Estimation of Antibacterial Properties of Chlorophyta, Rhodophyta and Haptophyta Microalgae Species

In this exploratory study, eight types of microalgae from different phyla (Chlamydomonas reinhardtii, Chlorella species, Haematococcus pluvialis, Porphyridium purpureum, Porphyridium cruentum, Isochrysis species, Isochrysis galbana, and Pavlova lutheri) were tested for their antibacterial activities against eight target pathogenic bacterial strains. The agar well diffusion method and broth micro dilution assay were conducted to estimate the antibacterial activity. Microalgae cell-free supernatants, exopolysaccharides (EPS), water, and organic solvent extracts were used for inhibition analysis. EPS extracted from P. lutheri showed activity against Bacillus subtilis and Pseudomonas aeruginosa. Inhibition zone diameters of 14-20 mm were recorded on agar plates, while the minimum inhibitory concentrations in the broth micro dilution assay were $0.39-25mg\;ml^{-1}$. During this study, haptophyte microalgae, Isochrysis species, and P. lutheri extracts showed the highest activity against most of the tested pathogenic bacterial strains, while most of the extracts were active against the important foodborne pathogen P. aeruginosa. This study showed promising results regarding important microalgae phyla, which will further aid research related to extracts and exploitation of bioactive metabolic compounds in the food and pharmaceutical industries.

<P>Fish-paste products, also known as fish cakes or surimi-based products, are worldwide favorites. Surimi, a wet protein concentrate of fish muscle, is used as an intermediate raw material to produce surimi seafood. The flavor, texture, taste, shelf-life, and market value of surimi-based products depend on the source of the fish meat, type of applied heat treatment, and additives used to prepare the surimi. While preparing surimi with chemical additives, several problems have been observed, such as a lack of unique characteristics, inferior acceptability, and poor functionality. Various types of fish-paste products have been developed by using different ingredients (e.g., vegetables, seafood, herbs and oriental medicines, grains and roots including carrots, and functional food materials). However, a systematic review of fish-paste products prepared using natural food additives has not yet been performed. Therefore, the quality characteristics of fish-paste products and their functionalities were elucidated in this study. With the increasing demand for surimi seafood products, the functional properties, physiochemical properties, and shelf-life of surimi-based products need to be improved. This review will aid the preparation of new value-added products in the surimi industry.</P>

Measuring Mechanical Cell Properties of Paclitaxel-treated MCF-7 Cells in a Monolayer

Cytoskeleton is the key element of cell proliferation. It regulates the size and shape of the cells, and it allows cell growth, division and locomotion. Cytoskeleton-affecting anti-cancer drugs work with interfering the proliferation by polymerization or depolymerization of cytoskeleton. To elucidate the mechanical impact of these drugs on cancer cells, we have measured the viscoelastic cell properties of MCF-7 cells in a monolayer with a ‘narrow gap rheometer’ after treatment with hydroxyurea and paclitaxel. Paclitaxel impedes depolymerization of the microtubule, which may have an impact on the cell mechanics. It acts cytotoxically at sufficiently high concentrations during the division cycle. To study living cells, we prevent the cells from division by using hydroxyurea, which arrests the cell cycle. To access the cell mechanics on the cell scale, we have improved the precision of commercial rheometers by about two orders of magnitude. Here, we focus on the impact of paclitaxel on the storage and loss moduli at different concentrations.
