        Butyrate regulates leptin expression through different signaling pathways in adipocytes

        Leptin is an adipocytokine that regulates body weight, and maintains energy homeostasis by promoting reduced food intake and increasing energy expenditure. Leptin expression and secretion is regulated by various factors including hormones and fatty acids. Butyrate is a short-chain fatty acid that acts as source of energy in humans. We determined whether this fatty acid can play a role in leptin expression in fully differentiated human adipocytes. Mature differentiated adipocytes were incubated with or without increasing concentrations of butyrate. RNA was extracted and leptin mRNA expression was examined by Northern blot analysis. Moreover, the cells were incubated with regulators that may affect signals which may alter leptin expression and analyzed with Northern blotting. Butyrate stimulated leptin expression, and stimulated mitogen activated protein kinase (MAPK) and phospho-CREB signaling in a time-dependent manner. Prior treatment of the cells with signal transduction inhibitors as pertusis toxin, Gi protein antagonist, PD98059 (a MAPK inhibitor), and wortmannin (a PI3K inhibitor) abolished leptin mRNA expression. These results suggest that butyrate can regulate leptin expression in humans at the transcriptional level. This is accomplished by: 1) Gi protein-coupled receptors specific for short-chain fatty acids, and 2) MAPK and phosphatidylinositol-3-kinase (PI3K) signaling pathways.

        Synthesis of Some New 4,5-dihydro-6-(4-methoxy-3-methylphenyl)-3(2H)-pyridazinone Derivatives

        4,5-Dihydro-6-(4-methoxy-3-methylphenyl)-3(2H)-pyridazinone derivative계 화합물의 합성연구를 수행하였다. 첫 번째로, 4,5-dihydro-6-(4-methoxy-3-methylphenyl)-3(2H)-pyridazinone (1)은 o-cresyl methyl ether와 succinic anhydride를 Friedel-Crafts 아실화 반응을 통하여 얻은 다음에, 고리화 반응을 통하여 합성하였다. 얻어진 화합물 1을, NaOEt조건 하에서, 방향족 알데히드와 반응시켜서 4-substituted benzyl pyridazinones (3a-d)을 합성하였으며, 화합물 1을 탈수소화반응을 시켜서 화합물 4를 얻었다. 한편 pyridazine 5는 화합물 1과 1,3-diphenyl-2-propen-1-one을 Michael 첨가반응을 이용하여 합성하였다. N-Dialkylaminomethyl 화합물 6a-b는 pyridazinone 1과 formaldehyde 및 2차 amine을 바능시켜서 얻은 반면에, 화합물 7은 pyridazinone 1을 반응시켜서 얻었으며, 화합물 8은 pyridazinone 3b를 phosphorus oxychloride 와 반응시켜서 얻었다. The present study describes the synthesis of 4,5-dihydro-6-(4-methoxy-3-methylphenyl)-3(2H)-pyridazinone derivatives. The synthesis of the first target compound, 4,5-dihydro-6-(4-methoxy-3-methylphenyl)-3(2H)-pyridazinone (1), was achieved by Friedel-Crafts acylation of o-cresyl methyl ether with succinic anhydride and subsequent cyclization of the intermediary g-keto acid with hydrazine hydrate. Condensation of compound 1 with aromatic aldehydes in the presence of sodium ethoxide affords the corresponding 4-substituted benzyl pyridazinones (3a-d). The dihydropyridazinone 1 underwent dehydrogenation upon treatment with bromine/acetic acid mixture to give (4). Pyridazine (5) has been synthesized upon the reaction of pyridazinone (1) with 1,3-diphenyl-2-propen-1-one under the Michael addition reaction. N-dialkylaminomethyl derivatives 6a-b have been obtained from the reaction of pyridazinone 1 with formaldehyde and secondary amine, whereas reaction of 1 with formaldehyde gives N-hydroxymethyl derivative (7). This study also includes the synthesis of the 3-chloropyridazine derivative 8 in excellent yield by heating pyridazinone 3b in phosphorus oxychloride. The behaviour of the chloro derivative toward sodium azide, benzyl amine and anthranilic acid was also studied. The proposed structures of the products were confirmed by elemental analysis, spectral data and chemical evidence.

        Role of protease inhibitors and acylation stimulating protein in the adipogenesis in 3T3-L1 cells

        Treatment of AIDS (HIV) and hepatitis C virus needs protease inhibitors (PI) to prevent viral replication. Uses of PI in therapy are usually associated with a decrease in body weight and dyslipidemia. Acylation stimulating protein (ASP) is a protein synthesized in adipocytes to increase triglycerides biosynthesis, for that the relation of PI and ASP to adipogenesis is tested in this work. ASP expression was increased during 3T3-L1 differentiation and reached a peak at day 8 with cell maturation. Addition of PI during adipocytes differentiation dose dependently and significantly (p < 0.5) inhibited the degree of triglycerides (TG) accumulation. Moreover, presence of ASP (450 ng/mL) in media significantly (p < 0.5) stimulated the degree of TG accumulation and there was additive stimulation for ASP when added with insulin (10 μg/mL). Finally, when ASP in different doses (Low, 16.7; Medium, 45 and High, 450 ng/mL) incubated with a dose of ×150 PI, ASP partially inhibited the PI-inhibited adipogenesis and TG accumulation. The results in this study show that PI inhibit lipids accumulation and confirm role of ASP in TG biosynthesis and adipogenesis.

        New CMOS Fully Differential Transconductor and Application to a Fully Differential Gm-C Filter

        A new CMOS voltage-controlled fully-differential transconductor is presented. The basic structure of the proposed transconductor is based on a four-MOS transistor cell operating in the triode or saturation region. It achieves a high linearity range of ± 1 V at a 1.5 V supply voltage. The proposed transconductor is used to realize a new fully-differential Gm-C low-pass filter with a minimum number of transconductors and grounded capacitors. PSpice simulation results for the transconductor circuit and its filter application indicating the linearity range and verifying the analytical results using 0.35 μm technology are also given.

        Nonlinear transient analysis of FG pipe subjected to internal pressure and unsteady temperature in a natural gas facility

        This study investigates the response of functionally graded (FG) gas pipe under unsteady internal pressure and temperature. The pipe is proposed to be manufactured from FGMs rather than custom carbon steel, to reduce the erosion, corrosion, pressure surge and temperature variation effects caused by conveying of gases. The distribution of material graduations are obeying power and sigmoidal functions varying with the pipe thickness. The sigmoidal distribution is proposed for the 1st time in analysis of FG pipe structure. A Two-dimensional (2D) plane strain problem is proposed to model the pipe cross-section. The Fourier law is applied to describe the heat flux and temperature variation through the pipe thickness. The time variation of internal pressure is described by using exponential-harmonic function. The proposed problem is solved numerically by a two-dimensional (2D) plane strain finite element ABAQUS software. Nine-node isoparametric element is selected. The proposed model is verified with published results. The effects of material graduation, material function, temperature and internal pressures on the response of FG gas pipe are investigated. The coupled temperature and displacement FEM solution is used to find a solution for the stress displacement and temperature fields simultaneously because the thermal and mechanical solutions affected greatly by each other. The obtained results present the applicability of alternative FGM materials rather than classical A106Gr.B steel. According to proposed model and numerical results, the FGM pipe is more effective in natural gas application, especially in eliminating the corrosion, erosion and reduction of stresses.

        Biology of the mason bee Osmia latreillei (Hymenoptera: Megachilidae) under artificial nesting conditions in Egypt

        The mason bee Osmia (Helicosmia) latreillei Spinola is one of the dominant species of bees throughout Egypt, and can be found commonly in the Suez Canal Region. The species visits numerous plant species of the family Asteraceae and is considered the most important pollinator of certain crops such as sunflower. This species and some other solitary cavity nesting bees are threatened by fragmentation of their nesting habitats. Several attempts were made to establish and propagate O. latreillei, and success was achieved for re-nesting it in various artificial materials such as wood and polystyrene with rolled paper straws. These artificial nests were established at the conservation area of the Bee Research Centre, Suez Canal University, in order to study the nesting behavior and biology of O. latreillei under artificial conditions and to document their natural enemies. Biologically, there was no significance difference between the life cycle of O. latreillei under natural and artificial nesting conditions. This solitary bee was univoltine, individuals active only during Spring, and attacked by diverse enemies, with Stelis (Stelis) murina Pérez and chrysidid wasps considered the two most major pests of this wild bee species.

        Analysis of crack occurs under unsteady pressure and temperature in a natural gas facility by applying FGM

        Cracking can lead to unexpected sudden failure of normally ductile metals subjected to a tensile stress, especially at elevated temperature. This article is raised to study the application of a composite material instead of the traditional carbon steel material used in the natural gas transmission pipeline because the cracks occurs in the pipeline initiate at its internal surface which is subjected to internal high fluctuated pressure and unsteady temperature according to actual operation conditions. Functionally graded material (FGM) is proposed to benefit from the ceramics durability and its surface hardness against erosion. FGM properties are graded at the radial direction. Finite element method (FEM) is applied and solved by ABAQUS software including FORTRAN subroutines adapted for this case of study. The stress intensity factor (SIF), temperatures and stresses are discussed to obtain the optimum FGM configuration under the actual conditions of pressure and temperature. Thermoelastic analysis of a plane strain model is adopted to study SIF and material response at various crack depths.

        Effects of Ultrasound, Laser and Exercises on Temporomandibular Joint Pain and Trismus Following Head and Neck Cancer

        Objective To compare the effects of low intensity ultrasound (LIUS), traditional exercise therapy (TET), low level laser therapy (LLLT) and TET on temporomandibular joint (TMJ) pain and trismus following recovery from head and neck cancer (HNC). Methods Sixty participants following, who had experienced HNC, were randomly allocated to three groups of 20 people each. Each group received different therapy. Group A received LIUS and TET; group B received LLLT and TET; while group C received TET. All 60 participants were evaluated under the visual analog scale (VAS), the University of Washington Quality of Life questionnaire (UW-QOL) and the Vernier caliper scale (VCS) at the beginning of the therapies and after 4 weeks. Results ANOVA test revealed significant improvements across all three groups with outcomes of p<0.05. The results of the UW-QOL questionnaire showed a significant difference between groups A, B and C in favor of group A (p<0.05). The VAS results showed a more improvement in group A as compared to group B (p<0.05), while there was no statistical difference between groups B and C (p>0.05). The VCS results showed more improvement for the individuals in group B as compared to those in group C (p<0.05), while there was minimal difference between groups A and B (p>0.05). Conclusion The LIUS and TET are more effective than LLLT and/or TET in reducing TMJ pain and trismus following HNC.
