Cloning and characterization of a novel gene with alternative splicing in murine mesenchymal stem cell Line C3H/10T1/2 by gene trap screening

( Ming Ke Wang ),( Hui Qin Sun ),( Fan Jiang ),( Jing Han ),( Feng Ye ),( Tao Wang ),( Yong Ping Su ),( Zhong Min Zou ) 생화학분자생물학회 (구 한국생화학분자생물학회) 2010 BMB Reports Vol.43 No.12

A novel gene, designated mgt-6, containing four splicing variants, was isolated from a gene trap clone library of C3H/10T1/ 2 cells transfected with retroviral promoterless gene-trap vector, ROSAFARY. The transcript variants were differentially expressed in murine tissues and cell lines and differentially responded to diverse stimuli including TGF-β1 and mitogen-activated protein kinase (MAPK) inhibitors. The mgt-6 gene encoded a protein of 37 or 11 amino acid residuals with cytoplasmic distribution. However, when C3H/10T1/2 cells were treated with 5-azacytidine, the protein translocated into cell nucleus as indicated by fused LacZ or C-terminally tagged EGFP. Our preliminary results suggest that further study on the role of mgt-6 gene in cell transformation and differentiation may be of significance. [BMB reports 2010; 43(12): 789-794]

      • Decreased risks of internal malignancy in patients with vitiligo: a 10-year population-based cohort study

( Jung Min Bae ),( Han Mi Jung ),( Hyuck Sun Kwon ),( Sook Jung Yun ),( Hee Su Kim ),( Byung Cheol Park ),( Joung Soo Kim ),( Soo Hong Seo ),( Dong-youn Lee ),( You Chan Kim ),( Hyang Joon Park ),( Ke 대한피부과학회 2017 대한피부과학회 학술발표대회집 Vol.69 No.2

Background: Recently, the autoimmune process of vitiligo has been further identified by the demonstration of CD8+T cells playing a major role in spreading vitiligo. Although the decreased risks of skin cancers in patients with vitiligo has been found, the risks of internal malignancy have not been assessed yet. Objectives: To determine the risks of internal malignancy in patients with vitiligo. Methods: A population-based cohort study was conducted using the Korean NHI Claims database from 2007 to 2016. All patients with vitiligo aged ≥20 years were identified, and age- and sex- matched controls selected from patients without vitiligo. Outcomes of interests were the incidences of internal malignancy since the enrollment. Results: A total of 101,078 patients with vitiligo and 202,156 controls without vitiligo were enrolled. The overall risk of internal malignancy was 612.9 and 708.9 per 100,000 person-years in vitiligo patients and controls, respectively, and was significantly decreased in patients with vitiligo (HR 0.857, 95% CI 0.823-0.893) after adjustment for potential confounders. In subgroup analyses, vitiligo was significantly associated with the reduced cancer risks of stomach (HR 0.838), colon and rectum (HR 0.617), lung (HR 0.748), ovary (HR 0.617), and leukemia (HR 0.592) (all P < 0.003). Conclusion: The autoimmune nature of vitiligo might protect patients with vitiligo from the development of various internal malignancies.

      • Development of Standard Radiation Sources for Verification of Classification System of Nuclear Decommissioning Waste

        So Jung Shim,Seung Su Shin,Young-Ku Choi,Sang-Han Lee,Minju Lee,Ke Chon Choi 한국방사성폐기물학회 2022 한국방사성폐기물학회 학술논문요약집 Vol.20 No.1

This study is about the production of radiation sources of simulated concrete and soil reference materials to verify the validity of the quality establishment and measurement of the detector (HPGe) of the radioactive soil and concrete waste classification system, which is being developed to quickly and accurately classify nuclear decommissioning waste. Specific activity of gamma nucleus among radioactive wastes is evaluated using gamma spectroscopy. At this time, in order to verify the validity and reliability of measuring equipment, it shall be a standardized substance of the same medium as nuclear decommissioning waste (chemical ingredients, particles, density, etc.) in order to correct the energy and efficiency of gamma nuclide analysis equipment. The CRM used for the detector’s energy correction used a 1 L Marinelli beaker standard correctional radiation source consisting of 10 radioactive isotopes. In order to correct efficiency, in accordance with the production and certification process of the Korea Standards and Research Institute, it has produced artificial simulated radioactive concrete similar to nuclear decommissioning waste (30% for cement, 60% for regulation and 10% for bentonite). The radioactive homogeneity of the simulated concrete reference materials was evaluated using dispersion analysis (ANOVA) in accordance with ISO Guide 35, while 137Cs and 60Co of concrete reference materials were able to obtain homogeneous measurements both in and between bottles. The self-absorption rate of the simulated concrete reference material was determined by the MCNP computer simulation measurement method, and the self-absorption correction coefficients of 137Cs and 60Co were assessed at 0.995 and 0.996, respectively, and the standard value for the radiation of the simulated concrete reference material was calculated on the weighted average of the measurements of 20 samples. The uncertainty about the reference value was calculated by combining measurement uncertainty (Type B evaluation), bottle to bottle standard deviation, and uncertainty within bottle by modifying the formula suggested in ISO Guide 35. The concentration of 137Cs and 60Co of reference materials was divided into high-speed measurement mode and precision measurement mode in consideration of the self-disposal standard. The reference value and uncertainty of expansion among reference materials for high-speed measurement mode were rated at 1,032.7 ± 64.0 Bq·kg?1and 1,083.7 Bq·kg?1, respectively. The standard value and expansion uncertainty for 137Cs and 60Co among reference materials for precision measurement mode were rated at 113.7 ± 10.0 Bq·kg?1 and 122.3 ± 10.3 Bq·kg?1, respectively.

Ring-Type Rotary Ultrasonic Motor Using Lead-free Ceramics

( Chang Hyo Hong ),( Hyoung Su Han ),( Jae Shin Lee ),( Ke Wang ),( Fang Zhou Yao ),( Jing Feng Li ),( Jung Ho Gwon ),( Nguyen Van Quyet ),( Jin Kyung Jung ),( Wook Jo ) 한국센서학회 2015 센서학회지 Vol.24 No.4

Ultrasonic motors provide high torques and quick responses compared to their magnetic counterparts; therefore, they are widely used in small-scale applications such as mobile phones, microrobots, and auto-focusing modules in digital cameras. To determine the feasibility of lead-free piezoceramics for ultrasonic motor applications, we fabricated a ring-type piezoceramic with a KNN-based lead-free piezoceramic (referred to as CZ5), intended for use in an auto-focusing module of a digital camera. The vibration of the lead-free stator was observed at 45.1 kHz. It is noteworthy that the fully assembled lead-free ultrasonic motor exhibited a revolution speed of 5-7 rpm, even though impedance matching with neighboring components was not considered. This result suggests that the tested KNN-based piezoceramic has great potential for use in ultrasonic motor applications, requiring minimal modifications to existing lead-based systems.

Disturbance Suppression and Contour Following Accuracy Improvement: An Adaptive PI-Type Sliding Mode Nonlinear Extended State Observer Approach

        Yen-Chun Chen,Yan-Rou Cai,Ming-Yang Cheng,Ke-Han Su 한국정밀공학회 2023 International Journal of Precision Engineering and Vol.24 No.3

The main idea behind the conventional extended state observer is that the total disturbance including nonlinearity, modeling error, and external disturbance, etc., is regarded as an augmented state variable which can be estimated using system input/output, system parameters and a nonlinear function. In order to further improve estimation accuracy, this paper proposes an adaptive PI-type sliding mode nonlinear extended state observer (APISMESO). In particular, a nonlinear function that contains an integral term is used in the proposed observer structure to reduce estimation error, especially when dealing with static friction and high-frequency noise often existing in servomechanisms. Moreover, an adaptive law is designed to provide on-line estimation of system parameters. In addition, the stability of the complementary sliding mode control scheme with the proposed APISMESO is investigated using the Lyapunov stability theory. Several contour following experiments are performed to compare the performance of the proposed APISMESO with that of several existing variants of extended state observers. Experimental results reveal that the proposed APISMESO outperforms the other variants of extended state observers also tested in the experiment.

A brief review on relaxor ferroelectrics and selected issues in lead-free relaxors

        Ahn, Chang Won,Hong, Chang-Hyo,Choi, Byung-Yul,Kim, Hwang-Pill,Han, Hyoung-Su,Hwang, Younghun,Jo, Wook,Wang, Ke,Li, Jing-Feng,Lee, Jae-Shin,Kim, Ill Won 한국물리학회 2016 THE JOURNAL OF THE KOREAN PHYSICAL SOCIETY Vol. No.

<P>Relaxor ferroelectricity is one of the most widely investigated but the least understood material classes in the condensed matter physics. This is largely due to the lack of experimental tools that decisively confirm the existing theoretical models. In spite of the diversity in the models, they share the core idea that the observed features in relaxors are closely related to localized chemical heterogeneity. Given this, this review attempts to overview the existing models of importance chronologically, from the diffuse phase transition model to the random-field model and to show how the core idea has been reflected in them to better shape our insight into the nature of relaxor-related phenomena. Then, the discussion will be directed to how the models of a common consensus, developed with the so-called canonical relaxors such as Pb(Mg1/3Nb2/3)O-3 (PMN) and (Pb, La)(Zr, Ti)O-3 (PLZT), are compatible with phenomenological explanations for the recently identified relaxors such as (Bi1/2Na1/2)TiO3 (BNT)-based lead-free ferroelectrics. This review will be finalized with a discussion on the theoretical aspects of recently introduced 0-3 and 2-2 ferroelectric/relaxor composites as a practical tool for strain engineering.</P>

      • Human Telomerase Gene and High-Risk Human Papillomavirus Infection are Related to Cervical Intraepithelial Neoplasia

Zhao, Xu-Ye,Cui, Yongm,Jiang, Shu-Fang,Liu, Ke-Jun,Han, Hai-Qiong,Liu, Xiao-Su,Li, Yali Asian Pacific Journal of Cancer Prevention 2015 Asian Pacific journal of cancer prevention Vol.16 No.2

Our aims were to evaluate the clinical performance of human telomerase RNA gene component (hTERC gene) amplification assay with high-risk human papillomavirus (HR-HPV) DNA test of Hybrid Capture 2 DNA test (HC2), for the detection of high grade cervical precancerous lesions and cancer (CIN 2+). In addition, the association shown between hTERC gene amplification and HPV DNA test positive in women with and without cervical neoplasia was assessed. There were 92 women who underwent cytology, HR-HPV DNA test, hTERC gene amplification test, colposcopy and biopsy. We compared the clinical performance of hTERC gene test along with HR-HPV DNA test of women with colposcopy and routine screening. The samples were histology-confirmed high-grade cervical intraepithelial neoplasia (CIN 2) or worse (CIN2+) as the positive criterion. The test of hTERC gene showed the hTERC gene amplification positivity increased with the severity of histological abnormality and cytological abnormality. The test of hTERC gene showed higher specificity than HR-HPV DNA test for high-grade lesions (84.4% versus 50%) and also higher positive predictive value (90.4% versus 76.5%). Our results predicted that hTERC gene amplification demonstrated more specific performance for predicting the risk of progression and offer a strong potential as a tool for triage in cervical cancer screening, with the limited sensitive as HR-HPV DNA test.
