      • Isolation and Transcriptional Expression of CuZn Superoxide Dismutase from Codonopsis lanceolata

        To investigate the defense mechanism against the abiotic stress, a cDNA clone encoding a CuZn superoxide dismutase (CuZnSOD) protein was isolated from a cDNA library prepared from tabroot mRNAs of Codonopsis lanceolata. The eDNA, designated ClSODCc, is 799 nucleotides long and has an open reading frame of 459 bp with a deduced amino acid sequence of 152 residues. The deduced amino acid sequence of ClSODCc matched to the previously reported CuZnSODs. Consensus amino acid residues (His-45, -47, -62, -70, -79, -119 and Asp-82) were involved in Cu-, Cu/Zn-, and Zn- binding ligands. The deduced amino acid sequence of ClSODCc showed high homologies (82%-86%) regardless of species. Expression of ClSODCc by oxidative stress was increased up to 1 h after treatment and declined gradually. Much earlier and stronger expression of ClSODCc was observed in the cold stress treatment.

        Physical and chemical Effects on the sonication Treatment of chitosan solution

        물리적 방법을 이용한 키토산 올리고당의 제조에 관한 기초 자료를 제시하기 위하여 키토산용액을 20kHz의 초음파로 처리하였으며, 초음파 처리 효과에 미치는 처리 조건의 영향을 조사하였다. 키토산 용액의 고유점도는 초음파 처리 5분까지는 급속하게 감소하였고 그 이후로는 서서히 감소하였다. 키토산 용액의 부피가 작을수록 초음파 처리효과는 컸으며, 키토산 용액의 온도는 초음파 처리에 별다른 효과가 없는 것으로 나타났다. 5분간 초음파 처리 후의 점도는 용매의 종류에 따라 큰 차이가 없었으나, acetate buffer의 경우 초음파 처리효과가 가장 컸다. pH가 높을수록 초음파 처리효과는 크게 나타났으며, 이온강도와 염의 종류에 따른 효과는 나타나지 않았다. 이상의 결과에서, 용액의 부피는 10~20ml, 온도는 20~30℃, 용액의 pH는 4.5, 그리고 용매의 종류는 acetate buffer로 하는 것이 최적 초음파 처리조건으로 추정된다. As the first step of studies related to production of chitooligosaccharides by physical methods, chitosan solution were sonicated with 20kHz and various treatment effects were examined to present fundamental data of sonicated chitosan solution. Intrinsic viscosity of chitosan solution sharply decreased from 3.76dl/g to 2.90dl/g until 5 minutes of sonication and then slowly decreased. With low volume of chitosan solution, sonication was very effective and temperature of chitosan solution slightly affected the efficiency of sonication. In case of changing the solvent, no significant differences were observed on the effect of sonication, however, acetate buffer had highest sonication effect among various solvents. The sonication effect was increased as the increasement of the value of pH, on the contrary, ionic strength and type of counterions showed no effect on sonication, With these results, we assumed that optimal sonication treatment would be as follows, solution volume was 10 ~20ml, temperature range was 20~30℃ , pH value of solution was 4.5 and type of solvent was acetate buffer

      • Immunohistochemical Study of Myogenic Sarcomas

        Using immunohistochemical staining with monoclonal antibodies to muscle cell components, histogenetical grouping and differential diagnosis of myogenic sarcoma could be possible. However immunohistochemical staining properties of myogenic sarcomas are not well defined in the formalin-fixed, paraffin-embedded tissue. We attempted to differentiate leiomyosarcoma from rhabdomyosarcoma. Twenty-nine formlin fixed myogenic sarcomas were investigated by means of immunohistochemical technique with the use of monoclonal antibodies panel against vimentin, desmin and myoglobin. In addition, the degree of differentiation of these sarcomas was investigated in connection with the immunoreactivity and intensity of immunohistochemical stains. The results were as follows; 1. Vimentin was detected in nine (60%) out of fifteen cases of leiomyosarcoma and in seven (50%) out of fourteen cases of rhabdomyosarcoma. 2. Desmin was detected in seven (47%) out of fifteen cases of leiomyosarcoma and in eight (57%) out of fourteen cases of rhabdomyosarcoma. 3. Myoglobin was detected in eleven (79%) out of fourteen cases of rhabdomyosarcoma. In all leiornyosarcomas, myoglobin was not detected. 4. The immunoreactivity and intensity of these sarcomas with anti-desmin and anti-myoglobin were related to the degree of tumor cell differentiation, but vimentin was more frequently detected in the poorly differentiated tumor cells. The above results suggest that demonstration of vimentin, desmin and myoglobin in the formalin-fixed, parnffin-embedded tissue may be a helpful adjunct tool in the diagiosis of myogenic sarcoma.

      • Superplastic Forming of Combustion Chamber

        <P>The major advantage of superplastic forming (SPF) technology is that it can form integral and complex components in simple operation, since it is possible to form one or more sheets of superplastic grade metal into single surface tools by relatively low gas pressure. Duplex steel contains two phases in nearly equal proportions which can suppress grain growth at a high temperature, like superplastic Ti-6Al-4V, and many duplex stainless steels with fine grained microstructures show superplastic behavior. In this study, superplastic forming technology was developed to fabricate a duplex stainless steel sheet for the outer surface of liquid propellant combustion chamber. Superplasticity of this alloy was investigated and forming methodology was analyzed and developed. The experimental results show a complex configuration of aerospace component was successfully fabricated by superpalstic forming of a sheet of duplex stainless steel.</P>

      • The investigation of Landfill Gas Generation and Safety for Land Reclamation at Kooui Landfill Site in Seoul City

        This inverstigation was carried was carried out to analyze the generation and composition of landfill gas generated from inserted pipe wells into the underground by boring operation and also study the undecomposed refuse characteristics by open-cut test, after 5 years from the elapse of landfill for land reclamiation, at Kooui Landfill Site in Seoul City. The experimental results from this study were summerized as follows. 1) from the open cut test, approximately 66.3% of organic matters in filled refuse has been decomposed with respect to the refuse composition of 1979, and the undecomposed organic matters remained in underground were mostly belonged to plastics, synthetic textiles woods and very few papers. 2) In the landfill gas generated from inserted pipe wells the compositions of CH and CO were measured in the range of 50 to 60% (V/V) and 20 to 35%(V/V), respectively, and the rates of landfill gas generation were determined to 7 1/min in minimum at the centural area of landfill site whereas be 160 1/min in miximum at the perimetric area adjescent to Han River express way and Kangbyon Subway Station. Therefore it is suggested that the former would be in stabilization stage in certain degree in this study, while the latter is still going to gradually decompose the underground refude to produse methane gas. 3) The change of underground temperature was linearly decreased to 16∼18℃ up to the depths of 10m with respect to 26∼32℃ of ambient temoerature. Lowering the underground temperature was caused by the wash-out phenomena of Han River water, although the anaerobic decomposition of refuse is being moderately progressed in the underground. 4) The general molecular formula of organic waste remained in underground was experimentally determined such as C ₁₂H_(26)O_(9)N_(0.1)S_(0.002).

        New vaccines against influenza virus

        <P>Vaccination is one of the most effective and cost-benefit interventions that prevent the mortality and reduce morbidity from infectious pathogens. However, the licensed influenza vaccine induces strain-specific immunity and must be updated annually based on predicted strains that will circulate in the upcoming season. Influenza virus still causes significant health problems worldwide due to the low vaccine efficacy from unexpected outbreaks of next epidemic strains or the emergence of pandemic viruses. Current influenza vaccines are based on immunity to the hemagglutinin antigen that is highly variable among different influenza viruses circulating in humans and animals. Several scientific advances have been endeavored to develop universal vaccines that will induce broad protection. Universal vaccines have been focused on regions of viral proteins that are highly conserved across different virus subtypes. The strategies of universal vaccines include the matrix 2 protein, the hemagglutinin HA2 stalk domain, and T cell-based multivalent antigens. Supplemented and/or adjuvanted vaccination in combination with universal target antigenic vaccines would have much promise. This review summarizes encouraging scientific advances in the field with a focus on novel vaccine designs.</P>

      • Inhibition of LPS-induced inflammatory biomarkers by ethyl acetate fraction of <i>Patrinia scabiosaefolia</i> through suppression of NF-κB activation in RAW 264.7 cells

        <P><I>Patrinia scabiosaefolia</I> (PS) has been used for curing various types of inflammatory-related disorders. However, the precise mechanism of the anti-inflammatory activity of PS remains unclear. Here, we investigated the anti-inflammatory effects of several fractions isolated from the PS in RAW 264.7 macrophages. The results indicated that the ethyl acetate fraction of PS (EAPS) concentration highly suppressed lipopolysaccharide (LPS)-induced nitric oxide (NO) and IL-6 productions without a cytotoxic effect on RAW 264.7 cells. EAPS inhibited the expressions of LPS-induced iNOS and COX-2 protein and their mRNA in a dose-dependent manner. Particularly, EAPS suppressed the level of nuclear factor-κB (NF-κB) activity, which was linked with the suppression of LPS-induced phosphorylation of p65 at serine 276 and p65 translocation into nuclei, but not MAPK signaling. In addition, treatment with <B>EAPS</B> inhibited the production of TNF-α in LPS-injected mice and suppressed the production of IL-6 and TNF-α in LPS-stimulated splenocytes from BALB/c mice. Therefore, we demonstrate here that <I>Patrinia scabiosaefolia</I> potentially inhibits the biomarkers related to inflammation through the blocking of NF-κB p65 activation, and it may be a potential therapeutic candidate for the treatment of inflammatory diseases.</P>

      • Pick-and-Put Simulation with a Redundant Manipulator under Teleoperation

        Operator-in-the-loop simulation is applied to pick-and-put operation with a redundant manipulator. The concept of supervisory control has been used to switch between teleoperation and autonomous operation: autonomous operation for fine, local motion control, and teleoperation for large, gross motion and force control. The simulation includes the manipulator's kinematics and its open-and closed-chain dynamics under position and force control. They are integrated with a pair of joystick hand controllers and computer graphics. Since the hand controllers specify the desired position and orientation of the end-effector, the inverse kinematics adopted an iterative use of the resolved-motion rate control in which the constraint Jacobian is constructed on-line and is used in the pseudo-inverse method. An efficient recursive algorithm is then used for the manipulator dynamics. Rear-time simulation is carried out on a two-processor IRIS workstation, with one processor for dynamics and control computation and the other for graphic visualization. The simulation shows a potential for previewing the planned operations, examining task performance, investigating control strategy, and evaluating the overall system integration.
