      • Comprehensive Bioinformation Analysis of the MRNA Profile of Fascin Knockdown in Esophageal Squamous Cell Carcinoma

        Background: Fascin, an actin-bundling protein forming actin bundles including filopodia and stress fibers, is overexpressed in multiple human epithelial cancers including esophageal squamous cell carcinoma (ESCC). Previously we conducted a microarray experiment to analyze fascin knockdown by RNAi in ESCC. Method: In this study, the differentially expressed genes from mRNA expression profilomg of fascin knockdown were analyzed by multiple bioinformatics methods for a comprehensive understanding of the role of fascin. Results: Gene Ontology enrichment found terms associated with cytoskeleton organization, including cell adhesion, actin filament binding and actin cytoskeleton, which might be related to fascin function. Except GO categories, the differentially expressed genes were annotated by 45 functional categories from the Functional Annotation Chart of DAVID. Subpathway analysis showed thirty-nine pathways were disturbed by the differentially expressed genes, providing more detailed information than traditional pathway enrichment analysis. Two subpathways derivated from regulation of the actin cytoskeleton were shown. Promoter analysis results indicated distinguishing sequence patterns and transcription factors in response to the co-expression of downregulated or upregulated differentially expressed genes. MNB1A, c-ETS, GATA2 and Prrx2 potentially regulate the transcription of the downregulated gene set, while Arnt-Ahr, ZNF42, Ubx and TCF11-MafG might co-regulate the upregulated genes. Conclusions: This multiple bioinformatic analysis helps provide a comprehensive understanding of the roles of fascin after its knockdown in ESCC.

      • Protein-protein Interaction Network Analyses for Elucidating the Roles of LOXL2-delta72 in Esophageal Squamous Cell Carcinoma

        Lysyl oxidase-like 2 (LOXL2), a member of the lysyl oxidase (LOX) family, is a copper-dependent enzyme that catalyzes oxidative deamination of lysine residues on protein substrates. LOXL2 was found to be overexpressed in esophageal squamous cell carcinoma (ESCC) in our previous research. We later identified a LOXL2 splicing variant LOXL2-delta72 and we overexpressed LOXL2-delta72 and its wild type counterpart in ESCC cells following microarray analyses. First, the differentially expressed genes (DEGs) of LOXL2 and LOXL2-delta72 compared to empty plasmid were applied to generate protein-protein interaction (PPI) sub-networks. Comparison of these two sub-networks showed hundreds of different proteins. To reveal the potential specific roles of LOXL2- delta72 compared to its wild type, the DEGs of LOXL2-delta72 vs LOXL2 were also applied to construct a PPI sub-network which was annotated by Gene Ontology. The functional annotation map indicated the third PPI sub-network involved hundreds of GO terms, such as "cell cycle arrest", "G1/S transition of mitotic cell cycle", "interphase", "cell-matrix adhesion" and "cell-substrate adhesion", as well as significant "immunity" related terms, such as "innate immune response", "regulation of defense response" and "Toll signaling pathway". These results provide important clues for experimental identification of the specific biological roles and molecular mechanisms of LOXL2-delta72. This study also provided a work flow to test the different roles of a splicing variant with high-throughput data.

      • Shortest Path Analyses in the Protein-Protein Interaction Network of NGAL (Neutrophil Gelatinase-associated Lipocalin) Overexpression in Esophageal Squamous Cell Carcinoma

        NGAL (neutrophil gelatinase-associated lipocalin) is a novel cancer-related protein involves multiple functions in many cancers and other diseases. We previously overexpressed NGAL to analyze its role in esophageal squamous cell carcinoma (ESCC). In this study, a protein-protein interaction (PPI) was constructed and the shortest paths from NGAL to transcription factors in the network were analyzed. We found 28 shortest paths from NGAL to RELA, most of them obeying the principle of extracellular to cytoplasm, then nucleus. These shortest paths were also prioritized according to their normalized intensity from the microarray by the order of interaction cascades. A systems approach was developed in this study by linking differentially expressed genes with publicly available PPI data, Gene Ontology and subcellular localizaton for the integrated analyses. These shortest paths from NGAL to DEG transcription factors or other transcription factors in the PPI network provide important clues for future experimental identification of new pathways.

        Highly efficient hydrogen evolution catalysis based on MoS<sub>2</sub>/CdS/TiO<sub>2</sub> porous composites

        <P><B>Abstract</B></P> <P>Efficient production of hydrogen through visible-light-driven water splitting mechanism using semiconductor-based composites has been identified as a promising strategy for converting light into clean H<SUB>2</SUB> fuel. However, researchers are facing lots of challenges such as light absorption and electron-hole pair recombination and so on. Here, new sheet-shaped MoS<SUB>2</SUB> and pyramid-shaped CdS <I>in-situ</I> co-grown on porous TiO<SUB>2</SUB> photocatalysts (MoS<SUB>2</SUB> CdSTiO<SUB>2</SUB>) are successfully obtained <I>via</I> mild sulfuration of MoO<SUB>3</SUB> and CdO coexisted inside porous TiO<SUB>2</SUB> monolith by a hydrothermal route. The scanning electron microscopy and transmission electron microscopy results exhibit that the MoS<SUB>2</SUB> CdSTiO<SUB>2</SUB> composites have average pore size about 500 nm. The 3%MoS<SUB>2</SUB> 10%CdSTiO<SUB>2</SUB> demonstrated excellent photocatalytic activity and high stability for a hydrogen production with a high H<SUB>2</SUB>-generation rate of 4146 μmol h<SUP>−1</SUP> g<SUP>−1</SUP> under visible light irradiation even without noble-metal co-catalysts. The super photocatalytic performance of the visible-light-driven hydrogen evolution is predominantly attributed to the synergistic effect. The conduction band of MoS<SUB>2</SUB> facilitates in transporting excited electrons from visible-light on CdS to the porous TiO<SUB>2</SUB> for catalytic hydrogen production, and holes to MoS<SUB>2</SUB> for inhibiting the photocorrosion of CdS, respectively, leading to enhancing the efficient separation of electrons and holes.</P> <P><B>Highlights</B></P> <P> <UL> <LI> MoS<SUB>2</SUB>-CT photocatalysts have been successfully synthesized by two-step method. </LI> <LI> The porous structure can enhance photogenerated electron-hole pairs separation. </LI> <LI> The 3% MoS<SUB>2</SUB>-CT shows an excellent H<SUB>2</SUB> evolution rate of 4146 μmol h<SUP>−1</SUP> g<SUP>−1</SUP>. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        Sarcopenia, which is characterized by progressive and generalized loss of skeletal muscle mass and strength, has been well described to be associated with numerous poor postoperative outcomes, such as increased perioperative mortality, postoperative sepsis, prolonged length of stay, increased cost of care, decreased functional outcome, and poorer oncological outcomes in cancer surgery. Multimodal prehabilitation, as a concept that involves boosting and optimizing the preoperative condition of a patient prior to the upcoming stressors of a surgical procedure, has the purported benefits of reversing the effects of sarcopenia, shortening hospitalization, improving the rate of return to bowel activity, reducing the costs of hospitalization, and improving quality of life. This review aims to present the current literature surrounding the concept of sarcopenia, its implications pertaining to colorectal cancer and surgery, a summary of studied multimodal prehabilitation interventions, and potential future advances in the management of sarcopenia.

        Effect of Argon Gas Pressure on Residual Stress, Microstructure Evolution and Electrical Resistivity of Beryllium Films

        The residual stress in beryllium films fabricated on K9 substrates by using magnetron sputtering deposition is measured by using a curvature method and is theoretically estimated by using the Nix and Clemens (NC) model. The experimental results indicate that the 1.3-μm-thick film is always in a tensile state for pressure variations in the range from 0.4 to 1.2 Pa. When the sputtering gas pressure is increased, the average stress increases at first, after which it decreases by a remarkable amount. The observed descending trend of the tensile stress when the sputtering gas pressure is beyond 0.6 Pa is mainly attributed to the grain size in the film being larger than that in the film when the pressure is below 0.6 Pa. The maximal residual stress of 552 MPa at a sputtering gas pressure of 0.6 Pa is close to the tensile strength (550 MPa) of the corresponding beryllium bulk material and is about 8 times smaller than that calculated by using the N-C model. In addition, the surface morphologies of the as-fabricated films reveal fibrous grains while the cross-sectional morphologies are characterized by a coarsening of columnar grains. The measured electric resistivity of each film strongly depends on its porosity and the sizes of its grains.

      • Common Variations of DNA Repair Genes are Associated with Response to Platinum-based Chemotherapy in NSCLCs

        Aim: Individual differences in chemosensitivity and clinical outcome of non-small-cell lung cancer (NSCLC) patients may be induced by host inherited factors. We investigated the impact of XPD Arg156Arg, XPD Asp312Asn, XPD Asp711Asp and XPD Lys751Gln gene polymorphisms on the efficacy of platinum-based chemotherapy in NSCLC patients. Methods: A total of 496 were consecutively selected from the Affiliated Hospital of Nantong University between Jan. 2003 and Nov. 2006, and all patients were followed-up until Nov. 2011. The genotyping of XPD Arg156Arg, XPD Asp312Asn, XPD Asp711Asp and XPD Lys751Gln was conducted by duplex polymerase-chain-reaction with the confronting-two-pair primer methods. Results: Individuals with XPD 312 C/T+T/T and XPD 711 C/T+T/T exhibited poor responses to chemotherapy when compared with the wild-type genotype, with adjusted ORs(95% CI) of 0.67(0.38-0.97) and 0.54(0.35-0.96), respectively. Cox regression showed the median PFS and OS of patients of XPD 312 C/T+T/T genotype and XPD 711 C/T+T/T genotype to be significantly lower than those with wild-type homozygous genotype. Conclusion: We found polymorphisms in XPD to be associated with response to platinum-based chemotherapy in NSCLC, and our findings provide information for therapeutic decisions for individualized therapy.

        CBX7 Rejuvenates Late Passage Dental Pulp Stem Cells by Maintaining Stemness and Pro-angiogenic Ability

        BACKGROUND: Ever-growing tissue regeneration causes pressing need for large population of stem cells. However, extensive cell expansion eventually leads to impaired regenerative potentials. In this study, chromobox protein homolog 7 (CBX7) was overexpressed to rejuvenate late passage dental pulp stem cells (DPSCs-P9). METHODS: The recruitment of copper ions (Cu2?)-activated hypoxia-inducible factor-1a (HIF-1a) to the CBX7 gene promoter was confirmed by chromatin immunoprecipitation assay. Functions subsequent to Cu2?-induced or recombinant overexpression of CBX7 on proliferation, multipotency, odontoblastic differentiation and angiogenesis were investigated in vitro, while murine subcutaneous transplantation model was used to further detect the effects of Cu2?-induced CBX7 overexpression in vivo. RESULTS: Our data displayed that CBX7 overexpression maintain proliferation and multipotency of DPSCs-P9 almost as strong as those of DPSCs-P3. Both gene level of odontoblast-lineage markers and calcium precipitation were nearly the same between CBX7 overexpressed DPSCs-P9 and normal DPSCs-P3. Moreover, we also found upregulated expression of vascular endothelial growth factor in DPSCs-P9 with CBX7 overexpression, which increased the number of capillary-like structures and migrating co-cultured human umbilical vein endothelial cells as well. These findings indicate CBX7 as an effective factor to rejuvenate late passage stem cells insusceptible to cell expansion. Cu2? has been proved to achieve CBX7 overexpression in DPSCs through the initiation of HIF-1a-CBX7 cascade. Under Cu2? stimulation since P3, DPSCs-P9 exhibited ameliorated regenerative potential both in vitro and in vivo. CONCLUSION: Long-term stimulation of Cu2? to overexpress CBX7 could be a new strategy to manufacture large population of self-renewing stem cells.

        A fiber beam element model for elastic-plastic analysis of girders with shear lag effects

        This paper proposes a one-dimensional fiber beam element model taking account of materially non-linear behavior, benefiting the highly efficient elastic-plastic analysis of girders with shear-lag effects. Based on the displacement-based fiber beam-column element, two additional degrees of freedom (DOFs) are added into the proposed model to consider the shear-lag warping deformations of the slabs. The new finite element (FE) formulations of the tangent stiffness matrix and resisting force vector are deduced with the variational principle of the minimum potential energy. Then the proposed element is implemented in the OpenSees computational framework as a newly developed element, and the full Newton iteration method is adopted for an iterative solution. The typical materially non-linear behaviors, including the cracking and crushing of concrete, as well as the plasticity of the reinforcement and steel girder, are all considered in the model. The proposed model is applied to several test cases under elastic or plastic loading states and compared with the solutions of theoretical models, tests, and shell/solid refined FE models. The results of these comparisons indicate the accuracy and applicability of the proposed model for the analysis of both concrete box girders and steel-concrete composite girders, under either elastic or plastic states.
