        Nuclear Pore Complex Protein Sequences Determine Overall Copolymer Brush Structure and Function

        The transport of cargo across the nuclear membrane is highly selective and accomplished by a poorly understood mechanism involving hundreds of nucleoporins lining the inside of the nuclear pore complex (NPC). Currently, there is no clear picture of the overall structure formed by this collection of proteins within the pore, primarily due to their disordered nature. We perform coarse-grained simulations of both individual nucleoporins and grafted rings of nups mimicking the in vivo geometry of the NPC and supplement this with polymer brush modeling. Our results indicate that different regions or blocks of an individual NPC protein can have distinctly different forms of disorder and that this property appears to be a conserved functional feature. Furthermore, this block structure at the individual protein level is critical to the formation of a unique higher-order polymer brush architecture that can exist in distinct morphologies depending on the effective interaction energy between the phenylalanine glycine (FG) domains of different nups. Because the interactions between FG domains may be modulated by certain forms of transport factors, our results indicate that transitions between brush morphologies could play an important role in regulating transport across the NPC, suggesting novel forms of gated transport across membrane pores with wide biomimetic applicability.

        In Situ Dry Matter, Nitrogen and Phosphorous Disappearance of Different Feeds for Ruminants

        Four feeds, three concentrates (rice bran, soybean meal and flaked corn) and one forage (corn silage) were incubated in four ruminally fistulated Holstein steers over three one week periods in a 3${\times}$4 incomplete latin square design where steers served as blocks and feeds as treatment. The objectives of the study were to investigate in situ DM, N and P degradability characteristics of feeds in order to assess availability of these nutrients by ruminants. In each period, all feeds were incubated in quadruplets (corn silage in triplicates) in the rumen of each steer in a reverse order for 3, 6, 9, 12, 18, 24 and 48 h. The DM 'a' fraction was higher and lower (p<0.001) in corn silage and rice bran respectively. Although corn silage contained the lowest (p<0.01) DM 'b' fraction, flaked corn contained the highest. Rate of DM degradation of flaked corn and corn silage were half (p<0.05) of the rate of DM degradation of either rice bran or soybean meal. Potential or effective DM degradability (p<0.05 to 0.001) at various passage rates were the lowest for rice bran and the highest for soybean meal. Corn silage N 'a' and 'b' was the highest and lowest, respectively (p<0.01). N 'c' of corn silage and rice bran was higher (p<0.001) than other feeds. Potential N degradability was the lowest in flaked corn (p<0.05). P 'a' was high (p<0.01) for corn silage and rice bran. P 'b' fraction was very high (p<0.001) in soybean meal but was absent in corn silage. Availability of DM (p<0.01 or 0.001), N (p<0.001) and P (p<0.05) differed between feeds at various passage rates except P availability at k=0.02 per h (p>0.05). The results demonstrate that the availability of DM, N and P by ruminants depends on feed as well as categories of animal.

        Effect of Variety and Stage of Maturity on Nutritive Value of Whole Crop Rice Silage for Ruminants: In situ Dry Matter and Nitrogen Degradability and Estimation of Metabolizable Energy and Metabolizable Protein

        The effect of eight varieties of whole crop rice silage (WCRS) harvested at four stages of maturity were investigated for in situ DM and N degradability, ME and MP yield and content in an 8${\times}$4 factorial experiment. The varieties were Akichikara, Fukuhibiki, Habataki, Hamasari, Hokuriku 168, Kusanami, Tamakei 96 and Yumetoiro. Hamasari and Kusanami were forage varieties while all others were grain varieties. Forages were harvested on 10, 22, 34 and 45 days after flowering, ensiled and kept in airtight condition. Between 45 and 49 days after ensiling, silages opened, chopped and milled green to pass through 4 mm screen. Samples were incubated in the rumen of two Holstein steers for 0, 3, 6, 9, 12, 24, 48, 72 and 96 h over eight 4 d periods. Bags at 0 h were washed in a washing machine. Variety affected DM (p<0.001: except 'a+b', p<0.01) and N (p<0.001) degradability characteristics of WCRS. Stages of maturity also affected DM (p<0.001: except 'a+b', p<0.05; 'c', p<0.08) and N (p<0.01: except 'c', p<0.05) degradability characteristics of WCRS. Interactions between variety and stages of maturity occurred in all DM (p<0.001) and N (p<0.001) degradability characteristics except (p>0.05) for DM 'b', DM 'c', DM 'a+b' nd N 'c'. Effective DM degradability was higher in grain varieties than forage varieties and degradability increased with maturity. N availability decreased only slightly with maturity. Variety was the key factor for N degradability characteristics of WCRS since variety accounted for most of the total variation for degradability characteristics. Both ME and MP content and yield were higher (p<0.001) in grain varieties, and they increased (p<0.001) with the maturity. The results clearly demonstrated that the grain type varieties contained higher ME and MP content than forage varieties, and increase in maturity increases both ME and MP content of WCRS.

        Effect of Variety and Stage of Maturity on Nutritive Value of Whole Crop Rice, Yield, Botanical Fractions, Silage Fermentability and Chemical Composition

        The effect of eight varieties of grain and forage type whole crop rice (Oryza sativa L Japonica) each harvested at four stages of maturity were investigated for morphology and yield, proportion of botanical fractions, fermentatability and chemical composition in an $8{\times}4$ factorial experiment. All crops were sown in 1997 at Saitama Prefecture, Japan under identical condition and harvested on 10, 22, 34 and 45 days after flowering in 1998. Total DM yield of forage type varieties was similar to that of the highest yield of grain type varieties. However, while yield of forage type varieties was attributed to higher proportion of straw than head, the reverse was in the case of grain type varieties. Yield in line with the proportion of head increased (p<0.001), but in contrast proportion of straw decreased (p<0.001) with the increase in maturity. Silage fermentability of grain type varieties was better than forage type varieties. Fermentability improved with the increase (p<0.001) in maturity suggesting that the moisture content should be reduced to improve fermentation quality. Forage type varieties contained higher (p<0.001) ash, crude fat (EE), organic cell wall (OCW) and acid detergent fiber (ADF), but contained lower crude protein (CP), organic cell content (OCC), CP in OCC and nitrogen-free cell wall extract (NCWFE) than the grain type varieties. The ash, CP, EE, Oa (60% digestible OCW), Ob (40% digestible OCW), OCW, ADF and acid detergent lignin (ADL) decreased (p<0.001), but OCC and NCWFE increased (p<0.001) with the increase in maturity. It is concluded that stage of maturity not only increases yield and proportion of head, but also improved the fermentation quality and increases quality chemical composition (except CP) of whole crop rice. Forage type varieties may be as good as grain type varieties in terms of yield, but fermentation quality and chemical composition may not be as good as that of grain type varieties.

        Estimation of Nutritive Value of Whole Crop Rice Silage and Its Effect on Milk Production Performance by Dairy Cows

        The nutritive value and utilization of whole crop rice silage (WCRS), Hamasari, at yellow mature stage was determined by three studies. In first study, chemical composition, in vivo digestibility and metabolizable energy (ME) content of WCRS was determined by Holstein steers. WCRS contains 6.23% CP, its digestibility is 48.4% and estimated TDN is 56.4%. Its ME content was 1.91 Mcal/kg DM. Gross energy (GE) retention (% of GE intake) in steers is only 22.7% most of which was lost through feces (44.7% of GE intake). It takes 81 minutes to chew a kg of WCRS by steers. In another study, the effect of Hamasari at yellow mature stage at three stages of lactation (early, mid and late lactation) and two levels of concentrate (40 or 60%) on voluntary intake, ME content and ME intake, milk yield and composition using lactating Holstein dairy cows were investigated. Total intake increased with the concentrate level in early and mid lactation, but was similar irrespective of concentrate level in late lactation. WCRS intake was higher with 40% concentrate level than with 60% concentrate. ME intake by cows increased with the concentrate level and WCRS in early lactating cows with 40% concentrate can support only 90% of the ME requirement. Milk production in accordance with ME intake increased with the increase in concentrate level in early and mid lactating cows but was similar in late lactating cows irrespective of concentrate level. Fat and protein percent of milk in mid and late lactating cows were higher with for 60% concentrate than 40%, but reverse was in early lactating cows. Solids-not-fat was higher with for 60% concentrate than 40% concentrate. Finally in situ degradability of botanical fractions such as leaf, stem, head and whole WCRS, Hamasari at yellow mature stage was incubated from 0 to 96 h in Holstein steers to determine DM and N degradability characteristics of botanical fractions and whole WCRS. Both DM and N solubility, rate of degradation and effective degradability of leaf of silage was lower, but slowly degradable fraction was higher compared to stem and head. Solubility of DM and N of stem was higher than other fractions. The 48 h degradability, effective degradability and rate of degradation of leaf were always lower than stem or head. In conclusion, voluntary intake of silage ranged from 5 to 12 kg/d and was higher with low levels of concentrate, but milk yield was higher with high levels of concentrate. Fat corrected milk yield ranged from 19 to 37 kg per day. For consistency of milk, early lactating cows should not be allowed more than 40% whole crop rice silage in the diet, but late lactating cows may be allowed 60% whole crop rice silage.

        This paper presents a circuit of the quasi-resonant DC link to achieve soft-switching three phase inverter using intelligent IGBT power module. The soft-switching operation in this circuit is confirmed simulation and experimental results. Its conductive noise is measured for electrical AC motor drive as compared with that of the conventional hard switching inverter.<br/>
