nanopowders were studied by X-ray diffraction (XRD) and a transmission electron microscope (TEM). The results showed that the nanopowders having a typical spinel structure are ultrapure and nano-sized. Due to the hindering effect of the polyacrylamide network, the average grain size of the MgAl2O4spinel is approximately 20 nm. Moreover, it is confirmed that the optimal sintering temperature for synthesizing the MgAl2O4spinel ceramic nanopowders is 900 oC, which is about 600-1000 oC lower than that of the traditional solid-state method.

        Adsorption of metal ions with biochars derived from biomass wastes in a fixed column: Adsorption isotherm and process simulation

        Biochars were derived from wood and water caltrop shell waste for the adsorption of Cr(III) and Cu(II) insolutions. In the competitive adsorption, Cu(II) had a good affinity for the water caltrop shell and blendedbiochars. The blended biochar was a mixture of wood and water caltrop shell biochars. Each biocharadsorption equilibrium datafit well with the Freundlich model. The effectiveness of the adsorption of Cr(III) and Cu(II) solutions utilizing biochar is proven to be effective in continuous adsorption columns, andthe adsorption performance is verified with model simulation.

        A Correlation between the Severity of Lung Lesions on Radiographs and Clinical Findings in Patients with Severe Acute Respiratory Syndrome

        Objective: The purpose of this study was to quantify lesions on chest radiographs in patients with severe acute respiratory syndrome (SARS) and analyze the severity of the lesions with clinical parameters. Materials and Methods: Two experienced radiologists reviewed chest radiographs of 28 patients with SARS. Each lung was divided into upper, middle, and lower zones. A SARS-related lesion in each zone was scored using a four-point scale: zero to three. The mean and maximal radiographic scores were analyzed statistically to determine if the scorings were related to the laboratory data and clinical course. Results: Forward stepwise multiple linear regression showed that the mean radiographic score correlated most significantly with the number of hospitalized days (p < 0.001). The second most significant factor was the absolute lymphocyte count (p < 0.001) and the third most significant factor was the number of days of intubation (p = 0.025). The maximal radiographic score correlated best with the percentage of lymphocytes in a leukocyte count (p < 0.001), while the second most significant factor was the number of hospitalized days (p < 0.001) and the third most significant factor was the absolute lymphocyte count (p = 0.013). The mean radiographic scores of the patients who died, with comorbidities and without a comorbidity were 11.1, 6.3 and 2.9, respectively (p = 0.032). The corresponding value for maximal radiographic scores were 17.7, 9.7 and 6.0, respectively (p = 0.033). Conclusion: The severity of abnormalities quantified on chest radiographs in patients with SARS correlates with the clinical parameters.
