      • Population Pharmacokinetics with Genetic Variants of SLCO1B1 of Rifampin in Korean Tuberculosis Patients

        Background Rifampin (RIF) is a backbone drug of tuberculosis (TB) treatment. However, little is known for the influences of SLCO1B1 on RIF pharmacokinetics (PK) in the Asian population. The aim of this study was to investigate the effect of SLCO1B1 variants on RIF PK parameters in Korean TB patients. Methods Clinical data, SLCO1B1 genotype, and drug concentrations from a multicenter prospective cohort study conducted at 20 clinical centers in Korea were utilized for the analysis. The data was divided randomly into a training dataset (80%) and a test dataset (20%). Nonlinear mixed effect analyses were employed to develop population PK modeling and simulation. The final model was internally validated by prediction-corrected visual predictive checks and non-parametric bootstrapping and externally validated using test dataset. Results In total, 879 TB patients (560 males and 319 females) were included in our study. The one-compartment disposition model with first-order absorption and elimination, and combination of additive and proportional residual error best described the RIF PK of the training data. Bodyweight and SLCO1B1 phenotype, classified as a normal, intermediate, and low function, significantly affected the apparent clearance (CL/F). The mean of RIF clearance of the normal SLCO1B1 function group is 18.3% and 14.1% higher than the intermediate and low function group (7.63 L/h to 6.45 L/h and 6.69 L/h, respectively). The findings indicated that the efficacy and safety of an intensified RIF initial dosing strategy could be partially based on SLCO1B1 phenotype classification. Conclusions The SLCO1B1 phenotype could partially explain the variability of RIF PK on Korean TB patients. Ensuring the proper RIF model- informed initial dose and dosing adjustment based on SLCO1B1 phenotype may improve the treatment outcome of the TB patients.

        Isolation and Cytotoxic Potency of Endophytic Fungi Associated with Dysosma difformis, a Study for the Novel Resources of Podophyllotoxin

        Endophytic fungi are promising sources for the production of podophyllotoxin-an important anticancer compound, replacing depleted medical plants. In this study, the endophytes asso- ciated with Dysosma difformis-an ethnomedicinal plant species were isolated to explore novel sources of podophyllotoxin. Fifty-three endophytic fungi were isolated and identified by morphological observation and ITS-based rDNA sequencing, assigning them to 27 genera in 3 divisions. Fusarium was found the most prevalent genus with a colonization frequency of 11.11%, followed by Trametes (9.26%) and Penicillium (7.41%). Phylogenetic trees were constructed for the endophytic fungi community in two collection sites, Ha Giang and Lai Chau, revealing the adaptation of the species to the specific tissues and habitats. Cytotoxic activity of endophytic fungal extracts was investigated on cancer cell lines such as SK-LU-1, HL-60, and HepG2, demonstrating strong anti-cancer activity of six isolates belonging to Penicillium, Trametes, Purpureocillium, Aspergillus, and Ganoderma with IC50 value of lower than 10 mg/mL. The presence of podophyllotoxin was indicated in Penicillium, Trametes, Aspergillus and for the first time in Purpureocillium and Ganoderma via high-performance liquid chromatography, which implied them as a potential source of this anti- cancer compound.

        Polymerization of vinyl pivalate in supercritical carbon dioxide and the saponification for the preparation of syndiotacticity-rich poly(vinyl alcohol)

        This paper reports a successful free-radical dispersion polymerization of vinyl pivalate in supercritical carbon dioxide using poly(1H,1H-dihydroperfluorooctyl acrylate) and dimethylsiloxane-g-pyrrolidone carboxylic acid (Monasil PCA) as stabilizers. In both cases, no spherical polymer particles were obtained, which might be due to the high solubility of poly(vinyl pivalate) (PVPi) in supercritical carbon dioxide. The effects of the reaction time, pressure,temperature, monomer loading, stabilizer concentration and initiator concentration on the polymer yield and molecular weight of PVPi were also investigated. In addition, the formation of poly(vinyl alcohol) via saponification of the resulting PVPi is described.

      • Characterization of Ag-Doped p-Type SnO Thin Films Prepared by DC Magnetron Sputtering

        <P>Crystalline structure and optoelectrical properties of silver-doped tin monoxide thin films with different dopant concentrations prepared by DC magnetron sputtering are investigated. The X-ray diffraction patterns reveal that the tetragonal SnO phase exhibits preferred orientations along (101) and (110) planes. Our results indicate that replacing Sn<SUP>2+</SUP> in the SnO lattice with Ag<SUP>+</SUP> ions produces smaller-sized crystallites, which may lead to enhanced carrier scattering at grain boundaries. This causes a deterioration in the carrier mobility, even though the carrier concentration improves by two orders of magnitude due to doping. In addition, the Ag-doped SnO thin films show a p-type semiconductor behavior, with a direct optical gap and decreasing transmittance with increasing Ag dopant concentration.</P>

        An Empirical Study of Perceived Organizational Support and Affective Commitment in the Logistics Industry

        The study examines the impact of organizational rewards, procedure justice, and perceived supervisor support on perceived organizational support, and examine the impact of perceived organizational support on affective commitment to the organization in the logistic enterprises as well. Quantitative research is applied to measure relationships by regression analysis with SPSS. The research data was collected by convenient method from 180 employees who work in different departments in the logistics industry. The study results in the model 1 found that organizational rewards, procedure justice, and perceived supervisor support have a positive relationship to the perceived organizational support. The study results in the model 2 also found that perceived organizational support has strong impact on the affective commitment to the organization. The study results contribute to both management theory and management practice. For the management theory aspect, the authors suggest that perceived organizational support should be considered the key antecedent of affective commitment about which researchers should pay more attention as a concept. Based on the research results, the authors also recommend for the management practice that managers should pay attention to the implementation of rewards, procedure justice, and supervisor support to increase the perceived organizational support and affective commitment in the logistic enterprises.

        Supercritical fluid mediated synthesis of poly(2-hydroxyethyl methacrylate)/Fe<sub>3</sub>O<sub>4</sub> hybrid nanocomposite

        <P><B>Graphical abstract</B></P><P>We successfully developed an easier and more effective method to prepare a magnetic polymer nanocomposite by in situ radical polymerization of 2-hydroxyethyl methacrylate in the presence of the surface-modified Fe<SUB>3</SUB>O<SUB>4</SUB> nanoparticles with a coupling agent in supercritical CO<SUB>2</SUB>. FE-TEM pictures showed that the nanoparticles were well dispersed in the polymer matrix. The incorporation of Fe<SUB>3</SUB>O<SUB>4</SUB> in the nanocomposite was confirmed by FT-IR, XRD and XPS. Fe<SUB>3</SUB>O<SUB>4</SUB> nanoparticles were first surface-modified with a silane coupling agent methacryloxypropyltrimethoxysilane (MPTMS), which provides a reactive CC bond and can copolymerize with 2-hydroxyethyl methacrylate (HEMA). After immobilization of the silane coupling agent, polymer chains were successfully grafted onto the surface of Fe<SUB>3</SUB>O<SUB>4</SUB>, resulting in the formation of core–shell nanostructure.         Multi-Strain Probiotics Enhance the Bioactivity of Cascara Kombucha during Microbial Composition-Controlled Fermentation

        Kombucha is a widely consumed fermented tea beverage with diverse health benefits. In a previous study, we demonstrated that the use of cascara as a substrate results in a special kombucha beverage with high bioactivity. Traditional kombucha fermentation using a symbiotic culture of bacteria and yeast (SCOBY) can lead to inconsistent product quality because of the lack of control over microbial composition. We successfully isolated and identified yeast and bacteria, including Saccharomyces cerevisiae, Komagataeibacter rhaeticus, and Lactobacillus brevis that are appropriate starter cultures for cascara kombucha fermentation. We also demonstrated that a supplementation with lactic acid bacteria (LAB) and a mixture of S. cerevisiae and K. rhaeticus resulted in higher total polyphenol and flavonoid content of cascara kombucha compared with the traditionally fermented product using SCOBY as the inoculum. The free radical scavenging activity, inhibitory effects on α-amylase, tyrosinase activity, and antibacterial properties of cascara kombucha were also enhanced as a result of LAB supplement. These findings provide valuable insights into the controlled microbiological composition required for the fermentation of cascara kombucha, thereby ensuring consistent quality and enhanced bioactivity of the product. Further, the use of cascara as a substrate for kombucha production not only offers various health benefits and biological effects, but also repurposes by-products from the coffee industry, which contributes to sustainable development and is eco-friendly.

        Exploration of genetic diversity of Bacillus spp. from industrial shrimp ponds in Vietnam by multi-locus sequence typing

        Bacillus is a diverse genus consisting of more than 200 species with extensive genetic diversity. Their beneficial effects in industrial shrimp farming have been well documented. However, little is known about the biodiversity of the Bacillus spp. in this aquaculture system. Taxonomic analysis by 16S rRNA sequencing does not always allow species-level identification of Bacillus spp. In this study, 26 Bacillus isolates from two industrial Litopenaeus vannamei shrimp ponds in Bac Lieu Province, Vietnam, were analyzed for their genetic diversity by multi-locus sequence typing (MLST). A total of 22 sequence types were identified and segregated into four distinct clusters, corresponding to B. subtilis, B. velezensis, B. siamensis, and B. licheniformis. Bacillus subtilis and B. velezensis accounted for more than 73% of the Bacillus isolates. Notably, the MLST scheme exhibited high discriminatory power and might be further simplified to be a convenient method to identify species of the genus Bacillus.
