RAPD와 핵 리보솜 DNA ITS2 염기서열에 의한 한국산 전나무속 식물의 유전적 변이 및 계통유전학적 분석

We analyzed the genetic variation and phylogenetic relationship among fourteen taxa of GenusAbies distributed in Korea using random amplified polymorphic DNA (RAPD) and internal transcribed spacer (ITS) 2 sequences of the nuclear ribosomal DNA (rDNA). RAPDs were used for identification of selected taxa by amplification using single 10-mer primers of arbitrary sequence. Fourteen taxa were clearly classified with 10 arbitrary primers, which generated 102 useful polymorphic bands that were evaluated according to the presence or absence of the fragments of equal size. The genetic distance between A. koreana for. chlorocarpa and A. koreana for. rubrocarpa was 0.127, showing the closest among any other pairs. By comparing the base sequences in the ITS2 region of the nuclear rDNA, genetic relationship was assesed among selected taxa. It showed twenty six sites of point mutation (base substitution) in 243 base pairs (bp) of ITS2 region. The G+C contents of ITS2 region were ranged from 58.3% to 61.7%. The value of nucleotide divergence among selected taxa was ranged from 0.4% to 9.8%. These results showed that RAPD and ITS2 sequence analysis was a useful tool for elucidating phylogenetic relationship and selection of specific marker in Abies species.
번식생리 및 내분비 : 제주흑우 동결정액 제조 시 Amide 계열의 동결보호제가 동결 융해 후 정자의 성상에 미치는 영향

본 연구는 희소 한우인 제주흑우의 정액 동결을 위하여 동결보호제로서 glycerol과 amide 계열의 동결보호제인 5%의 DMA, DMF 그리고 7%의 MF를 이용하여 동결 융해 후 정자의 운동성, 생존율, 정자막 온전성 및 정자의 첨체 양상의 변화 조사를 위하여 수행하였다. 제주흑우의 정액을 동결 한 결과 glycerol과 DMF를 사용하였을 때 운동성은 64.00±9.62와 59.00±5.48로 DMA와 MF에 비해 유의적으로 높았으며(p<0.05), 생존율 역시 glycero과 DMF 처리구에서 58.25±6.63과 53.05±3.77로 유의적으로 높게 나타났다(p<0.05). 그러나 glycerol과 DMF 처리구 간의 유의적 차이는 나타나지 않았다. 정자막 온전성 검사(HOST)에 있어서도 역시 glycerol과 DMF 처리에서 정자 미부의 팽창 비율 역시 45.12±25.08과 44.95±8.58로 유의적으로 높았다(p<0.05). 정자의 첨체막 양상 변화에 있어 F pattern의 비율이 MF 처리시 다른 처리구에 비하여 유의적으로 낮았으며(p<0.05), B pattern의 비율은 DMA, DMF 및 MF의 사용시 glycerol에 비하여 유의적으로 증가하였다(p<0.05). AR pattern의 비율에 있어서는 DMF를 제외한 amide의 사용은 F pattern의 감소와 조기 첨체반응에 의한 유의적인 AR pattern의 증가를 볼 수 있었다(p<0.0.5). 그러나 DMF의 사용은 glycerol 보다 AR pattern의 비율이 유의적으로 낮았다(p<0.05). 동결 융해 후 시간에 따른 정자의 생존율과 정자 미부 팽창 비율에 있어서 융해 초기에는 glycerol에서 DMF 보다 높은 생존율과 정자 미부 팽창율을 나타냈으나, 1시간 이후부터는 DMF에서 glycerol 보다 높은 생존율과 정자 미부 팽창율을 나타내었다. 이러한 결과는 희소 가축의 생식세포 보존 및 유전자원 확립을 위한 중요한 자료가 될 것이며, 제주흑우 및 희소 가축의 안정적이고 효율적인 동결 정액 제조 연구에 있어 중요한 정보를 제공할 것으로 평가된다. The objective of this study was to examine the effect of amides as a cryoprotectant for semen cryopreservation in Korean Jeju Black Bull. The semen was cryopreserved with extenders containing 5% dimethyl acetamide(DMA), 5% dimethyl formamide (DMF), 5% methyl formamide(MF) or 7% glycerol. Post-thawed sperm were evaluated for sperm motility, viability, acrosome integrity and membrane integrity. Post-thawed sperm motility was significantly higher(p<0.05) in glycerol and DMF(64.00%± 9.62 and 59.00%±5.48, respectively) than DMA and MF(50.00%±3.24 and 44.00%±4.18, respectively). Sperm viability wassignificantly higher(p<0.05) in glycerol and DMF(58.25%±7.35 and 53.05%±3.77, respectively) than others. However, for sperm motility and viability, there were no differences among glycerol and DMF. Also, swelling sperm ratio by hypo-osmetic selling test(HOST) was significantly increased(p<0.05) in glycerol and DMF treatments(45.12%±25.08 and 44.95%±8.58, respectively). The percentage of capacitated sperm assessed by CTC staining, F pattern was lower(p<0.05) in DMF than others. B pattern was increased(p<0.05) in DMA, DMF and MF when compared with glycerol. AR pattern ratio was decreased(p<0.05) in glycerol and DMF when compared with DMA and MF. These results suggested that amides performed better and could be used as a cryoprotectant for semen freezing of Korean Jeju Black Bull.
RCR - RAPD 분석에 의한 황칠나무(Dendropanax morbifera)와 일본황칠나무(D. trifidus)의 계통 유전학적 유연 관계

Phylogenetic relationship between Dendropanax morbifera and D. trifidus was analysed by calculating the genetic distance coefficient and constructing the phylogenetic tree among sixteen individuals based on PCR-RAPD. Sixteen individuals were branched off two clusters; cluster I formed by Tok 1, 2, Nag 4, Pog 1, 2, Hae 1, 2 and Wan 2, while cluster II contained Nag 1, 2, 3, MHa 1, 2, Tsu 1, 2 and Wan 1. Between Hae 1 and Hae 2, it showed the nearest value of genetic distance coefficient as 0. 026, while the furthest between MHa 1 and Hae 1, 2 as 0. 333. These results mean that morphological classifications between two taxa are not available, it is neccessary to take a new look at characteristic difference in these taxa by various fields, For that reason, much more informations on the DNA level e needed as well.

Ribosomal DNA 의 ITS 염기서열에 의한 자생 왕벚나무와 재배 왕벚나무의 유연관계

It has long been disputed whether Prunus yedoensis was originated from Mt. Halla in Cheju-do or Cheju-do originated taxon is the same as the cultivated one from Japan. By comparing the base sequences in the ITS 1, ITS 2 and 5.8s coding region of the nuclear ribosomal DNA, genetic relationship was assesed between P. yedoensis Matsumura-Native and P. yedoensis Matsumura-Cultivar. There showed three sites of point mutation (base substitution) and seven insertions or deletions in 236 bp of ITS 1 but no differences in 164 bp of 5.8s coding region. On the other hand, it was shown that four sites of point mutation (base substitution) and seven insertions or deletions in 233 bp of ITS 2. The content of (G+C) in ITS 1 region was 59.4% in natives and 60.2% in cultivar and in ITS 2, 64.0% in natives and 64.3% in cultivar, respectively. But the 5.8s coding region was identical as 56.3% in each taxon. The idea that P. yedoensis Matsumura-Native would be different from the P. yedoensis Matsumura-Cultivar would be strongly substantiated from the similar results obtained in this studies with those of previous studies.
P031 : Assessment of the quality of life in autoimmune blistering skin disease patients

Background: Autoimmune blistering skin diseases such as pemphigus vulgaris, pemphigus foliaceus, bullous pemphigoid and epidermolysis bullosa acquisita substantially affect patient`s daily life and psychosocial well-being. Objectives: The aim of this study was to discover the factors that can influence the QOL of autoimmune blistering skin disease patients by comparing the QOL in patients with autoimmune blistering skin disease and in healthy controls. Methods: 40 autoimmune bullous skin disease patients and 40 healthy controls were interviewed using the Korean version of Skindex-29 and clinical severity of the disease was assessed by the Physician Global Assessment (PGA) index. Results: The total, symptom, function and emotion scores of Skindex-29 were significantly higher in the patients with autoimmune blistering skin disease (48.22, 52.63, 44.46 and 49.33, respectively) than in the healthy controls (25.52, 27.50, 24.38 and 25.28, respectively) (p<0.001). High PGA indexhad negative correlation with QOL and QOL was more damaged if patients had low levels of satisfaction to treatment. Conclusion: The results show that autoimmune blistering skin disease can harmfully affect patients` QOL. Also, disease severity and low satisfaction to treatment are important factors reducing QOL. Newer biological treatment including rituximab is anticipated to make a progress in treatment efficacy and QOL of patients with autoimmune bullous disease.

Background: Oral isotretinoin is well-known effective drug for acne vulgaris, but it commonly causes dryness of skin and mucous membrane. Gamma-linolenic acid (GLA) is known for improving moisturizing effect and reducing transepidermal water loss (TEWL). Objectives: To investigate the clinical efficacy of GLA in the prevention of dryness of skin and mucous membrane and in the efficacy of improvement of acne lesions when concomitantly administered with isotretionin in patients with acne vulgaris. Methods: 89 Korean acne patients between Korean acne grading system (KAGS) grade 2 and 6 were enrolled and randomized to receive isotretinoin (20 mg/day) either with or without GLA (320 mg/day) for 12 weeks. Results: The decrease of the skin hydration and increase of the TEWL of the lip and cheek were less distinct in the combination (experimental) group than in alone (control) group. VAS score for dryness was lower in combination group (p=0.017). The acne lesions (papules, comedos, pustules) were reduced in both groups. In particular, only the numbers of papules were significant between two groups (p=0.043). The decrease of the KAGS and increase of the patient satisfaction for the treatment were more definite in the experimental group. Conclusion: We found that GLA could be effectively used to reduce xerosis in acne patients treated with isotretinoin. The improvements in acne lesions, especially papules, were better treated in combination group.
경피증양 만성 피부 이식편 대 숙주질환에서 광범위하게 발생한 근막염

Fasciitis can occur very rarely with sclerotic-type chronic cutaneous graft-versus-host disease (GVHD). A 54-year-old woman presented with painful skin tightness on upper and lower limb with limited range of movement. She was diagnosed with chronic myelocytic leukemia 5 years ago and underwent allogeneic bone marrow transplantation. Histopathologically, the interlobular septum of subcutis and fascia were remarkably thickened with fibrosis and moderate inflammatory infiltrates accompanying the dilated lymphatic channels with considerable leakage of lymph fluids. Herein, we report a case of extensive fasciitis as a manifestation of chronic GVHD associated with poor prognosis. (Korean J Dermatol 2013;51(7):544∼547)
