Abnormally expressed microRNAs (miRNAs) and genes have been found to play key roles in esophageal squamous cell carcinoma (ESCC), but little is known about the underlying mechanisms. The aim of this paper was to assess inter-relationships and the regulatory mechanisms of ESCC through a network-based approach. We built three regulatory networks: an abnormally expressed network, a related network and a global network. Unlike previous examples, containing information only on genes or miRNAs, the prime focus was on relationships. It is worth noting that abnormally expressed network emerged as a fault map of ESCC. Theoretically, ESCC might be treated and prevented by correcting the included errors. In addition, the predicted transcription factors (TFs) obtained by the P-match method also warrant further study. Our results may further guide gene therapy researchers in the study of ESCC.

This study aims to identify the relationship between the norm of emotion expression of the coaches working at sports facilities and emotional labor, exhaustion of emotion and the intention to change jobs. The objects of research are the 230 coaches working at sports facilities located in Seoul Metropolitan City and Gyeonggi–do Province. For the verification of the hypotheses established in this study, frequency analysis, exploratory factor analysis, reliability analysis, confirmatory factor analysis and structure equation model analysis were conducted with PASW 18.0 and AMOS 18.0 data processing programs. The following are the results of the analyses. Firstly, the norm of emotion expression has meaningful influence on the surface behavior and the internal behavior that are the sub factors of emotional labor. Secondly, while the surface behavior, which is a sub factor of emotional labor, has meaningful influence on exhaustion of emotion, it does not have influence on internal behavior. Thirdly, exhaustion of emotion has meaning influence on the intention to change jobs.
Adverse effect of DEHP exposure on the serum insulin level of Balb/c mice

Di(2-ethylhexyl) phthalate (DEHP) is a common indoor pollutant in the world, which may cause lots of harmful effects in human including diabetes according to epidemiological studies. To explore the underlying role of DEHP in diabetes-like symptoms, Balb/c mice were chose to be the experimental animals in this paper. They were separated as eight groups as follows: (1) saline+normal diet (vehicle control), (2) 10 mg/kg.day DEHP+normal diet, (3) 50 mg/kg.day DEHP+normal diet, (4) 250 mg/kg.day DEHP+normal diet, (5) streptozotocin (STZ)+high fat diet (diabetes model), (6) 10 mg/kg.day DEHP+ STZ+high fat diet, (7) 50 mg/kg.day DEHP+STZ +high fat diet, (8) 250 mg/kg.day DEHP+STZ+high fat diet. The biomarkers of this experiment include four groups: (1) general indicates: body weight and drinking water, (2) blood biomarkers: serum insulin and fasting glucose, (3) pathological examination: pancreas section and kidney section, and (4) biomarkers of oxidative stress: reactive oxygen species (ROS) and malondialdehyde (MDA) in liver cells. Our study results demonstrate that: (1) at our treatment levels DEHP cannot directly induce diabetes, but reduce serum insulin level in DEHP-exposed non-STZ-treated animals, (2) pathological examination finds that these is a dose-dependent damage in the pancreas in DEHP- exposed STZ-treated groups, and (3) the oxidative mechanism may be involved in this pathological process.

This study`s objective is to examine the influence of the fitness center chief`s servant leadership on the trainer`s self-leadership. To achieve this objective, 213 trainers from 30 fitness centers were surveyed. The documented surveys filled out by the subjects were collected and processed statistically for research purpose. Questionnaire was used as the measurement tool. The questionnaire had a total of 30 questions consisting of 4 social demographic questions, 10 servant leadership questions, and 16 self-leadership questions. For verification, 5% was the set significance level and a statistical program was used to analyze the correlation between the fitness center chief"s servant leadership and the trainer`s self-leadership and conduct multiple regression analysis. The deducted conclusion is the following. First, the chief`s servant leadership affected the constructive thinking strategy of the trainer`s self-leadership. Second, the chief`s servant leadership affected the natural reward strategy of the trainer`s self-leadership. Third, the chief`s servant leadership affected the behavior focused strategy of the trainer`s self-leadership.

A DNA fragment containing 2,079 base pairs from Bacillus circulans CGMCC 1416 was cloned using degenerate PCR and inverse PCR. An open reading frame containing 981 bp was identified that encoding 326 amino acids residues, including a putative signal peptide of 31 residues. The deduced amino acid sequence showed the highest identity (68.1%) with endo-β-1,4-D-mannanase from Bacillus circulans strain K-1 of the glycoside hydrolase family 5 (GH5). The sequence encoding the mature protein was cloned into the pET-22b(+) vector and expressed in Escherichia coli as a recombinant fusion protein containing an N-terminal hexahistidine sequence. The fusion protein was purified by Ni2+ affinity chromatography and its hexahistidine tag cleaved to yield a 31-kDa β-mannanase having a specific activity of 481.55 U/mg. The optimal activity of the purified protein, MANB48, was at 58℃ and pH 7.6. The hydrolysis product on substrate locust bean gum included a monosaccharide and mainly oligosaccharides. The recombinant MANB48 may be of potential use in the feed industry.

Background: Many clinical trials have been conducted to evaluate sorafenib for the treatment of advanced NSCLC, but the results for efficacy have been inconsistent. The aim of this study was to evaluate the efficacy and safety of sorafenib in patients with advanced NSCLC in more detail by meta-analysis. Methods: This meta-analysis of randomized controlled trials (RCTs) was performed after searching PubMed, EMBASE, ASCO Abstracts, ESMO Abstracts, and the proceedings of major conferences for relevant clinical trials. Two reviewers independently assessed the quality of the trials. Outcomes analysis were disease control rate (DCR), progression- free survival (PFS), overall survival (OS) with 95% confidence intervals (CI) and major toxicity. Subgroup analysis was conducted according to sorafenib monotherapy, in combination with chemotherapy or EGFR-TKI to investigate the preferred therapy strategy. Results: Results reported from 6 RCTs involving 2, 748 patients were included in the analysis. Compared to sorafenib-free group, SBT was not associated with higher DCR (RR 1.31 (0.96- 1.79), p=0.09), PFS (HR 0.82 (0.66-1.02), p=0.07) and OS (HR 1.01 (0.92-1.12), p=0.77). In terms of subgroup results, sorafenib monotherapy was associated with significant superior DCR and longer PFS, but failed to show advantage with regard to OS. Grade 3 or greater sorafenib-related adverse events included fatigue, hypertension, diarrhea, oral mucositis, rash and HFSR. Conclusions: SBT was revealed to yield no improvement in DCR, PFS and OS. However, sorafenib as monotherapy showed some activity in NSCLC. Further evaluation may be considered in subsets of patients who may benefit from this treatment. Sorafenib combined inhibition therapy should be limited unless the choice of platinum-doublet regimen, administration sequence or identification of predictive biomarkers are considered to receive better anti-tumor activity and prevention of resistance mechanisms.
Road-friendliness of Fuzzy Hybrid Control Strategy Based on Hardware-in-the-Loop Simulations

Purpose: In order to improve road-friendliness of heavy vehicles, a fuzzy hybrid control strategy consisting of a hybrid control strategy and a fuzzy logic control module is proposed. The performance of the proposed strategy should be effectively evaluated using a hardware-in-the-loop (HIL) simulation model of a semi-active suspension system based on the fuzzy hybrid control strategy prior to real vehicle implementations. Methods: A hardware-in-the-loop (HIL) simulation system was synthesized by utilizing a self-developed electronic control unit (ECU), a PCI-1711 multi-functional data acquisition board as well as the previously developed quarter-car simulation model. Road-friendliness of a semi-active suspension system controlled by the proposed control strategy was simulated via the HIL system using Dynamic Load Coefficient (DLC) and Dynamic Load Stress Factor (DLSF) criteria. Results: Compared to a passive suspension, a semi-active suspension system based on the fuzzy hybrid control strategy reduced the DLC and DLSF values. Conclusions: The proposed control strategy of semi-active suspension systems can be employed to improve road-friendliness of road vehicles.

Klotho, an antiaging protein, has been shown to play a protective role in renal tubular epithelial-mesenchymal transition (EMT) during the development of diabetic kidney disease (DKD). Long noncoding RNAs (lncRNAs) participate in the progression of EMT in many diseases. However, the effect of Klotho on lncRNAs during the development of DKD is still unknown. In this study, we found that Klotho overexpression in high-fat diet (HFD)- and streptozotocin (STZ)- induced DKD mice significantly inhibited the expression of lncRNA nuclear-enriched abundant transcript 1 (Neat1). We demonstrated that NEAT1 was significantly upregulated in both bovine serum albumin (BSA)-stimulated HK2 cells and mice with HFD- and STZ-induced diabetes. In addition, we observed that Klotho displays colocalization with NEAT1. Furthermore, overexpression of Klotho can inhibit the high expression of NEAT1 in BSA-stimulated HK2 cells, while silencing Klotho can further upregulate the expression of NEAT1. Silencing NEAT1 in HK2 cells resulted in inhibition of the EMT-related markers alpha smooth muscle actin (α-SMA) and vimentin (VIM) and the renal fibrosis-related markers transforming growth factor-β1 (TGF-β1) and connective tissue growth factor (CTGF). The effect of NEAT1 on DKD was partly mediated by regulation of the ERK1/2 signaling pathway. Finally, we found that silencing NEAT1 can reverse the activation of EMT and fibrosis caused by Klotho silencing in a manner dependent on the ERK1/2 signaling pathway. These findings reveal a new regulatory pathway by which Klotho regulates ERK1/2 signaling via NEAT1 to protect against EMT and renal fibrosis, suggesting that NEAT1 is a potential therapeutic target for DKD.
