        Background : Serum osteocalcin is synthesized by osteoblast and has been shown to be sensitive indicator of bone turnover inpatients with various metabolic bone disease. In renal osteodystrophy, serum osteocalcin is elevated due to decreased renal clearance and elevated level of PTH. This study was done to evaluate the usefulness of serum osteocalcin as a marker of bone metabolism and the correlation with other biochemical markers of bone metabolism. Methods : We measured serum osteocalcin, calcium, phosphorus, ALP(alkaline phosphatase) and PTH(parathyroid hormone) in 37 patients with end stage renal disease on hemodialysis. Osteocalcin was determined by radioimmunoassay and PTH was determined by radioimmunometric assay. Results : 1) The mean level of serum osteocalcin in ESRD patients was 233.8± 218.2ng/ml which was significantly higher than that of controls(p<0.0001). 2) The mean level of serum PTH in ESRD patients was 40.5± 43.8pg/ml was significantly higher than that of controls(p<0.005). 3) There was a significant positive correlation between the level of serum PTH, ALP and the level of serum osteocalcin in ESRD patients. 4) By using multiple regression, PTH is most reliable factor that affect to elevated level of serum osteocalcin ( beta coefficient = 0.687, Sig T<0.05). Conclusion : Serum osteocalcin as a marker of bone metabolism in ESRD patients is more useful than other biochemical marker such as serum calcium, phosphorus, ALP and PTH is a most reliable factor that affect to elevated level of serum osteocalin.

        삼출건비탕의 HPLC-PDA 동시 분석법 설정 및 세포독성

        Objectives: To develop and validate HPLC-PDA methods for simultaneous determination of seven constituents in Samchulkunbi-tang (SKT). Additionally, we investigated the cytotoxicity against BEAS-2B cell line and splenocytes of SKT. Methods: Reverse-phase chromatography using a Gemini C18 column operating at 40℃, and photodiode array (PDA) detection at 230, 254 and 280nm, were used for quantification of the three marker components of SKT. The mobile phase using a gradient flow consisted of two solvent systems. Solvent A was 1.0% (v/v) aqueous acetic acid and solvent B was acetonitrile with 1.0% (v/v) acetic acid. The cytotoxicity of SKT were measured by the CCK-8 assay method. Results: Calibration curves were acquired with r2>0.9999, and the relative standard deviation (RSD) values (%) for intra- and inter-day precision were less than 6.0%. The recovery rate of each compound was in the range of 86.89-109.78%, with an RSD less than 4.0%. The contents of seven compounds in SKT were 1.39-6.84mg/g. SKT had no cytotoxicity effect at 50-200ug/mL concentrations. Conclusions: The established method will be helpful to improve quality control and in vitro efficacy study of SKT.

      • Evaluation of Oral Subchronic Toxicity of Soshiho-Tang Water Extract: The Traditional Herbal Formula in Rats

        <P>Soshiho-tang (Xiao-chai-hu-tang in Chinese and Sho-saiko-to in Japanese) has been widely used for its various pharmacological effects, which include anti-inflammatory, antioxidant, antihepatic fibrosis, and antitumor properties. To evaluate the safety of Soshiho-tang water extract (SST), we tested its subchronic toxicity in male and female Crl:CD (SD) rats. Rats were orally treated with four different doses (0, 500, 1000, and 2000 mg/kg/day) of SST administered for 13 weeks. Mortality, clinical signs, body weight changes, food and water consumption changes, ophthalmology, urinalysis, hematological and biochemical parameters, gross findings, organ weights, and histological markers were monitored during the study. The SST treatment did not result in any toxicologically significant changes in mortality, clinical signs, body weights, food and water consumption, ophthalmoscopy, urinalysis, hematological and serum biochemical parameters, gross findings, organ weights, or histopathology. Histological analysis did not show any liver or kidney alteration. We concluded that the 13-week repeated oral administration of SST did not cause any adverse effects in rats at dosage levels of ≤2000 mg/kg/day. Under these experimental conditions, the no-observed-adverse-effect level was concluded to be 2000 mg/kg/day for both sexes.</P>

      • Inhibitory activity of Socheongryong-tang and its constituent components against the production of RANTES, eotaxin, eotaxin-3 and MMP-9 from BEAS-2B cells

        <P>Socheongryeong?tang (SCRT) is a herbal formula previously used to treat pulmonary diseases primarily caused by the common cold virus, including airway inflammation, asthma and allergy. The aim of the present study was to investigate the inhibitory effect of SCRT water extract and its 13 constituent components on chemokine and enzyme production in the human bronchial epithelium cell line BEAS?2B when induced by tumor necrosis factor?α and interleukin?4. The chemokines examined included regulated on activation of normal T?cell?expressed?and?secreted (RANTES), eotaxin and eotaxin?3. The SCRT water extract demonstrated a dose?dependent inhibition of RANTES, eotaxin, eotaxin?3 and matrix metalloproteinase?9 (MMP?9) in BEAS?2B cells. The 13 constituent compounds of SCRT water extract were quantitatively determined, and it was found that gallic acid, 6?gingerol and methyl eugenol produced the most potent inhibition of RANTES, eotaxin and eotaxin?3 as well as MMP?9 activity regardless of their concentration in SCRT water extract. Principal component analysis and hierarchical clustering analysis revealed that the inhibitory effect of these three compounds contributed to that of SCRT water extract. In conclusion, the results of the present study indicated that the inhibitory effects of SCRT on chemokine and enzyme production in BEAS?2B cells was associated with three of its constituent compounds, gallic acid, 6?gingerol and methyl eugenol. This therefore suggested the potential use of these compounds as anti?inflammatory agents.</P>

        IFN-γ Down-Regulates TGF-β1-Induced IgA Expression through Stat1 and p300 Signaling

        IFN-γ has been shown to either up- or down-regulate the expression of specific TGF-β1-induced target genes. We investigated the effect of IFN-γ on TGF-β1-induced IgA isotype expression. We found that IFN-γ inhibited not only TGF-β1-induced germ-line (GL) α transcription, but also IgA secretion by TGF-β1-stimulated murine B cells. Overexpression of Stat1 diminished TGF-β1-induced, Smad3/4-and Runx3-mediated GLα promoter activity. Overexpression of p300 also increased the promoter activity, while its effect was abrogated by co-transfected Stat1. Stat1 interfered with the Smad3:p300 interaction, likely due to a stronger Stat1:p300 binding affinity. These results indicate that Stat1 can inhibit GLα transcription through binding to p300. Further, overexpression of SOCS1, a JAK inhibitor,diminished the antagonistic effect of IFN-γ on TGF-β1-induced GLα transcription and IgA secretion. These results indicate that JAK/Stat1-mediated IFN-γ signaling antagonizes TGF-β1-induced GLα transcription, mainly through deprivation of p300 from Smad3, resulting in decreased IgA synthesis.

        아로마테라피가 간호대학생의 스트레스, 불안 및 우울에 미치는 영향

        본 연구는 아로마 손 마사지와 아로마 목걸이 향기 흡입법이 간호대학생의 스트레스, 불안 및 우울에 미치는 효과를 파악하기 위하여 편의추출 대조군 전후설계를 이용한 유사 실험연구이다. 연구는 J시 소재 대학교 간호학과 4학년에 재학 중인 44명을 대상으로 하였으며, 22명은 아로마 목걸이 향기 흡입군에, 22명은 아로마 손 마사지군에 소속되어 적용법을 교육 받고 1일 2회, 5일간 자가 적용하도록 하였다. 실험처치 전과 후 자가 응답 설문지로 스트레스, 피로 및 우울의 정도를 측정하였다. 연구결과, 아로마 손 마사지와 아로마 목걸이 향기 흡입법, 두 방법 모두 간호학과 4학년 재학생의 불안 및 우울을 감소시키는데 효과적인 방법임을 확인하였고, 스트레스는 아로마 손 마사지군에서만 유의한 감소를 보였다. 본 연구에서 아로마 손 마사지가 아로마 목걸이 향기 흡입법을 사용하는 것보다 스트레스, 불안 및 우울을 감소시키는데 더 효과적이었음을 확인하였으나, 간호대학생을 대상으로 아로마 손마사지를 적용한 선행 연구 논문은 찾아볼 수 없었으므로 효과 검증을 위한 유사 또는 반복연구를 제언한다. The purpose of this study was to examine and compare the effects of aromatherapy using aroma hand massage and aroma inhalation on stress, anxiety, and depression. It was a quasi-experimental study using a convenient sampling method. The participants were 44 4th-year nursing students in J city of Korea. The participants in the aroma hand massage group were given a bottle of blended essential oil with instructions for the hand massage, and the participants in the aroma inhalation group were given an aroma necklace with instructions for inhalation. A set of self-reported questionnaires were given before and after the intervention. After 5 days of intervention, the findings revealed that the levels of stress, anxiety, and depression of the aroma hand massage group had significantly decreased. On the other hand, the changes in the stress level of the aroma inhalation group showed no significance but the levels of anxiety and depression had decreased significantly. Therefore, the study indicated that both types of aromatherapy can be effective in reducing the levels of anxiety and depression of 4th-year nursing students but further studies are needed to clarify the effects of aroma inhalation on stress. Further similar or replicative studies are suggested to compare the effects of different types of aromatherapy.

        Palmul-tang, a Traditional Herbal Formula, Protects against Ethanol-induced Acute Gastric Injury in Rats

        Objectives: Palmul-tang (hachimotsu-to in Japanese and bawu-tang in Chinese) is a mixture of eight herbs. It is traditionally used for the treatment of anemia, anorexia, general weakness, and female infertility in China, Japan, and Korea. In this study, we investigated the protective effects of Palmul-tang water extract (PTE) against ethanol-induced acute gastric injury in rats. Material and Methods: Acute gastric lesions were induced by intragastric administration of 5mL/kg body weight of absolute ethanol to each rat. Control group rats were given PBS orally and the ethanol group (EtOH group) received absolute ethanol (5mL/kg) by oral gavage. The positive control group and the PTE group were given oral doses of omeprazole (50mg/kg) or PTE (400mg/kg), respectively, 2 h prior to the administration of absolute ethanol. The stomach of each animal was excised and examined for gastric mucosal lesions. To confirm the protective effects of PTE, we evaluated the degree of lipid peroxidation, the level of reduced glutathione (GSH), and the activities of the antioxidant enzymes catalase, glutathione-S-transferase, glutathione peroxidase, and glutathione reductase in the stomach. Results: PTE reduced ethanol-induced hemorrhage and hyperemia in the gastric mucosa. PTE reduced the increase in lipid peroxidation associated with ethanol-induced acute gastric lesions and increased mucosal GSH content and the activities of antioxidant enzymes. Conclusion: These results indicate that PTE protects gastric mucosa against ethanol-induced acute gastric injury by increasing antioxidant status. We suggest that PTE could be developed as an effective drug for the treatment of acute gastric injury.

      • Anti-Asthmatic Effects of an <i>Amomum compactum</i> Extract on an Ovalbumin (OVA)-Induced Murine Asthma Model

        <P>Despite ongoing intensive asthma research, the incidence of asthma is increasing worldwide. We investigated in this study the effects of <I>Amomum compactum</I> on ovalbumin (OVA)-induced asthma in a mouse model, and studied the possible mechanism for its anti-asthmatic action. Our data show that an <I>A. compactum</I> treatment markedly decreased the number of infiltrating eosinophils and the hypersecretion of mucus when compared with the effects on mice treated with OVA alone. The <I>A. compactum</I> treatment dose-dependently decreased the levels of reactive oxygen species (ROS) and T helper (Th)2 cytokines, including interleukin (IL)-4 and IL-5, in the bronchoalveolar lavage fluid (BALF), and a high dose of <I>A. compactum</I> effectively reduced the level of total immunoglobulin (Ig)E in the serum. Taken together, these data indicate that the administration of <I>A. compactum</I> may have potential therapeutic value when used as an adjuvant for the immunomodulatory treatment of allergic asthma.</P>

        아로마 손마사지가 산욕기 산모의 스트레스, 피로 및 우울에 미치는 영향

        본 연구는 산욕기 산모의 스트레스, 피로 및 우울에 미치는 효과를 파악하기 위하여 편의추출 대조군 전 · 후 시차설계를 이용한 유사 실험연구다. 연구대상은 J시 소재 2개 산후조리원에 입원한 출산 후 2주 이내 산욕기 산모 32명을 대상으로 하였으며, 실험 군(n=16)은 아로마 오일을 블렌딩한 마사지 오일, 대조군(n=16)은 아로마 오일을 블렌딩하지 않은 마사지 오일을 제공 받아 1일 2회, 5일간 자가 손 마사지를 실시하였다. 실험처치 전과 후 실험 군과 대조군에게 자가 응답 설문지로 스트레스, 피로 및 우울의 정도를 측정하였다. 연구 결과, 실험 군이 대조군보다 실험 후 스트레스, 피로와 우울 증상이 유의하게 감소하여, 아로마 손 마사지 요법이 연구대상자의 스트레스, 피로 및 우울을 감소시키는데 효과적인 방법임을 확인하였다. 따라서, 향후 산후 우울증 예방을 위한 프로그램 개발에 아로마 요법의 사용방안에 대한 노력이 요구된다. 또한, 본 연구의 한계점으로 연구 대상자의 수가 적어 연구의 결과를 일반화하는데 어려움이 있으므로, 산욕기 산모를 대상으로 하는 유사 또는 반복연구를 제언한다. The purpose of this study was to investigate the effect of aroma hand massages on stress, fatigue, and depression among postpartum mothers. It was a quasi-experimental study using a sampling method of the parallax of control and experimental group. The participants were 36 mothers admitted to 2 postpartum caring facilities located in J city of Korea. The aroma blended massage oil was given to the experimental group with the instructions on how to perform a hand massage and the control group was given massage oil without the blended aroma oil. After 5 days of intervention, the findings revealed that the levels of stress, fatigue, and depression among mothers belonging to the experimental group had significantly decreased, but the changes in the control group showed no significance. Therefore, the study indicated that the aroma hand massage was effective in reducing the stress, fatigue, and depression of mothers in the postpartum period, and there was a need for such program development using aroma hand massage to reduce the chance of postpartum depression. The limitation of this study was the relatively small sample size, thus further repetitive studies were suggested.
