        IFT46 plays an essential role in cilia development

        <P><B>Abstract</B></P> <P>Cilia are microtubule-based structures that project into the extracellular space. Ciliary defects are associated with several human diseases, including polycystic kidney disease, primary ciliary dyskinesia, left–right axis patterning, hydrocephalus and retinal degeneration. However, the genetic and cellular biological control of ciliogenesis remains poorly understood. The IFT46 is one of the highly conserved intraflagellar transport complex B proteins. In zebrafish, <I>ift46</I> is expressed in various ciliated tissues such as Kupffer׳s vesicle, pronephric ducts, ears and spinal cord. We show that <I>ift46</I> is localized to the basal body. Knockdown of <I>ift46</I> gene results in multiple phenotypes associated with various ciliopathies including kidney cysts, pericardial edema and ventral axis curvature. In <I>ift46</I> morphants, cilia in kidney and spinal canal are shortened and abnormal. Similar ciliary defects are observed in otic vesicles, lateral line hair cells, olfactory pits, but not in Kupffer׳s vesicle. To explore the functions of <I>Ift46</I> during mouse development, we have generated <I>Ift46</I> knock-out mice. The <I>Ift46</I> mutants have developmental defects in brain, neural tube and heart. In particular <I>Ift46</I>(−/−) homozygotes displays randomization of the embryo heart looping, which is a hallmark of defective left–right (L/R) axis patterning. Taken together, our results demonstrated that IFT46 has an essential role in vertebrate ciliary development.</P> <P><B>Highlights</B></P> <P> <UL> <LI> IFT46 over-expression induces apoptosis in developing zebrafish embryos. </LI> <LI> Loss of IFT46 displays typical phenotypes of ciliary defects in zebrafish and mice. </LI> <LI> IFT46 C-terminal domain is required for IFT46 function. </LI> <LI> IFT46 is not essential for axonemal dynein assembly in zebrafish. </LI> </UL> </P>

        식물 40종 고압용매 추출물의 통합적 항산화 능력 및 항균 활성

        제주, 전남 고흥, 경북 울릉도에 자생하는 식물자원 40종을 대상으로 고압용매 추출하여 항산화 활성과 항균활성을 측정하여, 식품산업에 응용할 천연소재를 탐색하였다. 총 페놀성 화합물 함량은 짚신나물이 174.4 ㎎ GAE/g로 자생식물 중 가장 높았고, 다음으로 물참나무, 서어나무, 붉나무가 각각 116.9, 113.3, 108.2 ㎎ GAE/g로 높은 함량을 나타내었다. 총고형분 함량에 대한 총 페놀 함량의 비율은 짚신나물이 72.6%로 가장 높았고, 서어나무(47.3%), 물참나무(46.4%), 자금우(40.2%), 바늘꽃(40.1%) 순으로 높은 비율을 나타내었다. 수용성 항산화 능력은 마가목이 5.81 mmol ascorbic acid equivalents/g로 가장 높았고, 다음으로 나도송이풀, 붉나무, 물참나무, 서어나무가 각각 3.96, 3.63, 3.63, 3.34 mmol ascorbic acid equivalents/g를 나타내었다. 지용성 항산화능력은 서어나무가 8.51 mmol Trolox equivalents/g로 가장 높았고, 다음으로 마가목, 세잎이질풀, 물참나무, 바늘꽃, 섬기린초가 각각 6.57, 5.68, 3.85, 3.83, 3.69 mmol Trolox equivalents/g를 나타내었다. 국내 자생식물 40종 추출물에 대하여 4종의 양식 넙치 질병 세균에 대한 항균활성을 측정한 결과 9종 즉, 바늘꽃, 독활, 짚신나물, 물참나무, 나도송이풀, 세잎이질풀, 서어나무, 섬기린초, 섬바디 만이 항균활성을 나타내었다. 짚신나물은 S. iniae에 대하여 높은 항균활성을 나타내었으며, 독활, 나도송이풀, 섬바디는 S. parauberis과 S. iniae에 대하여, 물참나무, 세잎이질풀, 서어나무는 V. ordalii에 대하여 높은 항균활성을 나타내었다. Forty natural plants collected in Jeju, Jeonnam-Goheung, and Gyeongbuk-Ulleung were extracted using a pressurized liquid. Extraction yields of total soluble solids and total phenolics (TP), and integral antioxidative capacity (IAC) were measured, and antimicrobial activity was tested against Streptococcus parauberis, Streptococcus iniae, Edwardsiella tarda, and Vibrio ordalii. Jipsinnamul showed the highest content of TP (174.4 ㎎ GAE/g), followed by Mulchamnamu (116.9), Seoeonamu (113.3), and Buknamu (108.2). Percent TP/TSS was high in Jipsinnamul (72.6%), Seoeonamu (47.3%), Mulchamnamu (46.4%), Jageumu (40.2%), and Baneulkkot (40.1%), respectively. Magamok, Nadosongipul, Buknamu, Mulchamnamu, and Seoeonamu showed 5.81, 3.96, 3.63, 3.63, and 3.34 mmol ascorbic acid equivalents/g of IAC of water-soluble substances, and Seoeonamu, Magamok, Seipijilpul, Mulchamnamu, Baneulkkot, and Seomgirincho showed 8.51, 6.57, 5.68, 3.85, 3.83, and 3.69 mmol Trolox equivalents/g of IAC of lipid-soluble substances, respectively. Only nine species such as Baneulkkot, Dokhwal, Jipsinnamul, Mulchamnamu, Nadosongipul, Seipijilpul, Seoeonamu, Seomgirincho, and Sumbadi of 40 selected plants showed the antimicrobial activity against four bacteria tested. Jipsinnamul showed the strong antimicrobial activity against S. iniae, while Dokhwal, Nadosongipul, and Sumbadi against S. parauberis and S. iniae, and Mulchamnamu, Seoeonamu, and Seipijilpul against V. ordalii.

        국문초록 세 편의 논문으로 구성된 본 연구에서는 아인슈타인의 7차원 통일장론 기하학에 관하여 연구한다. 제 1편과 제 2편에서는 X7의 대수학적 구조에 관하여 다루고, 제 3편에서는 X7의 미분기하학적 구조 (곧, 7차원 아인슈타인 접속)를 주로 다룬다. 본 논문에서는, 제 1종과 제 2종의 일반화된 7차원 리이만 다양체 X7에서 성립하는 매우 강력한 새로운 회귀관계식들을 유도한다. 이 관계식은 7차원 아인슈타인 접속을 이용가능한 텐서표시로 나타내는 연구(제3편)에서 매우 유용하게 이용된다. Abstract In a sequence of three papers, we study about 7-dimensional Einstein's unified field theory, with emphasis on the algebraic structure of X7 in the first two papers and on the differential geometric structure (mainly on the 7-dimensional Einstein's connection) in the third paper. In this second paper, we derive a new set of powerful recurrence relations which hold in a 7-dimensional generalized Riemannian manifold X7. Three relations will be used in our last paper to derive a surveyable tonsorial representation of 7-dimensional Einstein's connection in terms of the unified field tensor gλμ.

      • 연령별 투자자의 재무 태도가 부동산 및 주식 투자 의도에 미치는 영향 분석

        본 연구는 국내 가계자산비중이 비금융자산인 부동산에 편중되어 있다는 금융투자협회의 발표를 바탕으로 가계자산의 불균형을 해소할 수 있는 방안을 마련하기 위해 한국인의 투자 행태에 대한 이해를 높이고자 계획행동이론을 바탕으로 고안되었다. 투자자의 미래재무인식, 현재재무인식, 재무관심도, 재무목표와 같은 투자자의 재무태도가 부동산 및 주식 투자 결정에 영향을 미치며 그 영향도는 연령에 따라 다르다는 가설 하에 실제로 어떤 변수들이 가장 큰 영향을 미치는지 회귀분석을 통해 검증하였다. 이는 금융자산 투자에 있어 돈에 대해 개인이 가진 인식과 관심도 및 목표와 같은 요인이 투자자의 투자 결정에 어느 정도의 영향을 미치며 그 영향력은 연령에 따라 어떻게 달라지는지 구체적으로 살펴보는 데에 그 의의가 있다. 연구 방법은 먼저 선행 연구 검토 후 설문조사를 실시하였고 해당 자료는 SPSS 23 통계 패키지 프로그램을 사용하여 분석되었다. 재무태도 중 현재재무인식, 재무관심도, 재무목표는 부동산투자에 유의미한 영향을 미치는 것으로 나타났다. 특히 재무목표는 전 연령대에 영향을 미치고 있는 것으로 나타났는데, 그 영향력은 연령이 높아질수록 감소하였다. 30대에서는 재무목표가 부동산 투자에 큰 영향력을 보이는 반면, 50대에서는 재무관심도가 부동산투자에 큰 영향을 미치는 것으로 나타났다. 한편 재무태도가 주식투자결정에 미치는 영향과 이에 대한 연령대별 분석 실시 결과 재무목표만이 주식투자결정에 극명하게 큰 영향을 미치는 것으로 나타났다. 특히 30대의 경우 매우 큰 영향력을 보이고 있었으며 이후 50대, 40대 순으로 영향을 보이고 있었다. 요약해 보면 한국인들의 부동산 또는 주식 투자 의도를 갖게 하는 가장 큰 요인은 재무목표이다. 즉 결혼이나 자녀교육, 은퇴 등 미래를 위한 계획을 수립하고자 하는 것이 투자의 가장 큰 동기로 부동산과 같은 실물자산과 금융자산 투자에 나타난 연령별 특성을 감안한 정책마련이나 교육 등의 필요성도 시사한다고 하겠다. Based on the Korea Financial Investment Association's announcement that Korean household assets are concentrated in real estate, which is a non-financial asset, this study was designed based on the Theory of Planned Behavior by Ajzen to improve the understanding of Koreans' investment behavior in order to come up with a plan to resolve the imbalance in household assets. Under the hypothesis that investors' money attitudes, such as future financial perception, current financial perception, financial interest, and financial goals, affect real estate and stock investment intentions, and their impact varies according to age, regression analysis was used to verify which variables actually have the greatest impact. This is meaningful in examining in detail how factors such as individual perceptions, interests, and goals of money affect investors' investment intention and how their influence varies with age. As for the research method, a survey was conducted after reviewing previous studies, and the data were analyzed using the SPSS 23 statistical package program. In summary, the biggest factor that drives Koreans' intention to invest in real estate or stocks is their financial goals. In other words, the biggest motivation for investment is to plan for the future, such as one's own marriage, children's education, and retirement. Based on this founding, the government should prepare relevant policies to support each age group and provide appropriate publicity and education to persuade the investors that they can achieve their financial goals through investment in financial assets rather than real estate.

      • Preliminary <sup>19</sup> F-MRS Study of Tumor Cell Proliferation with 3′-deoxy-3′-fluorothymidine and Its Metabolite (FLT-MP)

        <P>The thymidine analogue 3′-deoxy-3′-[<SUP>18</SUP>F]fluorothymidine, or [<SUP>18</SUP>F]fluorothymidine ([<SUP>18</SUP>F]FLT), is used to measure tumor cell proliferation with positron emission tomography (PET) imaging technology in nuclear medicine. FLT is phosphorylated by thymidine kinase 1 (TK1) and then trapped inside cells; it is not incorporated into DNA. Imaging with <SUP>18</SUP>F-radiolabeled FLT is a noninvasive technique to visualize cellular proliferation in tumors. However, it is difficult to distinguish between [<SUP>18</SUP>F]FLT and its metabolites by PET imaging, and quantification has not been attempted using current imaging methods. In this study, we successfully acquired<I> in vivo </I><SUP>19</SUP>F spectra of natural or nonradioactive 3′-deoxy-3′-fluorothymidine ([<SUP>19</SUP>F]FLT) and its monophosphate metabolite (FLT-MP) in a tumor xenograft mouse model using 9.4T magnetic resonance imaging (MRI). This preliminary result demonstrates that <SUP>19</SUP>F magnetic resonance spectroscopy (MRS) with FLT is suitable for the<I> in vivo</I> assessment of tumor aggressiveness and for early prediction of treatment response.</P>

      • 부모의 양육태도와 아동의 사회성 발달과의 관계

        The purpose of this study was to examine the relationship between the child rearing attitudes of parents and the social development of their children and to examine the sex difference in those relationship. The subjects were 60 5-year-old children and their parents. The instruments for this study were the Diagnostic Examination of the Relationship between Parents and Children for Parent by Oh(1965), and the Examination of Social Maturity by Kim and Kim(1985). The data were analyzed with ANOVA. The results showed that the correlation between the type of parents' rearing attitudes and the children's social development was not statistically significant and that there were no difference in the social development between boys and girls.

        Influence of the Duration of Diabetes on the Outcome of a Diabetes Self-Management Education Program

        <P><B>Background</B></P><P>Diabetes education and lifestyle modification are critical components in controlling blood glucose levels of people with type 2 diabetes. Until now, available data on the effectiveness of education with respect to the duration of diabetes are limited. We investigated whether adherence to lifestyle behavior modification prompted by diabetes education was influenced by the duration of diabetes.</P><P><B>Methods</B></P><P>Two hundred and twenty-five people with type 2 diabetes were recruited for an intensive, collaborative, group-based diabetes education program with annual reinforcement. We divided the patients into two groups based on the duration of their diabetes prior to the education program (≤1 year [≤1Y] vs. ≥3 years [≥3Y]). Dietary habits, physical activity, and the frequency of blood glucose self-monitoring were evaluated with a questionnaire prior to education and at the follow-up endpoint.</P><P><B>Results</B></P><P>The mean follow-up period was 32.2 months. The mean hemoglobin A1c (A1C) value was significantly lower in the ≤1Y group. Self-care behaviors, measured by scores for dietary habits (<I>P</I>=0.004) and physical activity (<I>P</I><0.001), were higher at the endpoint in the ≤1Y group than in the ≥3Y group. Logistic regression analysis revealed that a longer diabetes duration before education was significantly associated with mean A1C levels greater than or equal to 7.0% (53 mmol/mol).</P><P><B>Conclusion</B></P><P>Diabetes duration influenced the effectiveness of diabetes education on lifestyle behavior modification and glycemic control. More-intense, regular, and sustained reinforcement with encouragement may be required for individuals with longstanding type 2 diabetes.</P>

        암컷 Sprague-Dawley 흰쥐에서 양측 난소절제술이 대뇌피질 및 해마의 serotonin 치에 미치는 영향

        Objective : The purpose of this study is to investigate the effect of chronic deprivation of estradiol on the serotonin level in Sprague-Dawley rat's cerebral cortex and hippocampus. Methods : One hundred of female Sprague-Dawley rats were taken care for seven to eight weeks, then they were randomly assigned into two group; fifty rats underwent bilateral oophorectomy & the remaining rats were the control group. Six months after oophorectomy, brain tissue was extracted from cerebral cortex and hippocampus and then homogenated: this was done in the both groups. The upper layer of the homogenated substance was taken and frozen and finally dried up. The next process was marking with PITC, quantification through HPLC and comparing with authentic serotonin. Protein quantity was measured by using Bradford method. Results : Comparison between the bilateral oophorectomy group and the control group for serotonin content in the cerebral cortex was 134.2±14.3 nmol/mg protein(mean±SEM) and 122.5±11.6 nmol/mg protein in each group. As for the serotonin content in the hippocampus was 148.8±13.9 nmol/mg protein and 132.9±13.7 nmol/mg protein in each group. It shows that, there was no significant difference in the serotonin level, in the two areas mentioned above. Conclusion : The effect of estradiol on the serotonin level may be dependent on the hormonal change duration. Chronic effect of hypoestrogenism might have no effect on tryptophan hydroxylase and monoamine oxidase, thus not changin the serotonin level. These data supported the previous findings that chronic hypoestrogenism had no effect on serotonin level.
