        Atypical parathyroid tumor: clinical and parathyroid hormone response to surgical treatment

        Purpose: Primary hyperparathyroidism (PHPT) is caused by typical adenoma (TA), multiglandular disease (MD), or parathyroid carcinoma (PC), and in a smaller percentage of cases by atypical parathyroid tumor (APT). The objective of this study is the retrospective analysis of clinical features and parathyroid hormone (PTH)/calcium response to surgery in patients who underwent parathyroidectomy for symptomatic PHPT with histological evidence of APT. Methods: We retrospectively reviewed our institutional experience in the management of PHPT from January 2016 to December 2021 focusing on those patients presenting APTs. We analyzed the clinical features of this disease and PTH/ calcium response to surgical treatment in APTs compared to the other pathological conditions causing PHPT. Results: In a cohort of 125 patients with PHPT we found 112 TAs (89.6%), 6 APTs (4.8%), 6 PCs (4.8%), and only 1 MD (0.8%). APTs in comparison to other parathyroid diseases showed peculiar features such as adhesion to the surrounding structures and a frequent intrathyroidal location, which may justify thyroid loboistmectomy adopted in most of the observed cases. APTs showed significantly higher preoperative PTH values compared to TA + MD and were relevant to PC. Conclusion: Due to its rarity, there is a lack of specific indications in the management of APTs. Biochemical features observed in APT and PC can be related to similar biological behavior. However, some specific features observed preoperatively in some cases of PHPT might suggest presence of an APT, which could be helpful mostly in surgical and postoperative management. Further studies are required to confirm the results of the present preliminary report.

      • Hytramycins V and I, Anti-<i>Mycobacterium tuberculosis</i> Hexapeptides from a <i>Streptomyces hygroscopicus</i> Strain

        <P>Thirty-five thousand actinomycete extracts were screened for anti-<I>Mycobacterium tuberculosis</I> (<I>M. tb</I>) activity, followed by C<SUB>18</SUB> cartridge fractionation of 37 prioritized extracts. Based on MICs against replicating and nonreplicating <I>M. tb</I>, and IC<SUB>50</SUB> values against Vero cells to generate selectivity indices, seven fractions from seven different strains were selected for further examination. When cultured in G.S.S. media and extracted with ethyl acetate, the <I>Streptomyces hygroscopicus</I> strain ECUM 14046 yielded an extract with promising anti-<I>M. tb</I> activity and a well-defined chromatographic profile. Fractionation by preparative HPLC and subsequent structure elucidation of two active fractions using 1D- and 2D-NMR and MS methods revealed the presence of two cyclohexapeptides, hytramycins V and I, each containing three unusual piperazic acid moieties. The use of <SUP>1</SUP>H iterative full spin analysis (HiFSA) on both hytramycins confirmed that quantum mechanics-simulated spectra match the experimental data, and all <I>J</I><SUB>H,H</SUB> and δ<SUB>H</SUB> values are consistent with the proposed structures. The absolute configuration of each amino acid moiety was determined by Marfey’s method. The MICs against replicating and, more importantly, nonreplicating <I>M. tb</I> fall into the range of some existing second-line anti-TB drugs, such as streptomycin and capreomycin, respectively.         Differential Effects of Typical and Atypical Neuroleptics on Mitochondrial Function In Vitro

        A series of typical (chlorpromazine, haloperidol and thioridazine) and atypical (risperidone, quetiapine, clozapine and olanzapine) antipsychotics were tested for effects on integrated bioenergetic functions of isolated rat liver mitochondria. Polarographic measurement of oxygen consumption in freshly isolated mitochondria showed that electron transfer activity at respiratory complex I is inhibited by chlorpromazine, haloperidol, risperidone, and quetiapine, but not by clozapine, olanzapine, or thioridazine. Chlorpromazine and thioridazine act as modest uncouplers of oxidative phosphorylation. The typical neuroleptics inhibited NADH-coenzyme Q reductase in freeze-thawed mitochondria, which is a direct measure of complex I enzyme activity. The inhibition of NADH-coenzyme Q reductase activity by the atypicals risperidone and quetiapine was 2-4 fold less than that for the typical neuroleptics. Clozapine and olanzapine had only slight effects on NADH-coenzyme Q reductase activity, even at 200 $\mu$ M. The relative potencies of these neuroleptic drugs as inhibitors of mitochondrial bioenergetic function is similar to their relative potencies as risk factors in the reported incidence of extrapyramidal symptoms, including tardive dyskinesia (TD). This suggests that compromised bioenergetic function may be involved in the cellular pathology underlying TD.

        Estimation of Genetic Parameters of Some Productive and Reproductive Traits in Italian Buffalo. Genetic Evaluation with BLUP-Animal Model

        In this study, the Italian milk recorded buffalo population from 1974 to 1996 was analysed with the purpose to estimate genetic and environmental variability and provide genetic parameters for the most important economic traits. High variability between herds was evident due to the poor knowledge of feeding requirements and husbandry technology in this species compared to cattle. Age at first calving was reduced by 57 days during the considered years following efforts made in better feeding and management from 1990; on the contrary, calving interval has increased by 17 days as a consequence of forcing buffaloes to calve in spring, in order to have the peak milk yield when milk is much better paid. Average milk yield increased by 1853 kg during these years, while lactation duration was reduced by 30 days. Season of calving has no effect on all traits. Calving order has a positive effect on milk yield especially because older cows produce more milk in shorter lactations. Heritability for the age at first calving and calving interval was 0.26 and 0.05 respectively. Heritability of productive traits, milk yield and duration of the lactation was 0.19 and 0.13 respectively, with repeatabilities of 0.40 and 0.26. Genetic trend for milk yield was 2.1 kg milk/year for the bulls and 1 kg for all population. The high genetic variability of milk production as well as duration of the lactation, indicates that there are good opportunities for genetic improvement when including these traits in a selection scheme. The low genetic trend registered over 15 years of recording activity can be explained by the fact that neither progeny testing was performed or selection schemes were implemented, due to the difficulties to use artificial insemination in buffalo.

        Essential Oil Composition of Teucrium divaricatum Sieb. ssp. villosum (Celak.) Rech. fil. Growing Wild in Lebanon

        Teucrium divaricatum Sieb. ssp. villosum (Celak.) Rech. fil. is a medicinal plant used in Lebanon. The hydrodistillation of the aerial parts of T. divaricatum yielded 0.27% (wt/wt) of essential oil; gas chromatography and gas chromatography/mass spectrometry analysis enabled the identification of 60 compounds representing 94.5% of the total oil. On the whole, the oil was constituted mainly by sesquiterpenes (64.6%), particularly sesquiterpene hydrocarbons (50.0%). The most abundant compounds were (E)-caryophyllene (30.1%) and caryophyllene oxide (6.1%). The in vitro activity of the essential oil against some microorganisms in comparison with gentamicin by the broth dilution method was determined. The oil exhibited a good activity as an inhibitor of growth of Gram-positive bacteria.

        SPECULATION AND MONETARY POLICY BEHAVIOUR IN THE 1992 CURRENCY CRISIS: THE ITALIAN CASE

        The paper proposes an explanation for the 1992 currency crisis as the result of monetary policy behaviour and private agents’ speculation. Our analysis reveals how speculators’ expectations and the behaviour of the monetary policy authority were formed on the widespread beliefs about the future value of income. We show that the real effects of monetary policy measures represent the link between the action of the Central Bank and speculation.

        Increased MHC class I expression on thyrodytes is evident in ATD (4, 21) and has been suggested to be an important causal factor in the development or perpetuation of disease expression (3). The hypothesis has been offered that the elevations result in abnormal presentation of thyroid antigens to immune cells, thereby breaking tolerance, and that this elicits the clonal expansion of autoreactive T cells associated with disease expression (3).

      • COUNTRY IMAGE AS SEGMENTATION TOOL IN THE EMERGING MARKETS: EVIDENCE FROM ITALY

        This paper analyses consumers’ perception of the country image of Italy across 7 emerging countries: Brazil, China, India, Indonesia, Russia, South Africa, and Turkey. Drawing from the international marketing and tourism literature (De Nisco, Papadopoulos and Heslop, 2017; Roth and Diamantopoulos, 2009), 4 main structural dimensions of the Italian country image were chosen as focus of investigation: general country image, product-country image, tourism destination image and cultural heritage image. The empirical research was based on a survey conducted on a sample of 4,550 respondents intercepted through an online panel. Two preliminary exploratory factor analysis (EFA) were ran to refine the scales and to summarize the data into structural factors. The first EFAs grouped measurement scales in 7 components: tourism destination image; general country image; product country image; product accessibility; evaluation of hedonic products; evaluation of utilitarian products; and evaluation of Italy as a tourism destination. The second EFA identified 3 distinct components of the cultural heritage image construct, i) art, literature, history, movies, ii) sociality, cooking, handcrafts and iii) traditions, expression, celebrations. Using the dimensions resulting from the EFA as data input, a latent class analysis was then employed. A 4-cluster solution emerged and the four segments reveal how the general country, product, tourism and cultural heritage components of place image are related and of how they jointly affect consumers’ intentions.         Effect of Annurca Apple Polyphenols on Human HaCaT Keratinocytes Proliferation

        Polyphenols have been demonstrated to have clear antioxidant activities in vitro. However, in complex biological systems, they exhibit additional properties, which are yet poorly understood. The apple is among the most consumed fruits worldwide, and several studies suggest that apple polyphenols could play a role in the prevention of degenerative diseases. The present study aimed at evaluating the Annurca apple polyphenol extract (APE) effects both proliferation and apoptosis on HaCaT cells. The data indicate that apple polyphenolic compounds had significant antiproliferative action on HaCaT cells. The fluorescence-activated cell-sorting analysis showed that APE induced cell apoptosis in a dose-dependent manner. Moreover, apple polyphenols induced apoptosis in epithelial cells by triggering a death receptor-associated extrinsic pathway p53-independent. APE was also capable of inducing morphological changes as evidenced by nuclear condensation. The cellular, morphological, and molecular data unequivocally demonstrated that induction of cellular apoptosis was mainly responsible for the previously observed antiproliferation-induced APE on HaCaT keratinocytes. Our experimental results suggest that apple polyphenols are a promising source from which a natural-based topical agent could be developed for skin diseases treatment.
