      • A Mathematical Study on the Growth Factor of the Antennal Segments in Anoplocnemis dallasi (Coreidae, Het.)

        摘 要 : 장수허리노린재의 後胚子成長期에 있어서 4個의 觸角節을 對象으로 제1齡 幼 부터 成 에 이르기까지의 길이를 測定하여 Holzinger & Harmann(1941)의 因子分析法과 Park(1977)의 變形法을 적용하여 成長因子를 分析한 結果 다음과 같은 結論을 얻었다: ① 4個의 觸角節은 2個의 成長因子가 區別된다. ②第1因子는 一般因子로소 成長促進因子였고, 第2因子는 群因子로서 作用하였다. ③2種의 因子를 結合해 보면 4個의 觸角節은 成長影響을 달리하는 第1節과 弟4節의 弟1群, 弟2群, 弟3節의 弟3群으로 區別할 수 있었으나 弟3群은 弟2群에 近似하였다. ④弟2節의 共通度가 가장 크고, 弟4節의 共通度가 가장 낮다. ⑤成長因子의 特性은 齡이 進前됨에 따라 增加한다(P<0.05). ⑥ 觸角 弟1節과 弟2節間의 分節과 觸角 弟3節과 弟4節間의 分節의 2個 分節이 先行되고, 觸角 弟2節과 弟3節間의 分節이 二次的으로 行해진다. Applying the Averoid Solution of Factor Analysis of Holzinger & Harmann(1941) and Park's techniques (1977) to the original values for instar in the four antennal segments of A. dallasi, it is summarized as follows: ①All the four antennal segments are found to have the two factors.②The first os acceptable as a general factor and a growth promoting factor, whereas the second factor is indicated as a group factor.③The four segment are classifiable into the three groups- The first group of the first and fourth segments, the second group of the second segment, and the third group, of the third segment. But the third group is similar to the second group in the influence of the factors. ④ The communality of the highest, while that of the fourth segment is the lowest. ⑤ The contribution of the growth factor is increased according to the development of the instars (P<0.05). ⑥ The segmentations between the first and second segments and between the third and fourth segments are precded, and followed by the segmentation between the second and third segments secondarily.

        <P>We have cloned the macrophage migration inhibitory factor (MIF)-like protein (Anisakis simplex (As)-MIF) from larvae of the whale worm (Anisakis simplex third-stage larvae). Asthma was induced in the mice using OVA/alum, with or without various concentrations of rAs-MIF treatment before OVA/alum challenge. Treatment with rAs-MIF coupled with OVA/alum during the challenge period induced a complete inhibition of eosinophilia and goblet cell hyperplasia within the lung and profoundly ameliorated the development of lung hyperreactivity. Also, rAs-MIF was shown to reduce profoundly the quantity of Th2-related cytokines (IL-4, IL-5, and IL-13) in the bronchial alveolar lavage fluid and allergen-specific IgG2a in sera. IL-10 and TGF-beta levels in the bronchoalveolar lavage fluid of the rAs-MIF-treated group were significantly higher than in the other groups. Additionally, CD4(+)CD25(+)Foxp3(+) T cells (regulatory T) were recruited to the spleen and lungs of the rAs-MIF-treated mice, but this recruitment was inhibited by anti-rAs-MIF Ab.</P>

        Progression-Free Survival: An Important Prognostic Marker for Long-Term Survival of Small Cell Lung Cancer

        Background: Small cell lung cancer (SCLC) is an extremely aggressive tumor with a poor clinical course. Although many efforts have been made to improve patients' survival rates, patients who survive longer than 2 years after chemotherapy are still very rare. We examined the baseline characteristics of patients with long-term survival rates in order to identify the prognostic factors for overall survivals. Methods: A total of 242 patients with cytologically or histologically diagnosed SCLC were enrolled into this study. The patients were categorized into long- and short-term survival groups by using a survival cut-off of 2 years after diagnosis. Cox's analyses were performed to identify the independent factors. Results: The mean patient age was 65.66 years, and 85.5% were males; among the patients, 61 of them (25.2%) survived longer than 2 years. In the multivariate analyses, CRP (hazard ratio [HR], 2.75; 95% confidence interval [CI], 1.25-6.06; p=0.012), TNM staging (HR, 3.29; 95% CI, 1.59-6.80; p=0.001), and progression-free survival (PFS) (HR, 11.14; 95% CI, 2.98-41.73; p<0.001) were independent prognostic markers for poor survival rates. Conclusion: In addition to other well-known prognostic factors, this study discovered relationships between the long-term survival rates and serum CRP levels, TNM staging, and PFS. In situations with unfavorable conditions, the PFS would be particularly helpful for managing SCLC patients.

        Treatment outcome and risk analysis for cataract after radiotherapy of localized ocular adnexal mucosa-associated lymphoid tissue (MALT) lymphoma

        Purpose: We retrospectively reviewed the results of radiotherapy for localized ocular adnexal MALT lymphoma (OAML) to investigate the risk factors of cataract. Methods: Sixty-seven patients with stage IE OAML treated with radiotherapy at Seoul St. Mary's Hospital from 2001 to 2016 were included. Median treatment dose was 30 Gy. Lens protection was done in 52 (76%) patients. Radiation therapy (RT) extent was as follows: superficial (82.1%), tumor mass (4.5%), and entire orbital socket (13.4%). The risk factors for symptomatic cataract were analyzed using the Cox proportional hazard model. Results: Median follow-up time was 50.9 months (range, 1.9 to 149.4 months). All patients were alive at the time of analysis. There were 7 recurrences and there was no local recurrence. Median time to recurrence was 40.4 months. There were 14 cases of symptomatic cataract. Dose >30 Gy had hazard ratio of 3.47 for cataract (p = 0.026). Omitting lens protection showed hazard ratio of 4.10 (p = 0.008). Conclusions: RT achieves excellent local control of ocular MALT lymphoma. Consideration of RT-related factors such as lens protection and radiation dose at the stage of RT planning may reduce the risk of RT-induced cataract after radiotherapy.

        Effects of ice-binding protein from Leucosporidium on the cryopreservation of boar sperm

        The aim of this study was performed to evaluate the effects of ice-binding protein from the arctic yeast Leucosporidium (LeIBP) supplementation on cryopreservation of boar sperm. The collected semen was diluted ($1.5{\times}10^8/ml$) in lactose egg yolk (LEY) and cooled at $5^{\circ}C$ for 3 h. The cooled semen was then diluted ($1{\times}10^8/ml$) in LeIBP containing LEY with 9% glycerol and maintained at $5^{\circ}C$ for 30 min. The semen was divided into six experimental groups (control, 0.001, 0.005, 0.01, 0.05 and 0.1 mg/ml of LeIBP). The straws were kept on above the liquid nitrogen ($LN_2$) vapors for 20 minutes and then plunged into $LN_2$. After thawing, computer-assisted sperm analysis was used for sperm motility and flow cytometry was performed to assess the viability, acrosome integrity (FITC-PSA/PI), ROS (DCF/PI), lipid peroxidation (BODIPY C11/PI) and apoptosis (Annexin V/PI), respectively. No significant responses were observed for sperm motility. However, sperm viability was significantly increased on 0.05 and 0.1 mg/ml of LeIBP groups compared to control (P < 0.05). In addition, acrosome integrity was significantly increases LeIBP groups (P < 0.05) and both ROS and lipid peroxidation level were lower in all LeIBP groups than those of control (P < 0.05). On the other hand, a significant higher apoptosis rate was observed in 0.05 and 0.1 mg/ml of LeIBP groups compared to control (P < 0.05). It can be assumed that a supplementation of LeIBP in boar sperm freezing extender is an effective method to increase the sperm qualities after cryopreservation.

        This study was performed to evaluate effects of Leucosporidium ice-binding protein (LeIBP) supplementation on boar (Duroc) sperm freezing. The collected semen was diluted (1.5×10⁸ /mL) in lactose egg yolk (LEY) and cooled at 5°C for 3 h. The cooled semen was then diluted (1×10⁸ /mL) in LeIBP containing LEY with 9% glycerol and maintained at 5°C for 30 min. The semen was divided into six experimental groups (control, 0.001, 0.005, 0.01, 0.05, and 0.1 mg/mL of LeIBP). The straws were stayed above the liquid nitrogen (LN2) vapors for 20 minutes and then plunged into LN2. After thawing, sperm parameters such as motility, viability, and acrosome integrity, intracellular reactive oxygen species (ROS), apoptosis, and undetected lipid peroxidation were determined. Computer assisted sperm analysis was used for sperm parameters and flowcytometry was performed to acrosome integrity (FITC-PSA/PI), ROS (DCFHDA/ PI), lipid peroxidation (BODIPY C11/PI), and apoptosis (Annexin V/PI), respectively. The differences were not in sperm motility, but in viability of 0.05 and 0.1 mg/mL groups compared to control (p<0.05). Higher acrosome integrity was observed in LeIBP groups compared to control (p<0.05), except for 0.001 group. Both ROS and lipid peroxidation level were lower in all LeIBP groups than that of control (p<0.05). Furthermore, lower apoptosis rate was observed in both 0.005 and 0.1 LeIBP groups compared to control (p<0.05). It can be postulated that a supplementation of LeIBP in boar sperm freezing extender is an effective method to increase the sperm qualities after cryopreservation.

        Effect of Temperature on the Growth of the Water Strider, Gerris paludum insularis (Gerridae, Heteroptera)

        The water strider, Gerris paludum insularis, was reared in a growth cabinet at two constant temperatures and ${\approx}20^{\circ}C$ under a 16:8 (L:D) photoperiod. Differences in head width among three temperatures of ${\approx}20^{\circ}C,\;25^{\circ}C\;and\;30^{\circ}C$ in G. paludum insularis, were compared. Temperature affected growth of head width of G. paludum insularis. There were highly significant differences among head width for three temperature regimes and the immature stages. It was found that head width is greater in $30^{\circ}C$ than $25^{\circ}C\;and\;{\approx}20^{\circ}C$. The growth quantity of the head width in ${\approx}20^{\circ}C$ shows the slowest increase, and $25^{\circ}C\;and\;30^{\circ}C$ were gradually increased in that order. The maximum ratio of growth quantity by each stage is in the first instar in all regimes. The growth rates of all regimes show an increase pattern.

      • S-598 Coix sprout extract (CSE) induces cell cycle arrest and apoptosis in human cervical cancer cells

        Cervical cancer is the second most common women carcinoma worldwide and the fourth leading cause of cancer-associated mortality in women. Coix lachryma-jobi L. var. ma-yuen Stapf (coix) is the most widely recognized medicinal herb due to its remedial effects in treat inflammation, dysfunctions of the endocrine system, warts, chapped skin, rheumatism, and neuralgia and also as a nourishing food. In this study, we investigated the anticancer effects of the coix sprout extract (CSE) in human cervical cancer HeLa cells. CSE significantly inhibited cell proliferation in a dose-dependent manner using a CCK-8 assay. CSE induced cycle sub G1/ S phase arrest and apoptosis in HeLa cells in a dose-dependent manner as detected by BrdU-FITC and Annexin V-FITC staining. In addition, apoptosis was evaluated using DAPI staining, which clearly demonstrated cell shrinkage and condensed nuclei. An investigation of the molecular mechanism of CSE reveals that induction of cell cycle arrest is associated with downregulation of cyclin D1, cyclin-dependent kinase (CDK)2, CDK4 and CDK6. Induction of apoptosis by CSE involves the upregulation of the apoptotic proteins p53, cleaved PARP, cleaved-caspase 3, 8, and the downregulation of the anti-apoptotic protein Bcl-2. But, Bax proteins expression was not changed in CSE treated cells. Furthermore, we observed that CSE reduced the phosphorylation of PI3K and AKT protein expression. The use of the AKT inhibitor, the expression of p-AKT and Bcl-2 was inhibited to a significant extent following treatment with both than with LY294002 alone. Accordingly, these finding suggested that CSE induces apoptosis through the PI3K/AKT pathway. Our results suggested that CSE may be considered as a therapeutic agent in cervical cancer.
