        섬유 및 혼화제를 이용한 황토 모르타르의 수축균열 제어

        A total of 8 hwang-toh mortar mixes with no cement were tested to evaluate the effect of different fibers and admixtures on the control of shrinkage cracks of such mortars. The fiber types added were Lyocell, Poly-Amid and PVA, while, CSA and organic polymer were selected as the admixture. The shrinkage crack resistance of hwang-toh mortar were examined from the unrestrained drying shrinkage strain, restrained drying shrinkage and plastic shrinkage. Test results revealed that the unrestrained drying shrinkage strain of hwang-toh mortar sharply increased at an early age showing approximately 2.5 times higher values than predictions obtained from ACI 209, regardless of the addition of fibers and admixtures. In addition, cracks owing to the restrained drying shrinkage developed at the only age of 1.6 days in all test mixes; as a result, the cracking grade of hwang-toh mortar was evaluated to be high. On the other hand, the addition of fibers required a value of fiber spacing above 0.0003 in order to enhance the tensile resistance capacity of hwang-toh mortar under the restrained drying shrinkage and reduce the cracking area under the plastic shrinkage. Overall, it could be concluded that moist curing at an early age and the addition of fiber with fiber spacing above 0.0003 are practically effective in reducing shrinkage crack of hwang-toh mortar.

        Memory Impairment in Estrogen Receptor α Knockout Mice Through Accumulation of Amyloid-β Peptides

        <P>Estrogen has been known to reduce the development of Alzheimer’s disease (AD). However, exact mechanisms are not clear. We investigated whether estrogen can increase amyloid-beta (Aβ) degradation and affects Aβ-induced memory impairment in an estrogen deficiency model. Estrogen receptor alpha (ERα) knockout mice and wild-type mice were intracerebroventricular (ICV) infused with Aβ (300 pmol) for 2 weeks. Cognitive function was then assessed by the Morris water maze test and passive avoidance test. In addition, Western blot analysis, immunostaining, immunofluorescence staining, ELISA, and enzyme activity assays were used to examine the degree of Aβ deposition in the brains of ERα knockout mice. In our present study, Aβ was accumulated more in the ERα knockout mice brain and greatly worsened memory impairment and glial activation as well as neurogenic inflammation.         Immunohistochemical study of the distribution of sodium-dependent vitamin C transporters in adult rat brain

        <P>Sodium-dependent vitamin C transporters (SVCTs) is known to transport the reduced form of ascorbic acid into the cell, whereas the oxidized form of vitamin C (VC) is moved through a facilitative sugar transporter, such as glucose transporter (GLUT). With regard to the distribution of SVCT1 and -2 within the various organs, they were reported to be expressed in different types of cells. Especially in the central nervous system, only SVCT2 mRNA was expressed mainly in neurons and some types of neuroglial cells. However, data on the expression of SVCT proteins in the brain are scant. Therefore, we tried to develop comprehensive data on the distribution of SVCT proteins in adult rat brain by using immunohistochemical techniques for the first time. In our study, SVCT2 immunoreactivities (IRs) were intensely localized in the neurons of cerebral cortex, hippocampus, and Purkinje cells of cerebellum, and much weaker SVCT2 IRs were found in the other brain regions. Judging from double-immunohistochemical data, most of the cells expressing SVCT2 IRs were likely to be neurons or microglia, even though the cells in choroids plexus or ependymal cells around the ventricles also exhibited SVCT2 IRs. Complete mapping of the distribution of SVCT2 IRs was available by using a semiquantitative method. The subcellular localization of SVCT proteins is necessary for understanding the exact role of the protein, so the current overall mapping of SVCT IRs in the rat brain could be the basis for further studies on related subjects. © 2006 Wiley-Liss, Inc.</P>

        Intraperitoneal (IP) vancomycin is widely used to treat gram-positive peritonitis associated with peritoneal dialysis. There have been 2 cases of red man syndrome, a vancomycin- specifi c non-immunologic reaction, associated with IP vancomycin. However, immune-mediated hypersensitivity reaction to IP vancomycin has not yet been reported. A 49-year-old woman on continuous ambulatory peritoneal dialysis developed her fi rst peritonitis episode. The patient was treated with IP vancomycin once per week for 4 weeks. She experienced mild itching throughout her body for 1 day after the first treatment. Whenever vancomycin was administered, generalized urticaria and a prickling sensation developed, and the intensity increased gradually; however, these symptoms improved after vancomycin was discontinued. An allergic skin test was performed 6 weeks after the previous urticarial episode, and intradermal skin test revealed a positive response to vancomycin. To our knowledge, this is the fi rst case report of IgE-mediated hypersensitivity reaction to IP vancomycin administration.         Legume and rhizobia symbiosis plays an important role in conversion of atmospheric dinitrogen to ammonia. On a global scale, thin interaction represent a key entry point for reduced nitrogen into the biosphere, and as a consequence this symbiosis in important in both natural and agricultural systems. Symbiotic development of nodule organ in triggered by chito-oligosaccharide signals(Nod factors) from the bacterium which are perceived by the legume root. Understanding the molecular and cellular processes that underlie Nod factor perception is one focus of legume biology. Although forward genetics has proved to be an important tool to elucidate key players in Nod factor perception, we still know relatively little regarding the functional networks of genes and proteins that connect the earliest steps of Nod factor perception to immediate downstream outcomes. To identify genes and proteins that link Nod factor perception to cellular and physiological responses we are taking a discovery-based strategy on large-scale transcriptome profiling using RNA sequencing in the roots of Medicago truncatula in response to Sinorhizobium meliloti. Functional characterization of a number of candidate genes is currently in progress to further examine their role in nodulation.

      • Dual color FISH karyotype of wild radish species

        Radish, Raphanus sativus L., is an annual vegetable of the family Cruciferae. Radish has RR genome with 18 somatic chromosome numbers (2n=2x=18). Until now, detailed karyotypic analysis is not only constructed only by conventional staining techniques but also other method. Fluorescence in situ hybridization (FISH) is a powerful molecular cytogenetic technique using chromosomal markers that reveal the positions of specific genes, such as ribosomal DNAs, thereby making it easy to identify individual chromosomes. We have constructed detailed karyotypes of four different local and wild varieties of radish, based on the chromosome arm length and fluorescence in situ hybridization (FISH) with the 45S rDNA and 5S rDNA as probes. As for the karyotype of radish, 9 pairs of chromosomes were extremely small in size with about 1 to 3 um in length at mitotic metaphase having metacentrics or submetacentrics. Three pairs of 45S rDNA signals and two pairs of 5S rDNA signals were observed in four radish species. One pair of 45S rDNA signal was located on terminal region of short arm chromosome, while two pairs were in interstitial region. Two pairs of 5S rDNA signals were located on interstitial region of chromosome. In conclusion, it was feasible to identify the radish by karyotype and physical mapping analyzed using ribosomal DNA.

        Molecular identification and expression analysis of bactericidal permeability-increasing protein/ LPS-binding protein (BPI/LBP) from black rockfish Sebastes schlegeli

        Bactericidal/permeability-increasing protein (BPI) and lipopolysaccharide-binding protein (LBP) are important components of the mammalian innate defence system against Gram-negative infections. The BPI/LBP cDNA was identified from the black rockfish ConA/PMA or LPS stimulated leukocyte cDNA library. The full-length BR-BPI/LBP cDNA was 2118 bp long and contained an open reading frame (ORF) of 1422 bp that encoded 473 amino-acid residues. The 5` UTR had a length of 57 bp, and the 3` UTR 639 bp. The molecular weight and theoretical isoelectric point (pI) values were calculated 51.4 kDa and 9.72, respectively. Compared with other known BPI or BPI/LBP peptide sequences, the most conserved regions of the black rockfish BPI/LBP peptide were found to be the BPI1 N-terminal, BPI2 C-terminal domains and a LPS binding domain. Phylogenetic analysis based on the deduced amino acid sequence revealed a homologous relationship between the BPI/LBP sequence of black rockfish and that of other teleosts. The black rockfish BPI/LBP gene was predominantly expressed in the PBLs, head kidney, trunk kidney and spleen. The expression of the black rockfish BPI/LBP molecule was induced in the peripheral blood leukocytes (PBLs) from 1 to 24 h following LPS stimulation, with a peak at 12 h post-stimulation.

        The immune-adjuvant effect and safety of recombinant CC chemokine 1 (rRbCC1) in rock bream, Oplegnathus fasciatus

        Adjuvants are immune enhancers that are often used in vaccination to augment the immune response of a vaccine, thereby enhancing the protective immunity against the targeted disease. In the present study, we used the recombinant protein, such as rRbCC1, this protein was produced from rock bream CC chemokine 1. To verify the adjuvant effects of this recombinant protein, the immune responses of rock bream to Streptococcus iniae (S. iniae) FKC vaccination, which alone or in combination with recombinant protein was analyzed and then also performed experimental challenge with live S. iniae. The result of serum agglutination titres was showed relatively low levels however, the efficacy of FKC vaccine still conferred protection against S. iniae. Moreover, the adverse effects result showed that no statistically significant difference was revealed between high concentration injected and non-injected fish groups, generally. The relative percent survival (RPS) of FKC + recombinant vaccination group was significantly higher than that of vaccinated group with FKC alone. After experimental challenge to the rock bream by injection with live bacteria (S. iniae), the FKC + rRbCC1 vaccination group was showed 87.0% RPS, however, the RPS of FKC alone vaccination was 68.2%. The results indicated that the recombinant protein as an adjuvant had a clear synergism to injection vaccine of rock bream.
