      • LC/MS/MS 분석법을 이용한 Amlodipine의 약물동태연구

        The aim of this study were to confirm the analysis method and also to estimate the pharmacokinetic parameters of amlodipine in human volunteers. In an open-label single-dose pharmacokinetic study, a group, consisting of 24 healthy volunteers, received single oral dose of 5mg amlodipine. Blood sample were taken for up to 120 hours. The concentration of amlodipine in these body fluids was determinated using a validated high-performance liquid chromatography(HPLC) method with tandem mass spectrometry. Amlodipine and ketoconazole, an internal standard, were extracted from plasma using ethyl acetate in the presence of 0.1M sodium carbonate. After drying the organic layer, the residue was reconstituted in mobile phase(acetonitrile : water = 70 : 30 v/v (0.1% formic acid)) and injected onto a Zorvax C18 column (2.1 × 100 mm, 3.5 ㎛ particles). The isocratic mobile phase was eluted at 0.2ml/min. The ion transitions monitored in multiple reaction-monitoring mode were m/z 410.10 → 294.95 and 532.11 → 81.95, respectively. The coefficient of variance of the assay precision was less than 12%, and the accuracy exceeded 99.1%. The limit of quantification was 0.1 ng/ml. The pharmacokinetic parameters were calculated with non-compartmental(AUC, C_(max), T_(max), CL_(t), V/F) and compartmental(K_(el), K_(a), t_(lag)) pharmacokinetic analysis using WinNonlin program. The estimated means of AUC_(0-120hr), C_(max) and T_(max) were 196.90 ± 5.02 ng·hr/ml, 3.36 ± 0.09 ng/ml and 10.44 ± 0.61 hr, respectively. The means of other pharmacokinetic parameters(V/F, CL_(t), K_(el), K_(a) and t_(lag)) were 1208.06 ± 50.61 L, 25.39 ± 0.65 L/hr, 0.2806 ± 0.0294 hr^(-1), 0.0210 ± 0.0008 hr-1 and 0.4574 ± 0.0635 hours, respectively.

        골아세포 MC₃T₃-E₁의 증식과 기능에 Insulin Like Growth Factor Ⅰ, Ⅱ 및 부갑상선 호르몬이 미치는 영향

        MC₃T₃-E₁골아세포군을 이용하여 IGF-Ⅰ과 Ⅱ를 투여하고 DNA합성과 골아세포기능을 측정하기 위해 PTH에 대한 C-AMP반응정도, alkaline phosphatase 활성도를 측정하여 다음과 같은 결과를 얻었다. 1) IGF-Ⅰ과 Ⅱ는 모두 DNA합성이 어느 농도까지는 증식효과를 나타내지만 그 이후에는 억제되는 양상을 보였다. 2) PTH에 대한 골아세포의 C-AMP반응으로 증가되었지만 IGF-Ⅰ과 Ⅱ의 투여시 농도변화에 따른 의미있는 변화는 없었다. 3) 본 실험의 조건에서 alkaline phosphatase 활성도는 증가되지 않았다. 이상의 결과로 MC₃T₃-E₁ 골아세포에 IGF-Ⅰ과 Ⅱ는 세포의 증식을 자극하여 PTH는 C-AMP생성을 촉진하는 효과가 있었다. The effect of insulin-like growth factor (IGF) Ⅰ and Ⅱ on cloned MC₃T₃-E₁ cells that had osteoblastic activity was examined by [³H] thymidine incorporation into DNA, cyclic AMP responsiveness to parathyroid hormone (PTH), and alkaline phosphatase activity. IGF Ⅰ and Ⅱ stimulated the growth and proliferation of osteoblastic cells at 20ng/㎖ and 50ng/㎖, respectively. We next examined the effect of IGF-Ⅰ and Ⅱ on the expression of osteoblastic activity. Adenylate cyclase response of PTH to IGF-Ⅰ and Ⅱ-treated cells showed maximal stimulation at 10 and 50㎚PTH, respectively, but IGF Ⅰ and Ⅱ induced limited activity of osteoblastic function. In conclusion, IGF-Ⅰ and Ⅱ were potent stimulators to the growth of osteoblastic cells but did not have an immediate effect on expression of osteoblastic function in this experiment.

        Now, on the spot of construction when one test the compression of concrete, by operating the loading rate vaguely it occurred many cases to bring the profitable results for the factory of remicon. Because the same compressive strength of concrete is influenced according to the difference of loading rate, its required the correct and available materials of the test. In this research its measured the difference and rate of change on compressive strength according to the loading rate, for the effective quality control of concrete construction. And it has the comparative analysis of the results about the compressive strength influences a high strength concrete and general concrete by using the testing machining. Finally, the purpose of this is to suggest the basic materials for more effective way of test and plan of use in order to get the proper strength of concrete and improvement of quality.

        봉합법을 이용한 중검술의 임상적 연구

        One of the most common cosmetic surgery, the blepharoplasty can be divided two method. there were incisional method and sutured method. Authors try to understand the anatomic difference between the oriental upper eyelids and apply the surgical techniques of constructing upper eyelid crease of oriental case effectively. And we discussed the postoperative complications of the blepharoplasty and proposed the solve of these problems. The sutured method of blepharoplasty was more simple technique than conventional incision method and had low occurrence of complications. Even though slight recurrence, suture, method was more esthetic due to no scar formation.

        Effect of ${\beta}$-Glucan Originated from Aureobasidium pullulans on Asthma Induced by Ovalbumin in Mouse

        The objective of this study is to detect the effect of beta-glucan derived from Aureobasidium pullulans SM-2001, a UV induced mutant of A. pullulans on the ovalbumin (OVA) induced allergic asthma. The test articles were orally administered to OVA-inducing asthmatic mice 4 days after sensitization for 13 days at 31.25, 62.5 or 125 mg/kg levels. Three days after the OVA sensitization, ten mice were selected per group based on body weight and were sacrificed three days after the OVA aerosol challenge. The changes on the body weight, lung weight, total leukocytes in peripheral blood and total cells in bronchoalveolar lavage fluid (BALF) were observed with changes on the lung histopathology and histomorphometry. The results were compared with dexamethasone (DEXA) 3 mg/kg intraperitoneally treated mice. The results showed increases of body weight after the OVA aerosol challenge, lung weight, total leukocytes and eosinophils in peripheral blood, total cell numbers, neutrophil and eosinophils in BALF were detected in the OVA control compared to sham control (non-OVA). However, these changes from asthmatic responses were significantly or dose-dependently decreased in the beta-glucan-dosing groups compared to those of the OVA control. Therefore, it is concluded that beta-glucan has favorable effects on asthmatic response induced by OVA. It was found that beta-glucan 125 mg/kg showed similar or slightly lower efficacy compared with DEXA 3 mg/kg.

        Analysis of the Risk Factors for Unfavorable Radiologic Outcomes after Fusion Surgery in Thoracolumbar Burst Fracture : What Amount of Postoperative Thoracolumbar Kyphosis Correction is Reasonable?

        Objective : The aims in the management of thoracolumbar spinal fractures are not only to restore vertebral column stability, but also to obtain acceptable alignment of the thoracolumbar junction (T-L junction) to prevent complications. However, insufficient surgical correction of the thoracolumbar spine would be likely to cause late progression of abnormal kyphosis. Therefore, we identified the surgical factors that affected unfavorable radiologic outcomes of the thoracolumbar spine after surgery. Methods : This study was conducted in a single institution from January 2007 to December 2013. A total of 98 patients with unstable thoracolumbar spine fracture were included. In these patients, fixation was done through transpedicular screws with rods by three surgical patterns. We reviewed digital radiographs and analyzed the images preoperatively and postoperatively during follow-up visits to compare the change of the thoracolumbar Cobb angle with radiologic parameters and clinical outcomes. The unfavorable radiologic group was defined as the patients who were measured as having greater than 20 degrees of thoracolumbar Cobb angle on the last follow-up, or who underwent kyphotic progression of thoracolumbar Cobb angle greater than 10 degrees from the immediate postoperative state to final follow-up, or who had overt instrument failure with/without additional surgery. We assessed the risk factors that affected the unfavorable radiologic outcomes. Results : We had 43 patients with unfavorable radiologic outcomes, including 35 abnormal thoracolumbar alignments and 14 instrumental failures with/without additional surgery. The multivariate logistic regression test showed that immediate postoperative T-L junction Cobb angle less than 10.5 degrees was a statistically significant risk factor, as well as the presence of osteoporosis (p=0.017 and 0.049, respectively). Conclusion : Insufficient correction of thoracolumbar kyphosis was considered to be a major factor of an unfavorable radiological outcome. The spinal surgeon should consider that having a T-L junction Cobb angle larger than 10.5 degrees immediately after surgery could result in an unfavorable radiological outcome, which is related to a poor clinical outcome.

      • The Anti-osteoporotic Effect of Aqueous Extracts of Gastrodiae Rhizoma <i>In Vitro</i> and <i>In Vivo</i>

        <P>The anti-osteoporotic effects of aqueous extracts of Gastrodiae Rhizoma (GR) were observed <I>in vitro</I> and <I>in vivo</I>. The effects on proliferation and alkaline phosphatase activity of primary osteoblasts, bone nodule formation, pit formation of osteoclasts and osteoclastogenesis were observed <I>in vitro</I>, and to observe the <I>in vivo</I> efficacy GR were orally administered once a day for 28 days to bilateral ovariectomy-induced osteoporosis mice at 125, 250 and 500 mg/kg. GR extracts enhanced the proliferation, differentiation, and bone nodule formation of primary cultured osteoblasts, but they inhibited the pit formation and the number of multinucleated osteoclast-like cells, osteoclastogenesis <I>in vitro</I>. As results of ovariectomy-induced osteoporotic process, dramatical decreases of bone weights and thickness of bone at epiphyseal regions, bone Ca and P contents, bone mineral density and failure load, serum Ca and P levels with increase of serum osteocalcin level. At histopathology-histomorphometry, dramatical decreases of trabecular and cortical bone masses were detected with classic histomorphometrical changes of bones including decrease of trabecular bone volume, thickness, length and number and cortical bone thickness and increase of osteoclast/bone perimeter. Hoevere, these estrogen-deficient osteoporotic changes were also dramatically and dose-dependently inhibited by treatment of all three different dosages of GR extracts. It was concluded that GR extracts has relatively good favorable effect to prevention and/or treatment of ovariectomy (OVX)-induced osteoporosis through osteoblast activation and inhibition of osteoclastogenesis and osteoclast activity.</P>

        Surface Imaging of Barley Aleurone Cell by Atomic Force Microscopy

        To observe and analysis ultra-microscopically barley aleurone cell surface, atomic force microscope (AFM) was used. Seed coat of early maturing germplasm, eam9, was dehulled and scanned by non-contact mode. We have obtained the high resolution topographic 3-dimensional image of barley aleurone layer with high resolution. These images showed the membrane proteins in barley aleurone cell. One channel protein and numerous peripheral or integral proteins were detected in a area of 100 ~mu~textrmm2 . Furthermore, we found that their widths were ranged from 50 to 750nm and lengths from 0 to 66 ~mu~textrmm . The thickness of aleurone layer was measured by scanning electron microscope. The thickness at early developmental stage was about 16 and then the aleurone cell enlarged upto 57 ~mu~textrmm ~mu~textrmm at least until 42 days after anthesis. In this study, we firstly reported on the ultrastructural AFM analysis of living aleurone cell as a biological specimen. It was clearly suggested that AFM will become an powerful tool for probing both the structural properties of biological samples

        Effects of Basal Stacking Faults on Electrical Anisotropy of Nonpolar a-Plane (<tex> $11\bar{2}0$</tex> ) GaN Light-Emitting Diodes on Sapphire Substrate

        <P>We report on the effects of basal stacking faults (BSFs) on the electrical anisotropy and the device characteristics of nonpolar a-plane GaN (1120) light-emitting diodes (LEDs) on r-plane (1102 ) sapphire substrates. The sheet resistance in the direction parallel to the c-axis [0001] is 18%-70% higher than the one in the direction parallel to the m-axis [1100 ]. The anisotropic conductivity of faulted a-plane GaN films can be explained by carrier scatterings from BSFs. It is also shown that the output power of nonpolar a-plane GaN LEDs are significantly influenced by the presence of BSFs, which laterally hampers the carrier transport in the n-GaN layer, especially in the direction parallel to the c-axis in faulted nonpolar nitride films.</P>
