      • Mechanistic Analysis of Taxol-induced Multidrug Resistance in an Ovarian Cancer Cell Line

        Objectives: To establish a taxol-resistant cell line of human ovarian carcinoma (A2780/Taxol) and investigate its biological features. Methods: The drug-resistant cell line (A2780/Taxol) was established by continuous stepwise selection with increasing concentrations of Taxol. Cell morphology was assessed by microscopy and growth curves were generated with in vitro and in vivo tumor xenograft models. With rhodamine123 (Rh123) assays, cell cycle distribution and the apoptotic rate were analyzed by flow cytometry (FCM). Drug resistance-related and signal associated proteins, including P-gp, MRPs, caveolin-1, PKC-${\alpha}$, Akt, ERK1/2, were detected by Western blotting. Results: A2780/Taxol cells were established with stable resistance to taxol. The drug resistance index (RI) was 430.7. Cross-resistance to other drugs was also shown, but there was no significant change to radioresistance. Compared with parental cells, A2780/Taxol cells were significantly heteromorphous, with a significant delay in population doubling time and reduced uptake of Rh123 (p<0.01). In vivo, tumor take by A2780 cells was 80%, and tumor volume increased gradually. In contrast, with A2780/Taxol cells in xenograft models there was no tumor development. FCM analysis revealed that A2780/Taxol cells had a higher percentage of G0/G1 and lower S phase, but no changes of G2 phase and the apoptosis rate. Expression of P-gp, MRP1, MRP2, BCRP, LRP, caveolin-1, PKC-${\alpha}$, Phospho-ERK1/2 and Phospho-JNK protein was significantly up-regulated, while Akt and p38 MARK protein expression was not changed in A2780/Taxol cells. Conclusion: The A2780/Taxol cell line is an ideal model to investigate the mechanism of muti-drug resistance related to overexpression of drug-resistance associated proteins and activation of the PKC-${\alpha}/ERK$ (JNK) signaling pathway.

        It is of great significance to study how to assess the railway tunnel damage loss quickly after earthquake for the earthquake relief and post-disaster reconstruction work. Hierarchical weighted synthesis method is the theoretical foundation of the system of earthquake disaster loss assessment. The system is based on windows operation system and VB language is selected as integrated development environment. Access is as external database, the automotive conversion of the earthquake damage reduction value without manual entering and automotive computing of Loss Assessment will be achieved. The computing outcome will be saved in excel. The software can provide scientific evidence and improve the efficiency of the loss assessment.

        Synthesis, Crystal Structure and Quantum Chemistry of a Novel Schiff Base N-(2,4-Dinitro-phenyl)-N'-(1-phenyl-ethylidene)-hydrazine

        A novel Schiff base N-(2,4-dinitro-phenyl)-N'-(1-phenyl-ethylidene)-hydrazine has been synthesized and structurally characterized by X-ray single crystal diffraction, elemental analysis, IR spectra and UV-vis spectrum. The crystal belongs to monoclinic with space group P21/n. The molecules are connected via intermolecular O-H···O hydrogen bonds into 1D infinite chains. The crystal structure is consolidated by the intramolecular N-H···O hydrogen bonds. weak intermolecular C-H···O hydrogen bonds link the molecules into intriguing 3D framework. Furthermore, Density functional theory (DFT) calculations of the structure, stabilities, orbital energies, composition characteristics of some frontier molecular orbitals and Mulliken charge distributions of the title compound were performed by means of Gaussian 03W package and taking B3LYP/6-31G(d) basis set. The time-dependent DFT calculations have been employed to calculate the electronic spectrum of the title compound, and the UV-vis spectra has been discussed on this basis. The results show that DFT method at B3LYP/6-31G(d) level can well reproduce the structure of the title compound.

        Two Androstane Derivatives from the Cultures of Fungus Marasmiellus ramealis (Bull.) Singer

        A new androstane derivative, 4β-methyl-15-oxa-14β-androstane-7-ene-4α-carboxylic acid (1) and a known one 4β-methyl-15-oxa-14β-androstane-7-ene-4α-hydroxyl (2) were isolated from the EtOAc extract of the cultures of the fungus Marasmiellus ramealis (Bull.) Singer. Their structures were elucidated on the basis of 1D and 2D NMR as well as MS spectroscopic data analysis. The inhibitory activity of two isolates against acetylcholinesterase (AChE) revealed that compound 1 exhibited definitely inhibitory activity.

        A remote long-term and high-frequency wind measurement system: design, comparison and field testing

        The wind field measurement of severe winds such as hurricanes (or typhoons), thunderstorm downbursts and other gales is important issue in wind engineering community, both for the construction and health monitoring of the wind-sensitive structures. Although several wireless data transmission systems have been available for the wind field measurement, most of them are not specially designed for the wind data measurement in structural wind engineering. Therefore, the field collection is still dominant in the field of structural wind engineering at present, especially for the measurement of the long-term and high-frequency wind speed data. In this study, for remote wind field measurement, a novel wireless long-term and high-frequency wind data acquisition system with the functions such as remote control and data compression is developed. The system structure and the collector are firstly presented. Subsequently, main functions of the collector are introduced. Also novel functions of the system and the comparison with existing systems are presented. Furthermore, the performance of this system is evaluated. In addition to as the wireless transmission for wind data and hardware integration for the collector, the developed system possesses a few novel features, such as the modification of wind data collection parameters by the remote control, the remarkable data compression before the data wireless transmission and monitoring the data collection by the cell phone application. It can be expected that this system would have wide applications in wind, meteorological and other communities.

        Production of Palm Oil-based Diacylglycerol Using Lecitase Ultra-catalyzed Glycerolysis and Molecular Distillation

        Diacylglycerol (DAG) was prepared viaglycerolysis of palm oil catalyzed by Lecitase Ultra (LU),a novel phospholipase from the fusion of lipase genes fromThermomyces lanuginose and phospholipase genes fromFusarium oxysporum. Glycerolysis was performed in asolvent-free system. The optimized reaction conditionswere: a glycerol/palm oil mole ratio of 7.5:1, initialsubstrate water content of 5%, substrate enzyme load of2%, reaction temperature of 40oC, and reaction time of 8 h. In a scale-up reaction, a DAG content of 59.5% in the lipidlayer was achieved. Through a two-step molecular distillation,the composition of the target product was 88.1% DAG,2.8% TAG, 9.0% MAG, and 0.1% FFA. The fatty acidcomposition of the DAG oil, determined using GC-MS,was enriched compared with the original palm oil.

        From Is to Ought : A Comparative Study on the Neo-Confucian Concept of ren and Care Ethics

        Most care ethicists deny that care has an abstract philosophical ground, and believe that the moral value of care is derived from our daily caring activities. But as a normative theory that attempts to replace the mainstream moral theory, simply describing the phenomenon of care from the third person cannot answer the question of why we “ought” to be caring. The Confucian concept of ren is considered to be compatible with care ethics in many aspects. Especially the moral ontology of ren in Neo-Confucianism which embodied in the thought of the unity of tian1) and human beings is a different kind of relational ontology from that in care ethics which can inspire care ethicists to overcome the theoretical gap between “is” and “ought”.

        RUNX1 Ameliorates Rheumatoid Arthritis Progression through Epigenetic Inhibition of LRRC15

        Leucine-rich repeat containing 15 (LRRC15) has been identified as a contributing factor for cartilage damage in osteoarthritis; however, its involvement in rheumatoid arthritis (RA) and the underlying mechanisms have not been well characterized. The purpose of this study was to explore the function of LRRC15 in RA-associated fibroblastlike synoviocytes (RA-FLS) and in mice with collagen-induced arthritis (CIA) and to dissect the epigenetic mechanisms involved. LRRC15 was overexpressed in the synovial tissues of patients with RA, and LRRC15 overexpression was associated with increased proliferative, migratory, invasive, and angiogenic capacities of RA-FLS and accelerated release of pro-inflammatory cytokines. LRRC15 knockdown significantly inhibited synovial proliferation and reduced bone invasion and destruction in CIA mice. Runt-related transcription factor 1 (RUNX1) transcriptionally represses LRRC15 by binding to core-binding factor subunit beta (CBF-β). Overexpression of RUNX1 significantly inhibited the invasive phenotype of RA-FLS and suppressed the expression of proinflammatory cytokines. Conversely, the effects of RUNX1 were significantly reversed after overexpression of LRRC15 or inhibition of RUNX1-CBF-β interactions. Therefore, we demonstrated that RUNX1-mediated transcriptional repression of LRRC15 inhibited the development of RA, which may have therapeutic effects for RA patients.
