<P><B>Abstract</B></P><P>This study was designed to evaluate the enhancement of antioxidant, antimicrobial, enzymatic, cytotoxic, and cognitive activities of <I>Codonopsis lanceolata</I> extracted by high pressure treatment followed by probiotic fermentation. Dried <I>C</I>. <I>lanceolata</I> samples were subjected to 400MPa for 20min and then fermented with <I>Bifidobacterium longum</I> B6 (HPE-BLF) and <I>Lactobacillus rhamnosus</I> (HPE-LRF) at 37°C for 7days. Compared to conventional extraction (CE-NF, 6.69mg GAE/g), the phenol amounts of HPE-BLF and HPE-LRF were significantly increased to more than 8mg GAE/g, while the lowest flavonoid contents were observed for HPE-BLF (0.44mg RE/mL) and HPE-LRF (0.45mg RE/mL) (<I>p</I><0.05). Cinnamic acid was the most abundant phenolic acid in the fermented <I>C</I>. <I>lanceolata</I>. The highest DPPH scavenging activities were observed for HPE-BLF and HPE-LRF, with minimum EC50 values of 1.26 and 1.18mg/mL, respectively. The HPE-BLF and HPE-LRF samples exhibited the most noticeable antimicrobial activities against <I>Staphylococcus aureus</I>, <I>Listeria monocytogenes</I>, <I>Salmonella</I> Typhimurium, and <I>Shigella boydii</I> (MICs<15mg/mL). The fermented <I>C</I>. <I>lanceolata</I> samples effectively inhibited α-glucosidase and tyrosinase activities and potentially improved a scopolamine-induced memory deficit in mice. The application of a fermentation process can effectively improve the biological and pharmacological activities of high-pressure-extracted <I>C</I>. <I>lanceolata</I> by increasing the extraction efficacy and inducing probiotic conversion. The results suggest that the combined treatment of HPE and a fermentation process could be used as alternative extraction method over CE.</P>

In this paper, we utilize hand gesture information to recognize hand gesture. Firstly, we can utilize human skin color information in YCbCr color model to detect human skin region, and then use edge detection algorithm to detect skin region edge, and confirm the rotundity rate constant for every region by comparing region length to area value. Because that for human hand, it contain very complex edge, so hand's rotundity rate is usually very large. Through this method we can easily confirm hand region and remove those small parts which we call it noise parts. Then we segment the hand region into 5 by 5 pieces and calculate pixel rate of every region to extract eigenvector of hand. We employed a recognition machine based on neural net to recognize the detected hand posture. For recognized hand posture, we then code it to make a corresponding command for synthesizing a sound. And for hand gesture, we can divide the input video into several frames, and in every frame, we can applied the hand posture recognition to recognize the information.

<P><B>Abstract</B></P> <P>Stretchable conductive fibers have attracted significant attention due to their ability to be directly woven into or stitched onto fabrics, making them ideal for use in the design of integrated wearable strain sensors. Here, we report on a highly stretchable multi-walled carbon nanotube (MWCNT)/Thermoplastic Polyurethane (TPU) fiber produced via a wet spinning process. The effects of MWCNT content and alignment on the structural, mechanical, electrical and strain-sensing properties of the composite fibers were investigated. The highest conductivity (6.77 S cm−1), tensile strength (28 MPa) and maximum elongation at break (565%) were obtained by controlling the MWCNT content. Gauge factor (GF) values were also affected by the content and MWCNT alignment in the composite fibers, as these parameters determine the change in the effective contact area and number of conductive paths available during stretching. The well-aligned MWCNT/TPU fiber showed a high GF value of 5200. Wearable strain sensors capable of obtaining real-time mechanical feedback for various human motion detections with different GFs and working strain ranges could be realized by controlling the MWCNT concentrations in the TPU matrix.</P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

In this paper, we present an algorithm which detects human hand by skin color information in YCbCr and HIS color model. And for confirming special human hand we use circle rate of region to detect hand region because human hand have complex edge than other region, thus circle rate of hand region is usually more greater. For the recognition of detected hand, we use the Hausdorff to tracking the hand region. And we employed a recognition method based on PCA algorithm to recognize the hand gestures. The experimental results show that an algorithm plays an efficient effort for hand gesture recognition.

<P>Intracellular Vitamin C (VC) is maintained at high levels in the developing brain by the activity of sodium-dependent VC transporter 2 (Svct2), suggesting specific VC functions in brain development. A role of VC as a cofactor for Fe(II)-2-oxoglutarate-dependent dioxygenases has recently been suggested. We show that VC supplementation in neural stem cell cultures derived from embryonic midbrains greatly enhanced differentiation toward midbrain-type dopamine (mDA) neurons, the neuronal subtype associated with Parkinson's disease. VC induced gain of 5-hydroxymethylcytosine (5hmC) and loss of H3K27m3 in DA phenotype gene promoters, which are catalyzed by Tet1 and Jmjd3, respectively. Consequently, VC enhanced DA phenotype gene transcriptions in the progenitors by Nurr1, a transcription factor critical for mDA neuron development, to be more accessible to the gene promoters. Further mechanism studies including Tet1 and Jmjd3 knockdown/inhibition experiments revealed that both the 5hmC and H3K27m3 changes, specifically in the progenitor cells, are indispensible for the VC-mediated mDA neuron differentiation. We finally show that in Svct2 knockout mouse embryos, mDA neuron formation in the developing midbrain decreased along with the 5hmC/H3k27m3 changes. These findings together indicate an epigenetic role of VC in midbrain DA neuron development. Stem Cells2015;33:1320-1332</P>

<P><B>Summary</B></P> <P>A destabilized tumor vasculature leads to limited drug delivery, hypoxia, detrimental tumor microenvironment, and even metastasis. We performed a side-by-side comparison of ABTAA (Ang2-Binding and Tie2-Activating Antibody) and ABA (Ang2-Blocking Antibody) in mice with orthotopically implanted glioma, with subcutaneously implanted Lewis lung carcinoma, and with spontaneous mammary cancer. We found that Tie2 activation induced tumor vascular normalization, leading to enhanced blood perfusion and chemotherapeutic drug delivery, markedly lessened lactate acidosis, and reduced tumor growth and metastasis. Moreover, ABTAA favorably altered the immune cell profile within tumors. Together, our findings establish that simultaneous Tie2 activation and Ang2 inhibition form a powerful therapeutic strategy to elicit a favorable tumor microenvironment and enhanced delivery of a chemotherapeutic agent into tumors.</P> <P><B>Highlights</B></P> <P> <UL> <LI> ABTAA-induced Tie2 activation and Ang2 inhibition normalize tumor vasculature </LI> <LI> Tumor vessel normalization leads to enhanced blood perfusion and drug delivery </LI> <LI> Tumor vessel normalization lessens hypoxia, acidosis, tumor growth, and metastasis </LI> <LI> Tie2 activation favorably alters the tumor microenvironment and immune infiltration </LI> </UL> </P> <P><B>Graphical Abstract</B></P> <P>[DISPLAY OMISSION]</P>

Selective inhibition of phosphodiesterase (PDE) 5 opened a new therapeutic approach for cardiovascular disorders. Therefore, the effect of PDE5 inhibition on the cardiac function should thoroughly be defined. The purpose of the present study was to define the effects of sildenafil, a selective inhibitor of PDE5, on the atrial cGMP efflux, atrial dynamics, and the release of atrial natriuretic peptide (ANP). By perfusing rabbit left atria to allow atrial pacing, changes in atrial stroke volume and pulse pressure, transmural extracellular fluid translocation, cGMP efflux, and ANP secretion were measured. SIN-1, an NO donor and soluble (s) guanylyl cyclase (GC) activator, and C-type natriuretic peptide (CNP), an activator of particulate (p) GC activator, were used. Sildenafil increased basal levels of cGMP efflux slightly but not significantly. Sildenafil in a therapeutic dose increased atrial dynamics (for atrial stroke volume, 2.84⁑1.71%, n=12, vs 0.71⁑0.86%, n=21; p<0.05) and decreased ANP release (9.02⁑3.36%, n=14, vs 1.35⁑3.25%, n=23; p<0.05), however, it had no effect on the SIN-1- or CNP-induced increase of cGMP levels. Furthermore, sildenafil in a therapeutic dose accentuated SIN-1-induced, but not CNP- induced, decrease of atrial pulse pressure and ANP release. These data indicate that PDE5 inhibition with sildenafil has a minor effect on cGMP levels, but has a distinct effect on pGC-cGMP- and sGC- cGMP-induced contractile and secretory function.
