      • Polymorphisms of TERT and CLPTM1L and the Risk of Hepatocellular Carcinoma in Chinese Males

        Background: Telomerase reverse transcriptase (TERT) and cleft lip and palate trans-membrane 1 like (CLPTM1L) genes located on chromosome 5p15.33 are known to influence the susceptibility to various cancers. Here, we examined the association of TERT and CLPTM1L single nucleotide polymorphisms (SNPs) with hepatocellular carcinoma (HCC). Materials and Methods: Genotyping of TERT SNP rs2736098 and CLPTM1L SNP rs401681 was performed using TaqMan allelic discrimination assays in a case-control study of 201 HCC cases and 210 controls in a Chinese male population. Odds ratios (ORs) and 95% confidence intervals (CIs) were estimated using logistic regression analyses. Results: Both the rs2736098 T allele of TERT and the rs401681 T allele of CLPTM1L were associated with a significantly increased risk of HCC (adjusted odds ratio [OR]=1.605, 95% confidence interval [CI]=1.164-2.213; adjusted OR=1.399, 95%CI=1.002-1.955, respectively). Individuals carrying both TERT and CLPTM1L risk genotypes had an even higher risk of HCC (adjusted OR=4.420, 95%CI= 2.319-8.425). The TERT rs2736098 T allele was also significantly associated with the level of the HCC clinical indicator alpha-fetoprotein (P=0.026). Conclusions: Our results show that genetic variants of TERT and CLPTM1L may contribute to HCC susceptibility in Chinese males.

        Model-free Adaptive Tracking Control for Networked Control Systems Under DoS Attacks and RTT Delays

        The model-free adaptive secure tracking control (MFASTC) problem of nonlinear networked control systems is explored in this paper with the consideration of DoS attacks and RTT delays. In order to compensate for DoS attacks and RTT delays, a new estimate value dependent attack compensation mechanism is proposed and a network-based predictive delay compensation mechanism is employed. The data-based design method need not any model or structure information of the system, and only depends on input/output data. In addition, the MFASTC algorithm is proposed to guarantee the output tracking error being bounded in the sense of mean square. Finally, an example with comparisons is used to show the effectiveness of the new algorithm.

        Tissue Quality Comparison Between Heparinized Wet Suction and Dry Suction in Endoscopic Ultrasound-Fine Needle Biopsy of Solid Pancreatic Masses: A Randomized Crossover Study

        Background/Aims: A high-quality sample allows for next-generation sequencing and the administration of more tailored precision medicine treatments. We aimed to evaluate whether heparinized wet suction can obtain higher quality samples than the standard dry-suction method during endoscopic ultrasound (EUS)-guided biopsy of pancreatic masses. Methods: A prospective randomized crossover study was conducted. Patients with a solid pancreatic mass were randomly allocated to receive either heparinized wet suction first or dry suction first. For each method, two needle passes were made, followed by a switch to the other method for a total of four needle punctures. The primary outcome was the aggregated white tissue length. Histological blood contamination, diagnostic performance and adverse events were analyzed as secondary outcomes. In addition, the correlation between white tissue length and the extracted DNA amount was analyzed. Results: A total of 50 patients were enrolled, and 200 specimens were acquired (100 with heparinized wet suction and 100 with dry suction), with one minor bleeding event. The heparinized wet suction approach yielded specimens with longer aggregated white tissue length (11.07 mm vs 7.96 mm, p=0.001) and less blood contamination (p=0.008). A trend towards decreasing tissue quality was observed for the 2nd pass of the dry-suction method, leading to decreased diagnostic sensitivity and accuracy, although the accumulated diagnostic performance was comparable between the two suction methods. The amount of extracted DNA correlated positively to the white tissue length (p=0.001, Spearman̕s ρ=0.568). Conclusions: Heparinized wet suction for EUS tissue acquisition of solid pancreatic masses can yield longer, bloodless, DNA-rich tissue without increasing the incidence of adverse events (ClinicalTrials.gov. identifier NCT04707560).

        Development and verification of a novel system for computed tomography scanner model construction in Monte Carlo simulations

        The accuracy of Monte Carlo (MC) simulations in estimating the computed tomography radiation dose is highly dependent on the accuracy of CT scanner model. A system was developed to observe the 3D model intuitively and to calculate the X-ray energy spectrum and the bowtie (BT) filter model more accurately in Monte Carlo N-particle (MCNP). Labview's built-in Open Graphics Library (OpenGL) was used to display basic surfaces, and constructive solid geometry (CSG) method was used to realize Boolean operations. The energy spectrum was calculated by simulating the process of electronic shooting and the BT filter model was accurately modeled based on the calculated shape curve. Physical data from a study was used as an example to illustrate the accuracy of the constructed model. RMSE between the simulation and the measurement results were 0.97% and 0.74% for two filters of different shapes. It can be seen from the comparison results that to obtain an accurate CT scanner model, physical measurements should be taken as the standard. The energy spectrum library should be established based on Monte Carlo simulations with modifiable input files. It is necessary to use the three-segment splicing modeling method to construct the bowtie filter model.

        중국 물류산업의 국민경제 기여도 분석

        본 연구는 중국 통계청이 발표한 산업연관표를 활용하여 중국 물류산업의 국민경제 기여도를 분석함으로써 향후 중국 물류산 업의 발전을 위한 정책적 자료로 활용하는데 목적이 있다. 이러한 목적을 달성하기 위하여 중국 국가 통계청이 2015년 발표한 <2015년 산업연관표 연장표>를 이용하여 기초 분류 42개 산업 중 교통운송, 창고업 및 우편을 물류산업으로 분류하고 물류산업에 대한 경제적 기여도를 분석하였다. 분석 결과는 중국 2015년 물류산업의 생산유발계수 열합계는 2.8734, 행 합계는 4.3510, 감응도 계수는 0.0439, 영향력 계수는 0.0290, 소득유발계수는 0.5162, 생산세 유발계수는 0.0726, 부가가치유발계수는 0.9976, 최종수요 10 억 위안이 발생할 경우 노동유발계수는 총1,105.2명(간접819.1명, 직접286.1명) 등을 보여주고 있다. 분석결과에 따라 본문의 시사 점은 다음과 같다. 첫째, 중국 물류산업은 전반적으로 제조업보다 낮아 물류산업이 아직 성숙되지 못하기 때문에 정부의 적극적인 지원이나 정책적인 우대가 필요하다. 둘째, 중국 물류산업의 전·후방연쇄효과 모두 1보다 작아 최종 수요적 원시 산업형 산업이 기 때문에 기술적 혁명이나 새로운 운영이념 및 모델이 필요하다. 셋째, 물류산업은 자본, 노동시장에 거대한 경제적 파급효과를 유발할 수 있기 때문에 법적인 규정과 제도의 완비와 시장에 대한 감독이 요구된다. 마지막으로 중국의 급속한 경제성장과 함께 제조업과 서비스업의 균형발전을 염두에 두고 선진국의 성공 경험을 벤치마킹해야 한다. The purpose of this study is to identify the status of the logistics industry and the economic ripple effect of investment in the logistics industry by analyzing the economic contribution of the Chinese logistics industry by using the interindustry relations table published by the National Statistical Office of China, and to use it as a policy data for the development of Chinese logistic industry in the future. In order to achieve this purpose, the transportation, warehousing and postal services among 42 basic industries were classified as logistics industries by using <the Extension Tables of Interindustry relations table 2015> announced by the National Statistical Office of China in 2017, and the contribution of Chinese logistics industry to national economy and the economic ripple effect of logistics industry investment were analyzed. The results of the analysis show that the heat sum of the production inducement coefficient of China’s logistics industry in 2015 was 2.8734, and the row total was 4.3510, the sensitivity coefficient was 0.0439, the influence coefficient was 0.0290, the income inducement coefficient was 0.5162, the production tax inducement coefficient was 0.0726, the value-added inducement coefficient was 0.9976, the value-added inducement coefficient was 0.9976, If the final demand of 1 billion occurs, the labor inducement coefficient is 1,105.2 (indirectly 819.1 and direct 286.1). The implications of the text according to the analysis results are as follows. First, the Chinese logistics industry as a whole is lower than the manufacturing industry, so the government’s active support or policy priority is needed because the logistics industry is not yet mature. Second, both the pre- and rear-end linkage effects of China’s logistics industry are smaller than 1, so technical revolution or new operational ideology and model are needed because it is the final demand primitive industrial type industry. Third, the logistics industry is required to complete legal regulations and systems and supervise the market because it can have huge economic ripple effects on capital and labor markets. Finally, with China’s rapid economic growth and balanced development of manufacturing and service industries in mind, the success experience of advanced countries should be benchmarked.

        A facile macroporous resin-based method for separation of yellow and orange Monascus pigments

        The yellow Monascus pigments (YMPs) namedmonascin and ankaflavin and the orange Monascus pigments(OMPs) named rubropunctatin and monascorubrinare two groups of bioactive components in a mixture statein the Monascus fermented products. In order to separatethese two groups of bioactive pigments, a facile macroporousresin-based method was developed. The weak-polarresin CAD-40 was selected from the seven tested macroporousresins as it revealed better properties for theadsorption and desorption of the YMPs and OMPs. Then,CAD-40 resin was used for column-chromatographicseparation. After eluted by 4 bed volumes of ethanol, theyellow group (monascin and ankaflavin) and the orangegroup (rubropunctatin and monascorubrin) were successfullyseparated and purified, with an increased content from49.3% and 44.2% in the crude pigment extract to 85.2%and 83.0% in the final products, respectively. This methodwould be helpful for the large-scale separation and purificationof Monascus pigment products with specificbioactivity.

        Caspase-3 inhibitor inhibits enterovirus D68 production

        Enterovirus D68 (EVD68) is an emerging pathogen that recently caused a large worldwide outbreak of severe respiratory disease in children. However, the relationship between EVD68 and host cells remains unclear. Caspases are involved in cell death, immune response, and even viral production. We found that caspase-3 was activated during EVD68 replication to induce apoptosis. Caspase-3 inhibitor (Z-DEVDFMK) inhibited viral production, protected host cells from the cytopathic effects of EVD68 infection, and prevented EVD68 from regulating the host cell cycle at G0/G1. Meanwhile, caspase-3 activator (PAC-1) increased EVD68 production. EVD68 infection therefore activates caspase-3 for virus production. This knowledge provides a potential direction for the prevention and treatment of disease related to EVD68.
