        Improved Work Function of Preferentially Oriented Indium Oxide Films Induced by the Plasma Exposure Technique

        The preferentially oriented In2O3 thin films were prepared on glass substrates by conventional magnetron sputtering with Ar+ plasma exposure at room temperature. Based on the x-ray diffraction, x-ray photoelectron spectroscopy, and UV photoelectron spectroscopy results, it was found that the Ar+ plasma exposure not only enhanced the low-temperature crystallization of In2O3 thin films, but also led to a dramatic improvement in the work function. Furthermore, it demonstrated that the shift mechanism of the work function in In2O3 thin films mainly combined with theelimination of oxygen defects and the change of the preferential orientation of In2O3 film surface.

        Cockroach as a Vector of Blastocystis sp. is Risk for Golden Monkeys in Zoo

        Blastocystis sp. is a kind of protozoa living in the intestinal tract of human and animals, which will cause intestinal diseases such as diarrhea, abdominal distension and vomiting. This paper was aimed to understand the infection of Blastocystis sp. In golden monkeys and the transmission path in North China. Thirty-seven feces samples from golden monkeys and 116 cockroach samples from Shijiazhuang Zoo were collected from July to October 2019 for PCR analysis of Blastocystis sp. Genetic diversity analysis was further conducted on the samples with positive PCR results. The results showed that the infection rate was 48.7% (18/37) in golden monkeys and 82.8% (96/116) in cockroaches, respectively. The genetic evolution analysis based on small subunit ribosomal RNA demonstrated that three subtypes (ST) of Blastocystis sp. including ST1, ST2, and ST3 existed in the intestinal tract of golden monkeys, while only ST2 was detected in the intestinal tract of cockroaches. This paper may provide supports for the quarantine and control of Blastocystis sp. for the zoo in Northern China.

        Influence of Particle Shape on the Density and Compressive Performance of Calcareous Sand

        Particle shape influence the bearing capacity of calcareous sand foundations in island reef engineering. An improved Computer Image Analysis method that combined with mathematical statistics was proposed to categorize and sort different shapes of calcareous sand particles within the size range of 5 – 10 mm. Three single-shape calcareous sand specimens (granular ᅠ ᅠshape, dendritic shape, long strip shape) were obtained. A new self-developed vibration device was used measuring the maximum density of calcareous, the shape and content as the variables in the orthogonal test. On this basis, the lateral compression tests were carried out to assess the influence of particle shape on the density, compressive performance of calcareous sand. Test results showed that better discriminability was achieved by taking mean flatness (EL/B) as the quantification parameter of the particle shapes. The average flatness EL/B of specimens is approximately normally distributed in the statistical results. The shape and content of sand particles had an obvious influence on the density of calcareous sand. The compressive properties of calcareous sands with different particle shapes have obvious differences. After loaded, the particle size distribution of specimen improved, the average flatness EL/B of the granular-shaped specimens increased, while that of the dendritic-shaped and the long-strip-shaped specimens decreased, the average flatness EL/B of the specimens tended to be uniform. The relative fracture rate Br and the load value Pmax satisfied the exponential function fitting relationship. Therefore, it can be argued that particle shape should be taken into consideration when optimizing the foundation design in island reef engineering to achieve better compressive performance of the foundation.

        Rolling Bearing Incipient Fault Detection via Optimized VMD Using Mode Mutual Information

        The complete failure of the rolling bearing is a deterioration process from the incipient weak fault to the severe fault, thus it is important to alarm when the incipient fault appear. This work presents a novel incipient bearing fault diagnosis framework using mode mutual information (MMI) based fitness function, variational mode decomposition (VMD), and cuckoo search (CS) algorithm. MMI based fitness function is proposed in order to obtain the optimal combinations of the VMD parameters. Therefore, the optimal parameters of VMD can be obtained by CS algorithm using proposed fitness function. Afterwards, a vibration signal is decomposed into a set of modes using the optimal VMD, and the kurtosis value of all modes are calculated. The envelop of the mode with maximum kurtosis value between modes and raw signal is computed as the input vector of the stacked denoised autoencoder (SDAE). Comparisons have been conducted via SDAE to evaluate the performance by using EMD and the fixedparameter VMD. The experimental results demonstrate that the proposed method is more effective in extracting the incipient bearing fault characteristics.

        Mechanical Performance of Prefabricated External Wall Panel under Horizontal Displacement

        In this paper, experiment and Finite Element Method (FEM) approaches are employed to study the mechanical performance of prefabricated external wall panel under lateral displacement. Tensile bond strength test and Z-shaped specimen direct shear test of the bonding interface of mortar to ceramsite concrete are performed, the normal and shear mechanical property of the interface is studied respectively. Scaled model test of prefabricated external wall panel under lateral displacement is then performed to obtain loaddisplacement curve, and mechanical behavior of prefabricated external wall panel in different stages is studied. Moreover, ABAQUS finite element analysis model for scaled model test panel is established based on previous study about bond interface and the results from experiment and FEM analysis are relatively consistent with each other. It is found that with the gradual increase in tangential contact stiffness of joint interfaces, the overall lateral stiffness of wall panels will approach complete joint consolidation conditions before the peak load. However, when the tangential contact stiffness of joint interfaces increases to a certain extent, there will be little change in initial lateral stiffness.

        Geometric Nonlinear Static Aeroelastic Characteristics Analysis of High-Aspect-Ratio Wing with Large Deformation

        To explore the geometric nonlinearity of high-aspect-ratio wing due to large deformation, the aerodynamics were solved by Navier–Stokes governing equation based on the loose coupling method of CFD/CSD. Considering the large displacement stiffness matrix of wing structure, the Newton–Raphson method is used to solve the nonlinear elastic deformation of the structure. A geometrically nonlinear static aeroelastic coupling calculation program is developed. During the coupling process, an improved elastic approximation method was used to update the aerodynamic mesh, and the data exchange on the coupling interface was realized by a fixed-volume interpolation method. The reliability of the proposed method was verified by HIRENASD wing model. Finally, the effects of geometric nonlinear static aeroelasticity on longitudinal aerodynamic characteristics and stability of swept wing with high aspect ratio at different angles of attack are studied using this method. The results show that the lift direction of a high-aspect-ratio wing is significantly deformed under the influence of aerodynamic loads. The effective angle of attack of the wing decreases with the whole deformation, and the degree of decrease increases from the root to the tip. The significant flexural and torsional deformation will change the lift coefficient and drag coefficient of the wing and reduce the margin of the static stability of the wing.

      • Potent Anticancer Effects of Lentivirus Encoding a Drosophila Melanogaster Deoxyribonucleoside Kinase Mutant Combined with Brivudine

        Objective: Deoxyribonucleoside kinase of Drosophila melanogaster (Dm-dNK) mutants have been reported to exert suicide gene effects in combined gene/chemotherapy of cancer. Here, we aimed to further evaluate the capacity of the mutanted enzyme and its potential for inhibiting cancer cell growth. Methods: We altered the sequence of the last 10 amino acids of Dm-dNK to perform site-directed mutagenesis and constructed active site mutanted Dm-dNK (Dm-dNKmut), RT-PCR and western bloting studies were used to reveal the expression of lentivirus mediated Dm-dNKmut in a breast cancer cell line (Bcap37), a gastric cancer cell line (SGC7901) and a colorectal cancer cell line (CCL187). [3H]-labeled substrates were used for enzyme activity assays, cell cytotoxicity was assessed by MTT assays, cell proliferation using a hemocytometer and apoptosis induction by thenannexin-V-FITC labeled FACS method. In vivo, an animal study was set out in which BALB/C nude mice bearing tumors were treated with lentivirus mediated expression of Dm-dNKmut with the pyrimidine nucleoside analog brivudine (BVDU, (E)-5-(2-bromovinyl)-(2-deoxyuridine). Results: The Dm-dNKmut could be stably expressed in the cancer cell lines and retained its enzymatic activity. Moreover, the cells expressing Dm-dNKmut exhibited increased sensitivity in combination with BVDU, with induction of apoptosis in vitro and in vivo. Conclusion: These findings underlined the importance of BVDU phosphorylated by Dm-dNKmut in transduced cancer cells and the potential role of Dm-dNKmut as a suicide gene, thus providing the basis for future intensive research for cancer therapy.

        Effects of Continuous Straw Returning on Soil Functional Microorganisms and Microbial Communities

        This study examined the changes in soil enzymatic activity, microbial carbon source metabolic diversity, and straw decomposition rates in paddy fields treated with 1, 2, or 3 years of straw returning (SR1–SR3). The soil’s ability to decompose straw and cellulolytic bacteria increased with the number of treatment years (1: 31.9% vs. 2: 43.9% vs. 3: 51.9%, P < 0.05). The numbers of Azotobacter, Nitrobacteria, cellulolytic bacteria, and inorganic phosphate bacteria increased progressively with the numbers of straw returning years. Cellulolytic bacteria and inorganic phosphate bacteria were significantly positively correlated with the decomposition rate (r = 0.783 and r = 0.375, P < 0.05). Based on 16S sequencing results, straw returning improved the microbial diversity of paddy soils by increasing unclassified bacteria and keeping dominant soil microorganism populations unchanged. The relative importance of individual microbial taxa was compared using random forest models. Proteobacteria, ammoniating bacteria, and potassium dissolving bacteria contributed to peroxidase activity. The significant contributors to phosphate monoesterase were Acidobacteriota, Desulfobacterota, ammoniating bacteria, cellulolytic bacteria, and potassium-dissolving bacteria. Proteobacteria, ammoniating bacteria, cellulolytic bacteria, and potassium-dissolving bacteria contributed to urease activity. Desulfobacterota, ammoniating bacteria, cellulolytic bacteria, and potassium-dissolving bacteria contributed to the neutral invertase activity. In conclusion, soil microbial community structure and function were affected within 2 years of straw returning, which was driven by the combined effects of soil organic carbon, available nitrogen, available potassium, and pH. With elapsing straw returning years, soil properties interacted with soil microbial communities, and a healthier soil micro-ecological environment would form.

        Identification and characterization of atypical porcine pestivirus genomes in newborn piglets with congenital tremor in China

        Recently, a novel atypical porcine pestivirus (APPV) in pig was reported. In this study, two APPV strains, APPV-China/GZ01/2016 (GZ01) and APPV-China/GD-SD/2016 (GD-SD), were identified in two newborn piglet herds with congenital tremor from China. The open reading frame of the two strains shared an 83.5% nucleotide identity. Phylogenetically, the APPV strains were placed into two groups: GZ01 belonged to group I and GD-SD belonged to group II. A high viral load was detected in the cerebellum (quantification cycles < 26). Further studies should be carried out to thoroughly elucidate the development of congenital tremors caused by APPV.

        Viruses, Bacteria and Helminths of Invasive Carp: Insights from an In Vitro Assay and a Survey with Native Fishes in a Large Midwestern River

        Pathogen introductions associated with aquatic invasive species threaten ecosystems and biodiversity worldwide. Bigheaded carps (BHC), including Silver Carp Hypophthalmichthys molitrix, Bighead Carp H. nobilis, and their hybrids, are prolific, invasive pests in central US rivers. However, little is known about pathogen effects on invading BHC or how BHC affect the disease risk profile for native fishes in receiving ecosystems. We therefore conducted, from May 2013-December 2014, a systematic pathogen survey for BHC and native fishes in the Wabash River watershed, Indiana, USA. We found Pseudomonas fluorescens, P. putida, and Salmonella enterica DNA in BHC as well as native fishes, although none of these bacteria were exclusively present in BHC. DNA from other bacterial taxa was detected only in native fishes and Common Carp Cyprinus carpio. No gastrointestinal helminths were detected in BHC, although they were common in most native fishes examined. We also conducted in vitro studies on BHC tissues (skin, gill, fin, and fry) and found high sensitivity to Largemouth Bass virus, viral hemorrhagic septicemia virus, and infectious pancreatic necrosis virus. We conclude that BHC are not heavily burdened by bacteria, viruses and parasites in the invaded study ecosystems, although they do harbor native bacteria and show potential for high sensitivity to endemic viruses.
