Isolation and Transcriptional Expression of CuZn Superoxide Dismutase from Codonopsis lanceolata

To investigate the defense mechanism against the abiotic stress, a cDNA clone encoding a CuZn superoxide dismutase (CuZnSOD) protein was isolated from a cDNA library prepared from tabroot mRNAs of Codonopsis lanceolata. The eDNA, designated ClSODCc, is 799 nucleotides long and has an open reading frame of 459 bp with a deduced amino acid sequence of 152 residues. The deduced amino acid sequence of ClSODCc matched to the previously reported CuZnSODs. Consensus amino acid residues (His-45, -47, -62, -70, -79, -119 and Asp-82) were involved in Cu-, Cu/Zn-, and Zn- binding ligands. The deduced amino acid sequence of ClSODCc showed high homologies (82%-86%) regardless of species. Expression of ClSODCc by oxidative stress was increased up to 1 h after treatment and declined gradually. Much earlier and stronger expression of ClSODCc was observed in the cold stress treatment.
Physical and chemical Effects on the sonication Treatment of chitosan solution

물리적 방법을 이용한 키토산 올리고당의 제조에 관한 기초 자료를 제시하기 위하여 키토산용액을 20kHz의 초음파로 처리하였으며, 초음파 처리 효과에 미치는 처리 조건의 영향을 조사하였다. 키토산 용액의 고유점도는 초음파 처리 5분까지는 급속하게 감소하였고 그 이후로는 서서히 감소하였다. 키토산 용액의 부피가 작을수록 초음파 처리효과는 컸으며, 키토산 용액의 온도는 초음파 처리에 별다른 효과가 없는 것으로 나타났다. 5분간 초음파 처리 후의 점도는 용매의 종류에 따라 큰 차이가 없었으나, acetate buffer의 경우 초음파 처리효과가 가장 컸다. pH가 높을수록 초음파 처리효과는 크게 나타났으며, 이온강도와 염의 종류에 따른 효과는 나타나지 않았다. 이상의 결과에서, 용액의 부피는 10~20ml, 온도는 20~30℃, 용액의 pH는 4.5, 그리고 용매의 종류는 acetate buffer로 하는 것이 최적 초음파 처리조건으로 추정된다. As the first step of studies related to production of chitooligosaccharides by physical methods, chitosan solution were sonicated with 20kHz and various treatment effects were examined to present fundamental data of sonicated chitosan solution. Intrinsic viscosity of chitosan solution sharply decreased from 3.76dl/g to 2.90dl/g until 5 minutes of sonication and then slowly decreased. With low volume of chitosan solution, sonication was very effective and temperature of chitosan solution slightly affected the efficiency of sonication. In case of changing the solvent, no significant differences were observed on the effect of sonication, however, acetate buffer had highest sonication effect among various solvents. The sonication effect was increased as the increasement of the value of pH, on the contrary, ionic strength and type of counterions showed no effect on sonication, With these results, we assumed that optimal sonication treatment would be as follows, solution volume was 10 ~20ml, temperature range was 20~30℃ , pH value of solution was 4.5 and type of solvent was acetate buffer
Prevalence and characteristics of isolated nocturnal hypertension in the general population

Background/Aims: Ascertaining the prevalence of isolated nocturnal hypertension (INHT) in the general population and identifying the characteristics of patients with INHT may be important to determine patients who should receive 24- hour ambulatory blood pressure (BP) measurements. This study aimed to evaluate the prevalence and characteristics of INHT in the general population. Methods: Of 1,128 participants (aged 20 to 70 years), we analyzed 823 who had valid 24-hour ambulatory BP measurements and were not on antihypertensive drug treatment. Results: The prevalence of INHT in the study was 22.8%. Individuals with INHT had a higher office, 24-hour, and daytime and nighttime ambulatory systolic and diastolic BPs compared to individuals with sustained day-night normotension. INHT was more prevalent in individuals with masked hypertension (MH) than in those with sustained hypertension (59.8% vs. 15.6%, p < 0.001). Among individuals with INHT, 92.6% had MH. Among individuals with office BP-based prehypertension, 34.5% had both INHT and MH. The prevalence of INHT was highest in individuals with office BP-based prehypertension. INHT was an independent determinant of MH after adjustment for age, sex, body mass index, diabetes, low-density-lipoprotein cholesterol, 24-hour systolic and diastolic BP, systolic and diastolic BP dipping, and systolic and diastolic BP non-dipping. Conclusions: The present study showed that INHT is not uncommon and is a major determinant of MH. Our findings strongly suggest the use of 24-hour ambulatory BP measurement for individuals within the prehypertension range of office BP owing to the high prevalence of INHT and MH in this population.
Epidemiological Studies of Avian Paramyxovirus Type 4 and 6 in Commercial Chicken Flocks in Korea

Avian paramyxovirus (APMV) type 4 and 6 were isolated during an avian influenza (AI) surveillance program of wild birds. This study also conducted experimental infection of wild-bird-origin APMV type 4 and 6 in specific pathogen free (SPF) chickens to study pathogenicity and transmission within domestic flocks. In addition, serological prevalence data of APMV type 4 and 6 in domestic fowls was conducted with chicken sera collected from 2007 to 2009 in order to understand infection status. The results of the animal experiment showed that APMV type 4 and 6 had the ability to infect chickens with sero-conversion and to transmit the virus from infected birds to contacted birds, but showed low pathogenicity. Serological tests revealed that APMV type 4 was widespread in the poultry industry, especially in layer flocks, but the positive rate for APMV type 6 was very low. This study concluded that wild bird-origin APMV type 4 and 6 could infect the chickens by inter-species transmission and the seroprevalence of APMV type 4 was quite high in Korean poultry. However, since almost all the chicken flocks had a high level of antibody titer against APMV type 1, there was possibility of cross reaction between APMV type 1 and 4, which made the interpretations more complicated. In order to understand infection status in the natural environment, additional study is necessary regarding the seroprevalence of APMV type 4 and 6 in the wild bird population.

ABSTRACT: Metabolic health and obesity are not stable conditions, and changes in the status of these conditions might lead to different clinical outcomes. We aimed to determine whether changes in metabolic health status or obesity over time have any effect on the risk of future diabetes.Nondiabetic individuals (n = 2692) from a population-based prospective cohort study with baseline and 2 follow-up examinations at 4-year intervals were included. Being “metabolically obese” (MO) was defined as being in the highest quartile of the TyG index (ln [fasting triglycerides (mg/dL) × fasting glucose (mg/dL)/2]), whereas falling into the lower 3 quartiles was regarded as being “metabolically healthy” (MH). Individuals were classified as “obese” (O) or “nonobese” (NO) using a body mass index of 25 kg/m as a cut-off. The risk of diabetes at year 8 was assessed according to changes of metabolic health status between year 0 and 4.Multivariate-adjusted relative risks (RRs) (95% confidence interval [CI]) of diabetes were significantly higher in individuals who retained the MONO phenotype (RR 3.72, 95% CI 2.10, 6.60) or who had progressed to MONO from the MHNO phenotype (RR 1.96, 95% CI 1.06, 3.61), whereas it was not significant in individuals who had improved to MHNO from the MONO phenotype (RR 0.67, 95% CI 0.26, 1.74) compared with individuals who retained the MHNO phenotype. In contrast, obese individuals had significantly higher RRs for diabetes, independent of changes in metabolic health status, whereas weight reduction resulted in a decreased risk of diabetes. Sensitivity analysis using the presence or absence of the metabolic syndrome as a definition of metabolic health revealed similar results.Changes in metabolic health status were an independent risk factor for future diabetes in nonobese individuals, whereas general obesity had a greater contribution to the risk of obese individuals developing diabetes. These observations might imply a different intervention strategy for diabetes prevention according to obesity status.
