        Processing and Assembly in vitro of Engineered Soybean b-Conglycinin Subunits with the Asparagine-Glycine Proteolytic Cleavage Site of 11S Globulins

        A short interdomain sequence between the N- and Cterminal domains of b-conglycinin, the major 7S seed storage protein of soybean, was selected as a target for insertion of amino acid residues specifically cleaved by an asparaginyl endopeptidase that processes 11S globulins into acidic and basic chains. Modified b- conglycinin subunits containing the proteolytic cleavage site self-assembled into trimers in vitro at an efficiency similar to that of the unmodified subunit. In contrast to the absence of cleavage of the unmodified subunits, however, the modified b-conglycinin trimers were processed by purified soybean asparaginyl endopeptidase into two polypeptides, each the size expected for the b-conglycinin N- and C-terminal domains, respectively. The cleavage did not alter the assembly of mutant b-conglycinins and the cleaved mutant trimers remained stable to further proteolytic attack. To examine the possibility of coassembly between the cleaved 11S and 7S subunits, in vitro processed mutant b-conglycinin subunits were mixed with native dissociated 11S globulin preparations. Reassembly at a high ionic condition did not induce the 7S subunits to interact with 11S subunits to form hexameric complexes.         Effect of the restorative technique on load-bearing capacity, cusp deflection, and stress distribution of endodontically-treated premolars with MOD restoration

        Objectives: To evaluate the influence of the restorative technique on the mechanical response of endodontically-treated upper premolars with mesio-occluso-distal (MOD) cavity. Materials and Methods: Forty-eight premolars received MOD preparation (4 groups, n = 12) with different restorative techniques: glass ionomer cement + composite resin (the GIC group), a metallic post + composite resin (the MP group), a fiberglass post + composite resin (the FGP group), or no endodontic treatment + restoration with composite resin (the CR group). Cusp strain and load-bearing capacity were evaluated. One-way analysis of variance and the Tukey test were used with α = 5%. Finite element analysis (FEA) was used to calculate displacement and tensile stress for the teeth and restorations. Results: MP showed the highest cusp (p = 0.027) deflection (24.28 ± 5.09 µm/µm), followed by FGP (20.61 ± 5.05 µm/µm), CR (17.72 ± 6.32 µm/µm), and GIC (17.62 ± 7.00 µm/µm). For load-bearing, CR (38.89 ± 3.24 N) showed the highest, followed by GIC (37.51 ± 6.69 N), FGP (29.80 ± 10.03 N), and MP (18.41 ± 4.15 N) (p = 0.001) value. FEA showed similar behavior in the restorations in all groups, while MP showed the highest stress concentration in the tooth and post. Conclusions: There is no mechanical advantage in using intraradicular posts for endodontically-treated premolars requiring MOD restoration. Filling the pulp chamber with GIC and restoring the tooth with only CR showed the most promising results for cusp deflection, failure load, and stress distribution.

        Simultaneous feeding of calcium butyrate and tannin extract decreased the incidence of diarrhea and proinflammatory markers in weaned piglets

        Objective: This study was conducted to investigate the effect of associating calcium butyrate with tannin extract, compared to an antimicrobial on the growth performance, incidence of diarrhea, intestinal histology, immune-expression of cyclooxygenase-2 (COX-2) and tumor necrosis factor α (TNF-α) in piglets. Methods: Seventy-two piglets (36 barrows and 36 gilts) weaned at 28±2 d and initial body weight of 7.17±1.07 kg were allocated to 3 treatments in a randomized complete block design with 8 replicates per treatment and 3 animals per experimental unit. Treatments were composed of NC, negative control: basal diet without additives; PC, positive control: basal diet + 40 mg/kg of colistin sulfate; or BT, basal diet + calcium butyrate + tannin extract. The butyrate and tannin inclusion levels were 0.15% in the pre-starter phase and 0.075% in the starter phase. Incidence of diarrhea was monitored daily, and on d 14 and 35 of experiment, 1 animal from each experimental unit was slaughtered to collect intestinal samples. Results: No significant differences were observed for growth performance. The butyrateand tannin-based additive resulted in reduced (p<0.05) incidence of diarrhea in piglets during d 1 to 14 and d 1 to 35 in comparison with the other treatments. Piglets that consumed the diet containing the calcium-butyrate and tannin showed a lower (p<0.05) crypt depth in the duodenum than those receiving the NC treatment at 14 d of experimentation. The BT treatment provided a lower (p<0.05) immune-expression of COX-2 at 14 d and TNF-α at 35 d in the duodenum. Conclusion: Association between calcium butyrate and tannin extract resulted in a significant decrease in the incidence of diarrhea and inflammatory process in the duodenum of piglets. Therefore, calcium-butyrate combined with tannin could be a part of an alternative program to reduce the use of antimicrobials in the diet of weaned piglets.         Ultrathin Polymer Fibers Coated with an Amorphous SiO2–CaO–P2O5 Bioactive Powders for Biomedical Applications

        The development of new biomaterials with improved properties is a trend in tissue regeneration. In this way, an innovative approach is employed in this work for obtaining polymer fibers coated with nanoparticles resulting from the simultaneous application of poly (lactic acid) (PLA)/polycaprolactone (PCL) electrospinning and bioactive particles from an amorphous multicomponent silica–calcium–phosphorus system (AMS) electrospraying. The osteogenesis was evaluated in vitro and in vivo using male rats, in which total protein, alkaline phosphatase, and biological performance through histological and histomorphometric analysis were discussed. The morphological results assessed by scanning electron microscopy showed a mesh of PLA/PCL fibers associated with AMS. The spraying of 17.44% of AMS particles in the PLA/PCL electrospun fibers decreased the Young’s modulus and tensile strength. However, the amount of AMS particles sprayed was enough to promote a reduction of 17.8% in the measured contact angle values. Phosphatase alkaline higher mean value was also observed in the fibers when in contact with the AMS, but nonstatistical difference was observed (p > 0.05). It was possible to observe the presence of mineralized nodules deposited on the bottom of the plate and between the fibers. The newly formed bone into defect filled with PLA/PCL-AMS fibers was higher than that observed in the control group. These findings suggest PLA/PCL-AMS fibers as a multifunctional composite system that may be attractive for both bone and dental tissue engineering applications.

        Acute and a 28-repeated dose toxicity study of commercial oleoresin from Copaifera sp. in rodents

        Copaifera spp. (copaiba) oleoresin is traditionally used as a medicinal compound since the 16th century, which is primarily indicated for its anti-inflammatory properties being widely sold in free fairs and medicinal herbal houses in Brazil. However, copaiba oleoresin toxicity information is still limited. In this study, we aimed to investigate the acute (in mice and rats) and 28 day—repeated doses (in rats) oral toxicity of commercial copaiba oleoresin. In the acute toxicity test, copaiba oleoresin oral administration at 2000 mg/kg dose did not induce mice or rat lethality. In the subacute toxicity tests, we administered copaiba oleoresin at daily doses of 25, 50, or 100 mg/kg for 28 days. We observed no toxicological effects in body weight gain, feed and water intake, gross necropsy, relative organ weight, histopathology, hematology, or biochemistry parameters. At the three copaiba oleoresin tested doses, we observed an alkaline phosphatase reduction, which might indicate hepatic protection. In conclusion, the commercial copaiba oleoresin has a low risk of toxicity and did not induce treatment-related adverse effects after short-term daily exposures to a dose two thousand times higher than folk use, showing safety doses for further pharmacological investigations.
