        Genome-Wide Association Analysis to Search for New Loci Associated with Lifelong Premature Ejaculation Risk in Chinese Male Han Population

        Purpose: Genetic factors play an indispensable role in the pathogenesis of lifelong premature ejaculation (LPE). The suscep-tibility genes/SNPs that have been discovered are very limited and can only explain part of the genetic effects of LPE. There-fore, discovering more genetic polymorphisms associated with the occurrence and development of LPE will help reveal the pathogenesis of LPE. Materials Materials and and Methods:Methods: We conducted a genome-wide association study of LPE in 486 Chinese male Han people (cases and controls). We used Gene Titan multi-channel instrument and Axiom Analysis Suite 6.0 software for genotyping. Imputation was performed by IMPUTE2 software and the 1000 Genomes Project (Phase3) was used as reference for haplotype. Finally, logistic regression analysis was performed on all loci that passed the quality control. The odds ratio and 95% confidence in-terval were calculated to determine the association between each SNPs and Chinese male Han population LPE risk. Results:Results: The results showed that a total of 33 genetic variants in 13 genes (LACTBL1 , SSBP3 , ACOT11 , LINC02486 , TMEM154, LINC01098, NONE, HCG27, HLA-C, TNFSF8, TNC, FAM53B, SULF2) have a suggestively significant genome-wide association with LPE risk (p<5×10-6). Conclusions:Conclusions: This study is the first to conduct a GWAS on LPE in Chinese male Han population 33 genetic polymorphisms have a suggestive genome-wide association with LPE risk. This study have provided data supplement for the genetic loci of LPE risk, and laid a scientific foundation for the pathogenesis and the targeted therapy of LPE.

        MLL4 Regulates the Progression of Non-Small-Cell Lung Cancer by Regulating the PI3K/AKT/SOX2 Axis

        Purpose Mixed-lineage leukemia protein 4 (MLL4/KMT2D) is a histone methyltransferase, and its mutation has been reported to be associated with a poor prognosis in many cancers, including lung cancer. We investigated the function of MLL4 in lung carcinogenesis. Materials and Methods RNA sequencing (RNA-seq) in A549 cells transfected with control siRNA or MLL4 siRNA was performed. Also, we used EdU incorporation assay, colony formation assays, growth curve analysis, transwell invasion assays, immunohistochemical staining, and in vivo bioluminescence assay to investigate the function of MLL4 in lung carcinogenesis. Results We found that MLL4 expression was downregulated in non–small cell lung cancer (NSCLC) tissues compared to adjacent normal tissues and tended to decrease with disease stage progression. We analyzed the transcriptomes in control and MLL4- deficient cells using high-throughput RNA deep sequencing (RNA-seq) and identified a cohort of target genes, such as SOX2, ATF1, FOXP4, PIK3IP1, SIRT4, TENT5B, and LFNG, some of which are related to proliferation and metastasis. Our results showed that low expression of MLL4 promotes NSCLC cell proliferation and metastasis and is required for the maintenance of NSCLC stem cell properties. Conclusion Our findings identify an important role of MLL4 in lung carcinogenesis through transcriptional regulation of PIK3IP1, affecting the PI3K/AKT/SOX2 axis, and suggest that MLL4 could be a potential prognostic indicator and target for NSCLC therapy.

      • Prognostic Analysis of Schistosomal Rectal Cancer

        Background: Schistosomiasis is an infectious disease that affects more than 230 million people worldwide, according to conservative estimates. Some studies published from China and Japan reported that schistosomiasis is a risk factor for colorectal cancer in Asia where the infective species is S. japonicum. Hoqwever, there have been only few reports of prognosis of patients with schistosomal rectal cancer SRC. Objectives: This study aimed to analyze differences in prognosis between SRC and non-schistosomal rectal cancer(NSRC) with current treatments. Materials and Methods: A retrospective review of 30 patients with schistosomal rectal cancer who underwent laparoscopic total mesorectal excision operation (TME) was performed. For each patient with schistosomal rectal cancer, a control group who underwent laparoscopic TME with non-schistosomal rectal cancer was matched for age, gender and tumor stage, resulting in 60 cases and controls. Results: Univariate analysis showed pathologic N stage (P=0.006) and pathologic TNM stage (P=0.047) statistically significantly correlated with disease-free survival (DFS). Pathologic N stage (P=0.014), pathologic TNM stage (P=0.002), and with/without schistosomiasis (P=0.026) were statistically significantly correlated with overall survival (OS). Schistosomiasis was the only independent prognostic factor for DFS and OS in multivariate analysis. Conclusions: The prognosis of patients with schistosomal rectal cancer is poorer than with non-schistosomal rectal cancer.

      • Prognostic Significance of the Mucin Component in Stage III Rectal Carcinoma Patients

        Background: Although mucinous adenocarcinoma has been recognized for a long time, whether it is associated with a poorer prognosis in colorectal cancer patients is still controversial. Many studies put emphasis on mucinous adenocarcinoma containing mucin component ${\geq}50%$. Only a few studies have analyzed cases with a mucin component <50%. Objectives: This study aimed to analyze the prognostic value of different mucin component proportions in patients with stage III rectal cancer. Materials and Methods: Clinical, pathological and follow-up data of 136 patients with the stage III rectal cancer were collected. Every variable was analyzed by univariate analysis, then multivariate analysis and survival analysis were further performed. Results: Univariate analysis showed pathologic T stage, lymphovascular invasion, and histological subtype were statistically significant for DFS. Pathologic T stage was significant for OS. Histological subtype and lymphovascular invasion were independent prognostic factors in multivariate analysis for DFS, and histological subtype was the only independent prognostic factor for OS. Survival curves showed the survival time of mucinous adenocarcinoma (MUC) was shorter than non-MUC (adenocarcinomas with a mucin component <50% and without mucin component). Conclusions: Histological subtype (tumor with different mucin component) was an independent prognostic factor for both DFS and OS. Patients with MUC had a worse prognosis than their non-MUC counterparts with stage III rectal carcinoma.

        CNC Corner Milling Parameters Optimization Based on Variable-Fidelity Metamodel and Improved MOPSO Regarding Energy Consumption

        In the corner milling process, processing energy consumption is a very important objective, since the energy efficiency of CNC machine is barely above 14.8%. Meanwhile, the excessive processing temperature will increase the thermal deformation of the product and leads to quality decline. Improper process parameters will lead to unnecessary high temperature and energy consumption. By optimizing the process parameters, the appropriate temperature and Specific Energy Consumption can be obtained. This study investigated into modeling Specific Energy Consumption and temperature in corner milling process using variable-fidelity metamodels. The adopted variable-fidelity metamodels are constructed by Hierarchical Kriging, in which 48 sets of low-fidelity data obtained from the AdvantEdge software simulation are used to reflect the trends of the metamodels, and 16 sets of high-precision data obtained from physical experiments are used to calibrate the trends. The experimental cost is reduced and the prediction accuracy is increased by making full use of both sets of data. An improved K-means Multi-objective Particle Swarm Optimization algorithm was adopted and applied on the multi-objective corner milling parameters optimization problem to find satisfactory specific energy consumption and temperature. The obtained Pareto solutions can provide guidance for selecting process parameters according to different requirements, such as reducing energy consumption or temperature.

        Influencing Factors Analysis of Degradation and Damage and the Effect on Steel Frame

        In order to discuss the influencing factors of damage and degradation and the effect on steel frame subject to strongearthquakes, firstly, a constitutive model was proposed for 3D finite element models to calculate the seismic behaviors of steelframe more accurately. The model was proved correct and applicable due to typical quasi-static tests. Then, based on theverified model, the damage and degradation factors were discussed by parametric analyses, including the panel zone, loadinghistory, loading amplitude, the width to thickness ratio and yield strength. The cumulative damage phenomena of steel frameconnections were further studied and the degradation distribution curve was obtained. Finally, the traditional beam elementmodel and proposed shell element model of structure were established by ABAQUS for time history analysis. The effect ofdamage and degradation on seismic behaviors of steel frames were compared and discussed. The tests and analysis resultsrevealed that: panel zone strength, loading amplitude and width-thickness ratios of plates had significant impact on damage anddegradation behaviors and failure modes of connections. Premature weld cracking would lead to earlier damage and degradationunder “strong to weak” earthquake, which would seriously affect the overall mechanical properties of the structure. Once localbuckling caused damage, the results of the beam element model and accurate shell element model were remarkably different. Beam element model without considering damage and degradation underestimated the structural deformations, which shouldbe paid more attention for design safety.

        Isoindolin-1-ones from the stems of Nicotiana tabacum and their antiviral activities

        In previous studies, several isoindolin-1-oneanalogs that exhibited signifi cant anti-tobacco mosaic virus(anti-TMV) activities were isolated from Nicotiana tabacum . Since gene-editing mutants provide a new sample for thediscovery of active metabolites, we focused on the stems ofYN-18–23 (a mutant N. tabacum for gene editing with thealkaloid metabolic pathway cultivated by Yunnan TobaccoCompany), which led to the isolation of four new ( 1–4 )and four known ( 5–8 ) isoindolin-1-ones. To the best of ourknowledge, nicindole C ( 3 ) is the fi rst subclass of isoindolin-1-one bearing a pentacyclic ketone, while nicindole D ( 4 )is the fi rst example of isoindolin-1-one bearing a methylpyridin-2-(1 H )-one moiety. Compounds 1–4 were testedfor their anti-TMV activities, and the results revealed thatcompounds 1 , 3 , and 4 exhibited high anti-TMV activities atconcentrations of 20 μM with inhibition rates of 48.6, 42.8,and 71.5%, respectively. These rates are higher than the inhibitionrate of the positive control (33.2%). The mechanisticstudy of compound 4 , which had the highest anti-TMV activityrevealed that increased potentiation of defense-related enzyme activities and downregulation of expression of theNtHsp70 protein may induce resistance in tobacco againstthe viral pathogen TMV. Molecular docking studies alsorevealed that the isoindolin-1-one substructure is fundamentalfor anti-TMV activity. The methyl-pyridin-2-(1 H )-onemoiety in compound 4 and the 2-oxopropyl groups in compounds1 and 3 at the N -2 position may increase inhibitoryactivities. This study of the structure–activity relationshipis helpful for fi nding new anti-TMV activity inhibitors. Tostudy whether the isoindolin-1-ones have broader antiviralactivities, compounds 1–4 were also tested for their antirotavirusactivities. Compound 4 exhibited high anti-rotavirusactivity with a therapeutic index (TI) value of 20.7. This TI value is close to that of the positive control (20.2).

        As an important nonlinear filter theory, particle filter is a heated issue in domestic and foreign researches. The option of importance density is one of the key steps of particle filter algorithm. The proper option of importance density can minish the negative influence of filter algorithm caused by degeneracy problem. This paper introduces several widely-used options of importance density systemically, and analyzes their features and applied perspectives respectively. The paper also advances a comprehensive method of importance density, analyzes its technical features, explores the adjudgement and improvement of this method based on various performance, and finally puts forward the necessary further study according to the engineer requirements.

        A new tactic analysis of broadcast sports video is proposed based on player trajectory information. Tactic analysis of sports video aims to recognize and discover tactic modes and match strategies that teams and personal players used in the games. Based on players and ball trajectory. First presents an interactive relationship analysis of temporal and spatial algorithm based on time fragment slice. According to the speed and distance of trajectory shape and the distance measurement and each trajectory in time fragment, to use graphical model was constructed to express tactical video game sports event. Through the analysis of each component fragment of interaction trajectory, the offensive incidents of tactical mode carried out hierarchical recognition of coarse to fine in football game video. In the coarse recognition process, the interaction mode is divided into a coordinated attack and personal attack. In fine recognition further, the coordinated attack mode is divided into interception attack and no interception attack. The personal attack mode is divided into direct attack and the dribbling-attack.

        Triptolide inhibits ovarian cancer cell invasion by repression of matrix metalloproteinase 7 and 19 and upregulation of E-cadherin

        Triptolide, a compound extracted from the traditional Chinese medicine preparation of Tripterygium wilfordii Hook F., has been reported to have anti-inflammatory and anti-cancer activities. However, its effect on ovarian cancer invasion is unknown. We observed that MMP7 and MMP19 expression increased in ovarian cancer tissue. Triptolide treatment inhibited the migration and invasion of ovarian cancer cells SKOV3 and A2780 at the concentration of 15 nM. We also observed that triptolide suppressed MMP7 and MMP19 promoter activity in a dose-dependent manner,down-regulating the expressions of these promoters on mRNA and protein level. Moreover, triptolide enhanced E-cadherin expression in ovarian cancer cells. In vivo, triptolide inhibited tumor formation and metastasis in nude mice, and suppressed MMP7 and MMP19 expression; it also enhanced E-cadherin expression in tumor in a dose-dependent manner. Over expression of MMP7 and MMP19, or suppression of E-cadherin expression partially abolished the inhibitory effect of triptolide on invasion of ovarian cancer cells. To summarize, triptolide significantly inhibited the migration and invasion of ovarian cancer cells by suppression of MMP7 and MMP19 and up-regulation of E-cadherin expression. This study shows that triptolide is a good candidate for the treatment of ovarian cancer and reduction of metastasis.
