A cis-1,2-dichloroethylene (cis-DCE)-degrading anaerobic bacterium, Clostridium sp. strain KYT-1, was isolated from a sediment sample collected from a landfill site in Nanji-do, Seoul, Korea. The KYT-1 strain is a gram-positive, endospore-forming, motile, rod-shaped anaerobic bacterium, of approximately 2.5~3.0 μm in length. The degradation of cis-DCE is closely related with the growth of the KYT-1 strain, and it was stopped when the growth of the KYT-1 strain became constant. Although the pathway of cis-DCE degradation by strain KYT-1 remains to be further elucidated, no accumulation of the harmful intermediate, vinyl chloride (VC), was observed during anaerobic cis-DCE degradation. Strain KYT-1 proved able to degrade a variety of volatile organic compounds, including VC, isomers of DCE (1,1-dichloroethylene, trans-1,2-dichloroethylene, and cis-DCE), trichloroethylene, tetrachloroethylene, 1,2-dichloroethane, 1,1,1-trichloroethane, and 1,1,2-trichloroethane. Strain KYT-1 degraded cis-DCE at a range of temperatures from 15 to 37℃, with an optimum at 30℃, and at a pH range of 5.5 to 8.5, with an optimum at 7.0.

Objective: The primary objective of this study was to evaluate the safety of niraparib 300 mg/dayin Japanese patients with platinum-sensitive, relapsed ovarian cancer in a maintenance setting. Methods: Phase 2, multicenter, open-label, single-arm study enrolled Japanese patients withplatinum-sensitive, relapsed ovarian cancer who had received ≥2 platinum-based regimens. The primary endpoint (incidence of grade 3 or 4 thrombocytopenia-related events within 30days after initial niraparib administration) was justified by the incidences of a global pivotalphase 3 study and its post-hoc safety analysis on thrombocytopenia, the major hematologicaladverse event of niraparib. The overall safety analysis examined other treatment-emergentadverse events (TEAEs). Results: Enrolled patients (n=19) had a median (min, max) body weight of 53.9 (40.8–79.1)kg; all but one patient weighed <77 kg. Most (94.7%) patients initially received niraparib300 mg/day but this decreased in subsequent cycles (mean±standard deviation doseintensity, 191.6±65.7 mg/day). In total, 6/19 (31.6%) patients experienced grade 3 or 4 thrombocytopenia-related events within 30 days of initial niraparib administration. Other common TEAEs included nausea, and decreased platelet or neutrophil counts. Noprogression-free or overall survival events occurred; only 1 of 4 response-evaluable patientshad a post-baseline tumor assessment (stable disease). Conclusion: The incidence of grade 3 or 4 thrombocytopenia-related events in Japaneseovarian cancer patients was similar to that in the corresponding non-Japanese study. Overall,the safety profile was acceptable and consistent with the known safety profile and previousexperience with niraparib. Trial Registration: ClinicalTrials.gov Identifier: NCT03759587

Clostridium bifermentans strain DPH-1 has already been found to dechlorinate perchloroethylene (PCE) to cis-dichloroethylene (cis-DCE) via trichloroethylene (TCE). In this study, our investigation on different culture conditions of this DPH-1 strain was extended to find a more efficient and cost effective growth medium composition for this DPH-1 strain in bioremediation practices. Temperature dependency of strain DPH-1 showed that the growth starting time and PCE degradation at 15oC was very slow compared to that of 30oC, but complete PCE degradation occurred in both cases. For the proper utilization of strain DPH-1 in more cost effective bioremediation practices, a simpler composition of an effective media was studied. One component of the culture medium, yeast extract, had been substituted by molasses, which served as a good source of electron donor. The DPH-1 strain in the medium containing molasses, in the presence of K2HPO4 and KH2PO4, showed identical bacterial multiplication (0.135 mg protein mL-1h-1) and PCE degradation rates (0.38 M/h) to those of the yeast extract containing medium.

Background/Aims: Multiple white and flat elevated lesions (MWFL) that develop from the gastric corpus to the fornix may be strongly associated with oral antacid intake. Therefore, this study aimed to determine the association between the occurrence of MWFL and oral proton pump inhibitor (PPI) intake and clarify the endoscopic and clinicopathological characteristics of MWFL. Methods: The study included 163 patients. The history of oral drug intake was collected, and serum gastrin levels and anti-Helicobacter pylori immunoglobulin G antibody titers were measured. Upper gastrointestinal endoscopy was performed. The primary study endpoint was the association between MWFL and oral PPI intake. Results: In the univariate analyses, MWFL were observed in 35 (49.3%) of 71 patients who received oral PPIs and 10 (10.9%) of 92 patients who did not receive oral PPIs. The occurrence of MWFL was significantly higher among patients who received PPIs than in those who did not (p<0.001). Moreover, the occurrence of MWFL was significantly higher in patients with hypergastrinemia (p=0.005). In the multivariate analyses, oral PPI intake was the only significant independent factor associated with the presence of MWFL (p=0.001; odds ratio, 5.78; 95% confidence interval, 2.06–16.2). Conclusions: Our findings suggest that oral PPI intake is associated with the presence of MWFL (UMINCTR 000030144).

Iron as well as aluminum is reported to accumulate in neurons with neurofibrillary tangles (NFTs) of Alzheimer's disease(AD) brain. Previously we demonstrated that aluminum(Ⅲ) shows phosphate-dependent binding with hyperphosphorylated τ(PHFτ), the major constituent of NFTs, thereby inducing aggregation of PHFτ. Herein we report that iron(Ⅲ) can also induce aggregation of soluble PHFτ to occur, iron in the oxidized state (Ⅲ) is essential since iron in the reduced state (Ⅲ) lacks such ability. Furthermore, iron (Ⅲ)-induced aggregation is reversed by reducing iron (Ⅲ) to iron (Ⅱ). Thus the iron-participating aggregation is mediated not only by τ phosphorylation but also by the transition of iron between reduced (Ⅱ) and oxidized (Ⅲ) states. Further incubation of insoluble PHFτ aggregates isolated from AD brain with reducing agents produced liberation of solubilized PHFτ and iron (Ⅱ), indicating that PHFτ in association with iron (Ⅲ) constitutes the insoluble pool of PHFτ. These results indicate that iron might play a role in the aggregation of PHFτ leading to the formation of NFTs in AD brain.
Acid-suppressive medication, a possible risk factor for asthma

Background: Acid-suppressive medication (ASM), such as proton pump inhibitors and histamine 2 receptor antagonists, are commonly used. However, concerns have been raised that they might cause allergic diseases, including asthma. We investigated the relationship between the use of ASM and the subsequent incidence of asthma. Methods: A retrospective cohort study was conducted using the data from health insurance claims and results of specific health checkups for metabolic syndrome from 3 health insurance societies April 2011-thru-March 2015. Among the 40-64-year-old subjects without asthma (5,915 men, 3,973 women), logistic regression analyses were performed to investigate the relationship between the use of ASM (stratified as none, 1-59 days/year, ≥60days/year) and subsequent incidence of asthma. The adjustment was made for age, smoking status, BMI, and registered “allergic rhinitis” and “reflux esophagitis” diagnoses in multivariate logistic regression analyses. Results: 213 (3.6%) men (median 47 [IQR, 11] years old) and 211 (5.3%) women (median 47 [IQR, 9]) developed asthma. In the whole cohort, both short- and long-term use of ASM was significantly related to asthma incidence (adjusted Odds Ratio 1.89 [95% CI, 1.40-2.57]; P <0.001, aOR 1.82 [95% CI, 1.15-2.89]; P = 0.01, respectively). In men, only short-term use of ASM was significantly related to asthma incidence (aOR 1.87 [95% CI, 1.21-2.92]; P = 0.005). In women, both short- and long-term use was significantly related to asthma incidence (aOR 1.92 [95% CI, 1.26-2.92]; P = 0.002, aOR 2.85 [95% CI, 1.50-5.40]; P = 0.001, respectively). Longer duration of ASM use was also significantly associated with asthma incidence after adjusting for confounders (P-values for trends <0.001 in women and the whole cohort). Results in subjects without registered “reflux esophagitis” were almost the same as in the main analyses. Conclusions: Acid- suppressive medication could be a risk factor for the subsequent incidence of asthma.

Study Design: Retrospective case series. Purpose: To investigate the oncological outcomes, including distant relapse, after en bloc spondylectomy (EBS) for spinal metastases in patients with a minimum of 2-year follow-up. Overview of Literature: Although EBS has been reported to be locally curative and extend survival in select patients with spinal metastases, detailed reports regarding the control of distant relapse after EBS are lacking. Methods: We conducted a retrospective review of 18 consecutive patients (median age at EBS, 62 years; range, 40–77 years) who underwent EBS for spinal metastases between 1991 and 2015. The primary cancer sites included the kidney (n=7), thyroid (n=4), liver (n=3), and other locations (n=4). Survival rates were estimated using the Kaplan–Meier method, and groups were compared using the log-rank method. Results: The median operative time and intraoperative blood loss were 767.5 minutes and 2,375 g, respectively. Twelve patients (66.7%) experienced perioperative complications. Five patients (27.8%) experienced local recurrence of the tumor at a median of 12.5 months after EBS, four of which had a positive resection margin status. Thirteen patients (72.2%) experienced distant relapse at a median of 21 months after EBS. The estimated median survival period after distant relapse was 20 months (95% confidence interval, 0.71–39.29 months). No association was found between resection margin status and distant relapse. Overall, the 2-year, 5-year, and 10-year survival rates after EBS were 72.2%, 48.8%, and 27.1%, respectively. Importantly, the era in which EBS was performed did not impact the oncological outcomes. Conclusions: Our results suggest that EBS by itself, even if margin-free, cannot prevent further dissemination, which occurred in >70% of patients at a median of 21 months after EBS. These results should be considered and conveyed to patients for clinical decision-making.
