Novel Associations between Related Proteins and Cellular Effects of High-Density Lipoprotein

Background and Objectives: Recent studies have examined the structure-function relationship of high-density lipoprotein (HDL). This study aimed to identify and rank HDL-associated proteins involved in several biological function of HDL. Methods: HDLs isolated from 48 participants were analyzed. Cholesterol efflux capacity, effect of HDL on nitric oxide production, and vascular cell adhesion molecule-1 expression were assessed. The relative abundance of identified proteins in the highest vs. lowest quartile was expressed using the normalized spectral abundance factor ratio. Results: After adjustment by multiple testing, six proteins, thyroxine-binding globulin, alpha-1B-glycoprotein, plasma serine protease inhibitor, vitronectin, angiotensinogen, and serum amyloid A-4, were more abundant (relative abundance ratio ≥2) in HDLs with the highest cholesterol efflux capacity. In contrast, three proteins, complement C4-A, alpha-2-macroglobulin, and immunoglobulin mu chain C region, were less abundant (relative abundance ratio <0.5). In terms of nitric oxide production and vascular cell adhesion molecule-1 expression, no proteins showed abundance ratios ≥2 or <0.5 after adjustment. Proteins correlated with the functional parameters of HDL belonged to diverse biological categories. Conclusions: In summary, this study ranked proteins showing higher or lower abundance in HDLs with high functional capacities and newly identified multiple proteins linked to cholesterol efflux capacity.

<P>Ring-type 33-mode piezoelectric energy harvesters (PEHs) were fabricated using Pb(Zr,Ti)O<SUB>3</SUB>- and (Na,K)NbO<SUB>3</SUB>-based piezoelectric ceramics. The relationship between the output performance of the PEH and the piezoelectric properties of the ceramics was investigated. Ring-type PEHs were selected for this investigation because they can be mounted on a specimen holder at the same position, ensuring structurally reliable output performance. The <I>d</I> <SUB>33</SUB>?×?<I>g</I> <SUB>33</SUB> value, which is a FOM derived from the theoretical output power of the 33-mode PEH, can explain the variation of the output power of the 33-mode PEH with a large specimen holder. However, since the variation of the output power was more accurately explained by the <I>d</I> <SUB>33</SUB>?×?<I>g</I> <SUB>33</SUB>?×?<img ALIGN='MIDDLE' ALT='${{k}_{eff}}^{2}$' SRC='http://ej.iop.org/images/0964-1726/27/11/115027/smsaae6a6ieqn3.gif'/> value, this was suggested as a new FOM of the output power of the ring-type 33-mode PEH with a large specimen holder.</P>

<P>A semicrystalline p-type thermoelectric conjugated polymer based on a polymer backbone of cyclopentadithiophene and benzothiadiazole, poly[(4,4′-(bis(hexyldecylsulfanyl)methylene)cyclopenta[2,1-<I>b</I>:3,4-<I>b</I>′]dithiophene)-<I>alt</I>-(benzo[<I>c</I>][1,2,5]thiadiazole)] (PCPDTSBT), is designed and synthesized by replacing normal alkyl side-chains with bis(alkylsulfanyl)methylene substituents. The sp<SUP>2</SUP>-hybridized olefinic bis(alkylsulfanyl)methylene side-chains and the sulfur-sulfur (S-S) chalcogen interactions extend a chain planarity with strong interchain packing, which is confirmed by density functional calculations and morphological studies, i.e., grazing incidence X-ray scattering measurement. The doping, electrical, morphological, and thermoelectric characteristics of PCPDTSBT are investigated by comparison with those of poly[(4,4′-bis(2-ethylhexyl)cyclopenta[2,1-<I>b</I>:3,4-<I>b</I>′]dithiophene)-<I>alt</I>-(benzo[<I>c</I>][1,2,5]thiadiazole)] (PCPDTBT) with ethylhexyl side-chains. Upon doping with a Lewis acid, B(C<SUB>6</SUB>F<SUB>5</SUB>)<SUB>3</SUB>, the maximum electrical conductivity (7.47 S cm<SUP>-1</SUP>) of PCPDTSBT is ∼1 order higher than that (0.65 S cm<SUP>-1</SUP>) of PCPDTBT, and the best power factor is measured to be 7.73 μW m<SUP>-1</SUP> K<SUP>-2</SUP> for PCPDTSBT with doping 9 mol % of B(C<SUB>6</SUB>F<SUB>5</SUB>)<SUB>3</SUB>. The Seebeck coefficient-electrical conductivity relation is analyzed by using a charge transport model for polymers, suggesting that the doped PCPDTSBT film has superb charge transport property based on a high crystallinity with olefinic side-chains. This study emphasizes the importance of side-chain engineering by using the sp<SUP>2</SUP>-hybridized olefinic substituents to modulate interchain packing, crystalline morphology, and the resulting electrical properties.</P> [FIG OMISSION]</BR>

<P>The prognosis of patients with stage III/IV NK/T-cell lymphoma (NTCL) is extremely poor. Although L-asparaginase (L-asp) is effective for NTCL, its significance has not been clearly demonstrated. In addition, there are few studies comparing treatment outcomes in stage III/IV NTCL. This study evaluated the efficacy of L-asp-based chemotherapy and prognostic factors in stage III/IV NTCL. Seventy patients with newly diagnosed stage III/IV NTCL were enrolled between January 2000 and February 2013. Patients received ifosfamide, etoposide, methotrexate, and prednisolone (IMEP) plus L-asp (N?=?22) or combination chemotherapy without L-asp (N?=?48) as a first-line treatment. Clinical prognostic factors, treatment outcomes, and prognostic scores were compared between the groups. After a median follow-up period of 12.8 months (range, 1.1-186.6 months), median overall survival (OS) and progression-free survival (PFS) were 11.3 and 5.6 months, respectively. Treatment outcomes were superior in patients treated with IMEP plus L-asp compared to those treated with chemotherapy without L-asp (overall response rate, 90.0 vs. 34.8 %, P?<?0.001; complete remission rate, 65.0 vs. 21.7 %, P?=?0.001). The OS and PFS were significantly higher for the IMEP plus L-asp group compared with the chemotherapy without L-asp group. In a multivariate analysis, the use of chemotherapy without L-asp was an independent predictor of reduced OS (hazards ratio (HR)?=?2.18, 95 % confidence interval (CI) 1.08-4.40; P?=?0.030) and PFS (HR?=?2.29, 95 % CI 1.22-4.29; P?=?0.010). IMEP plus L-asp is active against stage III/IV NTCL, and it is an independent predictor of improved survival.</P>

플라스틱 포장재는 식품, 의약품, 화장품 등의 내부 물질과 접촉하게 되면 이행이 발생할 수 있다. 이러한 이행은 제품의 관능, 물리적, 화학적 변화를 야기할 뿐만 아니라 독성을 가지는 화합물로 인하여 인체에 부작용을 끼칠 수 있다. 본 연구에서는 EU의 용기 이행 시험 표준에 근거하여 화장품 1차 포장재로 널리 사용되는 PP, LDPE, HDPE, PET(transparent & colored) 용기 5종에 대하여 overall migration 시험을 수행하였다. 수성, 산성, 지용성, 염기성 화장품을 대표할 수 있는 6종의 모사품을 사용하였고, 시료들을 40℃에서 10일간 보관 후에 분석하였다. 시험에 사용한 모든 용기들이 시험조건에서 EU 기준을 충족하는 것을 확인하였고(10 mg/dm² 또는 60 mg/kg 이하), 일부 폴리올레핀계 용기에서는 규제치 이하지만 산화방지제 등의 첨가제와 올리고머가 이행되는 것을 확인하였다. Migration can occur when plastic packaging materials contact with contents such as food, medicine, and cosmetics. As a result, the organoleptic, physical, and chemical changes of the product can arise and some toxic migrants can produce a risk for human health. In this study, overall migration was studied from PP, LDPE, HDPE, and PET(transparent & colored) packaging into 6 different cosmetic simulants (aqueous, acidic, basic, and fat-soluble solution) through incubation at 40℃ for 10 days. Under these conditions, it was confirmed that all plastic packaging complied with EU regulations, but some migrants such as antioxidants and oligomers were carried over from polyolefin packaging to 95% ethanol aqueous solution.

Followed by dismantling of waste LCD, for hydrometallurgical indium recovery from glass cullet leaching is the rudimentary stage. Although cullet-size reduction adds convenience for recycling, the determination of optimum size is rather essential from efficient leaching process development perspective. Hence, leaching efficiency of indium as function of cullet size were investigated. From our investigation, it has been concluded that size of waste LCD cullet inversely affects leaching efficiency of indium. For efficient leaching smaller cullet size has been recommended, hence, waste LCD needed to be crushed less than 1 mm. The comparison of indium leaching behavior using several acid solutions showed that all the acid solutions had similar leaching efficiency. Hence, the HCl can be recommended for industrial application perspective.

Our understanding of the differential effects between specific omega-3 fatty acids is incomplete. Here, we aimed to evaluate the effects of docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA) on T-helper type 1 (Th1) cell responses and identify the pathways associated with these responses. Naïve CD4<sup>+</sup> T cells were co-cultured with bone marrow-derived dendritic cells (DCs) in the presence or absence of palmitate (PA), DHA, or EPA. DHA or EPA treatment lowered the number of differentiated IFN-γ-positive cells and inhibited the secretion of IFN-γ, whereas only DHA increased IL-2 and reduced TNF-α secretion. There was reduced expression of MHC II on DCs after DHA or EPA treatment. In the DC-independent model, DHA and EPA reduced Th1 cell differentiation and lowered the cell number. DHA and EPA markedly inhibited IFN-γ secretion, while only EPA reduced TNF-α secretion. Microarray analysis identified pathways involved in inflammation, immunity, metabolism, and cell proliferation. Moreover, DHA and EPA inhibited Th1 cells through the regulation of diverse pathways and genes, including Igf1 and Cpt1a. Our results showed that DHA and EPA had largely comparable inhibitory effects on Th1 cell differentiation. However, each of the fatty acids also had distinct effects on specific cytokine secretion, particularly according to the presence of DCs. [BMB Reports 2021; 54(5): 278-283]
