        The extractability and stability of ginkgoflavonolglycosides under presence of several cellulase preparations were investigated. The enzymes used were macerosin, cellulase C and cellulase NC. The content variation of the glycosides was measured with HPLC method, using caffeic acid as an internal standard. The methanol extract of ginkgo leaf, containing the total flavonolglyco-sides of 4.46%, was used for the content comparison. By extraction with the enzymes, each or mixed, the peak levels of all the glycosides began to decrease after 1 or 2 hours. After 24 hour extraction, most of the glycosides were degraded to minor components. The flavonolglycosides in ginkgo leaf were also hydrolysed simply by the water extraction. After 24 hour extraction with water at 40°C, the peak levels of major glycosides were distinctly decreased. Rutin was hydrolysed by enzyme treatment or by ginkgo leaf itself. As a result, it was concluded that the commercially available cellulases and the ginkgo leaf itself contain the activities of β-glycosidase and α-rhamnosi-dase. Kaempferol-3-0-(6' -0-p-coumaroylglucosyl)-rhamnoside and four other ginkgo flavonolglycosides were not hydrolysed under the same condition."

      • 신생검증례에 의한 사구체신질환의 분석

        To elucidate the incidences and the clinical characteristics of glomerular diseases in our hospital, we reviewed the clinical records and histological findings of the 117 cases, which were diagnosed as one of the glomerular disease in renal biospy for about 6 years since 1991. In these 117 cases, 74 cases (63.2 %) fell into primary glomerulonephritis(GN), 18 cases (15.4%) into secondary GN, 2 cases (1.7 %) into end stage kidney, 4 cases (3.4%) into tubulointerstitial disease and the biopsy samples were inadequate for diagnosis in 19 cases. In primary glomerular diseases, most common pathologic diagnosis was IgA nephropathy (36.5 %}, followed by minimal change nephrotic syndrome (29.7%). The incidence of IgA nephropathy would be increased if we had biopsied every asymptomatic urinary abnormality (AUA) patients. Systemic lupus erythematosus (SLE) . hepatitis B accounted for most of secondary GN in our cases. One case of progressive scleroderma was included in this secondary GN. Minimal change nephrotic syndrome (22 cases) was the most common cause of nephrotic syndrome, followed by membranous GN (9 cases) and IgA nephropathy (8 cases). 2 cases of primary amyloidosis were presented with nephrotic syndrome. Nephrotic syndrome as presenting symptom in our IgA nephropathy patients was higher than in other studies, which may be due to indication bias for renal biopsy. We suggest that renal biopsy should be done in adult nephrotic syndrome and in SLE, because various kind of glomerular diseases can develop nephrotic syndrome in adult, renal biopsy can affect the treatment and prognosis in each patients and renal biopsy also determines the extent of renal involvement in SLE on which the treatment depends. In AUA urinary patients, renal biopsy could be reserved for the patients who show declining of GFR or rising of proteinuria. Because IgA nephropathy account for most of AUA. and there is no specific treatment for IgA nephropathy, and renal biopsy in those cases is helpful in ruling out the patients who need no treatment.

        Dual effects of isoflavonoids from <i>Pueraria lobata</i> roots on estrogenic activity and anti-proliferation of MCF-7 human breast carcinoma cells

        <P><B>Abstract</B></P> <P> <I>Pueraria lobata</I> root (PLR), well known as Kudzu root, has recently become commercially available in Western dietary supplements for menopausal symptoms. The scientific basis for its action has been attributed to the action of phytoestrogens. This study aimed to investigate the estrogen-like activity of isoflavonoids isolated from <I>P. lobata</I> root and their safety with respect to their effect on breast cancer cell proliferation. In an E-screen assay, crude MeOH extract of PLR significantly increased the proliferation of MCF-7 cells in a concentration-dependent manner. Among the four fractions obtained by solvent fractionation of MeOH extract, the <I>n</I>-BuOH fraction had significant estrogen-like activities at all concentrations tested. Phytochemical analysis of the <I>n</I>-BuOH fraction led to the isolation of 10 isoflavones (<B>1</B>–<B>10</B>), among which genistein (<B>10</B>) had significant estrogen-like activities at all concentrations tested. These activities were significantly enhanced by treatment with genistein and 17β-estradiol compared with 17β-estradiol alone, and this effect was mediated by decreased expression of estrogen receptor (ER)α and phospho-ERα in MCF-7 cells. In a cell cytotoxicity assay, genistein (<B>10</B>) exhibited significant cytotoxicity in both ER-positive MCF-7 and ER-negative MDA-MB-231 breast cancer cells. This cytotoxicity was characterized by the induction of apoptotic cells stained with annexin V conjugated with Alexa Fluor 488 and involved activation of mitochondria-independent and -dependent apoptosis pathways in MCF-7 cells.       • 활강 압박고 나사와 전자부 고정 금속판을 이용한 대퇴골 전자부 분쇄골절의 치료

        목적 : 대퇴골 전자부 분쇄 골절기 활강 압박고 나사만으로 치료한 결과와 활강 압박고 나사와 전자부 고정화 금속판을 함께 사용하여 치료한 결과를 비교 관찰하고자 하였다. 연구 대상 및 방법 : 1997년 6월부터 2000년 8월까지 대퇴골 전자부 골절로 입원하였던 환자로 전자 관통 골절(A2)과 대 전자부 하외측 피질골에 골절선이 동반된 전자간 골절(A3) 환자 중 대전자부 골절이 동반된 환자로 32명을 대상으로 하였으며 활강 압박고 나사만 사용한 활강 압박고 나사군 과 전자부 고정화 금속판을 같이 사용한 전자부 고정화 금속판군을 나누어서 단순 방사선 사진상 지연나사의 활강 정도와 대전자의 외측 전위 정도 및 골유합 기간을 추시하였다. 결과 : 모든 예에서 골유합이 되었으며 추시 단순 방사선 사진상으로 골유합이 되었을 때 측정한 지연 나사 활강 정도는 활강 압박고 나사군에서는 14.5mm, 전자부 고정화 금속판 군에서는 12.6mm이었다. 대퇴 대전자 평균 외측 전위 정도는 활강 압박고 나사군에서는 9.8mm, 전자부 고정화 금속판군에서는 1.2mm이었다. 결론 : 활강 압박고 나사와 전자부 고정화 금속판을 이용하여 대퇴골 전자간 분쇄 골절과 함께 대전자부 골절이 동반되어있는 경우의 치료시 방사선학적 추시상 지연나사의 활강 정도 및 대전자 외측 전위가 적었으며, 기존의 압박고 나사를 단독으로 사용하는 것에 비하여 수술시 얻은 골절 정복 및 내고정의 위치를 계속 유지하여 골유합을 시킬 수 있는 좋은 방법이라 사료된다. Purpose : The purpose of this study is to evaluate the results of treatment of comminuted femoral trochanteric fracture using dynamic hip screw(DHS) with trochanteric stabilizing plate(TSP) and DHS only. Materials and Methods : we analysed retrospectively 32 cases that has fracture extends over two or more levels of medial cortex(A2 of AO classification) and fracture extends through lateral cortex of femur(A3 of AO classicification) of femoral trochanteric fractures between 1997 and 2000. On simple AP radiograph of the DHS with TSP(n=16) and DHS only group(n=16), we reviewed bony union, slippage of lag screw, lateral displacement of greater trochanter. Result : Bony union was observed in all cases. When bony union is done in follow up radiograph, Mean slippage of lag screw is 14.5mm in DHS only group, 12.6㎜ in DHS with TSP group and mean lateral displacement of greater trochanter is 9.8㎜ in DHS only group, 1.2mm in DHS with TSP group. Conclusion : Use of DHS with TSP in comminuted femoral trochanteric fracture is lesser slippage of lag screw and lateral displacement of greater trochanter than DHS only used, and that is better method to maintain fracture reduction and internal fixation in treatment of comminuted femoral trochanteric fractures than DHS only.

        Effect of Methanol Extract from Cassia mimosoides var. nomame on Ischemia/Reperfusion-induced Renal Injury in Rats

        Objectives : The purpose of this study was to determine whether the methanol extract of Cassia mimosoides var. nomame Makino, a naturally growing plant in Korea, could prevent the renal-ischemia/reperfusion injury in a rat model or not. Methods : The radical scavenging activities of the extracts, and ascorbic acid as a positive control, were measured in vitro. At one hour after an intraperitoneal injection of the extract (400 mg/kg), renal ischemia/reperfusion injury was generated by 40 min clamping of the left renal artery in rats. After renal ischemia/reperfusion and 24 hr restoration of blood circulation, the serum creatinine concentration was measured. And the extent of epithelial cell injury and apoptosis was assessed by various staining technologies. The Bax/Bcl-2 ratio and activated caspase-3 were assessed by immunohistochemistry. Results : The extract showed a slightly lower level of radical scavenging activity than that of ascorbic acid. Compared to those of the vehicle-treated group, the extract-treated group displayed a significantly smaller tubular epithelial cell injury of 54% reduction in the outer medulla region and a lower serum creatinine concentration of 50% reduction. It seems that the reduction in cellular injury is due to the attenuation of the Bax/Bcl-2 ratio, and the inhibition of caspase-3 activation by the extract of Cassia mimosoides. Conclusions : Cassia mimosoides var. nomame Makino could be a good candidate for a prophylactic agent against the ischemia/reperfusion/induced kidney injury.

      • Electrospinning of polystyrene/dicyanopyrazine-linked porphyrin nanofibers.

        <P>We have fabricated fluorescing polystyrene/dicyanopyrazine-linked porphyrin (PS/4-TDCPP) nanofibers using the electrospinning technique. UV-vis spectroscopy shows a strong Soret band and two relatively weak Q bands from the PS/4-TDCPP films and fibers, and reveals that the 4-TDCPP molecules are homogeneously dispersed in the films and fibers. Scanning electron microscopy (SEM) reveals the effect of solvent and collecting distance on the morphology of the electro-spun PS/4-TDCPP fibers. Fibers spun from a 50% dimethlyformamide (DMF), 50% methylethylketone (MEK) solution have ultra-fine structures with an average diameter of 300 nm. In the case of fibers from pure DMF and DMF:MEK (1:3) solutions, beads are formed along the length of the fibers. Variation of the collecting distance from 20 to 30 cm does not induce significant differences in the morphology of the electro-spun PS/4-TDCPP fibers. However, at a collecting distance of 15 cm, many beads are formed along the fibers. Acid-sensing capability of the PS/4-TDCPP fibers is demonstrated by fluorescence microscopy.</P>

        Diagnostic Role of Bile Pigment Components in Biliary Tract Cancer

        Bile pigment, bilirubin, and biliverdin concentrations may change as a results of biliary tract cancer (BTC) altering the mechanisms of radical oxidation and heme breakdown. We explored whether changes in bile pigment components could help distinguish BTC from benign biliary illness by evaluating alterations in patients with BTC. We collected bile fluid from 15 patients with a common bile duct stone (CBD group) and 63 individuals with BTC (BTC group). We examined the bile fluid’s bilirubin, biliverdin reductase (BVR), heme oxygenase (HO-1), and bacterial taxonomic abundance. Serum bilirubin levels had no impact on the amounts of bile HO-1, BVR, or bilirubin. In comparison to the control group, the BTC group had considerably higher amounts of HO-1, BVR, and bilirubin in the bile. The areas under the curve for the receiver operating characteristic curve analyses of the BVR and HO-1 were 0.832 (p<0.001) and 0.891 (p<0.001), respectively. Firmicutes was the most prevalent phylum in both CBD and BTC, according to a taxonomic abundance analysis, however the Firmicutes/Bacteroidetes ratio was substantially greater in the BTC group than in the CBD group. The findings of this study showed that, regardless of the existence of obstructive jaundice, biliary carcinogenesis impacts heme degradation and bile pigmentation, and that the bile pigment components HO-1, BVR, and bilirubin in bile fluid have a diagnostic significance in BTC. In tissue biopsies for the diagnosis of BTC, particularly for distinguishing BTC from benign biliary strictures, bile pigment components can be used as additional biomarkers.

        Methanol Extract of Goat's-beard (Aruncus dioicus) Reduces Renal Injury by Inhibiting Apoptosis in a Rat Model of Ischemia-Reperfusion

        Interruption or prolonged reduction and subsequent restoration of blood flow into the kidney triggers the generation of a burst of reactive oxygen species (ROS), leading to injury in the tubular epithelial cells. In this study, we determined whether methanol extract of goat's-beard (Aruncus dioicus) (extract) could prevent this ischemia/reperfusion injury. When in vitro radical scavenging activity of the extract was measured using a DPPH radical quenching assay, the extract displayed slightly lower activity than ascorbic acid. One hour after administration of the extract (400 mg/kg) by intraperitoneal injection in rats, renal ischemia/reperfusion injury was generated by clamping the left renal artery for forty minutes, followed by 24 hr restoration of blood circulation. Prior to clamping the left renal artery, the right renal artery was removed. Compared with the vehicle-treated group, pretreatment with the extract significantly reduced the tubular epithelial cell injury by 37% in the outer medulla region, and consequently reduced serum creatinine concentration by 39%. Reduction in the cell injury was mediated by attenuation of Bax/Bcl-2 ratio, inhibition of caspase-3 activation from procaspase-3, and subsequent reduction in the number of apoptotic cells. Thus, goat's-beard (Aruncus dioicus) might be developed as a prophylactic agent to prevent acute kidney injury.

        A new s-adenosylhomocysteine hydrolase-linked method for adenosine detection based on DNA-templated fluorescent Cu/Ag nanoclusters

        <P><B>Abstract</B></P> <P>We herein describe a novel fluorescent method for the rapid and selective detection of adenosine by utilizing DNA-templated Cu/Ag nanoclusters (NCs) and employing s-adenosylhomocysteine hydrolase (SAHH). SAHH is allowed to promote hydrolysis reaction of s-adenosylhomocysteine (SAH) and consequently produces homocysteine, which would quench the fluorescence signal from DNA-templated Cu/Ag nanoclusters employed as a signaling probe in this study. On the other hand, adenosine significantly inhibits the hydrolysis reaction and prevent the formation of homocysteine. Consequently, highly enhanced fluorescence signal from DNA-Cu/Ag NCs is retained, which could be used to identify the presence of adenosine. By employing this design principle, adenosine was sensitively detected down to 19nM with high specificity over other adenosine analogs such as AMP, ADP, ATP, cAMP, guanosine, cytidine, and urine. Finally, the diagnostic capability of this method was successfully verified by reliably detecting adenosine present in a real human serum sample.</P> <P><B>Highlights</B></P> <P> <UL> <LI> A new fluorescent method for adenosine detection utilizing DNA-Cu/Ag NCs and SAHH. </LI> <LI> The experimental procedure to detect adenosine conducted in a quite simple manner. </LI> <LI> Adenosine is detected with high specificity over other adenosine derivatives. </LI> <LI> The practical utility is verified by determining adenosine in human serum. </LI> <LI> A new platform for the convenient biomolecular detection based on Cu/Ag NCs. </LI> </UL> </P>
