        Panaxadiol saponins treatment caused the subtle variations in the global transcriptional state of Asiatic corn borer, Ostrinia furnacalis

        Background: The lepidopteran Asiatic corn borer (ACB), Ostrinia furnacalis (Guenee), has caused huge economic losses throughout the Asian-Western Pacific region. Usually, chemical pesticides are used for the control, but excessive use of pesticides has caused great harm. Therefore, the inartificial ecotypic pesticides to ACB are extremely essential. In our previous study, we found that panaxadiol saponins (PDS) can effectively reduce the harm of ACB by causing antifeedant activity. Therefore, it is necessary to reveal the biological molecular changes in ACB and the functionary mechanism of PDS. Methods: We analyzed the global transcription of ACB with different PDS concentration treatment (5 mg/mL, 10 mg/mL, and 25 mg/mL) by high-throughput sequencing and de novo transcriptome assembly method. Results: PDS treatment could cause the changes of many gene expressions which regulate its signal pathways. The genes in peroxisome proliferator-activated receptor (PPAR) signaling pathway were significantly downregulated, and then, the downstream fatty acid degradation pathway had also been greatly affected. Conclusion: Through this experiment, we hypothesized that the occurrence of antifeedant action of ACB is because the PDS brought about the downregulation of FATP and FABP, the key regulators in the PPAR, and the downregulation of FATP and FABP exerts further effects on the expression of SCD-1, ACBP, LPL, SCP-X, and ACO, which leads to the disorder of PPAR signaling pathway and the fatty acid degradation pathway. Not only that, PDS treatment leads to enzyme activity decrease by inhibiting the expression of genes associated with catalytic activity, such as cytochrome P450 and other similar genes.

        A Spectrogram Based Local Fluctuation Feature for Fault Diagnosis with Application to Rotating Machines

        Rotating machines are one of the most common equipment in modern industry, the health condition of the equipment is closely linked to safety of workers and production eff ectiveness. Thus accurate and robust fault diagnostic approaches are vital to safety production. In practice, diagnostic accuracy is seriously aff ected by noises, especially in low signal-to-noise (SNR) ratio conditions, and the quality of fault features is positively link to the diagnosing accuracy. In consideration of distinguishable feature expression can improve diagnosing result and robust to wider range of experimental conditions, this paper presents a novel spectrogram based local fl uctuation feature (SLFF) for low SNR conditions. Firstly, signals are transformed into spectrograms. Then a feature extracting window bank is established on spectrograms for SLFF. At last, a support vector machine (SVM) is applied as a fault classifi er for evaluating the proposed feature. The proposed SLFF represents the basic spectral shape and variation which leads to robust and well distinguishable feature expression, the feature reveals the diff erences of spectral local variation trends between normal and fault types that can improve the discrimination under the infl uence of strong noises. The eff ectiveness of the proposed method has been proved experimentally in this paper.

      • Test Research on Influence Rule of Subsoiling Pressure to Subsoiling Quality

        Pneumatic subsoiling is a new subsoiling technology which uses the pneumatic fracturing principle. To conduct a research on the influence rules of subsoiling pressure to subsoiling quality, the method combining the soil resistivity test and CFD simulation test will be adopted to carry out a pneumatic subsoiling test through taking the increasing rate of soil porosity and soil surface uplift as evaluation index. The test results indicate that after pneumatic subsoiling, the soil porosity of plow pan increases obviously under different subsoiling pneumatic, effective break the plow pan; with the increase of bulk density of plow pan, the spread of the high pressure gas upwards in the plow pan shows a weakening trend and its spread towards horizontal direction shows a enhancing trend; for the plow pan with larger bulk density, the increasing rate of soil porosity and soil surface uplift will increase with the increasing of subsoiling pneumatic accordingly and for the plow pan with smaller bulk density, its regularity is not obvious. Simultaneously, the subsoiling pneumatic and its technical parameters fit for plow pan with different bulk density will be obtained to provide a basis for in-depth research of pneumatic subsoiling technology and development of Pneumatic subsoiling machine.

      • Study on Defects Edge Detection in Infrared Thermal Image Based on Ant Colony Algorithm

        Edge extraction is an important part in the detection of infrared thermal images. Ant colony algorithm has the characteristics of high efficiency, high noise suppression, and comprehensive information of edge information. The basic principle of ant colony algorithm is analyzed. An ant colony optimization algorithm for image edge detection is established. And to have defective parts for analysis of infrared thermography The ant colony algorithm and the classical Canny operator are compared and analyzed. The experimental results show that the algorithm has high efficiency, comprehensive information and high computational efficiency.

        Expression of Cdc25B mRNA in Duodenal Mucosa of Chicken

        Cdc25B is a mitotic regulator that might act as a starter phosphatase to initiate the positive feedback loop at the entry into mitotic (M) phase. In the present study, distribution of Cdc25B mRNA in duodenal mucosa of the chicken was demonstrated by means of in situ hybridization histochemistry (ISHH) using sense and antisense digoxigenin (DIG)-labeled RNA probes. The results showed that there were many labeled cells distributing in the duodenal mucosa of the adult chicken. Of these labeled cells, 81.60${\pm}$9.63% of Cdc25B mRNA positive cells was distributed in the basilar part and mid-portion of the intestinal gland and 36.21${\pm}$8.81% in the middle and basilar portion of villi of the small intestine of the chicken, respectively. Most of these labeled cells were positive in the regions of the stem cell and proliferation. The signals of ISHH decreased from basilar to upper part in the crypt of Lieberkuhn and weakened in the inferior villi of the duodenum. Moreover, the positive signals were both in the cytoplasm and cell nucleus. However, the labeled cells were negative in both the lamina muscularis mucosae and muscular layer. The results of ISHH suggested the existence of Cdc25B mRNA and vigorous proliferation activities in the duodenal mucosa of adult chicken, replenishing the cells which had sloughed off from the superior part of the villus. Our results provide some molecular evidence for a regular pattern of avian intestinal epitheliosis and functional partition and provide an approach to further study of the locations of Cdc25B in the chicken.

        The Expression Patterns of Cdc25A, Cdc25B, Sox2 and Mnb in Central Nervous System in Early Chicken Embryos

        The sense and antisense digoxigenin-labeled RNA probes of four genes, Cdc25A, Cdc25B, Sox2 and Mnb, were produced by using SP6 and T7 RNA polymerases, respectively, and in vitro transcription. Expression patterns of the four genes were detected by in situ hybridization in HH (Hamburger and Hamilton) stage 10 chick embryos. In general, expression patterns of the four genes were similar. mRNA of the four genes was mostly restricted to the entire CNS (central nervous system). All were confined to an identical region, neural tube, neural groove and caudal neural plate, corresponding to the notochord or spinal cord, but there was some distinction in specific region or in concentration, for example in somites. The overlap in expression at the same developmental stage in the CNS suggests that the four genes may be functional similar or related in CNS development. Expression patterns of the four genes support specific roles of these regulators in the developing CNS.
