        Maintaining Drosha expression with Cdk5 inhibitors as a potential therapeutic strategy for early intervention after TBI

        Traumatic brain injury (TBI) is a major cause of death and disability in adults. The pathological process of TBI involves a multifactorial cascade in which kinases have been proven contribute to interactions between relevant factors and amplification of signaling cascades. Cyclin-dependent kinase 5 (Cdk5) is a promising kinase that has been implicated in various brain disorders, including TBI. However, the mechanism by which Cdk5 induces neuronal damage remains unclear. Here, we show for the first time that Drosha, a key enzyme in microRNA biogenesis, is a pivotal substrate of abnormally activated Cdk5. Cdk5-mediated phosphorylation decreases Drosha expression and exacerbates nerve injury in TBI. We proved that maintaining Drosha expression via the administration of repurposed Cdk5 inhibitors that were previously studied in clinical trials is a promising approach for the early treatment of TBI. Together, our work identifies Drosha as a novel target for neuroprotective strategies after TBI and suggests Cdk5-mediated regulation of Drosha expression as a potential therapeutic strategy for early TBI intervention.

        In vitro differentiation of spermatogonial stem cells using testicular cells from Guangxi Bama mini-pig

        In this study, we attempted to establish a culture system for in vitro spermatogenesis from spermatogonial stem cells (SSCs) of Bama mini-pig. Dissociated testicular cells from 1-month-old pigs were co-cultured to mimic in vivo spermatogenesis. The testicular cells were seeded in minimum essential medium alpha (a-MEM) supplemented with Knockout serum replacement (KSR). Three-dimensional colonies formed after 10 days of culture. The colonies showed positive staining for SSC-associated markers such as UCHL1, PLZF, THY1, OCT4, Dolichos biflorus agglutinin, and alkaline phosphatase. Induction of SSCs was performed in a-MEM + KSR supplemented with retinoic acid, bone morphogenetic protein 4, activin A, follicle-stimulating hormone, or testosterone. The results showed that STRA8, DMC1, PRM1, and TNP1 were upregulated significantly in the colonies after induction compared to that in testis from 1-month-old pigs, while expression levels of those genes were significantly low compared to those in 2-month-old testis. However, upregulation of ACROSIN was not significant. Replacement of a-MEM and KSR with Iscove’s modified Dulbecco’s medium and fetal bovine serum did not upregulate expression of these genes significantly. These results indicate that SSCs of Bama mini-pig could undergo differentiation and develop to a post-meiotic stage in α-MEM supplemented with KSR and induction factors.

      • Anti-Diabetic Medications Do Not Influence Risk of Lung Cancer in Patients with Diabetes Mellitus: a Systematic Review and Meta-analysis

        Objectives: Several preclinical and observational studies have shown that anti-diabetic medications (ADMs) may modify the risk of lung cancer. We performed a systematic review and meta-analysis evaluating the effect of metformin, sulfonylureas (SUs), thiazolidinediones (TZDs), and insulin on the risk of lung cancer in patients with diabetes mellitus (DM). Materials and Methods: We conducted a systematic search of Pubmed and Web of Science, up to August 20, 2013. We also searched the Conference Proceedings Citation Index (CPCI) and China National Knowledge Infrastructure (CNKI) for abstracts from major meetings. Fixed or random effect pooled measures were selected based on heterogeneity among studies, which was evaluated using Q test and the I2 of Higgins and Thompson. Meta-regression was used to explore the sources of between-study heterogeneity. Publication bias was analyzed by Begg's funnel plot and Egger's regression test. Associations were assessed by odds ratios (ORs) with 95% confidence intervals (CIs). Results: A total of 15 studies (11 cohort, 4 case-control) were included in this meta-analysis. In observational studies no significant association between metformin (n=11 studies; adjusted OR=0.99, 95%CI: 0.87-1.12), SUs (n=5 studies; adjusted OR=0.98, 95%CI: 0.79-1.22), or TZDs (n=7 studies; adjusted OR=0.92, 95%CI: 0.75-1.13), insulin (n=6 studies; adjusted OR=1.13, 95%CI: 0.79-1.62) use and risk of developing lung cancer was noted. There was considerable inherent heterogeneity between studies not explained by study design, setting, or location. Conclusions: Meta-analysis of existing studies does not support a protective or harmful association between ADMs use and risk of lung cancer in patients with DM. There was considerable heterogeneity across studies, and future, well-designed, prospective studies would be required for better understanding of any association.

        The Relationship Between Preoperative Cervical Sagittal Balance and Clinical Outcome of Patients With Hirayama Disease Treated With Anterior Cervical Discectomy and Fusion

        Objective: Anterior cervical discectomy and fusion (ACDF) is a common surgical method used to treat patients with Hirayama disease. And sagittal balance indexes have been revealed to be predictors of clinical outcomes in patients with cervical diseases, but their relationships with ACDF-treated Hirayama disease outcomes remain unknown. The purpose of this study is to evaluate the relationship of preoperative cervical sagittal balance indexes and clinical outcomes in ACDF-treated Hirayama disease patients. Methods: Eighty patients with Hirayama disease treated by ACDF were reviewed retrospectively. Six cervical sagittal balance parameters were collected including Cobb angle, T1 slope, C1–7 sagittal vertical axis (SVA), C2–7 SVA, center of gravity of the head (CGH)-C7 SVA, range of motion. The recovery outcomes of the patients were divided into 2 groups by Odom score and the differences in recovery between the 2 groups were confirmed by electromyography. The correlation between imaging parameters and postoperative outcome was evaluated with logistic regression. The receiver operating characteristic (ROC) curve and area under the ROC curve (AUC) were used to evaluate the significant result of logistic regression and the optimal diagnostic value. Results: Only 2 parameters, Cobb angle and CGH-C7 SVA, showed statistical correlation with the postoperative outcome assessment by logistic regression. AUC of Cobb angle and CGH-C7 SVA were 0.559 and 0.702 respectively. The optimal predictive threshold was 1.50° and 5.40 mm, respectively. Conclusion: A larger Cobb angle and smaller CGH-C7 SVA seemed to correlate with a better postoperative outcome. These 2 factors could be used to predict the outcome of surgical treatment of Hirayama disease preoperatively.

        Molecular characterization and expression of ZAP-70 in Nile tilapia (Oreochromis niloticus) in response to Streptococcus agalactiae stimulus

        The 70 kDa zeta-chain associated protein kinase (ZAP-70) plays a critical role in effective signal transductions that are fundamental to T cell differentiation, proliferation and effector functions. In this paper, the ZAP- 70 gene of Nile tilapia, Oreochromis niloticus (designated as On-ZAP-70) was cloned and its expression pattern under the stimulation of Streptococcus agalactiae was investigated. Sequence analysis showed important structural characteristics required for TCRs signal transduction were detected in the deduced amino acid sequence of On-ZAP- 70, and the deduced genomic structure of On-ZAP-70 was similar to the known ZAP-70. In healthy tilapia, the On- ZAP-70 transcripts were mainly detected in the thymus, spleen, head kidney and gill. Moreover, there was a clear time-dependent expression pattern of On-ZAP-70 after immunization and the expression reached the highest level at 48 h in the spleen and head kidney, and at 72 h in the thymus, respectively. This is the first report on the expression of ZAP-70 induced by intracellular bacteria vaccination in teleosts. These findings indicated that On- ZAP-70 may play an important role in the immune response to intracellular bacteria in Nile tilapia.

        A SM2 Elliptic Curve Threshold Signature Scheme without a Trusted Center

        Threshold signature is very important in identity authentication and some other applications. In December 2010, Chinese Encryption Administration released the SM2 elliptic curve digital signature algorithm as the first standard of the digital signature algorithm in China. At present, the papers on the threshold signature scheme based on this algorithm are few. A SM2 elliptic curve threshold signature scheme without a trusted center is proposed according to the Joint-Shamir-RSS algorithm, the Joint-Shamir-ZSS algorithm, the sum or diff-SS algorithm, the Mul-SS algorithm, the Inv-SS algorithm and the PM-SS algorithm. The proposed scheme is analyzed from correctness, security and efficiency. The correctness analysis shows that the proposed scheme can realize the effective threshold signature. The security analysis shows that the proposed scheme can resist some kinds of common attacks. The efficiency analysis shows that if the same secret sharing algorithms are used to design the threshold signature schemes, the SM2 elliptic curve threshold signature scheme will be more efficient than the threshold signature scheme based on ECDSA.

        Optimal Network Defense Strategy Selection Based on Markov Bayesian Game

        The existing defense strategy selection methods based on game theory basically select the optimal defense strategy in the form of mixed strategy. However, it is hard for network managers to understand and implement the defense strategy in this way. To address this problem, we constructed the incomplete information stochastic game model for the dynamic analysis to predict multi-stage attack-defense process by combining Bayesian game theory and the Markov decision-making method. In addition, the payoffs are quantified from the impact value of attack-defense actions. Based on previous statements, we designed an optimal defense strategy selection method. The optimal defense strategy is selected, which regards defense effectiveness as the criterion. The proposed method is feasibly verified via a representative experiment. Compared to the classical strategy selection methods based on the game theory, the proposed method can select the optimal strategy of the multi-stage attack-defense process in the form of pure strategy, which has been proved more operable than the compared ones.

        Network Security Situation Assessment Method Based on Markov Game Model

        In order to solve the problem that the current network security situation assessment methods just focus on the attack behaviors, this paper proposes a kind of network security situation assessment method based on Markov Decision Process and Game theory. The method takes the Markov Game model as the core, and uses the 4 levels data fusion to realize the evaluation of the network security situation. In this process, the Nash equilibrium point of the game is used to determine the impact on the network security. Experiments show that the results of this method are basically consistent with the expert evaluation data. As the method takes full account of the interaction between the attackers and defenders, it is closer to reality, and can accurately assess network security situation.

      • Prediction of the compressive and tensile strength of HPC concrete with fly ash and micro-silica using hybrid algorithms

        Evaluating the impact of fly ash (FA) and micro-silica (MS) on the tensile (TS) and compressive strength (CS) of concrete in different ages provokes to find the effective parameters in predicting the CS and TS, which not only could be usable in the practical works but also is extensible in the future analysis. In this study, in order to evaluate the effective parameters in predicting the CS and TS of concrete containing admixtures and to present a fitted equation, the multivariate adaptive regression splines (MARS) method has been used, which could find a relationship between independent and dependent variables. Next, for optimizing the output equation, hybrid genetic algorithm (GA), particle swarm optimization (PSO), and grey wolf optimization (GWO) methods have been utilized to find the optimal conclusions. It could be concluded that for both predictions of CS and TS, all models have the coefficient of determination (R<sup>2</sup>) larger than 0.949 and 0.9138, respectively. Furthermore, between three hybrid algorithms, MARS-PSO could be proposed as the best model to obtain the most accuracy in the prediction of CS and TS. The usage of hybrid MARS-PSO techniques causes a noticeable improvement in the prediction procedure.

        Temporal gas temperature of atmospheric pressure air plasma

        In this paper, temporal gas temperature in plasma was measured by Rayleigh scattering in a passive way since synchronization was difficult due to the randomness of current pulses. The plasma was generated between a 10 mm pin-to-plane gap connected to a H.V DC voltage through a 130 MΩ resistor and a skin sample was placed on a grounded plate. Even the plasma can be touched by a human hand without any feeling of warmth, the peak temperature could be 337 K then decrease to 295 K over 60 μs at 1 mm. Moreover, the applied voltage dramatically affects peak current and the peak temperature. Therefore, the transient “high” temperature cannot be touched and the so-called “cold” plasma might not be “cold”.
