      • Influence of Ribosomal Protein L39-L in the Drug Resistance Mechanisms of Lacrimal Gland Adenoid Cystic Carcinoma Cells

        Background: Cancer constitutes a key pressure on public health regardless of the economy state in different countries. As a kind of highly malignant epithelial tumor, lacrimal gland adenoid cystic carcinoma can occur in any part of the body, such as salivary gland, submandibular gland, trachea, lung, breast, skin and lacrimal gland. Chemotherapy is one of the key treatment techniques, but drug resistance, especially MDR, seriously blunts its effects. As an element of the 60S large ribosomal subunit, the ribosomal protein L39-L gene appears to be documented specifically in the human testis and many human cancer samples of different origins. Materials and Methods: Total RNA of cultured drug-resistant and susceptible lacrimal gland adenoid cystic carcinoma cells was seperated, and real time quantitative RT-PCR were used to reveal transcription differences between amycin resistant and susceptible strains of lacrimal gland adenoid cystic carcinoma cells. Viability assays were used to present the amycin resistance difference in a RPL39-L transfected lacrimal gland adenoid cystic carcinoma cell line as compared to control vector and null-transfected lacrimal gland adenoid cystic carcinoma cell lines. Results: The ribosomal protein L39-L transcription level was 6.5-fold higher in the drug-resistant human lacrimal gland adenoid cystic carcinoma cell line than in the susceptible cell line by quantitative RT-PCR analysis. The ribosomal protein L39-L transfected cells revealed enhanced drug resistance compared to plasmid vector-transfected or null-transfected cells as determined by methyl tritiated thymidine (3H-TdR) incorporation. Conclusions: The ribosomal protein L39-L gene could possibly have influence on the drug resistance mechanism of lacrimal gland adenoid cystic carcinoma cells.

        A Convergent Study on Seoul‘s Image as a Tourism Destination using V-method

        This study investigated Seoul’s image as a tourism destination that Chinese tourists perceived, taking advantage of Chinese Social Media “Renren”. The study used visual method (V-method) to collect images and analyzed them in terms of convergent disciplinary approach. It searched Chinese word, “首尔”, in Renren social network. 526 Seoul’s photos were collected from 129 users’ mini-homepage. Destination planners and marketers should understand how the Chinese tourists perceive the Seoul’s image and recognize the contribution factors to representations of the city. It is recommended that more cultural experience should be offered to Chinese tourists.

        Subjectivities of Ethnic Minority Groups from Xishuangbanna, Yunnan, China toward Ethnic Tourism: Using Q-Methodology

        Ethnic tourism is utilized by many countries to improve the economic status. Ethnic tourism in Xishuangbanna started from the 1980s and grew rapidly during the past 2 decades. Although many researchers had stressed economic and socio-cultural impacts of ethnic tourism on ethnic minority people, little was done to identify their subjectivities. This research revealed the social and economic benefits from ethnic tourism, representing the identity of ethnic minority people. This research investigated the ethnic groups’ subjectivities toward ethnic tourism and identity in Xishuangbanna, Yunnan, China, using the Q-methodology. It was found that the community residents did not get much benefits regardless of the social welfare that ethnic tourism had brought about. Finally, this research also made several implications with regard to tourism management.

        The Effect of Electron-withdrawing Group Functionalization on Antibacterial and Catalytic Activity of Palladium(II) Complexes

        The design, synthesis, and structural characterization of two new palladium complexes based on Schiff base ligands is reported; $[Pd(L1)_2]$ (1) and $[Pd(L2)_2]$ (2), [HL1 = 2-((E)-(2,6-diethylphenylimino)methyl)-4,6-dibromophenol, L2 = (E)-N-benzylidene-2,6-diethylbenzenamine], which are obtained by functionalizing Schiff base ligands with or without electron-withdrawing groups. Both compounds are mononuclear structures. Comparisons are made to the compounds 1 and 2 to analyze and understand the effect of electron-withdrawing groups. Antibacterial activity studies indicate the electron-withdrawing groups on Schiff base ligands enhance antibacterial activity. Catalytic activity, however, is reduced due to the enhanced steric-hindrance of the electron-withdrawing groups. Electronic absorption and emission properties of HL1, L2, 1 and 2 are also reported.

        Epigallocatechin gallate has pleiotropic effects on transmembrane signaling by altering the embedding of transmembrane domains

        <P>Epigallocatechin gallate (EGCG) is the principal bioactive ingredient in green tea and has been reported to have many health benefits. EGCG influences multiple signal transduction pathways related to human diseases, including redox, inflammation, cell cycle, and cell adhesion pathways. However, the molecular mechanisms of these varying effects are unclear, limiting further development and utilization of EGCG as a pharmaceutical compound. Here, we examined the effect of EGCG on two representative transmembrane signaling receptors, integrin alpha IIb beta 3 and epidermal growth factor receptor (EGFR). We report that EGCG inhibits talin-induced integrin alpha IIb beta 3 activation, but it activates alpha IIb beta 3 in the absence of talin both in a purified system and in cells. This apparent paradox was explained by the fact that the activation state of alpha IIb beta 3 is tightly regulated by the topology of beta 3 transmembrane domain (TMD); increases or decreases in TMD embedding can activate integrins. Talin increases the embedding of integrin beta 3 TMD, resulting in integrin activation, whereas we observed here that EGCG decreases the embedding, thus opposing talin-induced integrin activation. In the absence of talin, EGCG decreases the TMD embedding, which can also disrupt the integrin alpha-beta TMD interaction, leading to integrin activation. EGCG exhibited similar paradoxical behavior in EGFR signaling. EGCG alters the topology of EGFR TMD and activates the receptor in the absence of EGF, but inhibits EGF-induced EGFR activation. Thus, this widely ingested polyphenol exhibits pleiotropic effects on transmembrane signaling by modifying the topology of TMDs.</P>

        Effect of pulse electrodeposition parameters on electrocatalytic the activity of methanol oxidation and morphology of Pt/C catalyst for direct methanol fuel cells

        <P><B>Abstract</B></P> <P>The electrodeposition technique for preparing direct methanol fuel cell electrodes has been developed to increase the Pt utilization and lower the Pt loading. The performance of the Pt/C electrode for methanol oxidation reaction (MOR) was optimized by adjusting the electrodeposition parameters such as applied electrical signal types, ratios of t<SUB>on</SUB>/t<SUB>off</SUB>, deposition temperatures, and electrolyte concentrations, systematically. Furthermore, the effects of two kinds of additives, i.e. polyethylene glycol (PEG) and sodium dodecyl sulfonate (SDS), on the catalytic performance and morphology of Pt catalyst were investigated for MOR by SEM, XRD, cyclic voltammetry and linear sweep voltammetry. The results show that the optimal Pt catalyst has been prepared by the square wave current method with t<SUB>on</SUB>/t<SUB>off</SUB> of 1 s/5 s at 30 °C in a 1.0 mmol L<SUP>−1</SUP> H<SUB>2</SUB>PtCl<SUB>6</SUB> solution with a 10<SUP>−4</SUP> mmol L<SUP>−1</SUP> PEG additive. Moreover, the effect of the additive type and amount on the formation mechanism of the Pt crystallite morphology has also been discussed.         The Presence of EpCAM-/CD49f+ Cells in Breast Cancer Is Associated with a Poor Clinical Outcome

        Purpose: It is well established that breast cancer stem cells (BCSCs) play an essential role in tumor invasion for both local and distant metastasis. The aim of this study was to establish whether BCSCs could act as a prognostic and clinical marker. Methods: We analyzed tumor tissues from 161 breast cancer patients. Dual immunohistochemistry and immunofluorescence were performed on all the slides, and we analyzed the relationship between EpCAM–/CD49f+ tumor cells and key clinical and prognostic factors. Results: Univariate survival analysis using the Kaplan-Meier method showed that the presence of EpCAM–/ CD49f+ tumor cells in breast cancer was significantly associated with shorter disease-free survival (DFS) and overall survival (OS). Multivariate analysis showed that the presence of EpCAM–/ CD49f+ cells was associated with shorter DFS (p=0.010; hazard ratio [HR], 2.070) and OS (p=0.002; HR, 3.235). Tumors containing EpCAM–/CD49f+ cells were also more likely to metastasize after initial surgery (p=0.048). Conclusion: Our study suggests that breast tumors containing EpCAM–/CD49f+ cells are more likely to undergo distant metastasis after initial surgery and are associated with a shorter DFS and OS.

        Development and validation of risk score for predicting spontaneous rupture of hepatocellular carcinoma

        Purpose: Spontaneous rupture is a potentially serious complication of liver cancer. A risk score was developed and validated for predicting spontaneous rupture based on a retrospective study. Methods: Multiple logistic regression analysis was used to study the relationship between clinical variables and spontaneous rupture. The independent rupture predictors were converted into a score based on the odds ratio. Predicted attributes of the developed scores were then verified using a dataset in 2019. Results: The incidence of spontaneous rupture was 5.5% from 2002 to 2019. A 10-point score (α-FP of ≥400 μg/L, 1; protrusion from liver surface, 2; ascites, 3; tumor size of >5 cm, 4) was derived for prediction of rupture and area under the receiver-operating characteristic curve was 0.9 (95% confidence interval, 0.87–0.92). When applying a cutoff value of 5 points or more, the specificity was 0.87 and the sensitivity was 0.84. A validation cohort consisting of 202 hepatocellular carcinoma patients reproduces the predictive, identification, and calibration characteristics. The observed rate of spontaneous rupture according to risk stratification of the score was 0.6% for those with a score of 0–4, 21.6% for a score of 5–7, and 36.4% for a score of 8–10 in the validation cohort. Conclusion: Here, based on routine clinical data, we determine the factors that affect prognosis and propose an effective tool for predicting spontaneous rupture, which may be useful in guiding priority treatment of high-risk patients or clinical routine preventive treatment.

        The Influence of Nano- and Micron-size of MXene Flakes on the Electrochemical Performance

        MXene, as a new type of two-dimensional material, has attracted much interest since it was discovered in 2011. However, only few articles discussed the eff ect of MXene fl ake size on its electrochemical performance. Here, a sand milling way is explored to produce nano-size MXene and the MILD method is used to prepare micron-size MXene (1 μm) as a comparison. Meanwhile, a mask-assisted interdigital micro-supercapacitors is prepared to explore the dependence of the electrochemical properties of MXene on their fl ake size. We show that nano-size MXene (200 nm) has a higher ionic conductivity as compared to normal micron-size MXene (1 μm). On the contrary, the larger fl ake size has higher electrical conductivities. As a result, the capacitance of micron-size MXene is better than nano-size MXene (200 nm) because the electrical conductivities are dominant. This research is helpful for further understanding of the infl uence of MXene fl ake size on its electrochemical performance.

        A RuO2IrO2 electrocatalyst with an optimal composition and novel microstructure for oxygen evolving in the single cell

        A highly active RuO2IrO2 electrocatalyst was developed via dip-coating/calcination method for oxygen evolution reaction (OER). The catalyst on Ti substrate with a 7/3 molar ratio between Ru and Ir showed the highest electrocatalytic activity for OER among composite samples in different molar ratios. Moreover, the properties of RuO2IrO2 grown on carbon paper were evaluated by proton exchange membrane water electrolysis single cell. Compared with the micron-particle structure of RuO2IrO2 catalyst on the Ti substrate, the catalyst grown on the carbon paper showed a novel nano dendrite shape and can be used directly as the gas diffusion electrode. Owing to the large surface area of the catalyst, the nano dendrite-shaped RuO2IrO2 catalyst exhibits excellent OER performance in the single cell. Furthermore, a cell voltage of 2.50 V is achieved under 200mA cm2 at 30 oC by using the optimal composition RuO2IrO2 (Ru : Ir=7/3) and the commercial 20% Pt/C as anode and cathode, respectively.
