        Quartz 자극에 의한 iNOS 발현의 자가조절

        목적 : Rat2 섬유모세포에서 α-quartz에 의해 유도된 NOS의 조절기전을 알아보기 위하여 본 연구가시도 되었다. 방법 : Rat2 세포에 있어서 α-quartz 자극에 의한 NO와 H₂O₂의 분비 및 iNOS의 발현을 관찰하였고, iNOS 억제제인 L-NIL과 H₂O₂ 억제제인Catalase를 전처리하고 마찬가지로 NO와 H₂O₂분비 그리고 iNOS의 단백발현 정도를 western blot-ting을 이용하여 관찰하였다. 결과 : o-quartz는 Rat2 세포에서 iNOS에 의한NO와 H₂O₂의 분비를 유도하였다. L-NIL은 R3t2세포에서 D-quartz에 의한 H₂O₂의 분비와 iNOS발현을 완전히 억제하였다. Catalase로 전처리 하였을 때 o-quartz로 유도된 iNOS에 의하여 생산증가된 H2Oa가 iNOS를 억제하는 autoinhibitorypathway가 차단되어 결과적으로 H₂O₂의 분비와 iNOS의 발현이 증가하였다. 결론 : Rat2 세포에서 α-quartz에 의하여 유도된iNOS는 H₂O₂의 분비를 촉진하고 이 H₂O₂는 iNOS기능을 조절하는 자가조절기전에 중요한 역할을 하는 것으로 생각된다. Objectives : This study was performed in order to investigate the molecular mechanism regulating nitric oxide synthase (NOS) induced by α-quartz in Rat2 fibroblast. Methods : α-quartz-induced nitric Oxide (NO) anti H₂O₂ formation anti α-quarts-induced iNOS protein expression in Rat2 fibroblast were monitored. With iNOS inhibitor (L-N6- (1-iminoethyl) lysine hydrochloride, L-NIL) or antioxidant (catalase), we observed NO and H₂O₂ formation and iNOS protein expression in Rat2 fibroblast stimulated with α-quartz. Results : α-quartz stimulated iNOS-induced NO and H₂O₂ formation in Rat2 fibroblast. L-NIL inhibited H₂O₂ formation and iNOS protein expression by α-quartz in Rat2 fibroblast. Pretreatment with catalase blocked the autoinhibitory pathway of iNOS by iNOS-induced H₂O₂ therefore H₂O₂ and NO production and iNOS protein expression were increased in Rat2 fibrobalst stimulated with α-quartz Conclusion : α-quartz-induced iNOS Stimulated H₂O₂ formation in Rat2 fibroblast. INOS-induced H₂O₂ by α-quartz plays an important role in the autoinhibition pathway for regulating the iNOS function in Rat2 fibroblast

        라오스의 벼 생산 현황과 문제점

        Rice is the single most important crop in Lao PDR. In the 2000 rice production year, total rice production in the Laos reached 2.2 million tons. This is regarded as sufficient to provide national self-sufficiency. The rice harvested area in 2000 was approximately 719,500ha and represented 80% of cropped land area. Approximately 82% of production was from wet season cropped and 18% was from the irrigated dry season. Grain yield per unit area vary from 1.68 to 4.39 ton/ha in the wet season environment. However, the highest grain yield (4.39 ton/ha) obtained from the dry season irrigated crop. Higher yields and reduced year variation in production can be expected with the further intensification of production systems in the low land environments. However, further imporvements in production will be dependent on higher levels of inputs and further alleviation of some the production constraints.

      • 실리카흄을 흔입한 고강도 콘크리트 개발

        콘크리트 구조물이 대형화됨에 따라 설계단면이 증대되어 상대적으로 많은 경제적 손실을 부담하게 된다. 따라서 단면을 감소시키면서도 소요의 하중에 안전한 구조물을 건설하기 위해서는 우선적으로 구조물 건설에 기초가 되는 고품질, 고내구성의 고강도 콘크리트 개발이 절실히 요구된다 본 연구에서는 일정 시멘트비 및 혼화재 비율하에서 소요의 워커빌리티가 확보되는 고강도 콘크리트를 시간과 온도의 변화에 따라 증기 양생하여, 압축, 인장, 휨강도 뿐만 아니라 파괴특성을 실험적으로 연구하였다. 시료 제작시 시멘트 입자 사이의 공극 및 불연속 영역을 충전하여 고밀도화하기 위한 콘크리트용 혼화재로 시멘트 비표면적이 상당히 작은 초미립 분말인 실리카흄을 이용하였다. 또한 AE감수제 및 고성능 유동화제를 사용하여 혼화재의 첨가에 의하여 발생될 수 있는 워커빌리티의 감소를 방지하였다. 실험결과에 의하면 일정 양생 지속 시간하에서 온도의 증가에 따라 콘크리트의 압축, 인장 및 휨강도가 전반적으로 증가하였다. 동일하게 일정 온도하에서 양생 지속시간이 커짐에 따라 강도들이 증가됨을 알 수 있었다. 따라서 상대적으로 높은 온도와 긴 지속 양생 조건하에서 파괴에 대한 저항력이 크게 나타났다. The compressible, tensible, and flexibleresistance of the high strength concrete is analyzed by the experimentation in the present study. For the test, we cure several samples with the silica fume as a mixture being become dense the spaces between the particle of cement under the variation of both the temperature and the curing-interval. Then, the superplasticizer and the ezcon are also used to satisfy the required workability for construction. The compressible, tensible, and flexible resistances to a stress are increased as increasing the temperature and the time interval for the curing. Therefore it is concluded that the overal fractural and mechanical properties is improved by mixing the silica fume into the cement.

      • 녹동균 세포외막 단백질 백신 CFC-1-101의 안정성 및 면역원성 검토 : 임상 제 Ⅰ/Ⅱa상 시험

        목적 : 제일제당에서는 녹농균의 세포외막 단밸질을 유효성분으로 하는 백신인 CFC-101을 개발하였으며, 동물시험에서 이 백신의 안전성과 유효성을 입증하였다. 본 연구에서는 이 녹농균 백신의 인체에 대한 안전성과 면역원성을 평가하는 동시에 인체 접종시의 최적 투여 용량을 결정하기 위하여 제 I/Ⅱa상 임상시험을 수행하였다. 방법 : 건강한 성인 남자를 피험자로 선별하여 각 용량군에 백신투여자 6명, 위약투여자 2명을 배정하였다. 백신 투여군은 0.25mg, 0.5mg 또는 1.0mg 용량의 녹농균 백신을 7일 간격으로 3회에 걸쳐 근육주사 하였으며, 위약 투여군에게는 세포외막 단백질을 제외한 동일한 성분을 투여하였다. 백신접종 후 국소적 또는 전신적인 반응의 발생여부를 관찰하고, 혈액시료를 체취하여 백신의 역가와 유효성을 검정하였다. 결과 : 녹농균 백신 CFC-101은 모든 접종자에서 양호한 내약성을 보였다. 또한 0.5mg 과 1.0mg 백신 투여군에서는 100%의 항체양전율을 나타내었다. 생성된 항체는 녹농균 세포외막단백질에 특이성을 보였고, 녹농균 감염에 대해 방어효능이 있었다. 결론 : 이와같은 결과로부터 이 녹농균 백신은 인체에 안전하게 투여할 수 있으며, 높은 항체 생성능으로 감염방어 효능을 보이고 0.5mg과 1.0mg이 최적용량인 것으로 판단되었다. Background : We developed a Pseudomonas aeruginosa outer membrane protein(OMP) vaccine CFC-101, and the prophylactic efficacy of which has been demonstrated in animal models. In order to evaluate the safety and immunogenicity of the P. aeruginosa vaccine, we carried out a phase I/Ⅱa clinical trial in healthy male volunteers. Methods : Groups of eight volunteers, including two placebo subjects, were vaccinated intramuscularly with three doses of 0.25, 0.5 or 1.0 mg of the vaccine at one week intervals. Sings of systemic and local reactions observed after vaccination were recorded for each vaccinee for 5 days. Physical examinations were performed on days 0, 1, 7, 8, 14, 15, 21, and 42, and clinical laboratory tests were done on days 0, 3, and 21. Blood samples for assay of serum antibody levels were obtained up to 42 days after the first vaccination. Results : The vaccine was generally well tolerated by all vaccinees, showing no significant side effects. In the three dosage groups, all vaccinees, except one receiving the 0.25 mg dose, showed significant elevation in serum IgG antibody titers against the vaccine proteins, indicating 100% seroconversion in 0.5 and 1.0 mg groups. The human antibodies induced by the vaccine were specific for P. aeruginosa OMPs, as confirmed by western blot analysis and immunoprecipitation assays. The capacity of the human antisera to enhance opsonophagocytic killing activity by polymorphonuclear leukocytes and to confer protection against P. aeruginosa infections indicates that the antibodies elicited by the vaccine have protective efficacy. Conclusion : We conclude that the P. aeruginosa OMP vaccine is safe and effective for human use and its optimal dose to be 0.5 or 1.0 mg.

        Background: Learning portfolio is a collection of evidence that learning has taken place. It has gained its reputation asa useful assessment tool in the education of health professionals. The purpose of this study is to describe the pharmacystudents` experiences and perceptions upon the introduction of a learning portfolio into the Introductory Pharmacy PracticeExperience course. Methods: Fifty five students from one pharmacy school who used a learning portfolio to documenttheir progress in the IPPE course participated in 16-item questionnaire exploring opinions and experiences oflearning portfolio preparation, assessment, and personal and professional development and reflection. Results: Most studentsagree that a learning portfolio is a valuable tool in promoting self-directed and reflective learning. However mostof them (46/55) also feel developing a portfolio is time-consuming, and when compared to their effort, an appropriatereward has not been given. Conclusion: To make the use of learning portfolios successful students should receive clearguidelines on their purpose, content and structure. Also the assessment criteria should be provided before the introductionof learning portfolio and their effort in developing learning portfolio should be rewarded.

      • The co-expression pattern of p63 and HDAC1: a potential way to disclose stem cells in interfollicular epidermis

        Background: Stem cell markers of interfollicular epidermis (IEF) have not been established thus far. Objectives: The aim of this study is to suggest a new way to disclose IFE-stem cells by combining the expression of histone deacetylases (HDAC) 1 and p63. Methods: Immunohistochemical staining of HDAC1 and p63 was performed in six normal human samples. Moreover, a skin equivalent (SE) model was treated with suberoylanilohydroxamic acid (SAHA, a HDAC inhibitor) to elucidate the role of HDAC1. Finally, rapidly adhering (RA) keratinocytes to type IV collagen, which have been identified to represent epidermal stem cells, were subjected to Western blot analysis with antibodies against HDAC1. Results: In normal samples, there was a minor subpopulation comprised of p63-positive and HDAC1-negative cells in the basal layers. The proportion of this subpopulation was decreased with age. In the SE model, SAHA treatment increased the epidermal thickness and number of p63-positive cells in a dose dependent manner. After SAHA treatment, the expression of differentiation markers was decreased, while that of basement membrane markers was increased. In a Western blot analysis, HDAC1 was not expressed in RA cells. Conclusion: The combination of p63-positive and HDAC1-negative expressions can be a potential new way for distinguishing epidermal stem cells.

      • Hypothyroid states mitigate the diabetes-induced reduction of calbindin D-28k, calretinin, and parvalbumin immunoreactivity in type 2 diabetic rats.

        <P>In this study, we investigated the differences in calbindin D-28k (CB), calretinin, (CR) and parvalbumin (PV) immunoreactivity in the hippocampus of Zucker diabetic fatty (ZDF) rats and Zucker lean control (ZLC) rats. In addition, we observed the effects of hypothyroidism on the levels of immunoreactivity of these proteins in ZDF rats. For this study, 7-week-old ZDF rats were used, and methimazole treatment was continued for 5 weeks to induce hypothyroidism. The animals were sacrificed at 12 weeks of age. ZDF rats showed increased blood glucose levels compared to those in ZLC rats. Methimazole intervention significantly reduced total and free T3 levels, and it ameliorated the increase of blood glucose levels in ZDF rats. In ZLC rats, CB, CR, and PV immunoreactivity was detected in regions of the hippocampus proper. In vehicle-treated ZDF rats, CB, CR, and PV immunoreactivity was significantly decreased in the hippocampus. However, in the methimazole-treated rats, CB, CR, and PV immunoreactivity was significantly increased compared to that in the vehicle-treated rats. These results suggest that hypothyroidism ameliorated the diabetes-induced reduction of CB, CR, and PV immunoreactivity in the hippocampus.</P>

      • Detailed Nodal Features of Cervical Tuberculous Lymphadenitis on Serial Neck Computed Tomography Before and After Chemotherapy: Focus on the Relation Between Clinical Outcomes and Computed Tomography Features

        OBJECTIVE:: To investigate the relation between clinical outcomes and nodal features on computed tomography (CT) in cervical tuberculous lymphadenitis (CTBL) before and after antituberculous chemotherapy. METHODS:: Fifty-six patients with CTBL underwent CT before and after a 6- or 12-month course of standard chemotherapy. Three radiologists evaluated the nodal features on serial CT retrospectively, including calcification (no/punctuate/large), necrosis (no/eccentric/central), perinodal infiltrations (no/localized/extensive), and enhancing patterns (no/peripheral/homogeneous). The clinical outcome was defined as “favorable” (n = 33) or “unfavorable” (n = 23) at the completion of chemotherapy. RESULTS:: All the features on the initial CT scan did not show a statistically significant difference between the favorable and unfavorable groups. On the final CT scan, absence of necrosis (P < 0.005), no infiltration (P < 0.005), no enhancement (P < 0.008), and central enhancement (P < 0.014) were more common in the favorable group, whereas large necrosis (P < 0.005), localized and extensive infiltration (P = 0.005, P < 0.005), and peripheral enhancement were more common in the unfavorable group (P < 0.005, P = 0.009). CONCLUSIONS:: Central necrosis, perinodal infiltration, and peripheral rim enhancement on the final CT scan showed differences between the 2 groups. These CT features reflecting inflammation can be useful findings for assessing treatment response.

      • Effects of long-term exposure to aluminum in the hippocampus in the type 2 diabetes model rats

        <P>We investigated the long-term effects of aluminum (Al) exposure in the hippocampus in Zucker diabetic fatty (ZDF) rats and Zucker lean control (ZLC) rats. Six-week-old ZLC and ZDF rats were randomly divided into Al- and non-Al-groups. They were sacrificed 27 weeks after Al exposure (2000 ppm) through drinking water. Al exposure did not affect physiological parameters such as the body weight and blood glucose levels, but the prolonged diabetic condition had significant effects on the body weight and blood glucose levels. To determine the effects of diabetes and Al exposure on the neural plasticity and inflammatory response in the hippocampus, we examined the levels of doublecortin (DCX), <I>N</I>-methyl-d-aspartate receptors (NMDAR1, NMDAR2A, and NMDAR2B), and ionized calcium-binding adapter molecule 1 (Iba-1) in the hippocampus. DCX immunohistochemical staining revealed that Al exposure significantly reduced neuronal differentiation in both ZLC and ZDF rats. In particular, ZDF rats showed significantly decreased DCX immunoreactive neuroblasts compared with ZLC rats after aluminum exposure. In contrast, the expression of postsynaptic NMDARs was altered only in ZDF-Al rats; the protein expression level of NMDAR1 was reduced, but that of NMDAR2B increased in the hippocampus. Iba-1-immunoreactive microglia with morphological changes, including increased cytoplasm and retracted processes, were detected in the long-term diabetic condition and in the case of the co-existence of diabetes and Al exposure. Al exposure aggravated the diabetes-induced reduction of neuroblast differentiation and NMDAR signaling and facilitated the morphological changes associated with inflammatory activation in microglia in the hippocampus. However, further studies are still needed to confirm these findings.</P>
