Cytophaga hutchinsonii can efficiently degrade crystalline cellulose, in which the cell surface cellulases secreted by the type IX secretion system (T9SS) play important roles, but the degradation mechanism remains unclear, and the anchor mechanism of cellulases on the outer membrane in C. hutchinsonii has not been studied. Here, chu_2177 was identified by transposon mutagenesis and was proved to be indispensable for cellulose utilization in C. hutchinsonii. Disruption of chu_2177 resulted in O-antigen deficiency and chu_ 177 could confer O-antigen ligase activity upon an Escherichia coli waal mutant, indicating that chu_2177 encoded the Ontigen ligase. Moreover, deletion of chu_2177 caused defects in cellulose utilization, cell motility, biofilm formation, and stress resistance. Further study showed that the endoglucanase activity was markedly decreased in the outer membrane but was increased in the culture fluid without chu_2177. Western blot proved that endoglucanase CHU_1336 was not located on the outer membrane but was released in the culture fluid of the Δ2177 mutant. Further proteomics analysis showed that many cargo proteins of T9SS were missing in the outer membrane of the Δ2177 mutant. Our study revealed that the deletion of chu_2177 affected the localization of many T9SS cargo proteins including cellulases on the outer membrane of C. hutchinsonii.
Neutronics analysis of JSI TRIGA Mark II reactor benchmark experiments with SuperMC3.3

Jozef Stefan Institute (JSI), TRIGA Mark II reactor employs the homogeneous mixture of uranium and zirconium hydride fuel type. Since its upgrade, a series of fresh fuel steady state experimental benchmarks have been conducted. The benchmark results have provided data for testing computational neutronics codes which are important for reactor design and safety analysis. In this work, we investigated the JSI TRIGA Mark II reactor neutronics characteristics: the effective multiplication factor and two safety parameters, namely the control rod worth and the fuel temperature reactivity coefficient using SuperMC. The modeling and real-time cross section generation methods of SuperMC were evaluated in the investigation. The calculation analysis indicated the following: the effective multiplication factor was influenced by the different cross section data libraries; the control rod worth evaluation was better with Monte Carlo codes; the experimental fuel temperature reactivity coefficient was smaller than calculated results due to change in water temperature. All the results were in good agreement with the experimental values. Hence, SuperMC could be used for the designing and benchmarking of other TRIGA Mark II reactors.

Temperature is an important environmental factor that can greatly influence the cultivation of Auricularia cornea. In this study, lignin peroxidase, laccase, manganese peroxidase, and cellulose in A. cornea fruiting bodies were tested under five different temperatures (20℃, 25℃, 30℃, 35℃, and 40℃) in three different culture periods (10 days, 20 days and 30 days). In addition, the V4 region of bacterial 16S rRNA genes in the substrate of A. cornea cultivated for 30 days at different temperatures were sequenced using next-generation sequencing technology to explore the structure and diversity of bacterial communities in the substrate. Temperature and culture days had a significant effect on the activities of the four enzymes, and changes in activity were not synchronized with changes in temperature and culture days. Overall, we obtained 487,694 sequences from 15 samples and assigned them to 16 bacterial phyla. Bacterial community composition and structure in the substrate changed when the temperature was above 35℃. The relative abundances of some bacteria were significantly affected by temperature. A total of 35 genera at five temperatures in the substrate were correlated, and 41 functional pathways were predicted in the study. Bacterial genes associated with the membrane transport pathway had the highest average abundance (16.16%), and this increased at 35℃ and 40℃. Generally, different temperatures had impacts on the physiological activity of A. cornea and the bacterial community in the substrate; therefore, the data presented herein should facilitate cultivation of A. cornea.
