N/A Background: Potential involvement of the angio-tensinogen gene(M235T) in the pathogenesis of essen-tial hypertension has been suggested by some inves- tigators. However, an association between M235T gene polymorphism and essential hypertension has been reported by some, but not by others. Since genetic diversity exists among different ethnic population, we addressed the question of whether there is an association between M235T gene poly-morphism and essential hypertension in the Korean. Methods: 100 patients with essential hypertension and 100 control subjects were recruited from out- patients at the Department of Internal Medicine, Soon-chunhyang Hospital, Chunan City. The criteria for hypertension was defined as systolic blood pressure higher than 160mmHg and/or diastolic blood pressure higher than 95mmHg. For detection of the M235T polymorphism at the angiotensinogen locus, the primer sequences were: sense primer : 5'-TGAAGGAG- AAGGTGTCTGCGGGA-3' and antisense primer : 5'- AGGACGGTGCGGTGAGAGTG-3'. The PCR product mixture was exposed to restriction enzyme Tthlll I and then submitted to electrophoresis in polyacryl-amide gel. Differences between the molecular variants of the gene in hypertensives and normotensives were com- pared by using the X²test. p<0.05 was considered statistically significant. The odds ratio and 95% confidence interval were calculated using Woolf's method. Results: Compared with the control subjects, hypertensives had higher values of three established risk factors for hypertension : age, BMI cholesterol (total and LDL). X analysis showed no difference in the distribution of genotype or allele frequency between the hypertensives and normotensives(X²=1.14, p=0.29). The crude odds ratio was 0.73 for CC over CT(95% confidence interval 0.41-1.30). The adjusted odds ratio with age, sex, lipid profilcs and BMI was 0.68 for CC over CT(9596 confidence interval 037-1,23). Conclusion : The molecular varient M235T of the angiotensinogen gene is not associated with essential hypertension in Korean population.

BACKGROUND: Health problems related to the lack of bone formation are a major problem for ageing populations in the modern world. As a part of the ongoing trend to develop natural substances that attenuate bone loss in osteoporosis, the effects of the edible brown alga Sargassum thunbergii and its active contents on adipogenic differentiation in 3T3-L1 fibroblasts and osteoblast differentiation inMC3T3-E1 pre-osteoblasts were evaluated. RESULTS: Treatment with S. thunbergii significantly reduced lipid accumulation and expression of adipogenic differentiation markers such as peroxisome proliferator-activated receptor gamma , CCAAT/enhancer- binding protein.. and sterol regulatory element binding protein 1c. In addition, S. thunbergii successfully enhanced osteoblast differentiation as indicated by increased alkaline phosphatase activity along raised levels of osteoblastogenesis indicators, namely bone morphogenetic protein-2, osteocalcin and collagen type I. Two compounds, sargaquinoic and sargahydroquinoic acid, were isolated fromactive extract and shown to be active by means of osteogenesis inducement. CONCLUSION: S. thunbergii could be a source for functional food ingredients for improved treatment of osteoporosis and obesity. (C) 2015 Society of Chemical Industry

Purpose: The development of compact tractors that can be used in dry fields, greenhouses, and orchards for pest control, weeding, transportation, and harvesting is necessary. The development and performance evaluation of power transmission units are very important when it comes to tractor development. This study evaluates the performance of a driving power transmission unit of a 50 kW multi-purpose narrow tractor. Methods: The performance of the transmission and forward-reverse clutch, which are the main components of the driving power transmission unit of multi-purpose narrow tractors, was evaluated herein. The transmission performance was evaluated in terms of power transmission efficiency, noise, and axle load, while the forward-reverse clutch performance was evaluated in terms of durability. The transmission’s power transmission efficiency accounts for the measurement of transmission losses, which occur in the transmission’s gear, bearing, and oil seal. The motor’s power was input in the transmission’s input shaft. The rotational speed and torque were measured in the final output shaft. The noise was measured at each speed level after installing a microphone on the left, right, and upper sides. The axle load test was performed through a continuous equilibrium load test, in which a constant load was continuously applied. The forward-reverse clutch performance was calculated using the engine torque to axle torque ratio with the assembled engine and transmission. Results: The loss of power in the transmission efficiency test of the driving power unit was 6.0-9.7 kW based on all gear steps. This loss of horsepower was equal to 11-18% of the input power (52 kW). The transmission efficiency of the driving power unit was 81.5-89.0%. The noise of the driving power unit was 50-57 dB at 800 rpm, 70-77 dB at 1600 rpm, and 76-83 dB at 2400 rpm. The axle load test verified that the input torque and axle revolutions were constant. The results of the forward-reverse clutch performance test revealed that hydraulic pressure and torque changes were stably maintained when moving forward or backward, and its operation met the hydraulic design standards. Conclusions: When comprehensively examined, these research results were similar to the main driving power transmission systems from USA and Japan in terms of performance. Based on these results, tractor prototypes are expected to be created and supplied to farmhouses after going through sufficient in-situ adaptability tests.

This study analyzed the antioxidant properties of Equisetum arvense and its effects on serum factor levels in mice fed a high-fat diet. The aim was to establish a new effective resource for biologically active materials. E. arvense stem and root extracts were obtained using deionized water at $95^{\circ}C$, and 70.5% ethanol at $85^{\circ}C$. These extracts were used to analyze the total phenolic compounds and antioxidant (ABTS, DDPH, and FRAP) activities. The effects of prepared ground samples were evaluated by feeding them to mice. E. arvense extracts showed strong antioxidant effects. The caffeic acid content was highest in the 70.5% ethanol extract of the vegetative stem, as determined by high-performance liquid chromatography. The blood concentrations of insulin and leptin were significantly lower in mice fed a high-fat diet supplemented with extracts of the root, reproductive stem, and vegetative stem of E. arvense than in mice fed only a high-fat diet. These results suggest that the polyphenolic compounds in E. arvense extracts exert various antioxidant effects. The stems and root of E. arvense can lower the blood levels of insulin and leptin, even after consumption of a high-fat diet.

The recombinant human interferon alpha 2α(rhIFN-a2a), expressed in Saccharomyrces cerevisiae, was purified from insoluble aggregates. The inclusion body of rhIFN-α was solubilized by guanidine salt in the presence of disulfide reducing agent. The refolding of denatured rhIFN-α2a was achieved by simple dilution. The authentic interferon alpha, which has two correctly matched disulfide bonds, was seperated from incompletely oxidized IFN-α and dimeric IFN-αa, by use of a CM-Sepharose column, followed by size exclusion columns at two different pH conditions. The purified protein has been subjected to detailed physicochemical characterization including sequence determination. Unlike other rhIFN-α2a from E. coli reported, the rhIFN-α2a from S. cereuisioe has no methionine residue at its N-terminus originating from the start codon, ATG. The pI of the protein was determined to be 6.05 with a single band in the pI gel, which demonstrated that the purified rhIFN-α was homogeneous. The structural study using circular dichroism showed that the protein retains its three dimensional structure in the wide range of pH conditions between pH 3 and 9, and only minor structural deformation was observed at pH 1.0.

Purpose: Kikuchi-Fujimoto disease (KFD) is a benign disease, which is characterized by a cervical lymphadenopathy with fever, and it often mimics malignant lymphoma (ML). 2-[$^{18}F$]fluoro-2-deoxy-D-glucose positron emission tomography/computed tomography ($^{18}F$-FDG PET/CT) is a powerful imaging modality for the diagnosis, staging and monitoring of ML, with the limitations including the nonspecific FDG uptake in infectious or inflammatory processes. This study compared clinical manifestations and PET/CT findings between KFD and ML patients. Methods: We retrospectively reviewed the medical records of 23 patients with KFD and 33 patients with ML, diagnosed histopathologically, between January 2000 and May 2013 at the Department of Pediatrics, Yeungnam University Medical Center. Among them, we analyzed the clinical manifestations, laboratory findings and characteristics, and the amount of $^{18}F$-FDG uptake between 8 KFD and 9 ML patients who had $^{18}F$-FDG PET/CT. Results: The $^{18}F$-FDG PET/CT maximum standardized uptake values ($SUV_{max}$) ranged from 8.3 to 22.5 (mean, 12.0) in KFDs, and from 5.8 to 34.3 (mean, 15.9) in MLs. There were no significant differences in $SUV_{max}$ between KFDs and MLs. $^{18}F$-FDG PET/CT with ML patients showed hot uptakes in the extranodal organs, such as bone marrow, small bowel, thymus, kidney, orbit and pleura. However, none of the KFD cases showed extranodal uptake (P<0.001). $^{18}F$-FDG PET/CT findings of KFD with nodal involvement only were indistinguishable from those of ML. Conclusion: Patients who had extranodal involvement on PET/CT were more likely to have malignancy than KFD.
