Are philosophical dialogues possible or relevant in catastrophic situations? What does a counselor know? Can philosophy help to understand catastrophic experience? Taking the Biblical parabola of Job as a model, I suggest to make a distinction between the long process of Job's reorientation in his world after the catastrophe, and the immediate post-catastrophic situation, in which Job is in great need of supporting others, and often has to deal with deceitful “friends”, those professional and non-professional helpers, who sometime “darken counsel” with “words without knowledge”. After the catastrophe Job will have to find his personal way in a new territory. A philosophical constructive dialogue in due course may give support at times of doubts and reassessments, offer options, enlarge and lighten the horizons of the searching Job. In the immediate post-catastrophic situation such conversations are not to the point. What is needed is a loving and humble support, with all the power that critical philosophy may provide, against Job's “friends”, their explanations and pretended knowledge. The paper is based on my observations in a neural rehabilitation center and conversations with inmates, professional stuff, family members and visitors. The inmates, mostly victims of road accidents, were at that time partially or completely paralyzed.
How to Train Philosophical Counselors

I explain my conception of philosophical experience and, on the basis of a proposed distinction between levels of philosophy, I deal with its place, in philosophical counseling and its importance in the training of counselees. My criteria for counseling that is really philosophical are, however, not only the level of the philosophical background of the counselor and the philosophical quality of the dialogues, but also the sincerity and moral responsibility that may justify the trust of a counselee in a philosophical counselor.
Stress-Inducible Gene Expression Pattern of the Athb-12, a Homeobox Gene in Arabidopsis

Athb-12 gene은 homeobox gene이고, homeodomain의 가까이에 leucine aipper motif를 가지고 있다. 그 gene은 abscisic acid (ABA)의 처리에 의해 발현이 유도되고, 여러 개의 putative ABRES와 DREs가 5' flanking region에 존재한다. 다양한 길이의 Athb-12 promoter부위와 β-glucuronidase(GUS) reporter gene를 fusion시킨 여러 constructs의 ABA, drought, salt stresses에 의한 발현량이 분석되었다. Transgenic plants에서의 GUS acclivity의 정량적 분석을 통해 Athb-12 gene expression level이 promoter의 길이에 비례적으로 증가함을 알았다. Yeast two-hybrid test를 통해 Athb-12의 HD-Zip은 yeast내에서 homodimer를 형성하지 않음을 밝혔다. 이 연구에서 보여진 결과들로 Athb-12 gene이 ABA에 의해 중계되는 water stress의 signaling pathway에 관여한다는 것을 확인하였다. The Athb-12 gene is a homeobox gene, and has a putative leucine zipper motif closley linked to the homeodomain. It was shown to be induced by exogenous abscisic acid (ABA), and several putative ABA-responsive elements and dehydration-responsive elements were found in its 5'- flanking region. The expression of the Athb-12 gene fused with β-glucuronidase (GUS) reporter gene in response to ABA, drought, and salt stresses was analyzed. The quantitative analysis of GUS activity in transgenic plants showed that Athb-12 gene expression level is in proportion to the length of the promoter. A yeast two-hybrid test revealed that the HD-Zip of Athb-12 does not form a dimer with itself in yeast. The results shown in this study verified that the Athb-12 gene may participate signaling pathway of water stress mediated by ABA.
애기장대의 Athb-12 유전자에 대한 불활성 돌연변이의 선발

애기장대의 homeobox gene인 Athb-12는 ABA에 의해 발현이 유도되는 것이 보고되어 water stress에 중요한 역할을 할 것으로 예상되었다. 이 유전자의 기능을 연구하기 위해, knockout mutants를 찾고자 하였다. T-DNA insertion에 의한 불활성화 돌연변이들의 pools을 이용하여 polymerase chain reaction(PCR)에 의해 pools을 screen하였다. 1차 screen에서 정확하게 Athb-12유전자에 insertion된 돌연변이체를 Southern blots 및 DNA sequencing으로 확인하였고, 이들을 2차 screen에서도 성공적으로 맞는 mutants를 선발하였다. 최종적으로 mutant를 찾아내면 Athb-12의 기능연구에 유용하리라 예상된다. Sine the Athb-12 gene, a homeobox gene from Arabidopsis, was reported to be expressed by ABA induction, it was expected to be important in plant response mechanism to water stress. Screening for knockout mutant for the gene was performed to study the function of the gene. Polymerase Chain Reactions(PCR) were used for screening pools of T-DNA inserted inactivated mutants. From the primary screening, the exact inserted mutant at the Athb-12 gene was found and confirmed by Southern blots and DNA sequencing. The correct mutant was successfully selected from the secondary screening, too. Finding the final insertion mutant will be useful in studying the function of Athb-12.
