Effects of gynosaponin TN-2 on L-DOPA-induced cytotoxicity in PC12 cells

Gynosaponins have pharmacological effects on 3,4-L-dihydroxyphenylalanine (L-DOPA)-related or dopamine-related neurological diseases; however, the neuroprotective functions of single compound of gynosaponins remain undefined. This study investigated the cytotoxic effects of gynosaponin TN-2 on L-DOPA in pheochromocytoma 12 cells. Gynosaponin TN-2, at 0.5−3 μM, did not exhibit cytotoxicity and protected against L-DOPA (100 and 200 μM)-induced cell death. Gynosaponin TN-2 (0.5 and 1.0 μM) inhibited the L-DOPA (100 and 200 μM)-induced sustained extracellular signal-regulated protein kinases 1 and 2 phosphorylation. Gynosaponin TN-2 at 0.5 and 1.0 μM also reduced L-DOPA (100 and 200 μM)-induced JNK1/2 phosphorylation and cleaved caspase-3 expression. These results suggested that gynosaponin TN-2 exerts protective effects on L-DOPA (100 and 200 μM)-induced apoptotic cell death by modulating extracellular signal-regulated protein kinases 1 and 2 activation in pheochromocytoma 12 cells.
Salmonella typhimurium에 의한 고관절염 1예

Salmonella septic arthritis is unusual and rare manifestation noted in fewer than 1% of salmonellosis. S. typhimurium is the most prevalent among the salmonella species from clinical specimens including bone and joint in United States and other countries, on the other hand, S. typhi is the most frequent organism in Korea. There has been no case report of septic arthritis caused by S. typhimurium in Korea. We report the first case of septic arthritis due to S. typhimurium in a patient with diabetes mellitus. The patient was a 43-year-old male. He had undergone sequestrectomy and iliac bone graft of the hip joint because of tuberculosis 16 years age, thereafter he took oral anti-tuberculous medication for a year. Six months before admission high fever and copious pus discharge were noted. And he was admitted to the our hospital via local clinic in Cheju Island, where new onset of diabetes mellitus was detected, for further proper managements. There was no history of Salmonella septicemia, dysenteric infection or pulmonary tuberculosis. Physical examination revealed warm, swollen and tender areas with two active draining sinuses on the anterolateral aspect of right hip joint, which was in fused state. And there were also an old operation scar and five healded draining sinuses. Cultures of pus obtained from right hip at reoperation grew group B Salomonella which was positive for H₂S, motility, citrate, fermentation of mannitol and sorbitol. Salmonella was finally identified as S. typhimurium having 1, 4, 5 and 12 of somatic (O) antigen, i on phase 1, 1 and 2 on phase 2 of flagella (H) antigens by Serological tests performing at National Institute of Health. In the other laboratory findings were as follows; WBC 12,000/㎕, Hb/Hct 15.2/46, platelets 510,000/㎕ on CBC, FBS/pp2h:330/482, glucosuria (3+), CRP(3+), Cryglobulin(-), anti-HIV Ab(-), 1:40 in both O and H titer of Widal tests, and HLA B27(-). AFB was not isolated on stained smears and cultures. He was treated with cefradine 1.0g and kanamycin 1.0g initially and chloramphenicol 1.0g daily for 6 weeks. And 8 U RI and 16 U human insulin(Novolyn N) were administrated for DM control. He responded to the treatment and he was retransferred to Cheju local clinic for continuous DM control.

An insecticidal crystal protein gene, crylA(c), from Bacillus thuringiensis HD-73 was integrated into the chromosome of a xanthan-producing bacterium, Xanthomonas campestris XP92. The crylA(c) gene expression cassette was constructed that placed the gene between the trc promoter and rmB transcriptional terminator. The lacl^q gene was also included to prevent the expression of crylA(c) gene in X campestris cells. Southern blot analysis confirmed the integration of the crylA(c) gene expression cassette in chromosome of X campestris XP92 transconjugant. Expression of the insecticidal crystal protein was confirmed by Western blot analysis and bioassay against the larvae of Hyphantria cunea (Lepidoptera: Arctiidae) and Plutella xylostella (Lepidoptera: Plutellidae).
Carrageenan 유도 관절염에 대한 灸律 藥鍼의 진통작용

Objectives: This study was produced to examine the effects of moxibustion that had been played important role to traditional oriental medical treatment on disease. Recently, it was reported that moxi-tar which is generated in the process of moxibustion as burning combustibles decreased nitric oxide(NO) and inducible NO synthase (iNOS) generation in cellular experiments. Methods: Carrageenan-induced arthritis rat model was used to test the effect of moxi-tar as a chronic pain model. Diluted moxi-tar was single injected in several acupoints or combined with electroacupuncture (1 ms, 2 Hz, and 2 mA) into contralateral ST36 acupoint for 30 min to assess the synergic effects. After the treatment, behavioral tests measuring stepping force were periodically conducted during the next 12 hours. Endogenous NO and iNOS, cyclooxygenase-2 (COX-2), and c-Fos protein expression in the spinal cord were examined on a rat model of carrageenan-induced arthritis. Results : After the induction of arthritis, rats subsequently showed a reduced stepping force of the affected limb for at least the next 4 days. The reduced stepping force of the limb was presumably due to a painful knee, since oral injection of indomethacin produced temporary improvement of weight bearing. Moxi-tar produced significant improvement of stepping force of the hindlimb affected by the arthritis lasting at least 9 hours. The magnitude of this improvement was equivalent to that obtained after an oral injection of 3 mg/kg of indomethacin and this improvement of stepping force was interpreted as an analgesic effect. Moxi-tar produced the improvement of stepping force of the affected hindlimb in a dose-dependent manner. Both NO production and iNOS, COX-2 protein expression increased by arthritis were suppressed by moxi-tar. Moxi-tar on combination with electroacupuncture (EA) produced more powerful and longer lasting improvement of stepping force of the hindlimb affected by the arthritis than either moxi-tar or EA did. Conclusion : The present study suggest that moxi-tar produces a potent analgesic effect on the chronic knee arthritis pain model in the rat and that moxi-tar-induced analgesia modulate endogenous NO through the suppression of iNOS/COX-2 protein expression.
Is the anatomical resection necessary for single hepatocellular carcinoma smaller than 3 cm?

Backgrounds/Aims: The superiority of anatomical resection (AR) for a small HCC remains controversial. In this study, we investigated the clinical outcomes after AR and non-anatomical liver resection (NAR) for single HCC smaller than 3 cm and the risk factors for HCC recurrence. Methods: A total of 116 consecutive patients who underwent liver resection for single HCC (<3 cm) between Jan 2006 and Dec 2015 were included in this study. The medical records of these patients were reviewed and analyzed retrospectively. Results: There was no significant difference in tumor recurrence and survival between AR and NAR group. Multivariate analysis showed that hepatitis B (p=0.035, HR=8.72), presence of satellite nodule (p=0.029, HR=3.97) and microvascular invasion (MVI) (p=0.039, HR=2.79) were independent risk factors for early recurrence within 1 year. The overall recurrence was independently related to the presence of satellite nodule (p=0.001, HR=4.98) and background liver cirrhosis (p=0.032, HR=1.96). In patients with MVI, HCC recurrence was significantly more frequent in width of safety margin <1 cm group than ≥1 cm group (p=0.049). Conclusions: The outcomes of NAR are comparable with those of AR in single HCC smaller than 3 cm. The presence of satellite nodule, MVI and hepatitis B are the independent risk factors for early recurrence, however overall recurrence is correlated with background liver cirrhosis and the presence of satellite nodule rather than pathobiologic factors in single HCC smaller than 3 cm. Hepatic resection with sufficient margin (≥1 cm) is recommended for decreasing risk of recurrence in patients with suspected MVI.

HVDC transmission lines must be designed to satisfy environmental regulations. Therefore it is necessary to pre-evaluate environmental problems for transmission line designer using prediction program. In this study, environment design software, for HVDC transmission lines was developed as a comprehensive window program. It can calculate electric field strength of ground surface, ion current density and charged voltage. This program will be usefully applied to the environmental friendly design and construction of HVDC transmission lines.

본 논문에서는 가정용 직류배전시스템 도입 시 가전기기 입력단 커패시터의 용량 산정 및 최적화 가능성 검증을 위하여 직류배전시스템용 가전기기 입력단 및 PWM 컨버터 출력단 사이의 관계를 전압방정식의 형태로 모델링 한다. 또한 모델링된 전압방정식을 이용하여 대용량 부하변화 시 발생가능 한 Sag 및 Surge 규격에 부합하는 커패시터의 용량을 산정하고 시뮬레이션을 통하여 검증한 뒤 직류배전시스템 가전기기 설계의 가이드라인을 제시한다.
