Real-time imaging and targeted drug delivery has been lately combined to enhance the therapeutic efficacy in cancer therapy. In this study, we prepared clickable doxorubicin nanoparticles (Dox NP) for theragnostic purposes. Dox conjugated with 8arm-PEG via disulfide bonds to form self-assembled nanoparticles. Dox NP were bi-functionalized with DBCO moiety for clusters formation with azide-modified quantum dots (QD) through copper-free click reaction and MMP cleavable linkers conjugated with methoxy-PEG to protect the Dox NP in circulation. At the high level of MMP-2 enzyme in tumor site, the fluorescence of QD was first quenched by Dox due to the clustering of Dox NP and QD via click reaction. Dox was subsequently released by cleavage of disulfide bonds resulting in recovery of QD fluorescence. Due to the quenching and recovering fluorescence behavior, our Dox NP is supposed to be employed as a molecular sensor for targeting and releasing of anticancer agents at tumor site.

This work studied the inhibitory effect of the combination of benzoate-intercalated hydrotalcite (HT-BZ) and Ce<sup>3+</sup>-loaded clay (Clay-Ce) on carbon steel (CS). HT-BZ was prepared by the co-precipitation method and Clay-Ce was fabricated by a cation exchange reaction. HT-BZ and Clay-Ce were assessed by scanning electron microscopy (SEM) and X-ray diffraction (XRD) coupled with zeta potential measurement. Electrochemical measurements coupled with scanning electron microscopy/energy-dispersive X-ray spectroscopy (SEM/EDX) were used for studying the inhibitory action of the mixture of HT-BZ and Clay-Ce on steel electrodes immersed in 0.1 M NaCl. For comparison, the inhibitory effect of HT-BZ or Clay-Ce alone was also evaluated. The results showed that HT-BZ combined with Clay-Ce provided synergistic inhibition of the CS substrate. The mixture of 0.5 g/L HT-BZ + 0.5 g/L Clay-Ce provided 93.5% inhibition efficiency. The protective mechanism of the HT-BZ + Clay-Ce mixture consisted of the reaction of released BZ and Ce<sup>3+</sup> and the deposition of HT-BZ and Clay-Ce structures on the CS substrate.

Objective: This study was conducted to evaluate the effects of diets containing different wet rice distillers' by-product (RDP) levels on growth performance, nutrient digestibility, blood profiles and gut microbiome of weaned piglets. Methods: A total of 48 weaned castrated male crossbred pigs, initial body weight 7.54±0.97 kg, and age about 4 wks, were used in this experiment. The piglets were randomly allocated into three iso-nitrogenous diet groups that were fed either a control diet, a diet with 15% RDP, or a diet with 30% RDP for a total of 35 days. Chromium oxide was used for apparent digestibility measurements. On d 14 and d 35, half of the piglets were randomly selected for hemato-biochemical and gut microbiota evaluations. Results: Increasing inclusion levels of RDP tended to linearly increase (p≤0.07) average daily gain on d 14 and d 35, and decreased (p = 0.08) feed conversion ratio on d 35. Empty stomach weight increased (p = 0.03) on d 35 while digestibility of diet components decreased. Serum globulin concentration decreased on d 14 (p = 0.003) and red blood cell count tended to decrease (p = 0.06) on d 35, parallel to increase RDP levels. Gene amplicon profiling of 16S rRNA revealed that the colonic microbiota composition of weaned pigs changed by inclusion of RDP over the period. On d 14, decreased proportions of Lachnospiraceae_ge, Ruminococcaceae_ge, Ruminococcaceae_UCG-005, and Bacteroidales_ge, and increased proportions of Prevotellaceae_ge, Prevotella_2, and Prevotella_9 were found with inclusion of RDP, whereas opposite effect was found on d 35. Additionally, the proportion of Lachnospiraceae_ge, Ruminococcaceae_ge, Ruminococcaceae_UCG-005, and Bacteroidales_ge in RDP diets decreased over periods in control diet but increased largely in diet with 30% RDP. Conclusion: These results indicate that RDP in a favorable way modulate gastrointestinal microbiota composition and improve piglet performance despite a negative impact on digestibility of lipids and gross energy.

This work examined the corrosion protection performance of benzoate loaded hydrotalcite/graphene oxide (HT/GO-BZ) for carbon steel. HT/GO-BZ was fabricated by the co-precipitation method and characterized by infrared spectroscopy, X-ray diffraction, and scanning electronic microscopy. The corrosion inhibition action of HT/GO-BZ on carbon steel in 0.1 M NaCl solution was evaluated by electrochemical measurements. The benzoate content in HT/GO-BZ was determined by UV-Vis spectroscopy. Subsequently, the effect of HT/GO-BZ on the corrosion resistance of the water-based epoxy coating was investigated by the salt spray test. The obtained results demonstrated the intercalation of benzoate and GO in the hydrotalcite structure. The benzoate content in HT/GO-BZ was about 16%. The polarization curves of the carbon steel electrode revealed anodic corrosion inhibition activity of HT/GO-BZ and the inhibition efficiency was about 95.2% at a concentration of 3g/L. The GO present in HT/GO-BZ enhanced the inhibition effect of HT-BZ. The presence of HT/GO-BZ improved the corrosion resistance of the waterborne epoxy coating.
Clustered CTLA-4 siRNA and CpG Nanoplexes for Cancer Immunotherapy

Cancer immunotherapy taking the advantages of adaptive immunity have gained increasing interests because it not only suppresses tumor development but induce generation of immunological memory cells and prevent tumor regeneration. Here, we fabricated nanoplexes (NP) composed of aizdo-PEG grafted branched polyethylenimine and CTLA-4 siRNA (siRNA NP) or CpG1826 (CpG NP) by electrostatic interaction. The two NPs were associated into nanoplex clusters (siRNA/CpG NCs) via click chemistry using an alkyne terminated MMP-2-cleavable peptide as a linker. The CpG NPs was demonstrated considerable BMDC activation with increased CD80 and CD86 surface expression. Likewise, splenocytes treated with siRNA NPs showed significant down-regulation of CTLA-4. When administered in vivo, the siRNA/CpG NCs was able to accumulated in tumor and be cleaved by MMP-2 overexpressed in tumor site. The dissociated NPs were released CpG and siRNA when uptaken by cells and activate immune system to combat tumors.

Photothermal therapy 와 polydynamic therapy 를 결합시켜 시너지 효과를 내는 방법이 최근 항암 치료에 많이 사용되고 있는 것처럼 본 연구에서도 두 가지 파장의 근적외선 빛(650 nm, 808 nm)을 이용해 시너지 효과가 발생함을 보았다. 여기에 쓰이는 입자를 만들기 위해 polydopamine (PDA)와 감광제인 Chlorin-e6 을 금 나노 입자(AuNP)에 차례로 코팅하였다(Ce6@PDA@AuNP). AuNP 는 808 nm 의 빛을 흡수하여 SPR 효과에 의해 열을 발생시키게 되고, Ce6 은 650 nm 의 빛을 흡수하여 ROS 를 발생시켜 표적 세포의 사망을 유발할 수 있다. 여러 분석들을 통해 이 입자의 특성들을 확인하였고, 한 파장의 근적외선 빛을 조사할 때 보다 두개 파장의 빛을 사용할 때 더 높은 온도 상승과 더 많은 Ce6 이 방출됨을 확인하였다. 이와 같은 행동 양상들이 세포 및 동물 실험에도 동일하게 적용되는지 확인해본 결과 pH 가 낮아질수록 조사된 파장의 종류에 따라 암세포에 더 큰 악영향을 줄 수 있음이 확인되어 결과적으로 이 나노 입자가 항암 치료에 사용이 가능하다고 말할 수 있다.
