      • Detection of Nosema ceranae from Bombus terrestris via Quantitative Real-Time PCR

        The bumblebee, Bombus terrestris, has played an important role as one of the alternative pollinators since the outbreak of honeybee collapse disorder. Recently, pathogens and parasites such as viruses, bacteria and mites, which affect the life span and fecundity of their host, have been discovered in B. terristris. In order to detect the microsporidian pathogen, Nosema Spp. in the field populations of B. terristris, we collected adults and isolated their genomic DNA for diagnostic PCR. The PCR primers specific for Nosema Spp. were newly designed and applied to gene amplification for cloning. Only small subunit ribosomal RNA(SSU rRNA) gene of N. ceranae was successfully amplified and sequenced among examined genes, which indicates that N. ceranae mainly infects the examined field population of B. terristris. To detect of SSU rRNA gene, two regions of SSU rRNA gene were selected by primary PCR analysis and further analyzed in quantitative real-time PCR(qRT-PCR). The qRT-PCR analysis demonstrated that SSU rRNA of N. ceranae was detected at concentrations as low as 0.85 ng/μl genomic DNA. This result suggests that the detection via qRT-PCR can be applied for the rapid and sensitive diagnosis of N. ceranae infection in the field population as well as risk assessment of B. terristris.

        골 어(La langue gauloise)와 골(Gaule) 주변 언어 간 혼효 현상 연구

        Cette étude, qui trouve place dans une diversité des inscriptions de la langue gauloise visant à révéler le contact des langues voisines dans l’antiquité et le haut moyen âge, tend à construire une cohérence entre, d’une part, la théorie de l’histoire de la langue gauloise, qui a connu différents aléas(L. Bonfante et G. Dottin) mais continue de s’avérer fécond et, d’autre part, la pratique des corpus des inscriptions dans la recherche archéologique(J. Pirson, M. Bat, J. De Hoz, M. Lejeune). Le contact des langues en Gaule élaboré par la guerre et la conquête implique de mettre en relation entre l’obstacle de la mer, spécialement la Méditerranée et de la terre. L’un des objectifs de cet article viserait alors la notion du contact des langues accompli par la conquête romaine et la promotion politico-administrative mais également par le contact réciproque dans le cadre économique, académique, culturel et religieux. Un deuxième objectif, plus contextuel, consisterait à tenter de classer ces phénomènes de contacts des langues et des cultures en Gaule, particulièrement en Gaule narbonnaise. La Gaule narbonnaise, proche de la Méditerranée, et occupée en plusieurs contacts des langues voisines dans l’antiquité et le haut moyen âge, a livré des vestiges écrits différents: une inscription gallo-grecque, une inscription gallo-romaine et les mots d’origine hébreu et germanique. Il est probable que plusieurs modèles linguistiques, différentes typologies des contacts de langues peuvent être élaborés, les objectifs visés et leurs zones de la promotion linguistique vers La Gaule dans le cadre du vulgarisme, l’archaïsme et la barbarisme. Le troisième objectif serait d’examiner l’influence de l’hébreu comme le grec antique en Gaule pour la diver sité des contacts des langues. En raison de la diversité des inscriptions, des réflexions sur le contact des langues en Gaule dans l’antiquité et le haut moyen âge sont amenées à se mettre en question à travers la guerre, la conquête et aussi la raison plus pacifique. Ces analyses nous montrent que le contact des langues pourrait être influencé par la Mer comme la Terre.

        Porphyromonas gingivalis 열충격 단백으로 면역한 백서에서의 치조골 파괴의 감소

        The present study has been performed to evaluate Porphyromonas gingivalis (P.gingivalis) heat shock protein(HSP)60 as a candidate vaccine to inhibit multiple bacteria-induced alveolar bone loss. Rats were immunized with P.gingivalis HSP60 and experimental alveolar bone loss was induced by infection with multiple periodonto -pathogenic bacteria. Post-immune rat anti-P.gingivalis HSP IgG levels were significantly elevated and have demonstrated highly significant inverse relationship with the amount of alveolar bone loss induced by multiple bacteria. Results from PCR detection of subgingival bacterial plaque indicated that the vaccine successfully eradicated the multiple pathogenic species. We concluded that P.gingivalis HSP60 could potentially be developed as a vaccine to inhibit periodontal disease induced by multiple pathogenic bacteria.

        Porphyromonas gingivali의 열충격단백-특이성 단클론항체의 개발

        Heat shock protein (HSP) is one of cellular protein commonly present in major periodontopathogenic bacteria as well as mammalian cells. The protein may play a role in the immunopathogenesis by modulating autoimmune reaction due to its high level of sequence homology between bacteria and human counterpart. Hence, identifying immunodomiant epitope of bacteria HSP that is cross-reactive to periodontopathogenic bacteria with a specificity to human HSP may comprise a critical strategy for development of a periodontal vaccine. The present study was performed to establish clones producing monoclonal antibody reactive to Porphyromonas gingivalis (p. gingivalis) HSP with a specificity to human HSP. 4 different hybridomas were cloned producing monoclonal IgG antibodies to P, gingivalis HSP and evaluated for their reactivity and specificity to other periodontopathogenic bacteria as well as to human HSP. These four monoclonal antibodies reacted with p. gingivalis HSP only with specificities to other bacteria tested and human HSP as well. The antigenic epitopes producing the 4 monoclonal antibody may be potentially developed as vaccine candidates. Further investigations are under way to identify more clones producing monoclonal antibodies reactive to P, gingivalis HSP and to other periodontopathogenic bacteria as well, while maintaining specificities to human counterpart.

        Pseudomembranous colitis is mainly caused by antibiotics and Clostridium difficile infection. But conditions such as gastrointestinal surgery, antacid medication, anti-neoplastic agent or immunosuppressive agent which influences the normal flora of colon can induce colitis without the administration of any antibiotics. We experienced a 13 year-old male who was taking low-dose methotrexate for juvenile rheumatoid arthritis complained diarrhea and abdominal pain for 3 weeks. Sigmoidoscopic findings revealed diffuse patch yellowish pseudomembranes on the rectum. Histologic finding was compatible to pseudomembranous colitis. His symptom was improved after stop taking methotrexate and the administration of metronidazole. If a patient treated with immunosuppressive agents or antineoplastic agents complains diarrhea, fever or abdominal pain and has not improved with conservative care, pseudomembranous colitis should be taken into account as a differential diagnosis and prompt treatment is required for better prognosis.         쇠미역사촌과 미역 포자체의 배양 및 야외 개체군의 생장 비교

        The effects of temperature, light, and salinity on the growth of Costaria costata and Undaria pinnatifida juveniles were examined in laboratory cultures. In a cultivation farm, the monthly yield and density were also investigated between December and April for C. costata and between December and March in 2007 and 2008 for U. pinnatifida. The relative growth rates (RGRs) were greater at 20~60 μmol photons m-2s-1 than at low (0~10 μmol photons m-2s-1) and high (100~180 μmol photons m-2s-l) irradiance levels. The optimal growth conditions for the two species were 17℃, 35 psu, 60 μmol photons m-2s-1, and a daylength of 12 h, indicating that C. costata and U. pinnatifida have very similar growth responses to temperature, light, and salinity. However, the growth responses of the two species to various environmental factors were different; C. costata grew faster than U. pinnatifida but the latter species grew well at low salinity. The monthly yield of C. costata and U. pinnatifida increased steadily over the study period, and it was maximal in March for both species, but the yield of U. pinnatifida was greater than that of C. costata.
